A Study on Improving Protein Quality of Wheat Bran by Fermentation with Aspergillus Oryzae (Aspergillus Oryzae를 이용(利用)한 밀기울단백질(蛋白質)의 질적(質的) 향상(向上)에 관(關)한 연구(硏究))

  • The purpose of this study was to investigate changes in nutritive quality of fermented wheat bran prepared by culturing with a microorganism, Aspergillus oryzae, in an attempt to improve the quality of protein in feedstuff. After incubation of wheat bran with Aspergillus oryzae, the contents of chemical composition, including amino acids, riboflavin and amino-nitrogen were increased, but the level of nitrogen free extract was reduced. The effects of supplementation of fermented wheat bran on the rat diets were evaluated by measuring growth rate, feed efficiency and biological values, such as NPU, PER and NPR. Sixty four male Sprague Dawley rats of 5-6 weeks of age were adopted for the feeding trial for 10 days period, and levels of dietary protein were set at 10%. The %contribution of protein from casein, wheat bran and fermented wheat bran for the other dietary treatments were, in the order, 50-50-0%, 50-30-20%, 50-20-30% and 50-0-50%, respectively. In the result of this study, no significant difference were observed in the amount of feed intake body weight gain and feed efficiency. On the whole, the values of NPU, PER and NPR of all the fermented wheat bran groups appeared to be higher than those of the group whose half of the protein was from wheat bran.

Role of a Burr Hole and Calvarial Bone Marrow-Derived Stem Cells in the Ischemic Rat Brain : A Possible Mechanism for the Efficacy of Multiple Burr Hole Surgery in Moyamoya Disease

  • Objective : This study investigates the role of a burr hole and calvarial bone marrow-derived stem cells (BMSCs) in a transient ischemic brain injury model in the rat and postulates a possible mechanism for the efficacy of multiple cranial burr hole (MCBH) surgery in moyamoya disease (MMD). Methods : Twenty Sprague-Dawley rats (250 g, male) were divided into four groups : normal control group (n=5), burr hole group (n=5), ischemia group (n=5), and ischemia+burr hole group (n=5). Focal ischemia was induced by the transient middle cerebral artery occlusion (MCAO). At one week after the ischemic injury, a 2 mm-sized cranial burr hole with small cortical incision was made on the ipsilateral (left) parietal area. Bromodeoxyuridine (BrdU, 50 mg/kg) was injected intraperitoneally, 2 times a day for 6 days after the burr hole trephination. At one week after the burr hole trephination, brains were harvested. Immunohistochemical stainings for BrdU, CD34, VEGF, and Doublecortin and Nestin were done. Results : In the ischemia+burr hole group, BrdU (+), CD34 (+), and Doublecortin (+) cells were found in the cortical incision site below the burr hole. A number of cells with Nestin (+) or VEGF (+) were found in the cerebral parenchyma around the cortical incision site. In the other groups, BrdU (+), CD34 (+), Doublecortin (+), and Nestin (+) cells were not detected in the corresponding area. These findings suggest that BrdU (+) and CD34 (+) cells are bone marrow-derived stem cells, which may be derived from the calvarial bone marrow through the burr hole. The existence of CD34 (+) and VEGF (+) cells indicates increased angiogenesis, while the existence of Doublecortin (+), Nestin (+) cells indicates increased neurogenesis. Conclusion : Based on these findings, the BMSCs through burr holes seem to play an important role for the therapeutic effect of the MCBH surgery in MMD.

  • Purpose: Phosphodiesterase (PDE) inhibitor increases the cellular content of cAMP, and cAMP suppresses connective tissue growth factor (CTGF) expression induced by TGF-${\beta}1$. Therefore, we investigated whether PDE inhibitor suppresses renal fibrosis without suppression of TGF-${\beta}1$. Materials and Methods : Renal interstitial fibrosis was produced by ligation of left ureter in Sprague-Dawley rats. Cilostazol, a selective PDE3 inhibitor, and dipyridamole, a hybrid PDE5, PDE6, and PDE8 inhibitor, were provided in drinking water for 7 days. In addition to the Masson-trichrome score of renal tissue, the concentration of fibronectin and TGF-${\beta}1$ in renal tissue- conditioned media was measured by ELISA. Results : Masson- trichrome score and fibronectin concentration were significantly lower in cilostazol-treated group compared to the control group (P<0.05). Though dipyridamole treatment seemed to suppress the Masson- trichrome score and fibronectin concentration too, the decrements were not statistically significant. There was no difference in TGF-${\beta}1$ concentration among the groups. Conclusion: A selective PDE3 inhibitor cilostazol suppresses renal fibrosis without alteration of TGF-${\beta}1$ expression. Effect of SipJeonDaeBo-Decoction on Blood Metal Level in Rats (십전대보탕(十全大補湯)을 투여한 흰쥐의 혈액중 금속농도변화에 관한 연구)

  • This study was to investigate the metal accumulation from SipJeonDaeBo-Decoction to rat blood of Sprague Dawley. 1. There were no significance in body weight, water dose feed ingestion quantity, liver, kidney, brain, bone weights between the control and the experimental groups. Under the experiment with drinking waters was no metal ${\sim}\;0.65mg/L$ detected. Metal level within feed found 0.0001-376.983mg/kg. 2. In the pack of SipJeonDaeBo-decoction, there detected no metal ${\sim}2.086mg/L$ 3. After P.O(per os) SipJeonDaeBo-decoction, As is detected $2.390{\pm}0.812mg/kg$ in blood; Cd $0.001{\pm}0.001mg/kg$, Co $0.003{\pm}0.001mg/kg$, Cr $0.432{\pm}0.234mg/kg$, Cu $1.013{\pm}0.373mg/kg$, Fe $426.293{\pm}114.842mg/kg$, no Hg, Mn $0.109{\pm}0.082mg/kg$, Ni $0.122{\pm}0.068mg/kg$, Zn $3.584{\pm}1.270mg/kg$. 4. The concentration of Hazardous heavy metal (As, Cd, Co, Cr, Hg, Ni, Pb) within blood control group is searched $0.488{\pm}0.138\;mg/l$; experiment I group $0.432{\pm}0.080mg/l$, experiment II group $0.588{\pm}0.213mg/l$. In the concentration of non hazardous heavy metal(Cu, Fe, Mn, Zn) control group $101.409{\pm}6.832mg/l$; experiment I group $96.062{\pm}5.732\;mg/l$, experiment II group $125.139{\pm}044.820mg/l$. 5. Correlation among every metal in blood Zn and Cr was 0.87956 ; Cd and As -0.02316, Pb and As -0.08738, Ni and As 0.07824, Mn and As 0.07824, Mn and Cd 0.04999. Briefly under the injection of SipJeonDaeBo-decoction, this study was defined within safety in blood level by P.O. during 10 days.

The Effect of Methylene Blue on Inducible Nitric-oxide Synthase in a Rat Model of Acute Lung Injury Induced by Paraquat (파라쿼트를 투여한 백서의 급성 폐 손상 모델에서 메틸렌블루 투여가 Inducible Nitric Oxide Synthase 유전자 발현에 미치는 효과에 관한 연구)

  • Purpose: This study was designed to determine if methylene blue inhibited the lipid peroxidation, the production of NO, and the gene expression of iNOS in acute lung injury induced by paraquat and if the inhibitory effect was dose dependent. Methods: Female Sprague-Dawley rats were divided into four groups: the control group, the group treated with paraquat only, the group treated with paraquat and a low dose of methylene blue (2 mg/kg), and the group treated with paraquat and a high dose of methylene blue (20 mg/kg). Methylene blue was administered via the jugular vein 1 h after paraquat administration, and animals were sacrificed 6 and 24 h after paraquat administration. Malondialdehyde (MDA) as lipid peroxidation, reduced glutathione (GSH) as an antioxidant defense, the plasma NO concentration, and the expression of iNOS mRNA in the lung tissue were measured Results: Lung MDA contents decreased, with no significant difference between the methylene-blue groups and the paraquat-only group. Lung GSH contents were significantly elevated at 24 h in the methylene-blue groups compared with the paraquat-only group. Plasma NO concentrations were significantly reduced at 6 and 24 h in the methylene-blue groups compared with the paraquat-only group. There was also a significant decrease in the plasma NO concentration at 6 h in the high-dose methylene-blue group compared with the low-dose methylene-blue group. The expression of iNOS mRNA in the lung tissue was slightly decreased in the methylene-blue groups. It was also markedly increased at 24 h in the paraquat-only group compared with the methylene-blue groups. The gene expression was relatively decreased in the high-dose methylene-blue group compared with the low-dose methylene-blue group. Conclusion: This study suggests that methylene blue has an inhibitory effect on the plasma NO concentration and the expression of iNOS mRNA in lung injury induced by paraquat. No inhibitory effect of methylene blue on lipid peroxidation or dose-dependent inhibitory effects were clearly shown.

Hypoglycemic Effects of Crude Extracts of Moutan Radicis Cortex (목단피 추출물의 혈당 강하 효과)

  • Hypoglycemie effect of Moutan Radicis Cortex (MRC) extract contained in Yukmijihuang-hwan was determined by investigating insulin-sensitizing and ${\alpha}-glucoamylase-suppressing$ actions. MRC was extracted with 70% ethanol, fractionated by XAD-4 column chromatography with mixture solvent of methanol and water, and utilized for hypoglycemic effect assay. Significant insulin sensitizing activities of MRC extracts were observed in 3T3-L1 adipocytes, giving MRC extracts with 1 ng/mL insulin reach glucose uptake level increased by 50 ng/mL of insulin alone. MRC methanol extracts of 20, 40, 60, and 80% suppressed ${\alpha}-glucoamylase$ activity in vitro. Peak serum glucose levels and area under curve were lower in Sprague Dawley male rats treated with MRC ethanol extract than those treated with cellulose in oral glucose tolerance test using 2 g dextrin/kg body weight. These data suggest MRC extracts contain effective insulin -sensitizing and ${\alpha}-glucoamylase-suppressing$ compounds for hypoglycemic activity.

Development of a Discogenic Pain Animal Model: Preliminary Study (추간판성통증 동물모델의 개발: 초기 연구)

  • Background: Discogenic pain can develop into chronic low back pain that is very difficult to treat effectively, because the pathogenesis of the disease still remains controversial. To clarify the pathogenesis, numerous animal models of intervertebral disc degeneration have been proposed in the literature, each with attendant advantages and disadvantages. The aim of this study was to determine the most efficacious method and dose of complete Freund's adjuvant (CFA) injection into intervertebral disc to develop a discogenic pain in a rat. Methods: CFA was injected into the L5-L6 or L4-L5 disc of male Sprague-Dawley rats in various conditions including a dose of CFA (10, 20, or 50 uL), drilling, injection site sealing using cyanoacrylate, and injection velocity. Sham animals were subjected to the same procedure, except for the CFA injection. Mechanical and heat allodynia were serially measured at both hindpaws until 8 weeks post-operatively. Serial MRI analyses were performed to observe degenerative changes of the discs. In addition, CGRP & Substance P-immunoreactivities (ir) in the superficial dorsal horn were evaluated at 4 weeks using immunohistochemistry. Results: Each condition provoked various problems such as development of hindpaw paralysis, CFA leakage, and no pain development. Mid-sagittal T2 MRI revealed no significant degenerative changes in the CFA injected disc. The CGRP-ir of the bilateral superficial dorsal horns at the level of L5-L6 was significantly increased in the CFA group. Conclusions: A total of 10 uL CFA injection into L5-L6 disc for a period of 10 minutes using a 26-gauge needle without drilling was the most efficacious way to develop discogenic pain animal model.

  • Cellulose has been widely applied into various medical fields including scaffolding, tissue engineering and tissue formation. In this study, we manufactured cellulose medical fiber from Styela clava tunics(SCT-CS) and analyzed the tensile strength, elongation at break, fluid uptake and surface morphology. And then, the biocompatibility and toxicity of SCT-CS were measured in Sprague-Dawley(SD) rats after the implantation for 30, 60 and 90 days. The level of tensile strength and fluid uptake were lower in SCT-CS than chromic catgut(CCG), while elongation at break level were maintained the higher in SCT-CS. Also, the roughness with pronounced surface patterns as a result of in vivo degradation was significantly greater in CCG than this of SCT-CS although these levels gradually appeared with time in both groups. After implantation for 90 days, SCT-CS and CCG was successfully implanted around muscle of thigh without any significant immune response. Furthermore, no significant alterations were measured in serum parameters and the specific pathological features induced by most toxic compounds for liver and kidney toxicity. Therefore, these results suggest that SCT-CS showing good biocompatibility and non-toxicity can be successfully prepared from cellulose powder of SCT as well as has the potential for use as a powerful biomaterial for medical sutures.

Effect of Hippocampectomy on Gastric Ulceration by Predicted Stress (예보된 스트레스에 의한 위궤양 발생에 미치는 뇌 해마 제거의 영향)

  • This study was under taken to investigate the influences of predictable or unpredictable stress upon gastric ulceration, and the hippocampectomy upon the ulceration order the stressful conditions. Sixty male albino rats(Sprague-Dawley strain) were divided equally into 3 groups: One was the hippocampal group(N=20) which received hippocampal ablation by suction, another was the cortical control group(N =20) which received partial cortical ablation over the hippocampus, and a third was the normal control group(N=20). Each group was further divided into two subgroups: One was the predicted subgroup(N=10) in which animals could predict the imminent stressful stimuli by hearing a sound(1,000 Hz, 2 sec in duration) 3 sec before the onset of the stress, and the other was the unpredicted subgroup(N=10). After starvation for 24 hours, but water ad libitum, each rat received the electric stimulation(3 mA, 60 Hz, 2 sec in duration, and once per minute in average) for 6 hours via a pair of electrodes attached on the tale. The electric stimulation served as the stress causing the gastric ulcer. Five hours after completion of stimulation, the stomach filled with the physiological saline was removed under deep anesthesia and spread out on a small glass plate. The numbers of the ulcer in each stomach were counted and the shape was examined under the dissecting microscope. Results obtained were as follows: 1. The mean numbers of the ulcer of the predicted subgroups were significantly larger than those of the unpredicted subgroups in the normal control and the cortical control groups, but there was no difference between the values of the two subgroups in the hippocampal group. 2. The mean numbers of the ulcer of the predicted subgroups in the normal control and the cortical control groups were larger(but not significant) than that in the hippocampal group. It is inferred from the above results that the prediction of the stress strengthens the effect of the stress on the gastric ulceration, and the hippocampus facilitates the effect of the prediction of the stress.

The Effect of ATP-sensitive Potassium Channel on R-PIA Induced Mechanical Antiallodynia in a Peripheral Neuropathic Rat (신경병증 통증 모델의 백서에서 R-PIA의 기계적 항이질통 효과와 ATP-감수성 칼륨 통로와의 연관성에 대한 연구)

  • Background: Nerve ligation injury may produce mechanical allodynia, but this can be reversed after an intrathecal administration of adenosine analogues. In many animal and human studies, ATP-sensitive potassium channel blockers have been known to reverse the antinociceptive effect of various drugs. This study was performed to evaluate the mechanical antiallodynic effects of spinal R-PIA (Adenosine A1 receptor agonist) and the reversal of these effects due to pretreatment with glibenclamide (ATP-sensitive potassium channel blocker). Thus, the relationship between the antiallodynic effects of R-PIA and ATP-sensitive potassium channel were investigated in a neuropathic model. Methods: Male Sprague Dawley rats were prepared by tightly ligating the left lumbar 5th and 6th spinal nerves and implantation of a chronic lumbar intrathecal catheter for drug administration. The mechanical allodynia was measured by applying von Frey filaments ipsilateral to the lesioned hind paw. And the thresholds for paw withdrawal assessed. In study 1, either R-PIA (0.5, 1 and $2{\mu}g$) or saline were administered intrathecally for the examination of the antiallodynic effect of R-PIA. In study 2, glibenclamide (2, 5, 10 and 20 nM) was administered intrathecally 5 min prior to an R-PIA injection for investigation of the reversal of the antiallodynic effects of R-PIA. Results: The antiallodynic effect of R-PIA was produced in a dose dependent manner. In study 1, the paw withdrawal threshold was significantly increased with $2{\mu}g$ R-PIA (P < 0.05). In study 2, the paw withdrawal threshold with $2{\mu}g$ R-PIA was significantly decreased almost dose dependently by intrathecal pretreatment of 5, 10 and 20 nM glibenclamide (P < 0.05). Conclusions: These results demonstrated that an intrathecal injection of ATP-sensitive potassium channel blockers prior to an intrathecal injection of adenosine A1 receptors agonist had an antagonistic effect on R-PIA induced antiallodynia. The results suggest that the mechanism of mechanical antiallodynia, as induced by an intrathecal injection of R-PIA, may involve the ATP-sensitive potassium channel at both the spinal and supraspinal level in a rat nerve ligation injury model.
