Inhibitory Effect of BCG Cell-Wall Skeletons (BCG-CWS) Emulsified in Squalane on Tumor Growth and Metastasis in Mice

  • The antimetastatic effect of BCG-CWS, which was emulsified in an oil-in-water form with either Drakeol 6VR mineral oil (BCG-CWS/DK) or squalane (BCG-CWS/SQA), on lung metastasis produced by highly metastatic murine tumor cells, Colon26-M3.1 carcinoma cells and B16-BL6 melanoma cells, was investigated in syngeneic mice. An intravenous (i.v.) administration of BCG-CWS (100 mg/mouse) 1 day after tumor inoculation significantly inhibited tumor metastasis of both Colon26-M3.1 carcinoma and B16-BL6 melanoma cells in experimental lung metastasis models. No differences in the antitumor activity of the two oil-based formulations (BCG-CWS/DK and BCG-CWS/SQA) were obverved. However, BCG-CWS/SQA administered through subcutaneous (s.c.) route was shown to be effective only when it was consecutively injected (3 times) after tumor inoculation. An in vivo analysis for tumor-induced angiogenesis shwed that a single i.v. administration of BCG-CWS/SQA inhibited the number of tumor-induced blood vessels and suppressed tumor growth. Furthermore, the multiple administration of BCG-CWS/SQA given at on week intervals led to a significant reduction in spontaneous lung metastasis of B16-BL6 melanoma cells in a spontaneous metastasis model. These results suggest that BCG-CWS emulsified with squalane is a potent inhibitory agent of lung metastasis, and that the anti metastatic effect of BCG-CWS is related to the suppression of tumor growth and the inhibition of tumor-induced angiogenesis.

  • This study was investigated to search for the effects of the structure of each component in four-component O/W microemulsion system on its formation region, droplet size and stablilty. The results was that the more number of OH site, the shorter carbon chain length of polyol, the larger formation region of microemulsion was showed. The small microemulsion droplet was obtained on condition that the polatry of oil was large and carbon chain length of hydrophobic group of surfactant was long. In using satrated hydrocarbon (such as liquid paraffin, squalane) as dispersed phase, the stability of microemulsion was better than aromatic oil phase.

  • Experiments of emulsion particles state of using polyglycerol ester emulsifier and the stability in accordance with the change of time were conducted using several oil. Experimental results confirmed that there is little difference in the stability and particle size depending on the type of oil. Most stable oil with polyglycerol ester is polar oil of silicon series and fatty acid ester oil, hydrocarbon oil of the nonpolar oil (Mineral oil, squalane, polydecene) was the most unstable state. And vegetable oils showed the stable form of particles with polyglycerol ester emulsifier.

  • This study was investigated optimal conditions which were necessary to prepare emulsion of four component systems by the Microfluidizer. Squalane, isopropyl myristate, PPG-15 stearyl benzoate ester were used as an oil and POE(20) sorbitan monolaurate, POE(20) sorbitan monopalmitate, POE(20) sorbitan monostearate, glycerine were selected as a cosurfactant and surfactant. The emulsion was affected by the operating pressure, the recycle frequency and oil, surfactant, cosurfactant phase. The operating pressure and recycle frequency increased, the droplet diameter of emulsion decreased and was constant later. In this study, the optimal pressure and recycle frequency were 1400 bar, 4 cycles. The droplet size was the smallest when the concentration of glycerine was 30 wt.%. As the oil phase is increased, the size is increased.

Physical Property and Extraction of Squalene and Alkoxyglycerol from Shark Liver Oil (상어간유에서 스쿠알렌과 알콕시글리세롤의 물성 및 추출)

  • A simple method has been successfully applied to extract squalene and its byproducts alkoxyglycerol from deep-sea shark liver oil. GC-MS was used to determine the extraction amount of the squalene and alkoxyglycerol, and the content of them in rough product and refined product were compared. The physical property of squalene was identified by measuring the pH value, peroxide value and iodine value. Under optimum extraction conditions, the amount of squalene and alkoxyglycerol increased 35.0% and 21.9%, respectively, while the amount of fatty acid decreased from 61% to 4%, especially, the amount of palmitic acid and oleic acid remarkably decreased. Large amount of peroxide and acid were removed from shark liver oil after refining process. Because squalene contains lots of double bond, so the value of iodine is much higher than squalane.

  • The o/w emulsions were prepared by lysolecithin as a biosurfactantsto to emulsify oils with squalane(SQ), liquid paraffin(LP), octylpalmitate(OP), octylstearate(OS), alkyl benzoate(AB), isostearyl benzoate(ISB). The droplets size and shape of o/w emulsions were investigated by laser light scattering, With dynamic light scattering hydrodynamic radius(Rh) of emulsion droplets was varied from 150m to 250m and critical concentration of oil In which the hydrodynamic radius(Rh) of emulsion droplets decreased and increased was found in the point of 0.5wt% oil concentration, and it was found increasing the polarity of oil deccreased the droplets, the droplets size of SQ(polar oil) were lower than SQ(nonpolar oil) With static light scattering radius of gyration(R$_{g}$) of emulusion droplets was to be calculated. From measurements of the ratio of R$_{g}$R$_{h}$ it was found that the shape of droplet of ISB, AB(polar oils) were sphere, for OP, OS(apolar oil) were oblate, for LP, SQ(nonpolar oil) were rod. The viscosity of emulsion in the form of rod was higher than that of emulsion in the form of sphere.e.e.

  • Recently, remarkable agreement has been reported between nonequilibrium molecular dynamics simulation and high-pressure Couette rheometry on squalane. We utilized the parameters obtained from this unique collaboration along with high-pressure viscometer measurements to calculate the elastohydrodynamic traction curve. A comparison with measured traction at 1.29 GPa shows excellent agreement, confirming the validity of the measurements and simulations. It should no longer be necessary to invoke a different rheological response to explain film thickness and traction.

  • Nanoemulsions have many specific characters compared with general emulsions in aspect of stability, rheological property, uniformity, high interfacial tension and so on. Therefore we study on nanoemulsions with changing various lecithins and oils using microfluidizer. As lecithins, we used saturated lecithin, unsaturated lecithin and hydrogenated phosphatidylcholine. Caprylic capric triglyceride, Squalane, Macadamia nut oil, Liquid paraffins, Dimethicone, and Cyclomethicone were used as oils. To identify nanoemulsions, we measured paticle size, zeta potential, turbidity and transmission electron microscope.

  • To investigate the effect on the antimicrobial activity against S.aureus ATCC 6538, E.coli KCTC 1039 and cell toxic level against transformed mouse fibroblast L929 in formula added with various concentrations of UVB blockers commonly used in cosmetic products, these experiments were carried out by preservative efficacy testing methods and in vitro cytotoxicity methods. The results obtained were as follow ; 1) Octyl Dimethyl PABA had a broad antibacterial spectrum against the Gram (+) and the Gram(-) bacteria at 5.84 % concentration, but not Octyl Methoxycinnamate. 2) Antibacterial activity was decreased in a combined UVB blocker system of squalane base. Especially, Octyl Dimethyl PABA was inactivated by Octyl Methoxycinnamate at 5.84% concentration to a large extents , but not 4-Methylbenzylidene Camphor. 3) Within in vitro cytotoxicity by use of mouse fibroblast L929 on UV-B blockers, NR assay was more excellent than MTT assay on quantitative
