Isolation and characterization of Staphylococcus hyicus subsp hyicus from chickens (닭으로부터 Staphylococcus hyicus subsp hyicus의 분리(分離) 및 그 특성(特性))

  • Isolation of Staphylococcus hyicus subsp. hyicus from the skin of healthy chickens was performed and biochemical characteristics of the isolates were examined. Staphylococcus hyicus subsp hyicus was isolated from 129(14.8%) of 874 healthy chickens. The rate of isolation of this organism from chickens was found to vary depending upon the poultry farms at the isolation rate of 0% to 38.7% and this organism was isolated more frequently from adult chickens of over 1 year old than young chickens. All of the isolates were negative for haemolysis on rabbit, sheep, pig and chicken blood agars, protein A and ${\beta}$-glucuronidase, but positive for coagulase using pig plasma, heat-stable DNase, phosphatase and Tween 80 hydrolysis. Hyaluronidase was positive in 97.7% of isolates, but their reaction was weaker than that of swine strains of Staphylococcus hyicus subsp hyicus. The isolates were biotyped on the basis of the reactions for protease, urease, nitrate, galactose, trehalose and lactose, and 11 biotypes were found among the 129 isolates of Staphylococcus hyicus subsp hyicus.

  • 돼지로 부터 분리한 Staphylococcus hyicus subsp hyicus 489주의 혈청형을 자가생산한 인자혈청을 이용한 평판응집반응으로 동정하였다. 분리균의 혈청형은 A(16.8%), C(20.4%), D(9.4%), E(7.2%), F 형(29.2%) 및 미분류형(17.0%)으로 동정되었으며, 돼지의 건강상태, 신체부위 및 연령에 관계없이 F형의 분포가 가장 높았다.

  • Swine plasminogen is not activated by staphylokinase of Staphylococcus aureus. In this study, the activation of swine plasminogen by staphylokinase of Staph hyicus subsp. hyicus was investigated and the effect of EDTA(disodium) on plasminogen activation was also studied. When the activation of swine plasminogen by staphylokinase of Staph hyicus subsp. hyicus was examined in fresh swine plasma, swine plasminogen could be weakly activated. However, when EDTA was added to the swine plasma, plasminogen activation was markedly enhanced, but this enhancement was not observed on bovine fibrin-dog plasminogen agar plate containing EDTA. Chicken and bovine plasminogens were not activated by staphylokinase of Staph hyicus subsp. hyicus. Using fresh swine plasma agar containing 0.07% EDTA, staphylokinase activity was detected in 96.3% of Staph hyicus subsp. hyicus strains isolated from pigs and in none of the chicken and bovine strains.

  • Strains of Staphylococcus hyicus subsp. hyicus were compared and differentiated by staphylococcin sensitivity. Strains were tested by the cross-streaking method on brain heart infusion agar. From 31 staphylococcin producers, a typing set of 9 producers was selected and used to type all of the strains. All 81 strains of Staph hyicus subsp. hyicus tested were typable and were differentiated into 12 different sensitivity patterns. Strains of the same serotype from different sources could be further subdivided by their staphylococcin sensitivity patterns. Other staphylococcus species could also be typed by use of the 9 standard strains of Staph hyicus subsp. hyicus.

  • A total of 113 Staphylococcus hyicus subsp. hyicus which had been isolated from healthy and diseased pigs were examined for their ability to produce staphylococcins. For selection of staphylococcin indicator strains, 80 strains were randomly selected from 113 strains, and were tested as indicator and as donator. The 8 indicators were chosen and used to type all of the strains. The ability to produce staphylococcin could be demonstrated in 43 strains (38.1%) of 113 Staph hyicus subsp. hyicus. The frequency of activity was higher in diseased pig strains (44.4%) than in healthy pig strains(36.8%). 43 staphylococcin producer strains were differentiated into 10 reaction types. The inhibitory effects of the staphylococcin (N27 and R 3) on other staphylococci differed by the staphylococcal species tested. All the gram-negative species were completely resistant to the three producing strains.

  • The Presence and quantity of protein A in Staphylococcus hyicus subsp hyicus isolates from pigs were determined by indirect hemagglutination test and enzyme-linked immunosorbent assay(ELISA). Cell-bound and extracellular protein A was demonstrated in 87.7% and 36.0% of 489 isolates, respectively, by indirect hemagglutination test. When contents of the protein A were estimated by ELISA method, all of the isolates that were positive to protein A in indirect hemagglutination test produced extracellular protein A of less than 1ng/ml. Most of the isolates produced cell-bound protein A of less than 1ng/ml, whereas 28 isolates produced 1 to 35ng/ml and 11 isolates produced 25 to 108ng/ml.

  • Isolation of Staphylococcus hyicus subsp. hyicus from piglets suffering from exudative epidermitis and healthy pigs was performed, and some properties of the isolates were examined. Twentry-six litters of exudative epidermitis observed in the field were sucking piglets with ages ranging from 1 to 5 weeks and most (73.1%) of these cases occurred in piglets between 2 and 3 weeks of age. Staphylococcus hyicus subsp. hyicus was isolated front 192(38.9%) of 494 healthy pigs. The rate of isolation of these organism from healthy pigs was found to vary greatly among farms at an isolation rate of 33.3% to 45.5% and this organism was isolated more frequently front sucking piglets below the age of 8 weeks than adult pigs. All of the isolates originated from the diseased and healthy pigs were positive in heat-stable DNase, Tween 80 hydrolysis, gelatinase, protease, but negative in clumping factor and hemelysin on sheep blood agar. There was no difference related to origin in the production of extracellular active substances. Typical lesions of the classic exudative epidermitis were produced by inoculation of Staphylococcus hyicus subsp. hyicus isolated from the diseased and healthy pigs. The pathogenic potentials of isolates from healthy pigs were no different from that of the isolates obtained from diseased pigs.

Identification of Staphylococcus hyicus subsp hyicus of swine and poultry origin by API STAPH-IDENT system (API STAPH-IDENT system에 의한 돼지 및 닭유래(由來) staphylococcus hyicus subsp hyicus의 동정(同定))

  • The API STAPH-IDENT system was evaluated as a means for identifying Staphylococcus hyicus subsp hyicus straints isolated from swine and poultry. Of 80 strains from swine, 68 (85%) were correctly identified by the API STAPH-IDENT system alone after 5 h of incubation. When results were determined after 24 h of incubation, the accuracy of this system alone was 93.8%. By additional tests in conjunction with the API STAPH-IDENT system, however, all 80 strains could be correctly identified. Of 120 strains from poultry, 87 (72.5%) required additional testing to achieve a correct identification, and 33 (27.5%) were incorrectly identified by this system after 5 h of incubation. After 24 h of incubation, 99 of 120 (82.5%) avian strains were incorrectly identified as Staph epidermidis owing to false-negative mannose and trehalose utilizations. Seventy-seven (96.3%) of swine strains were positive for ${\beta}-glucuronidase$, whereas all 120 strains recovered tram poultry were negative for it.

Production of staphylokinase in Staphylococcus hyicus subsp. hyicus strains of swine, poultry and bovine origin (돼지, 닭 및 소유래 Staphylococcus hyicus subsp. hyicus의 staphylokinase 산생능)

  • Staphylococcus hyicus subsp. hyicus strains isolated from pigs, chickens and cattle were examined for the production of staphylokinase after inhibition of staphylococcal proteases by two procedures with EDTA(disodium). In one, EDTA was added to the bovine fibrin-dog plasminogen agar medium in concentration of 0.07% and paper strips soaked in 2mg/ml soy bean trypsin inhibitor were then applied on the agar plates. In the other, paper strips soaked in 5% EDTA solution were applied on the bovine fibrin-dog plasminogen agar plates and the strains to be tested were then streaked at right angles with the strip. By these procedures, staphylokinase activity was detected in 8(88.9%) of 9 strains from diseased pigs and in 57(80.3%) of 71 strains from skin of healthy pigs, but not in any strains from skin of healthy chickens and milk samples of mastitic cattle. Additionally kinase activity in 9 Staphylococcus species and subspecies isolated from bovine intramammary infections was also tested by these procedures. Staphylokinase activity was detected in 74.2% of Staph aureus strains and in 25% of Staph xylosus strains.

  • The serological typing of staphylococcus hyicus subsp. hyicus isolated from pigs with exudative epidermitis and healthy pigs was attempted by slide agglutination technique with absorbed sera prepared from antisera against swine strains of the organism. Of 204 Staph. hyicus subsp. hyicus isolated, 202 (99.0%) were classified into 6 serotypes tentatively designated as A, B, C, D, E and F. The organisms were found to possess common antigens and a strain specific antigen. In 26 epizootics of exudative epidermitis, type A was found in 4(15.4%), type B in 12(46.2%), type C in 9(34.6%) and type E in 1(3.8%). All of the piglets of each affected litter were infected by serologically identical strain. In 178 strains isolated from the skin of healthy pigs, type A was found in 14.1%, type B in 23.0%, type C in 30.3%, type D in 1.7%, type E in 7.9% and type F in 21.9%, and 2 strains were untypable. There was a difference in the occurrence of serotypes of Staph. hyicus subsp. hyicus among the farms examined.
