The Effect of Addition of Potato Starch on the Frozen Dough (감자 전분의 첨가가 냉동 반죽에 미치는 영향)

  • This study was carried out to understand the effect of addition of potato search on the frozen dough. The characteristics of frozen dough were measured by the farinogram, the extensogram and the amylogram. The results of these measurements show that the dough added with starch has higher stability than the control. The physical and chemical change of the dough were measured in accordance with the period of the frozen storage. The dough added with starch showed smaller physical and chemical change than control, which means that the starch prevents the frozen dough from the deterioration during the frozen storage. It is supposed from this result that the starch protects the activity of yeast and the structure of gluten matrices from frozen damage. It is understood from this study that addition of potato starch into frozen dough improve the stability of the frozen dough.

Physicochemical Properties of Sodium Hypochlorite Oxidized Potato Starch (Sodium Hypochlorite로 산화된 감자 전분의 이화학적 특성)

  • Physicochemical properties of potato starches oxidized with sodium hypochlorite containing 1.5, 3.0, 4.5, and 6.0% active Cl/g starch at pH 7.0 were examined. Carboxyl group contents of oxidized potato starches were proportional to active chlorine concentration in use and water binding capacity, alkali number, and solubility increased with increasing the degree of oxidation. Blue value and iodine binding property indicated the change of amylose structure by the oxidation. Gelatinization trends obtained from transmittance and DSC thermograms showed that gelatinization temperature was lower as starch was oxidized higher. X-ray diffraction patterns and scanning electron micrographs implied oxidation might occur on the surface amorphous regions of starch granule.

Preparation and Characteristics of Chestnut Mooks (밤묵의 제조와 그 특성)

  • This study was undertaken to investigate preparation possibility of chestnut mooks added chestnut shell. Three types of chesetnut starches which were flesh starch(FS), inside shell containing starch(ISS), and shell containing starch(SCS) and acorn starch(AS) as control were used to preparation of mook. The crude tannin contents of chestnut starches was about 25% of that in acorn starch. In the textural properies, hardness of mooks was increased in proportion to the increasing concentration of starches. Hardness md cohesiveness of mooks with chesnut starches were higher than those of acorn starch. Color properties of mooks with OSS were similar to that of AS. Sensory evaluation by 9-point method indicated that the nooks with ISS had greater intensities in all investigation items. The total scores were higher in order of ISS, AS, FS and SCS. The mook with ISS had homogeneous and porous structure by SEM.

  • Morphological changes of starch granules from 12 different varieties of rice were examined by scanning electron microscopy during heating at 2.5% (w/v) concentration. Rice starch granules proceeded through a similar pattern of progressive morphological changes daring heating, regardless of variety. Rice starch granules began to swell radially in the initial stage of gelatinization and then undergo radial contraction and random tangential expansion to form complex structures in the latter stage of gelatinization temperature range. At higher temperatures, starch granules softened and melted into thin flat discs, and then stretched into thin filaments to form three-dimensional networks. These progressive morphological changes were reflected in the changes of swelling power, solubility and amylograph viscosity of starch. During the transition of melting or softening, swelling power, solubility and amylograph viscosity increased rapidly. The time of loss of granular structure of starch depended on gelatinization temperature range. The ratio of amylose to amylopectin was largely responsible fur the rate of melting or softening and the fineness of a three-dimensional filamentous network above the gelatinization temperature range. Therefore, both the gelatinization temperature range and amylose content of starch affect the rate of cooking, and amylose content of starch affects the final texture of cooked starch paste.

Studies for Processing Condition Optimization and Physicochemical Property of Resistant Starch (난소화성 전분 제조공정의 최적화 및 이화학적 특성 연구)

  • As a result of resistant starch yield depending on heating temperature, moisture content, storage temperature and heating-cooling cycle with RSM (response surface methodology), high amylose corn starch (46%) was appeared higher than normal corn starch in the yield (22%). At the high amylose corn starch, optimum conditions for resistant starch formation were 6 times of heating-cooling cycle, 108$^{\circ}C$ heating temperature and 67% moisture content at the 2$0^{\circ}C$ storage temperature, which resulted in 25% yield with these experiment conditions. Affecting factor for the resistant starch formation was arranged according to heating -cooling cycle, moisture content, heating temperature and storage temperature. Raw corn starch granule was destructive and appeared a porous reticular structure by the resistant starch formation. Color became dark and increased yellowness by caramelization during heating processing. Heating-cooling processing was the result of decreased hardness, cohesiveness, springiness and gumminess.

Influence of Different Transplanting Dates on Amylopectin Branch-chain-length and Pasting Properties of Rice Flour Varieties

  • This study was conducted to examine the influence of different transplanting dates on rice quality and starch properties (morphology and pasting properties) of rice varieties that may be used for the production of rice flour. Three rice flour varieties, 'Seolgaeng', 'Hangaru', and 'Milyang317', were transplanted on May 20, May 30, June 10, and June 20. The peak viscosity decreased with a delay in the transplanting date. However, the amylose content increased with a delay in the transplanting date, whereas that of protein decreased. Amylopectin short-branch chain content increased in the rice varieties that were transplanted on May 30. The morphology of the starch granule of the varieties was determined by SEM. No apparent external difference in the starch granules was observed for the different transplanting dates. These results indicate that the transplanting date influenced the amylopectin structure and pasting properties, which led to changes in the physicochemical characteristics of rice starch.

Comparison of Foliar Ultrastructure of 3 Dubautia species (Dubautia속(屬) 3종(種) 식물(植物)의 엽육조직(葉肉組織) 미세구조(微細構造) 비교연구(比較硏究))

  • The fine structure of palisade chloroplasts has been studied in the mature leaves of 3 Dubautia species (D. scabra var. leiophylla, D. knudsenii and D. scabra var. leiophylla${\times}$D. knudsenii) to explore variation at the ultrastructural level, since the parental species exhibit quite different morphological and anatomical features. Types of thylakoidal membrane systems, occurrence and distribution of phytoferritin-like structures, lipid droplets, starch grains, mitochondria and microbodies were examined. Four different types of thylakoidal membranes were found in D. scabra var. leiophylla, 2 rather uniform types in D. knudsenii and 3 intermediate types in their hybrid. D. scabra var. leiophylla and the hybrid were marked by statistically significant differences in mean numbers of thylakoids per granum, while no significant difference was found between D. knudsenii and the hybrid. Phytoferritin-like structures which were about $100-120{\AA}$ in diameter as a whole particle each were found in all 3 species. The amount and distribution of particles varied by species. Lipid droplets, plastoglobuli, and starch grains occurred in all 3 species, but the frequency of starch grains also varied with the species. More frequent and larger starch grains were observed in D. knudsenii than in the other two species. Microbodies, or peroxisome, were observed throughout all species. They occurred, either with or without crystalline inclusions, around the chloroplasts.

Molecular Structure of Sorghum and Waxy Sorghum Starches

  • Amylose contents and amylopectin chain architecture of sorghmn and waxy sorghum starches were determined and compared with those of other common cereal and tuber starches. Also, in vitro digestibility of sorghum starch was estimated using a novel methodology. The absolute amylose content of sorghum starch was similar to that of com and wheat starches. The side chain length distribution patterns for sorghum and waxy sorghum amylopectin were very similar to those of com and waxy com, respectively. The $k_{cat}/K_m$ values for sorghum and potato amylopectin did not show a significant difference. The kinetic parameters could be used as novel indicators for starch digestibility.

Modification of Sweet Potato $\beta$-Amylase with Periodate-Oxidized Soluble Starch (과요소산 산화 전분에 의한 고구마 $\beta$-아밀라제의 수식)

  • Sweet potato $\beta$-amylase is a tetrameric enzyme consisting of four identical polypeptide chains with a molecular weight of 5.6$\times$104, though most of the other $\beta$-amylases are monomeric enzymes. But, the relationship between subunit structure and catalytic function of the enzyme is not known. This study was done to know what the function of the subunit structure of the enzyme is. We obtained the monomer from the enzyme by the treatment of SDS, alkali pH buffer and urea. But the monomer had not activity. We tried to prepare the active monomer from the enzyme by the modification with periodate-oxidized soluble starch , In the result, we succeeded in isolating an active monomer as an oxidized soluble starch-conjugated form The active monomer had 57% of the original activity, 13.2% of the sugar and the molecular weight was estimated to be 5.4$\times$104. This results suggest that the tetrameric form of the enzyme is a most stable one and exists in nature, and the subunit structure of the enzyme Plays an important role in stabilization but not catalytic function.

Study of Molecular and Crystalline Structure and Physicochemical Properties of Rice Starch with Varying Amylose Content (아밀로오스 함량이 다른 쌀 전분의 분자 및 결정 구조와 이화학적 특성)

  • The in vitro digestibility and molecular and crystalline structures of rice starches (Seilmi, Dasan1, and Segoami) with differing amylose content were investigated. Segoami had the highest amylose content (30.9%), whereas Dasan1 had the lowest amylose content (21.2%). The molecular weight ($\bar{M}_w$) of amylose and amylopectin in Segoami was much lower than that of the other two rice starches. Segoami had the highest proportion (8.7%) of amylopectin short branch chains (DP 6-12) and the lowest proportion of B1 chains (DP 13-24). The relative crystallinity, intensity ratio of $1047-1022cm^{-1}$ (1047/1022) and gelatinization enthalpy followed the order: Segoami>Seilmi~Dasan1. Segoami showed substantially low pasting viscosity. Rapidly digestible starch (RDS), slowly digestible starch (SDS), and resistant starch (RS) contents showed the highest value in Seilmi, Dasan1, and Segoami, respectively. The expected glycemic index (eGI) of Segoami was lower than that of the other two rice starches. Overall results suggested that the digestibility of rice starch could be highly influenced by their molecular and crystalline structure.
