Experiments were conducted to investigate the effects of several hormones on the growth of suspension culture of carrot (Daucus carota L.) cells, where changes in pH and the amount of $NH_4-Nin$ the medium were observed with regard to growth. A treatment with 2,4-D at $10^{-5}M$ resulted in a highest rate of growth; the hormone at this concentration caused an increase in dry weight by about 40 to 50% over the control, measured at a stationary phase. It was thus indicated that 2,4-D at $10^{-5}M$ provided the optimal condition for the suspension culture. Changes in pH of the medium were found to be affected by hormonal treatments during the first 2-3 days following the inoculation, after which the pattern of pH changes in hormone enriched media paralleled that of the control. Subsequent changes of $NH_4-N and NO_3-N$ from the medium by the cells, and also by growth of the cells. The uptake of $NH_4-N$ by the cells did not appear to be influenced by hormonal treatments. At a stationary phase, a considerable amout of $NO_3-N$ played a more important role than $NH_4-N$ in the growth of the carrot cell suspension culture.
In order to utilize cinder melting slag as a filter media to control the quality of early rainwater, its environmental stability was verified by heavy metal elution experiment and improved by pre-treatment. Possibilities of improving its function as an absorbent was considered. Absorption characteristics of melting slag before and after the pre-treatment were analyzed by heavy metal equilibrium and stationary-phase column experiments, which in turn were analyzed by comparison experiment with activated carbon. As a result of heavy metal elution experiment, every metal item existed in a much lower amount than the criteria or was not detected, implying that there is no problem recycling it. Absorption equilibrium experiment showed that the time for pre-treatment melting slag to reach the equilibrium was reduced, while the absorbed amount was greatly increased. Stationary-phase column experiment assures us that the elimination rate was not changed much by influx rate, pH and the change in packing volume rate, indicating that this melting slag can be used not only as a filter media to control the quality of early rainwater but also in many areas of water-processing.

An isolate of Pseudomonas putida has been found to aggressively colonize root tips and induce plant resistance to Fusarium wilt. However, P. putida mutants lacking Fe-superoxide dismutase (SOD) or both FeSOD and MnSOD activities are less competitive in root tip colonization. In the current study, the growth of an FeSOD mutant was found to be more sensitive than that of the wild-type or a MnSOD mutant to oxidative stress imposed by paraquat treatment and culturing with the soil fungus Talaromyces flavus, which generates reactive oxygen species. Also, the loss of culturability with an aging stationary-phase culture was greater for a double SOD mutant than an FeSOD mutant, while no reduction in culturability was observed with the wild-type and a MnSOD mutant under the same protracted stationary-phase conditions. Accordingly, it was concluded that FeSOD activity is the major form of SOD in P. putida and plays an essential role in survival under stress conditions when increased oxidative stress is encountered.
The addition of glycine up to 0.5% (w/v) to Luria broth (LB) media on the secretion of levansucrase in a recombinant strain Escherichia coli JM109/pUPLK1 was observed to enhance the release of periplasmic proteins from the cell to the broth, without significantly affecting the cell growth rate and protein productivity. However, the glycine concentration at 1 % (w/v), the cell density attainable at the stationary phase fell to about 50% and the extracellular activity of levansucrase corresponded to about 80% of the total (extracellular plus intracellular) activity and increased by 2.6-fold, comparing to the cells grown in the absence of glycine. The increased pH at stationary phase accelerated the degradation of levansucrase. Maximal extracellular activity was attained when 1 % glycine was supplemented at the onset of strain growth.
A new HPLC chiral stationary phase (CSP 3) has been prepared by connecting N-phenyl N-propyl amide of (S)-naproxen to silica gel through the 6-methoxy-2-naphthyl group of (S)-naproxen. The new CSP has been applied in resolving a homologous series of N-(3,5-dinitrobenzoyl)-α-amino acid esters and a homologous series of N-(3,5-dinitrobenzoyl)-α-(4-alkylphenyl)alkylamines. The separation factors, α, for resolving a homologous series of N-(3,5-dinitrobenzoyl)-α-amino esters and a homologous series of N-(3,5-dinitrobenzoyl)-α-(4-alkylphenyl)alkylamines on the new CSP have been found to remain almost constant throughout the wide range of the length of the alkyl substituent of the analytes while those on the previously reported CSPs (CSP 1 and 2) which were prepared by connecting N-phenyl N-propyl amide of (S)-naproxen to silica gel through the N-propyl group increase or decrease continuously. These results are concluded to support the chiral recognition models which utilize the intercalation of the alkyl substituent of the racemic analytes between the adjacent strands of CSP 1 or 2 to rationalize the increasing or decreasing trends of separation factors.
A chiral stationary phase derived from (S)-N-(3,5-dinitrobenzoyl)leucine N-phenyl N-alkyl amide (CSP 2) was applied in separating the two enantiomers of various π-basic aromatic derivatives of leucine N-propyl amide in order to evaluate π-basic aromatic groups as an effective derivatizing group for the resolution of a-amino acids. Subsequently N-(3,5-dimethoxybenzoyl) group was found to be very effective as a π-basic aromatic derivatizing group. Based on these results, N-(3,5-dimethoxybenzoyl) derivatives of various a-amino N-propyl amides, N,N-diethyl amides and esters were resolved on the CSP derived from (S)-N-(3,5-dinitrobenzoyl) leucine N-phenyl N-alkyl amide (CSP 2) and the resolution results were compared with those on the CSP derived from (S)-N-(3,5-dinitrobenzoyl)leucine N-alkyl amide (CSP 1). The enantioselectivities exerted by CSP 2 were much greater than those exerted by CSP 1. In addition, racemic N-(3,5-dimethoxybenzoyl)-a-mino N,Ndiethyl amides were resolved much better than the corresponding N-(3,5-dimethoxybenzoyl)-a-mino N-propyl amides and esters on both CSPs. Based on these results, a chiral recognition mechanism utilizing the π-π donor-acceptor interaction and the two hydrogen bondings between the CSP and the analyte was proposed.

Colloids such as exogenous biocolloids in a bioremediation operation can enhance the transport of contaminant in ground water by reducing retardation effects. Because of their colloidal size and favorable surface conditions in addition to their low density, bacteria can act as efficient contaminant carriers. When mobile bacteria are present in a subsurface environment, the system can be treated as consisting of three phases: water phase, bacterial phase, and the stationary solid matrix phase. In this work, a mathematical model based on mass balances is developed to describe the facilitated transport and fate of a contaminant in a porous medium. Bacterial partition between the bulk solution and the stationary solid matrix, and the contaminant partition among the three phases are represented by the equilibrium relationships. Solutions were obtained to provide estimates of contaminant and bacterial concentrations. A dimensionless analysis of the transport model was utilized to estimate model parameters from the experimental data. The model results matched with experimental data of Jenkins and Lion (1993). The presence of mobile bacteria enhances the contaminant transport. However, bacterial consumption of the contaminant which serves as a bacterial nutrient, can attenuate the contaminant concentration.
This study was examined to investigate the time course behaviors of cell growth and sucrose consumption, and effects of various elicitors on ginsenoside production in batch suspension cultures of Panax ginseng Meyer. Suspended cells reached to the stationary phase at 12 days after innoculation. The maximum cell concentration was 14.7 g-DCW/L at 17 days. The highest cell growth rate was 0.59 g-DCW/L d. The sucrose used as a sole carbon source was hydrolysed to glucose and fructose in 4 days, then quickly utilized until the middle-log phase and consumpted completely at 16 days. Various elicitors were app1ied at 8 days from inoculation which is the middle-log phase. Among the elicitors tested, jasmonic acid was the most efficient to increase the ginseneside production, which was 1.5 times higher than control.
Lactobacillus acidophilus NCFM, Lactobacillus casei YIT 9018, Bifidobacterium longum 8001, and Bifidobacterium longum 8025 at the level of 106 cfu/$m\ell$ were cultured with 104 cfu/$m\ell$ of Escherichia coli O157:H7 KSC 109 or Salmonella typhimurium ATCC14028, in order to verify the effects of lactic acid bacteria and bifidobacteria on the growth of the pathogens. In the mixed culture of lactic acid bacteria with E. coli O157:H7 KSC 109, Growth inhibition and atypical microcolonies of E. coli O157:H7 KSC 109 were observed. The pathogens inoculated grew for 5 hors (pH 5.3), by the time L. acidophilus NCFM reached the exponential growth phase, and then the surviving pathogens were decreased to 101 cfu/$m\ell$ after 35 hours. When L. caseiYIT 9018 was grown with the pathogens, they grew for 10 hours (pH 4.6), by the time L. casei YIT 9018 reached the end of exponential growth phase, and then the surviving pathogens were decreased drastically. Up to the stationary growth phase of lactic acid bacteria, L. acidophilus NCFM exhibited stronger inhibition against the pathogens than L. casei YIT 9018 did, which might be attributed to its faster growth. Likewise bifidobacteria inhibited the growth of the pathogens tested, bifidobaceria was weaker in the inhibitory activity than lactic acid bacteria. When Bifidobacterium longum 8001 was cultured with the pathogens, E. coli O157:H7 KSC 109 was gradually ingibited at the stationary growth phase of bifidobacteria, atypical microcolonies were formed on Levine EMB medium after 48 hours, and Salmonella grew up to 106 dfu/$m\ell$, then was drastically ingibited at the exponential growth phage of Bifidobacterium longum 8001. But when Bifidobacteriuam longum 8025 was cultured with the pathogens, the pathogens grew to the same level of Bifidobacteriuam longum 8025 was cultured with the pathogens, the pathogens grew to the same lever of Bifidobacteriuam longum 8025 after 10 hours, then the surviving pathogens were decreased drastically.

A grid connected inverter must be operated as the main electricity source under an isolated condition caused by the grid problem. Conventionally, the dual loop controller is used for the grid inverter, and the controller is used for control under the stand-alone mode. Generally, the PI(Proportional - Integral) controller is highly efficient under a synchronous reference frame, and stable control can be available. However, in this synchronous frame-based control, high-quality DSP is required because many sinusoidal calculations are necessary. When the PI control is conducted under a stationary frame, the controller constructions are made simple so that they work even with a low-price micro controller. However, given the characteristics of the PI controller, it should be designed with the phase of reference voltage considered. Otherwise, the phase delay of the output voltage can occur. Although the current controller also has a higher bandwidth than the voltage controller, distortion of the voltage is difficult to avoid only by the rapid response of the PI controller, as a sudden load change can occur in the nonlinear load. In this study, a new control method that solves the voltage controller bandwidth problem and rapidly copes with it even in the nonlinear load situation is proposed. The validity of the proposed method is proved by simulation and experimental results.
