Chnracterization and Inhibitory Activity on Staphylococcus aureus of a Bacteriocin Produced by Lactobacillus plantarum KU107 (Lactobacillus plantarum KU107이 생산하는 박테리토신의 특성 및 Staphylococcus aureus 억제 작용)

  • A bacteriocin producing lactic acid bacteria was isolated from ground beef and the strain was identified as Lactobacillus plantarum ssp. by use of API carbohydrate fermentation pattern and physiological tests. The bacteriocin produced by L. plantarum KU107 exhibited a good spectrum of activity against foodborne pathogens including Bacillus cereus, Escherichia coli, Listeria ivanovii, Listeria monocytogenes, Pseudomonas aeruginosa, Pseudomonas chlororaphis, Staphylococcus aureus, Staphylococcus intermedius, Salmonella typhimurium and Yersinia enterocolitica. The bacteriocin was active over a wide pH range and stable of heat treatment, and inactivated by treatment with proteases. A bacteriocin from L. plantarum KU107 was effetive in reducing S. aureus in tryptic soy broth. On the ground beef containing S. aureus was added with the crude bacteriocin, S. aureus was inhibited during storage period at 4$\^{C}$.

Chemical Composition and Antimicrobial Activity of Essential Oil Extracted from Eucalyptus citriodora Leaf

  • Eucalyptus oil is a rich source of bioactive compounds with a variety of biological activities and is widely used in traditional medicine. Eucalyptus citriodora is cultivated for the production of essential oils. However, the mode of antibacterial action of essential oils from E. citriodora is not well-known. This study aimed to determine the chemical components, microbial inhibitory effect, and mechanism of action of the essential oil from E. citriodora. The oil was extracted from E. citriodora leaves by hydro-distillation and the chemical components were analyzed using gas chromatography-mass spectrometry. The antibacterial activities of eucalyptus oil against gram-positive bacteria (Bacillus subtilis, Staphylococcus aureus, and Staphylococcus intermedius) and gram-negative bacteria (Escherichia coli and Pseudomonas aeruginosa) were screened by disc diffusion method and quantitative analysis was conducted by the microdilution method. The mechanism of action of the extracted essential oil was observed using SEM and analyzed by SDS-PAGE. The major components of E. citriodora oil were citronellal (60.55 ± 0.07%), followed by dl-isopulegol (10.57 ± 0.02%) and citronellol (9.04 ± 0.03%). The antibacterial screening indicated that E. citriodora oil exhibited prominent activity against all tested strains. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) against B. subtilis were 0.5% and 1.0%, respectively. The MIC and MBC concentrations against S. aureus, S. intermedius, E. coli, and P. aeruginosa were 1% and 2%, respectively. As observed by SEM, the antibacterial mechanism of E. citriodora oil involved cell wall damage; SDS-PAGE revealed decrease in protein bands compared to untreated bacteria. Thus, E. citriodora oil showed significant antimicrobial properties and caused cellular damage.

  • The result of API staph-ident system was compared with cellular fatty acid composition for the identification of Staphylococcus species isolated from cattle. Isolated strains from cattle were correctly identified to S aureus, S intermedius, S hyicus, S simulans, S saprophyticus, S epidemis, S sciuri and S xylosus by API staph-ident system. The correlation between bacterial cellular fatty acid profile and Staphylococcus species isolated to API STAPH-IDENT system were. In conclusion, the result presented indicate that Staphylococci can be indentified to the species level by the cellular fatty acid profiles. Moreover, computerized fatty acid profile correlative anaylsis can be applied for determining identify of Staphylococcus species.

Antibacterial Efficacy of Chitosan against Staphylococcus intermedius in Dogs (개의 표재성 농피증에서 분리된 Staphylococcus intermedius에 대한 키토산의 항균효과)

  • The antibacterial efficacy of 0.1% (w/v) chitosan solution against Staphylococcus intermedius isolated from a dog with superficial pyoderma was evaluated in vitro and in vivo. The exposure time for the 0.1% chitosan solutions at different pH to be able to eliminate the bacterial cells and the effect of pH of the solutions on antibacterial activity was tested at the same time in vitro. The antibacterial activity of chitosan was compared to other antibacterial agents including 2.5% benzoyl peroxide, 0.5% chlorhexidine acetate, 0.1% chitosan solution combined with 2.5% benzoyl peroxide and chitosan combined with 0.5% chlorhexidine using a modified detergent scrub quantitative technique in 10 adult mongrel dogs in vivo. They were able to eliminate a number of bacteria after the exposure time of 10 minutes at varying degrees according to the pH of the solutions. The antibacterial activity of chitosan was inversely affected by pH with higher activity at lower pH value. The 0.1% chitosan solution was also efficacious against Staphylococcus intermedius in vivo. The combinations of chitosan with benzoyl peroxide and with chlorhexidine were shown to exert higher activity when compared to those of chitosan alone and benzoyl peroxide or chlorhexidine alone. The 0.1% chitosan solution was considered to be efficacious against Staphylococcus intermedius isolated from a dog with superficial pyoderma in both in vivo and in vitro and have a potential for the clinical applications in the treatment or pyoderma in dogs.

Distribution and Antimicrobial Susceptibility of Bacteria in the Oral Cavity of Smokers or Non-Smokers (흡연자와 비흡연자간의 구강 내 세균 분포 및 항균제 감수성)

  • It is well known that smoking as well as drinking is a factor of stomatopathy, however there are few investigations about comparison of oral flora between smokers and non-smokers. In this study, we isolated the oral flora of 30 smokers and 30 non-smokers and cultured them on blood agar plates. The isolated pathogenic microorganisms were tested for antibiotic susceptibility and resistance using the Kirby-Bauer antibiotic testing method. Each colony was stained using the Gram staining method and was identified by an automatic identifier, known as the VITEK system. We isolated 41 colonies from smokers' oral cavity, and they were sorted as 63% of Gram-positive cocci, 29% of Gram-negative cocci, 3% of Gram-positive bacilli, and 5% of Gram-negative bacilli by gram staining, whereas 38 colonies were isolated from non-smoters' oral cavity, and their proportions were 55% of Gram-positive cocci, 26% of Gram-negative cocci, 3% of Gram-positive bacilli, and 16% of Gram-negative bacilli. The VITEK system revealed specific distribution of bacteria species that Streptococcus mutans (6/41), Gemella morillorum (6/41), Streptococcus oralis (2/41), Streptococcus pneumoniae (1/41), Staphylococcus aureus (3/41), Streptococcus anginosus (1/41), Streptococcus intermedius (1/41), Streptococcus uberis (1/41), and Streptococcus sanguinis (1/41) in smokers oral cavity whereas Streptococcus sanguinis (8/38), Staphylococcus aureus (1/38), Staphylococcus auricularis (1/38), Streptococcus uberis (1/38), Streptococcus intermedius (1/38), Streptococcus mutans (1/38), and Streptococcus oralis (1/38) in those of non-smokers'. Three cases of Staphylococcus aureus from smokers produced Beta-lactamase and were identified methicillin-resistance Staphylococcus aureus (MRSA). However one case of Staphylococcus aureus from non-smoker did not produce Beta-lactamase and was sensitive to methicillin. In conclusion, the distribution of oral flora was different between smokers' and non-smokers' oral cavity, especially Gemella morillorum and MRSA were predominantly found in smoker's oral cavity. These results are useful in the treatment and prevention of patients with stomatopathy caused by smoking.

Antimicrobial Efficiency in the Fermented Slurry of Unpolished Rice (현미 발효 슬러리의 항균활성)

  • Unpolished rice (UR) is considered to be a healthy alternative to white rice when coping with chronic diseases. In the present study, the fermented slurry of unpolished rice (FSUR) was evaluated with respect to its antimicrobial activities and biochemical characteristics, including the quantities of sugar, total soluble sugar, organic acids, free amino acids, pH, and physiological activity. The antimicrobial efficiency of FSUR was assessed using the paper disc-agar diffusion method. FSUR exhibited strong antimicrobial activity against six pathogenic bacterial strains (Staphylococcus aureus, Escherichia coli, Listeria monocytogenes, Pseudomonas aeruginosa, Salmonella typhimurium, and Yersinia enterocolitica) and two fermentation strains (Gluconacetobacter intermedius and Lodderomyces elongisporus). The antimicrobial activity of FSUR was higher than the commercial antibiotics, carbenicillin ($50{\mu}g/ml$) and tetracycline ($50{\mu}g/ml$) against S. aureus, E. coli, L. monocytogenes, P. aeruginosa, S. typhimurium, Y. enterocolitica, and L. elongisporus. Also FSUR had a high antioxidant activity. The microorganisms were isolated from FSUR using tryptic soy broth and yeast extract-peptone-dextrose agar media. The isolated microorganisms were characterized using physiological and biochemical analyses as well as by 16S rRNA gene sequencing and phylogenic analysis. 16S rRNA gene sequence analysis showed that the isolated microorganisms had a high similarity to G. intermedius, Lactobacillus casei, Lactobacillus plantarum, and Acetobacter peroxydans.

Antibiotic Resistance of Bacterial Isolates from Nasal Discharges of Dogs with Respiratory Diseases (개의 비루에서 분리한 원인균의 항생제 내성)

  • Bacterial pathogens were isolated from 36 dogs with respiratory signs, that were submitted to Veterinary Clinics in Jeju, including Veterinary Medical Teaching Hospital in Cheju National University. Of 36 isolates, 16 (44.4%) bacterial pathogens were Gram-positive and 20 (55.6%) were Gram-negative. Gram-positive bacteria identified with API Staph were 12 S. intermedius (33.3%), 2 S. aureus (5.6%), 1 S. haemolyticum (2.8%), and 1 S. xylosus (2.8%). Gram-negative organisms identified with API 20E or API NE included 8 Bordetella bronchiseptica (22.2%), 6 Escherichia coli (16.7%), 4 Pasteurella spp. (11.1%), 1 Enterobacter intermedius (2.8%), and 1 Oligella ureolytica (2.8%). Both Staphylococcus spp. isolates and Gram-negative pathogens were resistant to one or more antibiotics, including ampicillin (AM), amoxicillin/clavulanic acid (AMC), chloramphenicol (C), cefazolin (CZ), erythromycin (E), gentamicin (GM), kanamycin (K), lincomycin (L), oxacillin (OX), trimethoprim/sulfamethoxazole (SXT), and tetracycline (TE). All Staphylococcus spp. were susceptible to AMC, OX and VA, while many isolates were highly resistant to L (87.5%), E (68.8%), P (62.5%), and AM (56.3%). Antibiotic-resistant patterns of staphylococcal isolates were shown ranges from single to 9-resistant patterns. Resistant rates to antibiotics of Gram-negative bacteria were usually higher than those of Staphylococcus spp. in this study. Most Gram-negative bacteria were highly resistant to L (90.0%), AM (85.0%), E (85.0%), P (85.0%), OX (80.0%), and CZ (75.0%). B. bronchiseptica isolates showed 5 to 8 antibiotics-resistant patterns and Pasteurella spp., 2 to 8-resistant patterns. In particular, all 6 E. coli isolates were resistant to more than 9 different kinds of antibiotics, including one strain resistant to all antibiotics tested.

Chemosystematic and Phenotypic Characterization of Gram-positive Bacteria from Coastal Seawater, Korea (한국 근해에서 분리한 그람양성 세균의 화학 분류학적 및 표현형적 특성)

  • Twenty-five halotolerant gram-positlve bacteria were isolated from the coastal seawater 01 Cheju Island and Incheon J&yakdo Chemosystematic and phenotypic characteristics were used to iuvestigate the taxonomic position of these bacteria. According to their chemosystematic characteristics, the twenty-tive isolates were divided into 4 groups. Group 1 bacteria possesed 40.1 to 49.9 inol% m DNA G+C content, menaquinone-7 as a major quinone, and meso-Alpm as a diamino acid of peptidoglycan. Group 1 tam were identified as Bacilluspumilis, Bacillus lichenifbrrizis, Bacillus megaterium, Bncill~rsubtilis. Group 2 bacteria possessed 63.9 to 66.4 mol% and MK-8. They were all in the genus Arth~obaclm-. Group 3 bacteria possessed 31.0 to 37.6 mol% and MK-7. They were identified as Staphylococcus haeniol.viicvs, Siaph~~lococc~is sapropl~j~ticns, and Siaphylococcus intermediru.. Group 4 bacterium possessed 72.0mol% and MK-8 and was identified as ~Lficrococcus ltdtezm. Bacillus species accounted for 68% of h e total isolates.

Microfloras of otitis externa and normal external ear canals in dogs (개에서 외이도염 및 정상외이도에서 분리한 세균총에 대한 연구)

  • This study was conducted to examine the outbreak rate and the causative agents of otitis externa in 26 dogs (49 ears ; 23 dogs = bilateral, 3 dogs = unilateral), and the normal microfloras of external ear canal in 68 dogs(133 ears ; 65 dogs = bilateral, 3 dogs = unilateral ) in Taegu, 1997. The breed, living environment, sex, age and season distribution of otitic dogs were as follows : Dogs with erect and hairy ears(42.3%), pendulous and hairy ears(38.5%), indoor(92.3%), female(65.4%) and below one year old(38.5%) were more prevalent. According to season, otitis externa was mainly occurred between July and October. The major causative agents of canine otitis externa were Malassezia pachydermatis (32.7%), Staphylococcus aureus (26.5%) and S intermedius (16.3%). In the microorganism isolated 39 otitic ear canals, single infection was 53.8% and mixed infection was 46.2%. The normal microfloras of canine external ear canal were fungi including M pachydermatis, Aspergillus spp, Microsporum canis, Alternaria spp, Verticillium spp and Yeast, and bacteria including Staphylococcus spp(10 species including S xylosus), Bacillus spp, Corynebacterium spp, Listeria spp, Actinomyces pyogenes and Escherichia coli. No growth was 34.6%.

Effect of Green Tea Extract on Healing of Contaminated Wound in Dogs (개의 피부 감염창 치유에 대한 녹차 추출물의 국소도포 효과)

  • The purpose of this study is to assess healing effect of the green tea on Staphylococcus contaminated wound. On the back of 7 dogs, 6 full-thickness skin wounds($2cm{\times}2cm$) were made and Staphylococcus intermedius was inoculated. Each wound was applied with the 2%(experimental group I), 1%(experimental group II) extract of green tea and normal saline(control group I) after inoculation. The wound contraction rate of the experimental group I and II was higher than that of the control group in entire period. And from the $6^{th}\;to\;22^{nd}$ day, the wound contraction rate of the experimental group I and II was significantly higher than that of the control group(p<0.05). The occupation rate of wound healing in the experimental group I was higher than that of the control group from $0\;to\;9^{th}$ day except from $1^{st}\;to\;2^{nd}$ day and also higher than that of the experimental group II from $0\;to\;3^{rd}$ day except from $1^{st}\;to\;2^{nd}$ day. And the occupation rate of wound healing in the experimental group II was higher than that of the control group from $1^{st}\;to\;8^{th}$ day except from $2^{nd}\;to\;3^{rd}$ day. According to concentration of green tea extract increased, the wound contracted more vigorously in earlier period. The tensile strength of the experimental group I and II was significantly higher than that of the control group(p<0.05). And that of the experimental group I was also higher than in the experimental group II(p<0.05). The number of S. intermedius in the experimental group I and II was decreased more rapidly than that of the control group. From the $4^{th}$ to the $11^{th}$ day, the number of S. intermedius in the experimental group I was significantly lower than that of the experimental group II and the control group. And from the $4^{th}$ to the $7^{th}$ day, the number of S. intermedius in the experimental group was significantly lower than that of the control group. The histopathological findings of the experimental group I and II were similar to those of the control group on the $1^{st}\;and\;4^{th}$ day. On $14^{th}$ day, reepithelialization was completed in the experimental group I and II except the control group. Higher wound contraction rate, better bacteriocidal action, higher tensile strength and better histopathological findings were observed in the experimental group I and II than in the control group. These results suggest that the topical application of green tea extract can promote contaminated wound healing in dogs.
