Supplemented Effect of Salicornia herbacea Extract Powder on Preparation and Quality Characteristics of Fermented Milk Product. (유산발효유 제조 및 품질특성에 미치는 함초 추출물 첨가의 영향)

  • Fermented milk was prepared from skim milk supplemented with Salicornia herbacea extract powder (SHEP) at the levels of 0∼0.4% (wi v) and was fermented with Lactobacillus acidophilus, Streptococcus thermophillus and Bifidobacterium longum. Quality characteristics of prepared fermented milk were evaluated for acid production, visible cell numbers, viscosity and sensory property during fermentation at 37$^{\circ}C$ for 6 hr. Supplementation of 0.1% SHEP stimulated the growth of lactic acid bacteria which showed the highest number of viable cell counts (9.23 log CFU/ml), and also enhanced the acid production which was pH 4.23 and titratable acidity 0.64%, and increased the viscosity (1,365 cps) after 6 hr incubation. The sensory scores of fermented milk supplemented with 0.1% SHEP were higher than other supplemented contents in taste, texture, flavor, aftertaste and overall acceptability. When the storage abilities of fermented milk supplemented SHEP at 6$^{\circ}C$ for 12 days were evaluated, its quality-keeping properties were relatively good in the fermented milk supplemented with 0.1% SHEP.

Probiotic Potential of Indigenous Bacillus sp. BCNU 9028 Isolated from Meju (메주로부터 분리한 토착 Bacillus sp. BCNU 9028의 프로바이오틱스로서 이용 가능성)

  • Spore-forming bacteria are being used as probiotic supplements for human and animal use, due to their low pH stability and ability to survive the gastric barrier. In this study, the BCNU 9028 strain was screened from meju, a Korean fermented soybean food starter. Biochemical and physiological characteristics, as well as 16S rDNA sequence analyses, indicate that this strain belongs to the genus $Bacillus$. $Bacillus$ sp. BCNU 9028 showed a 92% survivability at pH 2.5 and could also withstand 0.3% ox bile. Furthermore, it was postulated that $Bacillus$ sp. BCNU 9028 could prevent biofilm formation and adherence of food-borne pathogens such as $Listeria$ $monocytogenes$, $S.$ $aureus$ and $E.$ $coli$ on the basis of its autoaggregation and coaggregation capacity with food-borne pathogens. It was shown that BCNU 9028 has good abilities to adhere to the intestinal tract from its hydrophobic character (63.3%). The $Bacillus$ sp. BCNU 9028 strain especially elicited antibacterial activity against both Gram-positive and -negative pathogens. These findings suggested that the $Bacillus$ sp. BCNU 9028 strain could be used as a potential probiotic.

Selection of Lactic Acid Bacteria suitable for Manufacture of Freeze-dried Coffee (동결건조커피 제조에 적합한 유산균 균주 선발)

  • Probiotic functional foods are known to have various functional effects such as intestinal regulation, modulation of immune system, reduction of allergies, and lowering of cholesterol. The purpose of this study was to select probiotic strain that is most suitable for freeze-dried coffee for the development of functional coffee products. The survival rate of probiotics, at drinking condition of coffee, at acid, at bile and after freeze-dried in coffee were measured on 1 strain isolated from commercial freeze-dried coffee, 8 strains used as fermented milk starter, 1 Bifidobacterium and 1 Bacillus coagulans. Bacillus coagulans showed the highest survival rate from $2.4{\times}10^7cfu/g$ to $2.0{\times}10^7cfu/g$ especially after freeze-drying. The results at drinking condition of coffee, at acid tolerance, at bile tolerance and at storage test showed significantly better survival rate of Bacillus coagulans than that of control (Lactobacillus casei). Especially, Bacillus coagulans showed 3.8-fold higher survival rate at acid tolerance (pH 1, 120 minutes) than control. Thus, the lactic acid-producing Bacillus coagulans is characterized as a probiotic strain suitable for functional coffee formulation and commercialization.

Effects of Lactitol and Tributyrin on Growth Performance, Small Intestinal Morphology and Enzyme Activity in Weaned Pigs

  • One hundred and sixty crossbred pigs ($6.62{\pm}0.36kg$) weaned at day $18{\pm}1$ were used to investigate the effects of lactitol and tributyrin on performance, small intestinal morphology and enzyme activity. The pigs were assigned to one of five dietary groups (4 pens/diet with 8 pigs/pen) and were fed the negative control diet or the negative control diet supplemented with 10 g/kg glutamine (as a positive control), or 3 g/kg lactitol (${\beta}$-D-galactopyranosyl-($1{\rightarrow}4$)-D-sorbitol), or 5 g/kg tributyrin (butanoic acid 1,2,3-propanetriyl ester), or 3 g/kg lactitol+5 g/kg tributyrin. Body weight and feed intake were measured weekly during the 4-week study. On day 7, four pigs per dietary treatment were sacrificed to examine small intestinal morphology and enzyme activity. The results showed that: (1) Compared with the negative control diet, the positive control diet improved weight gain and feed efficiency during weeks 1-2 and over the entire study (p<0.05), and also decreased duodenal and ileal crypt depth (p<0.05), but did not alter intestinal enzyme activity (p>0.05). Lactitol improved feed efficiency during weeks 3-4 and over the entire study (p<0.05), but did not improve weight gain and feed intake, intestinal morphology or enzyme activity (p>0.05). Tributyrin improved weight gain and reduced feed/gain during weeks 3-4 and over the entire study. Tributyrin significantly decreased crypt depth in the duodenum and ileum, and increased duodenal lactase and ileal maltase activity (p<0.05). Lactitol+tributyrin increased weight gain during weeks 3-4 and over the entire study, and improved feed efficiency during weeks 1-2 and 3-4 and over the entire study (p<0.05). Lactitol+tributyrin increased the jejunal villus height, and decreased the duodenal and ileal crypt depth (p<0.05). Lactitol+tributyrin also increased jejunal lactase and sucrase activity (p<0.05). (2) Compared with the positive control, tributyrin improved weight gain and reduced feed/gain during weeks 3-4 (p<0.05), decreased the ileal crypt depth, and improved the duodenal lactase and sucrase activity (p<0.05). Lactitol+ tributyrin improved weight gain during weeks 3-4, improved feed efficiency during weeks 3-4 and over the entire study, increased the ileal villus height, and increased jejunal lactase, sucrase and maltase activity (p<0.05). These results showed that tributyrin improved performance, intestinal morphology and enzyme activity, while the effect of lactitol was very limited. These results also showed that, compared with glutamine, tributyrin was more effective in improving intestinal morphology and enzyme activity, and tributyrin exerted a superior effect in improving performance as weaning progressed. These observations suggest that, as a chemical for repairing intestinal atrophy, glutamine and tributyrin should be used in the first and second periods of the starter phase, respectively.

Preparation and Characteristics of Kimchi Tablet (김치 타블렛의 제조와 품질특성)

  • In order to prepare kimchi tablet(K7), the kimchi was fermented with crushed materials and lactic acid bacterial starter. The effect of raying methods (hot air-drying : HAD, freeze drying: FD), moulding pressure (100, 150 and 200 kg/cm2) and moulding time (0, 1, 2, 3 and 4 min) of kimchi tablet on residual solid, characteristics of filtrate, color, textual properties, hygroscopicity and sensory quality were investigated. No great difference showed in residual solid of the tablet prepared with freeze dried kimchi (FD-KT), but the tablet prepared with hot air-dried kimchi (HAD-KT) decreased markedly with an increase in moulding pressure. Residual solid of FD-KT was lower than that of HAD-KT. The pH and redness for HAD-KT and FD-KT were similar. Total sugar. acidity and lightness of FD-KT as compared with HAD-KT were higher, while higher max, yield, strength and hardness were found for HAD-KT. Color, tactile, flavor, chewing taste, acceptability and hygroscopicity of FD-KT were generally higher than those of HAD-KT. Hygroscopicity of HAD-KT decreased with an increase in moulding pressure in md time, but that of FD-KT increased. Overall qualities of FD-KT and HAD-KT showed best in 200 kg/㎠ of moulding pressure, 3min of moulding time.

Physiological Characteristics and GABA Production of Lactobacillus plantarum K74 isolated from Kimchi (김치에서 분리한 Lactobacillus plantarum K74의 GABA 생산 및 생리적 특성)

  • Aminobutyric acid (GABA), a major inhibitory neurotransmitter in the central nervous system of animals, has several physiological effects including anti-hypertensive, diuretic, tranquilizing, and anti-stress properties, in humans. The purpose of this study was to investigate Lactobacillus plantarum K74, which was isolated from kimchi and selected as a strain with a high ability to produce GABA, to develop a new starter culture for fermented milk production. L. plantarum K74 produced $134.52{\mu}g/mL$ GABA in MRS broth containing 1% MSG, $212.27{\mu}g/mL$ GABA in MRS broth containing 2% MSG, and $234.63{\mu}g/mL$ GABA in MRS broth containing 3% MSG. The optimum growth temperature of L. plantarum K74 was $34^{\circ}C$, reaching a pH of 4.4 after 18 hours of growth. L. plantarum K74 was most sensitive to novobiocin out of 16 different antibiotics tested, and was most resistant to kanamycin and polymyxin B. L. plantarum K74 did not produce ${\beta}$-glucuronidase, a carcinogenic enzyme, and was comparatively tolerant to bile juice and low pH. Furthermore, it displayed resistance to Escherichia coli, Salmonella typhimurium, and Staphylococcus aureus at rates of 54.9%, 46.3%, and 0.7%, respectively.

Anti-Inflammatory Effects of Fermented Milk Supplemented with Heat-Killed Enterococcus faecalis EF-2001 Probiotics (Enterococcus faecalis EF-2001 유산균 사균체 첨가 발효유의 항염증 효과)

  • This study was conducted to verify the physiological activity of heat-killed Enterococcus faecalis EF-2001 probiotics in fermented milk. The anti-inflammatory effects of fermented milk supplemented with different concentrations (0, 100, and 500 ㎍/mL) of E. faecalis EF-2001 were determined using MTT assay and nitric oxide inhibition assay. The MTT assay was performed using RAW 264.7 cells. Results revealed that the rates of cytotoxicity and cell survival decreased significantly with increase in the concentration of heat-killed probiotics (p<0.05). Moreover, fermented milk supplemented with 100 ㎍/mL EF-2001 (EFM1) and the fermented milk supplemented with 500 ㎍/mL EF-2001 (EFM2) exhibited higher nitric oxide inhibition than normal fermented milk (NFM). Additionally, EFM2 significantly reduced the ratio of prostaglandin E2 compared to NFM (p<0.05). In conclusion, the treatment sample showed higher anti-inflammatory activity than NFM. The findings of this study could be used as a basic guideline for manufacturing of NFM supplemented with heat-killed probiotics.

Study on Performance and Meat Characteristics in Korean Native Commercial Chicken I. Study on Performance in Korean Native Commercial Chicken by Feeding System by Feeding System (한국재래닭의 육용실용계의 발육 및 육질특성 구명 연구 1. 사료 급여체계에 따른 한국재래닭 육용실용계의 발육능력)

  • This experiment was carried out to investigate the effect of feeding system on performance in Korean Native Commercial Chicken. A total 864 birds produced from (Cornish ♂ X (Korean Native Chicken ♂ XRhode Island Red♀)♀ ] crossbreeds in National Livestock Research Institute, for 16 weeks. Feeding system of T1 and T$_2$ were same types from hatch to 8 weeks, starter diets(O~4 weeks, mash, ME 3,100kcal, CP 22.94%), grower diets(4~8 weeks, crumble, ME 3,100kcal, CP 19.31%). Nutrient content of finisher diets of T$_1$(pellet, ME 3,200kcal, CP 20.44%) was higher than T$_2$(mash, ME 3,100kcal, CP 14.88%) in order to improve meat quality for 8~16 weeks. Fertility and hatchability of Korean Native Commercial Chicken was 83.9% and 69.7%, respectively. Viabilities of T$_1$ and T$_2$ at 0, 4, 8, 12 and 16 weeks were 98.8%, 97.9%, 96.5% and 99.1%, 95. 8%, 92.8%, 90.3%, respectively. The viability of 0 to 8 weeks was not significantly in feed treatments, but 12 and 16 weeks was significantly T$_1$ higher than T$_2$(P<0.05). Body weights of T$_1$and T$_2$ at 4, 8, 12 and 16 weeks were 551g, 1,379g, 2,441g, 3,056g and 554g, 1,360g, 2,254g, 2,956g, respectively. The body weight of 0 to 8 weeks was not significantly feed treatments but 12 and 16 weeks was significantly T1 higher than T$_2$(P<0.05). Feed conversion of T$_1$ and T$_2$ to 4, 8,12 and 16 weeks were 1.91, 2.28, 3.34, 4.23 and 1.90, 2.28, 3.53, 4.46, respectively. The feed conversion of 0 to 8 weeks was not significantly feed treatments but 12 and 16 weeks was significantly T$_1$ lower than T$_2$(P<0.05). The ME intake 1 bird per 1 day of T$_1$ and T$_2$were 3S9kcal, 357kca1, respectively, not significantly feed treatments but CP intake were 24.8g, 20.3g, respectively. T$_2$ was lower than T$_1$(P$_1$ and T$_2$were 13,426kca1, 13,819Ykcal, respectively, not significantly feed treatments but CP requirement per kg body weight gain were 928g, 763g, respectively, T$_2$ was lower than T$_1$(P<0.05).

Effects of Inoculated Diet with Paecilomyces japonica on Broiler Performance (눈꽃동충하초균주(Paecilomyces japonica) 접종사료 급여가 육계의 생산성에 미치는 영향)

  • This study was carried out to investigate the effects of the inoculated diet with Paecilomyces japonica on weight gain, feed efficiency and cordycepin contents in the breast and thigh meats of broilers. One hundred and twenty broiler chicks were divided into four dietary groups ; 1) control (basal diet), 2) T1 (substituted with 5% inoculated diet), 3) T2 (substituted with 10% inoculated diet) 4) T3 (substituted with 20% inoculated diet) during 4 weeks. Cordycepin contents of inoculated diet with Pacilomyces japonica was significantly higher than before inoculation (P<0.05). Body weight gain and feed efficiency were highest in T1 group when fed starter diet (P<0.05) but these were lowest in T3 group fed when fed finished diet (P<0.05). Body weight gain and feed efficiency were the lowest in T2 group for overall period than other groups. Cordycepin contents of breast and thigh meats were significantly higher in Pacilomyces japonica groups than control at 5 weeks of age (P<0.05). However, it was similar among the T1, T2 and T3 groups. These results indicated that body weight gain and feed efficiency tended to increase in birds fed replaced 5% inoculation diet Cordycepin contents of breast and thigh meats would be increased significantly in birds fed inoculated diet (P<0.05).

Ac/Ds-mediated gene tagging system in rice

  • Transposon-mediated insertional mutagenesis provides one of the most powerful tools for functional studies of genes in higher plants. This project has been performed to develop a large population of insertional mutations, and to construct databases of molecular information on Ds insertion sites in rice. Ultimate goals are to supply genetic materials and information to analyze gene function and to identify and utilize agronomically important genes for breeding purpose. Two strategies have been employed to generate the large scale of transposon population in a Japonica type rice, Dongjin Byeo; 1) genetic crosses between Ac and Ds lines and 2) plant regeneration from seeds carrying Ac and Ds. Our study showed that over 70% of regenerated plants generally carried independent Ds elements and high activity of transposition was detected only during regeneration period. Ds-flanking DNA amplified from leaf tissues of F2 and T1 (or T2) plants have been amplified via TAIL-PCR and directly sequenced. So far, over 65,000 Ds lines have been generated and over 9,500 Ds loci have been mapped on chromosomes by sequence analysis. Database of molecular information on Ds insertion sites has been constructed, and has been opened to the public and will be updated soon at http://www.niab.go.kr. Detailed functional analysis of more than 30 rice mutants has been performed. Several Ds-tagged rice genes that have been selected for functional analysis will be briefly introduced. We expect that a great deal of information and genetic resources of Ds lines would be obtained during the course of this project, which will be shared with domestic and international rice researchers. In addition to the Japonica rice, we have established the tagging system in an rice line of indica genetic background, MGRI079. MGRI079 (Indica/Japonica) was transformed with Agrobacteria carrying Ac and Ds T-DNA vectors. Among transgenic lines, we successfully identified single-copy Ds and Ac lines in MGR1079. These lines were served as ‘starter lines’ to mutagenize Indica genetic background. To achieve rapid, large scale generation of Ds transposant lines, MGR1079 transformants carrying homozygous Ac were crossed with ones with homozygous Ds, and $F_2$seeds were used for plant regeneration. In this year, over 2,000 regeneration plants were grown in the field. We are able to evaluate the tagging efficiency in the Indica genetic background in the fall.
