Efficacy of a vaccine against Streptococcus parauberis infection in starry flounder Platichthys stellatus Pallas

  • Starry flounder, which are recently increasingly cultured in Korea, are known to highly vulnerable to Streptococcus parauberis infection. Five groups of starry flounder (n=30 for each group) were vaccinated with S. parauberis formalin-killed whole cells by intraperitoneal injection at a final concentration of 0, 0.01, 0.1, 1 and 10 mg $fish^{-1}$. Specific antibody production of 1 and 10 mg $fish^{-1}$ administered groups significantly increased at four weeks post immunization. All vaccinated groups showed higher survival rates than a control group when five groups of fish were challenged with S. parauberis at a dose of $1.14{\times}10^4$ cfu $fish^{-1}$ and $1.14{\times}10^2$ cfu $fish^{-1}$, respectively. In particular, 0.1 or higher concentrations of formalin killed bacterial cells are able to confer the fish high protection against S. parauberis infection.

Influence of Background Color and Substratum on the Blind-side Hypermelanosis in Starry Flounder Plathchthys stellatus (강도다리(Platichthys stellatus) 흑화 발현에 미치는 수조색깔 및 자갈기질의 영향)

  • To study the influence of background color and substratum on hypermelanosis of starry flounder, we compared the daily food intake (DFI), the feed efficiency (FE), the survival, the growth, the ratio of pigmented area on the blind side and the ratio of hypermelanic fish duplicately reared for 180 days in dark-green FRP aquarium (control), white FRP aquarium together with dark-green substratum. The ratio of pigmented area on the blind side was significantly higher at the dark-green group than at the white group. DFI, FE and growth were higher in the dark-green substratum. Pigmented area rate and ratio of hypermelanic fish were significantly higher at the dark green group than at the high dark-green substratum. The results suggest that bright tank color and substratum bottom could inhibit the hypermelanosis.

  • We have investigated lysozyme activity and protectivity of starry flounder, Platichthys stellatus against olive flounder pathogenic bacteria, Edwardsiella tarda, Vibrio ichthyoenteri and Streptococcus iniae in varying salinities, water temperatures and stocking density. Starry flounders were susceptible to E. tarda but not V. ichthyoenteri and S. iniae. Under low salinity condition, the lysozyme activity was decreased a little compared to the control but not significant. The physiological and immune activities were normal up to 26 ℃ culture temperature and 100% stocking density; they were compromised from 29℃ and 200%, respectively.

Quality Characteristics of Starry Flounder Platichthys stellatus Meat Reared in Different Salinity (저염분 해수사육 강도다리 Platichthys stellatus의 체성분 특성)

  • This study was performed to evaluate the food value of starry flounder Platichthys stellatus reared in different salinity. Results of salinity experiment in starry flounder clearly showed that there were no significant difference among cohesiveness and hardness except springiness. Proximate composition of muscle, crude lipid at the salinity 0 psu was significantly higher than those 15 psu and 33 psu, but there was no significant difference in terms of crude protein, moisture and ash. While variations in some amino acid composition, serine and threonine contents were observed among fishes reared in different salinity of water, but no significant difference among the other amino acid contents. Muscle fatty acid composition showed significant difference among C16:0, C16:1n-7, C20:1n-9, C22:5n-3 and C24:0. Results of a sensory evaluation clearly indicated that there were no significant difference in odor and color of fish muscle reared in different salinity. But taste, texture and overall acceptability showed the lowest values in 0 psu.

  • The starry flounder Platichthys stellatus, like all flatfish, exhibits conspicuous lateral asymmetry in numerous traits, most obvious of which is the migration of one eye to the other side of the head during metamorphosis. Additional changes related to eye migration include asymmetrical pigmentation, and a behavioral shift from larvae that exhibit upright, open-water swimming to juveniles and adults that lie on the ocean floor, eye side up. However, the morphology of these juveniles has been quite plastic in recent years, a phenomenon which is thought to be related to a diverse suite of semi-intensive and intensive larviculture methods. The cause of morphological abnormalities in the farmed flatfish is poorly understood. In the present study, we observe the features of morphological specificity and abnormality of immature fish (mean total length 23 cm) and survey the occurrence frequency of the specificity and abnormality of juvenile (mean total length 6.70 cm) in artificial culture facility. We find 2 types of abnormality (e.g., albino in ocular side and hypermelanosis in blind side) and 1 type of specificity (e.g., lateral polymorphism). These considerably differ from normal individuals (has sinistral eye and pigmented on only one side) by several characteristics (dextral eye, ocular side albinism, blind side hypermelanosis). The incidences of albinism, hypermelanosis, and body reversal are $10.1{\pm}2.56%$, $91.7{\pm}1.7%$, and $13.1{\pm}1.1%$, respectively. These suggest that these morphometric and morphological differences occur more in artificial environment during and just after metamorphosis.

  • A feeding trial was conducted to investigate the effects of dietary fish meal replacement by a blend of lysine cell mass, corn protein concentrate and poultry by-product meal on the growth and blood chemistry of the starry flounder Platichthys stellatus. The fish meal replacer (FMR) was prepared to have the same level of protein as fish meal (FM). With a commercial diet as a positive control, five experimental diets (basal, FM42, FM32, FM22 and FM12) were formulated to contain 52% protein and 10% lipid. The dietary FM levels decreased from 52% (basal) to 42, 32, 22 and 12% with concomitant increase in the FMR to 10, 20, 30, 40 and 50%, respectively. Juvenile starry flounder with an average body weight of 177.3 g were randomly distributed in each (30 fish/tank) of 18 plastic tanks ($139{\times}99{\times}54cm$). After a 45-day feeding trial, the survival rate ranged from 95.6% (FM22) to 100% (control and FM42), while the weight gain of the fish groups varied from 49.7 to 58.4 g. The results clearly revealed that starry flounder can grow well on a diet containing low FM (12%) with a high level of FMR (50%) without any adverse effects.

Molecular Characterization and Expression Pattern of Na+-K+-2Cl- Cotransporter 2 (NKCC2) in the Intestine of Starry Flounder Platichthys stellatus after Bacterial Challenge

  • We identified the $Na^+-K^+-2Cl^-$ cotransporter 2 (NKCC2) cDNA isoform from starry flounder, Platichthys stellate. The NKCC2 cDNA encoded a polypeptide of 1,043 amino acids representing 12 putative transmembrane domains based on the bioinformatic topology prediction. In addition, starry flounder NKCC2 possessed highly conserved residues within transmembrane domain 4, known as an essential site for its function. End-point reverse transcription-polymerase chain reaction analysis revealed that the NKCC2 transcript was moderately expressed only in the anterior and posterior intestines and the rectum. The NKCC2 mRNA level in the rectum, but not in other segments, was significantly induced 3 days post Streptococcus parauberis challenge, indicating that excess salt may be transported into the rectum. Taken together, our data indicate that an S. parauberis infection could tip the intestinal fluid balance in favor of fluid accumulation, indicating that bacterial pathogens can interfere with intestinal osmotic balance and normal mucosal immune homeostasis.

Morphological Analysis of Blind-Side Hypermelanosis of the Starry Flounder, Platichthys stellatus during Early Development

  • In Pleuronectiformes, blind-side malpigmentation (hypermelanosis) is common in cultured flatfishes, and is economically important. To understand the mechanism of blind-side hypermelanosis in flatfishes, we examined when the malpigmentation initially occurred, and studied how the symptoms proceeded during early development of the starry flounder, Platichthys stellatus. To assess quantitative pattern changes of blind-side skin, we observed morphological development of the whole body from 22 (total length [TL] $10.0{\pm}0.2$ mm and body weight [BW] $8.8{\pm}0.57$ mg) to 110 days (TL $23.4{\pm}0.7$ mm, BW $193.6{\pm}23.3$ mg) after hatching (DAH), and also examined the malpigmented area rate of blind-side skin and the malpigmented fish ratios. The experimental animals were reared in fiberglass-reinforced plastic (FRP) tanks in water at a temperature of $18.9{\pm}1.9^{\circ}C$ and salinity of $32.6{\pm}0.6$ psu and were fed with rotifer and Artemia nauplii from 22 to 48 DAH, and with A. nauplii and commercial feed from 49 to 110 DAH. As results, the first staining patch seen by the naked eye was observed around the area between the anus and pelvic fin or caudal edge of the trunk at 80 DAH (TL $20.6{\pm}0.5$ mm, BW $112.5{\pm}8.8$ mg). The pigmented area and the pigmented fish ratios were significantly increased from 80 to 110 DAH. These results indicated that malpigmentation on the blind side of starry flounder was initially observed at about 2 cm in length and 100 mg in weight, and the pigmented domain on the blind-side skin was continually broadened by the differentiation of pigmented cells (melanophores and xanthophores) with growth.

Morphological Characteristics of the Hybrids of Female Flounder Paralichthys olivaceus and Male Starry Flounder Platichthys stellatus (넙치 Paralichthys olivaceus와 강도다리 Platichthys stellatus 잡종의 형태적 특징)

  • Morphological characteristics of the cross-bred $F_1$ hybrid between female Paralichthys olivaceus and male Platichthys stellatus were studied. The hybrids showed common biological characteristics of both parents, but showed more morphological similarity to P. stellatus than P. olivaceusin the following biometries: higher ratios of body length to body height (56.3%), head length to snout length (20.6%), and head length to upper jaw length (34.2%). They also showed some significant biometric characteristics similar to P. stellatus, such as ratios of predorsal length (10.5%), prepectoral length (32.2%), preventral length (29.8%), and preanal length (28.0%) to body length. The hybrids had black spots on the dark brownish-green area near the eyes, and they lacked the small bony tubercles that area characteristic of Platichthys stellatus. The hybrids had unique bands: 6~20 bands on the dorsal fin and 5~10 bands on the anal fin that gradually lost their coloration towards the fin margins. The number of fin rays were 60~66 for dorsal fin, 40~51 for anal fin, 18~21 for caudal fin, and 11~12 for pectoral fin. The gills of the hybrids contained 13~18 gill rakers and 73~98 gill filaments. The vertebrae of the hybrids numbered 34~37, with 2~3 vertebrae fused from the start of the caudal vertebrae.

Reproductive Capacity in Starry Flounder Platichthys stellatus from Uljin Marine Ranching Area, Korea (울진 바다목장 해역에 서식하는 강도다리(Platichthys stellatus)의 번식능력)

  • We investigated the reproductive capacity of starry flounder Platichthys stellatus from the Uljin (UJ) marine ranching area and compared it to that of P. stellatus from the coastal waters of Pohang (PH). In UJ, female gonadosomatic index (GSI) peaked in October ($3.14{\pm}0.87$) and male GSI was high in October and December. In PH, female GSI peaked in January ($18.64{\pm}2.15$) while male GSI began to increase in October and remained high until March. Most ovaries of UJ females were immature with perinucleus oocytes, although the testes of UJ males were ripe in January. Both the ovaries and testes of PH starry flounders were ripe from January to March. The plasma estradiol-$17{\beta}$ ($E_2$) levels of UJ females were highest in October ($4.09{\pm}1.90$ ng/mL) although the testosterone (T) levels of UJ males were highest in December ($3.81{\pm}0.78$ ng/mL) and decreased gradually until April. The $17{\alpha}$,$20{\beta}$-dihydroxy-4-pregnen-3-one ($17{\alpha}20{\beta}P$) levels of UJ females were not detected. The E2 levels of PH females were highest in December ($36.25{\pm}33.07$ ng/mL) and $17{\alpha}20{\beta}P$ levels were highest in March ($5.51{\pm}0.95$5 ng/mL). The T levels of PH males were highest in December ($4.03{\pm}1.34$ ng/mL) and decreased gradually until October. Taken together, these results suggest that most females from UJ did not reach maturation with a spawning period that was considered to be between December and January.
