Differences of Germination and Emergence rate among Isogenic Lines of Waxy and Hulless Gene Combinations in Barley (보리의 찰ㆍ메 및 피ㆍ과성 동질유전자 계통내 발아 및 출아율 차이)

  • On the purpose to increase the cultivation stability of the hulless waxy barley which is unstable in the northern region of Korea, the effects of genes of hulled /hulless and waxy /nonwaxy on seedling emergency was studied by using four isogenic lines of hulled -waxy, hulless -waxy, hulled -nonwaxy and hulless -nonwaxy barley. The germination rates of hulless lines lowered by four to six percent compared with those of hulled counterparts with one day's rainfall just before harvest, and were much deteriorated with raining prolonged. The gaps between emergency rates of hulless lines and their counterparts were greater than those of germination rates, and the differences increased as the seeding depths were deepened. The emergency rates of hulless lines were affected in the order of seeding depth, rotation speed of thresher and preharvest rainfall. Waxy endosperm gene did not show any significant effects on the damage of grain during threshing. It seems that the embryos of hulled lines were less damaged than those of hulless ones during threshing due to the protecting effects by husks. When grains were threshed with thresher of 600rpm or 900rpm and dehulled with dehuller of 750rpm, any significant depreciations such as broken seeds and decreased germination rate did not occured in hulled lines. But the grains of hulless lines received some damages with the rotation speeds, especially germination rates decreased seriously when the rotation speed of de huller was high. Prehavest sprout rates of the lines incorporated with the waxy genes were higher than those with nonwaxy genes whereas the genes of hulled and hulless did not give any significant effects.

Microbial and Pathogenic Contamination of Ready-to-eat Fresh Vegetables in Korea (한국에 유통중인 신선편이 채소류의 미생물 품질 및 병원성 세균의 오염도 조사)

  • The purpose of this study was to evaluate microbiological contamination of fresh vegetables in Korea. Twenty types of vegetables were tested for total aerobic bacteria, coliforms, Escherichia coli, yeast and mold, and pathogenic bacteria such as Staphylococcus aureus, Listeria monocytogenes, Bacillus cereus, Salmonella, E. coli O157:H7, Cronobacter sakazakii, Shigella, and Campylobacter. Levels of total aerobic bacteria and coliform on 20 vegetables were between 3.74 and 8.04 log CFU/g, and 0.16 and 5.02 log CFU/g, respectively. The highest contamination levels of total aerobic bacteria were observed on water dropwort, sprouts, mungbean sprout, and ballflower root. There was no significant difference (p>0.05) in microbial contamination levels of total aerobic count, coliform, E. coli, yeast and mold between organic and nonorganic vegetables. When isolation methods using selective agars were applied, L. monocytogenes, B. cereus, Salmonella and Campylobacter were isolated from some fresh vegetable samples. Results of API kit tests showed that L. monocytogenes was identified on Chinese cabbage, cucumber, soybean sprouts, and iceberg lettuce while Salmonella was identified on Korean leek. Furthermore, Campylobacter jejuni was also identified in more than 50 of the 100 samples. However, when positive samples from API kit were tested for real-time PCR or 16S rRNA sequencing method, only B. cereus from perilla leaf, carrot, water dropwort, and sprouts showed positive results. These results indicate that selective agar and API kit detection methods might result in false positive results for some pathogens. Therefore, studies need to improve isolation or confirmation methods for such pathogens.

The Effect of Cultivars, Cultivation Periods and Regions of Chicory on the Storability of Chicon During MA Storage (치커리의 재배일수, 재배지역 그리고 품종이 치콘의 MA 저장성에 미치는 영향)

  • Chicons are kinds of sprout vegetables which forced from the roots of chicory (Cichorium intybus L.). Experiments were performed to investigate the proper cultivation period and region of chicories which are 3 different cultivars for improving storability of chicon. The fresh weight of chicon packed with 20 ${\mu}m$ LDPE film decreased less than 0.5% during the storage at $10^{\circ}C$ in all treatments, and Chuncheon region treatment and 'Focus' cultivar treatment showed higher fresh weight loss than the others. And it decreased higher, as the cultivation periods was longer. The oxygen content in film was from 8% to 17% during storage at $10^{\circ}C$. There was not any significantly different oxygen content among cultivar treatments and cultivation region treatments, but 100 days' cultivation period treatment showed the highest content than the others. The carbon dioxide content in film showed around 3% during storage at $10^{\circ}C$. It also did not influenced by cultivars and cultivation regions, but 100 days' cultivation period treatment showed the lowest content than the others. The ethylene content in film was changed dramatically from 2 to 14 ${\mu}l{\cdot}l^{-1}$, and it showed around 8 ${\mu}l{\cdot}l^{-1}$ as approaching to 21 days after storage at $10^{\circ}C$. The visual quality of chicon deteriorated below marketability level from 12 days after storage at $10^{\circ}C$, and that of 'Metafora' cultivars decreased the highest. That of Pyeongchang region treatment was kept higher, and 'Vintor' cultivar treatment showed highest visual quality than the others which were grown in Pyeongchang region. The firmness of chicon increased, as the cultivation period was longer regardless of cutlivars and cultivation regions. The russet spotting that is ethylene injury symptom of chicon appeared at the latter term of storage and was higher in Chuncheon region treatment. Conculsionally, chicory that is for producing chicon should be grown more than 120 days in Kangwon region and grown in high-land region like Pyeongchang to improve the storability of chicon.

Development of Immunochromatography for Detection of Escherichia coli O157:H7 Contaminated in Meats and Sprouts (육류 및 새싹채소에 오염된 Escherichia coli O157:H7의 신속검출을 위한 면역크로마토그래피법의 개발)

  • Escherichia coli O157:H7 has been considered as a significant food-borne pathogen since its role in causing hemorrhagic colitis and hemolytic uremic syndrome in humans was recognized. In this study, we developed an immunochromatography (ICG) assay for the detection of E. coli O157:H7. E. coli O157:H7 monoclonal antibody (EC MAb) and colloidal gold were conjugated and its specificity was determined by the ICG treated with EC MAb and antimouse IgG at test and control lines, respectively. The detection limit of the ICG was $1{\times}10^5$ CFU/mL, and no crossreactivity was observed to other E. coli strains and major food-borne pathogens. To determine the minimum enrichment time for the ICG, meats and sprouts were inoculated with $1{\times}10$ CFU/100 ${\mu}L$ of E. coli O157:H7. After enrichment time of 10 and 2 h for meats and sprouts, respectively, up to $1{\times}10$ CFU/100 ${\mu}L$ of E. coli O157:H7 could be detected by ICG.

Artificial Cultivation of Tricholoma giganteum Collected in Korea (I) - Morphological Charateristics of Fruitbody and Environmental Condition in Habitat of T. giganteum - (한국산 왕송이버섯의 인공재배 (I) - 자실체 형태적 특징과 발생지 환경조사 -)

  • The result of study on the morphological feature of fruitbody, soil characteristics and enviromental condition of habitat of Tricholoma giganteum in Korea was as follows: The Pileus was $5.5{\sim}28.0\;cm$ in diameter and $1.5{\sim}3.7\;cm$ in thickness, indicating of yellowish white, beige, or ivory color. The lamellae were $18{\sim}20\;mm$ in diameter; the stipe was $9.0{\sim}35.7\;cm$ long and $1.0{\sim}3.0\;cm$ wide. The spores were $3.5{\sim}4.8{\times}5.7{\sim}7.4\;{\mu}m$ in size, ellipsoid, hyaline and the spore deposit white. The basidia were $6.1{\sim}7.0{\times}32.2{\sim}39.2\;{\mu}m$ in size. Cheiloystidia were $3.5{\sim}4.4{\times}30.5{\sim}33.1\;{\mu}m$ in size. Pileipellis were $3.3{\sim}4.4{\times}33.0{\sim}55.0\;{\mu}m$ wide and had clamp connection. Stipitipellis were $2.2{\sim}3.3{\times}88.0{\sim}93.1\;{\mu}m$ in size. Texture of soil in habitate of T. giganteum was silty loam, and organic materials content and available phosphate content in this soil were high as comparision to general soil in Korea. In the time of the mushrooms sprout, the room temperature indicated $25{\sim}27^{\circ}C$, their humidity showed $80{\sim}83%$, and the brightnesses were 328 Lux.

Evaluation of Dietary Manganese Intake in Korean Men and Women over 20 Years Old (20세 이상 일부 성인남녀의 망간 섭취상태 평가)

  • This study was peformed to estimate manganese intake and the major food source of manganese in Korean adults. The 354 subjects aged over 20 years were measured anthropometrics and dietary intake using 24-hour recall method. Daily intake and the major food sources of manganese were calculated using manganese database of food composition tables in Korea, USA and Japan. The average age, height, weight and BMI were 54.6years, 165.7cm, 67.2kg and $24.5kg/m^2$ for men and 53.8 years, 153.7cm, 59.1kg and $24.9kg/m^2$ for women, respectively. The daily energy and manganese intake of men were significantly higher than those of women (1740.9 kcal vs. 1432.6 kcal; p<0.001, 3.7mg vs. 3.2mg; p<0.01). However, daily manganese intake per 1000kcal between men and women was not significantly different (2.2mg/1000kcal vs. 2.3mg/1000kcal). Daily manganese intakes from each food group were 1.9mg from cereals, 0.5mg from vegetables, 0.4mg from pulses and 0.2mg from seasonings. The 20 major food sources of dietary manganese were rice, soybean, sorghum, Kimchi, tobu, wheat flour, red pepper powder, small red bean, glutinous millet, soybean paste, potato, Ramyeon, green pepper, noodle, buckwheat Naengmyeon, soybean sprout, laver, watermelon, perilla seeds powder and soy sauce. Manganese intake from these 20 foods was 74.0% of the total dietary manganese intake. In conclusion, daily manganese intake of the subject was 3.4mg (2.2mg/1000 kcal) and met adequate intake of manganese. The mai or food sources of manganese were cereals, pulses, and vegetables such as rice, soybean, sorghum, Kimchi and tobu.

In vitro Antioxidant and Anti-inflammatory Effects of Ethanol Extracts from Sprout of Evening Primrose (Oenothera laciniata) and Gooseberry (Actinidia arguta) (달맞이순과 다래순 에탄올 추출물의 in vitro 항산화효과 및 항염증효과)

  • To investigate the biological benefits of Korean traditional vegetables, anti-oxidative and anti-inflammatory effects of ethanol extracts from blanched and dried sprouts of evening primrose (Oenothera laciniata, OL) and gooseberry (Actinidia arguta, AA) were measured. Total polyphenol and flavonoid contents of OL were higher than those of AA; OL contained 60.4 mg tannic acid/g dry weight and 31.9 mg rutin/g dry weight, while AA contained 33.0 mg tannic acid/g dry weight and 20.3 mg rutin/g dry weight. The $IC_{50}$ value for DPPH radical scavenging activity was $58.2{\mu}g/mL$ for OL ethanol extract and $122.1{\mu}g/mL$ for AA ethanol extract. The reducing power upon $500{\mu}g/mL$ of ethanol extract treatment was as strong as $52.1{\mu}g$ ascorbate eq./mL for OL and $45.3{\mu}g$ ascorbate eq./mL for AA. Regarding anti-inflammatory effects, inhibition rate against 5-lipoxygenase (LOX) and cyclooxygenase (COX)-2 activities were 29.5% and 79.5% for OL, as well as 11.5% and 39.1% for AA, respectively at a concentration of $250{\mu}g/mL$. Lipopolysaccaride ($1{\mu}g/mL$)-treated RAW 264.7 macrophage cells subjected to OL ethanol extract at various concentrations ($0{\sim}25{\mu}g/mL$) showed significantly reduced synthesis of nitrite oxide (NO), prostaglandin (PG) E2, and IL-6 in a dose-dependent manner without cytotoxicity, although TNF-${\alpha}$ synthesis was not affected. In conclusion, both OL and AA sprouts showed strong antioxidative activity, whereas OL showed very strong anti-inflammatory activity via effective reduction of NO, PGE2, and IL-6 synthesis in LPS-activated macrophage cells.

Studies on the Prevention of Menace to Public Health by the Ornamental Tree -Especially in South Area of Korea- (조경수목(造景樹木)에 의(依)한 공해방지(公害防止)에 관(関)한 연구(硏究) -특(特)히 남부지역(南部地域)을 중심(中心)으로-)

  • These studies were carried out to know the measure of damage having an effect on the ornamental trees owing to sulfurous acid gas($So_2$) which injures much forest trees among air pollutions in south area of Korean and to know the difference of smoke resistance on the five representative species among them and therefore to establish the preventive measure of air pollutions in industrial complex of south area of Korea by the ornamental trees. The reaction formula utilized was $Na_2So_2+H_2So_4$ $Na_2So_4+H_O+So_4$. The results obtained were as follows. 1) The ornamental trees surveyed in the industrial complexes of Ul-San, Chang-Won and Po-hang belonged to 51 species, 161,699 trees in all. The most five species among 51 sorts of woody plants were Populus tomentiglandulosa, Buxus microphylla, Forsythia korenan, Juniperus chinensis and Pinus densiflora. 2) The relation between the concentration of sulfurous acid gas($So_2$) and the damage rate upon investigation in each species: In case of low concentration the destructive quantities of cells of woody plants, and the more increased the concentrations, the more increased the amount of damage. 3) The difference of smoke resistance among five species showed to be the strongest in Buxus micorphylla, the second rand Pupulus tomentiglandulosa, the third rand Forsythia koreana, the fourth rank Juniperus chinensis and the weakest in Pinus densiflora. 4) The ornamental trees of industrial complex expecially in south area of Korea in future should be planted gradually broad-leaved trees which have strong sprout, smoke resistance and thick leaved rather than comifers. 5) I think that the area of city forest in industrial complex requires about $50m^2$ per head of population and about 100m width of green belt between industrial area and residential district.

Changes in Antioxidant Contents and Activities of Adzuki Beans according to Germination Time (발아시간에 따른 팥의 항산화성분 및 항산화활성의 변화)

  • The purpose of this study was to evaluate the yield, antioxidant content, and antioxidant activity of adzuki beans according to germination time. Cultivated varieties were Vigna angularis var. Nipponensis cv. Chungju-pat (CJP), and Yeonduchae (YDC), and Vigna radiata (L.) R. Wilczek cv. Dahyeon (DH). The moisture, crude protein, calcium, and magnesium contents of YDC significantly changed with increasing germination time, whereas potassium, natrium, and aluminium contents did not significantly change. Sprout yield, total polyphenol, flavonoid, and tannin contents of ethanolic extracts from adzuki and mung beans significantly increased with increasing germination time. Total polyphenol contents of ungerminated CJP, YDC, and DH were 1.96, 2.68, and 2.02 mg/g, and those of CJP and YDC germinated for 144 h were 3.33 and 3.47 mg/g, respectively. Total flavonoid content of adzuki beans substantially decreased with increasing germination time. Total tannin content substantially increased with increasing germination time, and YDC showed higher contents (0.85 mg/g) sample germinated for 120 h. DPPH radical scavenging activities of CJP and YDC substantially decreased with increasing germination time, whereas that of DH increased. ABTS radical scavenging activities of ungerminated CJP, YDC, and DH were 4.57, 6.51, and 2.82 mg/g, respectively, and increased after germination for 72~120 h.

Growth Response and Nutrient Content of Cowpea Sprouts Based on Growth Temperature and Genetic Resources (재배온도 및 유전자원에 따른 동부나물 생장반응 및 영양성분 변화)

  • The purpose of this study was to establish the optimal growth temperature and to select genetic resources for production of cowpea sprouts. Seowon was treated between $15^{\circ}C$ and $30^{\circ}C$ at intervals of $3^{\circ}C$ to investigate growth temperature. Twelve resources, including Seowon, IT154149, IT154153, Tvu7426, and Tvu7778, were used for cultivating sprouts at a temperature of $27^{\circ}C$. The yield ratio of cowpea sprouts was highest at $27^{\circ}C$ (657%), and was reduced when growth temperature was decreased. The hard seed rate was lower when the growth temperature was increased. Vitamin C content was highest at $24^{\circ}C$ (2.85 mg/g), ranged between 2.15 and 2.29 mg/g at other growth temperatures, and increased with the length of the growth period. The inorganic component content of cowpea sprouts did not vary based on growth temperature, while the amino acid content increased with increasing growth temperature between $15^{\circ}C$ and $24^{\circ}C$, and then subsequently decreased as growth temperature rose from $24^{\circ}C$ to $30^{\circ}C$. IT154153 had the highest yield ratio of cowpea sprouts per genetic resource (647%), followed by Seowon (615%), and Tvu7426 (608%). Genetic resources with a higher yield ratio had smaller seeds, a thinner seed coat, and superior germinability. The inorganic components found at highest concentrations in the cowpea sprouts were potassium, magnesium, calcium, sodium, iron, molybdenum, and zinc (in that order). In comparison to raw seeds, the protein, calcium, zinc, molybdenum, and iron content in the cowpea sprouts was higher, while the content of aluminum and boron was lower.
