  • New type of syngas generator based on the partial oxidation of biogas in volumetric permeable matrix reformers was suggested as an effective, adaptable and relatively simple way of syngas and hydrogen production for various low-scale applications. The use of biogas as an energy source reduces the chance of possible emission of two greenhouse gases, $CH_4$ and $CO_2$, into the atmosphere at the same time. Its nature of being a reproducible energy source makes its use even more attractive. Parametric screening studies were achieved as air ratio, biogas component ratio, input gas temperature, Steam/Carbon ratio. As the air ratio was low, the production of the hydrogen and carbon monoxide increased in the condition that 3D matrix reformer maintains the stable driving. As it was the simulation biogas in which the carbon dioxide content is high, the flammable range became narrow. And the flammable range was extended if the injected gas was preheated. The stable driving was possible in the low air ratio. The amount of hydrogen production was increased as S/C ratio increased.

  • This paper has reviewed the China Sericulture history in the 20th century. On the basis of the current sericulture production, sericulture area distribution, problems and challenges, it has been forecasted that China sericulture will be developed as following points: 1) On a stable basis, cocoon production increased gradually to a suitable amount; 2) Quality cocoon production bases established to form a regionalized, industrialized and specialized sericulture structure; 3) With increase of householding rearing capacities, rearing facilities improved; 4) Production efficiency raised and silk product quality improved through technical innovation; 5) New and diversified products developed from Sericultural resources with new and high technologies.

  • For production of thermophilic and alkaline protease, Bacillus sp. strain AP-5 was isolated from a compost. The production of the protease was reached at maximum for 4 days at $55^{\circ}$ in standing culture. Chitin and Cellulose as carbon source, and Skim Milk as nitrogen source were favorable for the production of the enzyme. Optimal temperature and optimal pH of the enzyme was $55^{\circ}$ and 11, respectively. Metal ion didn't effect on the enzyme activity, the protease was very stable at heat treatment of 30 min at $55^{\circ}$.

  • In order to realize a stable rubbing process for the liquid crystal panel, the authors investigated the quantitative evaluation method of rubbing process uniformity. The proposed method focuses on the relationship between the image quality of the LCD panel for gray scale images and the rubbing uniformity. The proposed method indicates rubbing uniformity using quantitative parameters of spots and hairlines on the LCD panel.

  • It is very important to get a stable and large-capacity organic effusion source for achievement of OLED mass-production equipment. We present an organic effusion source with film uniformity less than ${\pm}$ 5%, the material charge volume, 300cc for $400{\times}400\;mm^2$ substrate. The fine metal shadow mask alignment technology, one of the color forming technique, also have to support more accurate and fast operating in mass-production. In this paper, we will describe the OLED mass-production equipment with the large volume effusion source and the precision shadow mask alignment technique.

Use of Flp-Mediated Cassette Exchange in the Development of a CHO Cell Line Stably Producing Erythropoietin

  • The feasibility of the use of Flp-mediated cassette exchange in the development of a CHO cell line, which produces erythropoietin (EPO) stably and largely, was investigated. A stable, high enhanced green fluorescence protein (EGFP)-producing clone was screened by extensive flow cytometric analysis. An EPO expression unit was targeted into the premarked locus of the stable parental clone by Flp-mediated cassette exchange and a correctly targeted clone (FC28T7) was obtained. The EPO production of FC28T7 was proven to be stable in long-term culture. Furthermore, the Flp-mediated cassette exchange did not alter the stable parental clone's characteristics concerning transgene expression level and stability. Taken together, the data obtained here indicated that the establishment of CHO cell lines stably producing a desired protein is achievable using Flp-mediated cassette exchange.

Production of L-tryptophan by Genetically Engineered Klebsiella pneumoniae (유전자 조작된 Klebsiella pneumoniae에 의한 L-tryptophan의 생산)

  • The optimum conditions for the production of tryptophan using a recombinant Klebsiella pnuemoniae phe A tyr A trp R/pSC 101-$trp^{+}$ and its plasmid stability during tryptophan production were studied. The optimum temperature was $37^{\circ}C$ and the specific growth rate was 1.05$h^{-1}$ at $37^{\circ}C$. Tryptophan production was increased by glucose fed-batch culture, and tryptophan was accumulated to 0.175 g/l after 36 hrs. This amount was about 1.2 and 1.6 times greater than that obtained from batch culture and flask culture, respectively. The stability of the strain in fed-batch cu1ture was greatly different from that in repeated flask culture. After 6 generation, 95% of total cells was stable in repeated flash culture, but in fed-batch culture only 50% was stable.

Transesterification Using the Cross-Linked Enzyme Aggregate of Photobacterium lipolyticum Lipase M37

  • Biodiesel is methyl and ethyl esters of long-chain fatty acids produced from vegetable oils or animal fats. Lipase enzymes have occasionally been used for the production of this biofuel. Recently, biodiesel production using immobilized lipase has received increased attention. Through enhanced stability and reusability, immobilized lipase can contribute to the reduction of the costs inherent to biodiesel production. In this study, methanol-tolerant lipase M37 from Photobacterium lipolyticum was immobilized using the cross-linked enzyme aggregate (CLEA) method. Lipase M37 has a high lysine content (9.7%) in its protein sequence. Most lysine residues are located evenly over the surface of the protein, except for the lid structure region, which makes the CLEA preparation yield quite high (~93%). CLEA M37 evidences an optimal temperature of $30^{\circ}C$, and an optimal pH of 9-10. It was stable up to $50^{\circ}C$ and in a pH range of 4.0-11.0. Both soluble M37 and CLEA M37 were stable in the presence of high concentrations of methanol, ethanol, 1-propanol, and n-butanol. That is, their activities were maintained at solvent concentrations above 10% (v/v). CLEA M37 could produce biodiesel from olive oil and alcohols such as methanol and ethanol. Additionally, CLEA M37 generated biodiesel via both 2-step methanol feeding procedures. Considering its physical stability and reusability, CLEA M37 may potentially be used as a catalyst in organic synthesis, including the biodiesel production reaction.

Characterization of Bacteriocin Production by Lactococcus lactis LAB3113 Isolated from Kimchi

  • A lactic acid bacterium LAB3113, isolated from traditionally fermented Kimchi was found to produce bacteriocin whose activity was very specific toward lactobacilli and not effective against any other bacteria. Lactobacilli affected by the inhibitory substance included Lactobacillus delbrueckii-lactis, L. johnsonii, L. gsseri, and L. curvatus. Based upon biochemical and physiological characteristics, LAB3113 was classified as Lactococcus lactis, and its bacteriocin was named as lactococcin K3113. Lactococcus lactis. LAB3113 produced bacteriocin at th early stage of growth and the concentration of the bacteriocin did not decrease even after alt stationalry phase. Optimal temperature of bacteriocin production was $25^{\circ}C$ at the initial pH 7.0. Partially purified lactococcin K3113 was completely inactivated by protease, but not affected by lipase, lysozyme and RNase. The bacteriocin was very heat-stable even after autoclaving for 20 min. It was also stable in pH changes, an was not affected by th presence of solvents. lacotococcin K3113 appeared to act in bactericidal mode against L. delbrueckii-lactis ATCC4797. Molecular weight of lactococcin K3113 was calibrated as 10,500 dal by SDS-PAGE an activity staining. Lactococcus lactis LAB3113 had four residential plasmids of 3.7kb, 11.2kb, 15.5kb, and 48kh in molecular sizes. Plasmid profile analysis of mutant strain revealed that 15.5 kb plasmid was re-sponsible for the production of lactococcin K3113 and its immunity to the bacteriocin.

  • This Purpose of this study is to develop a system for stable production without troubling side effects, scalp and skin, and sensitivity to temperature and humidity, but the ozone-cream at a constant effective value and the durability of peroxide safe discharge tube. Furthermore, olive oil and ozone is reacted with the wish to apply to the manufacturing technology. If peroxide value of ozonized olive oil exceeded 1200 meg/kg, there may be microbial disinfection. However, to be produced with a minimum of risk because they look to the scalp and skin sensitization is the purpose of the present study. And also the durability of the discharge tube was tested during the production unit for stable production. And the experimental results, Peroxide value of the ozonized olive oil was measured to less than 1300 meg/kg, also it showed a stable result of the discharge tube.
