In vitro Anti-fungal Activity of Various Hydroxylated Fatty Acids Bioconverted by Pseudomonas aeruginosa PR3

  • The in vitro anti-fungal activity of hydroxylated fatty acids obtained from microbial conversion by Psuedomonas aeruginosa PR3 using ricinoleic acid(RA), eicosadienoic acid(EDA) and conjugated linoleic acid(CLA) as substrates, was investigated. Bioconverted hydroxylated fatty acids showed different anti-fungal activities potentials against the range of phytopathogenic fungi such as Botrytis cinerea, Rhizoctonia solani, Fusarium oxysporum, Sclerotonia sclerotiorum, Colletotricum capsici, Fusarium solani and Phytophthora capsici. RA and EDA showed up to 50% fungal mycelial inhibition at the concentration of $5{\mu}l\;ml^{-1}$. RA, EDA and CLA also exhibited anti-fungal activities with minimum inhibitory concentration(MIC), ranging from 500 to $1000{\mu}g\;ml^{-1}$. Screening was also carried out using varied concentrations of bioconverted RA and EDA for determining the anti-fungal effect on the spore germination of different fungi. Bioconverted RA and EDA showed a considerable degree of spore germination inhibition.

  • Medicinal plants play important roles in controlling plant diseases as one of the safest and ecofriendly methods. These plants have been used in the form of crude extracts as well as active principles in vitro and under field conditions to control plant diseases. Among the active principles, alkaloids have shown significant antifungal activity. We have investigated the effect of two alkaloids viz., (-)-corydahnine and (-)-isocorypahnine isolated from Corydalis chaerophylla, against spore germination of some plant pathogenic and saprophytic fungal spores. Significant inhibition of spore germination at 100 ${\mu}g/ml$ was seen against Curvularia penniseti, Curvularia sp. and Colletotrichum gloeosporioides by (-)-corydahnine but (-)-isocorypalmine was also effective against fungi included in the experiment.

Chemical Responses and Fitness-Related Characteristics of Dichlofluanid-Resistant and -Sensitive Botrytis cinerea Isolates (Dichlofluanid 저항성 및 감수성 잿빛곰팡이병균(Botryits cinerea)의 약제 반응과 생태 적응력과 관련된 특성)

  • Nine hundred and ninety-two isolates of Botrytis cinerea were obtained from infected strawberries, tomatoes and cucumbers in Taejon, Gongju, Puyo, Nonsan and Kimhae in Korea. Six hundred forty-two (64.7%) isolates were benomyl resistant (BR), 245 (24.7%) were procymidone resistant (PR), and 105 (10.6%) were dichlofluanid resistant (DR). In the minimum inhibitory concentration (MIC) test, DR isolates showed mycelial growth on the PDA incorporated with 100 or 500 $\mu\textrm{g}$/ml of dichlofluanid, while dichlofluanid sensitive (DS) isolates did not grow on the PDA incorporated even with 10 $\mu\textrm{g}$/ml of dichlofluanid. Chemical concentrations for inhibition of spore germination were much lower than those for inhibition of mycelial growth. IC50 values, the effective concentrations for 50% inhibition of spore germination, for DR were 0.11~0.29 $\mu\textrm{g}$/ml, whereas they were 0.04~0.09 $\mu\textrm{g}$/ml for DS isolates. In comparison of fitness-related characteristics such as virulence, sclerotial formation, and sporulation, DR isolates were inferior to DS isolates. However, mycelial growth was little different between DR isolates and DS isolates.

In vitro Antifungal Activities of Fungicides against Japanese Plum Fruit Anthracnose Fungi (자두 탄저병균에 대한 살균제의 활성)

  • BACKGROUND: In order to select a fungicide that can effectively control anthracnose disease in Japanese plum fruit, mycelial growth inhibition effect and spore germination inhibition effect of six fungicides were tested in vitro against six isolates of Colletotrichum acutatum and five isolates of C. gloeosporioides that were isolated from diseased Japanese plum fruit. METHODS AND RESULTS: Inhibitory effects of fungicides on mycelial growth were investigated after inoculating each isolate on potato dextrose agar amended with four discriminatory concentrations of each fungicide for 7 days at $25^{\circ}C$. For spore germination inhibitory effect, each isolate of the Colletotrichum spp. was cultured in potato dextrose agar for 7-14 days at $25^{\circ}C$. After adjusting the concentration of spores of each isolate to $1{\times}10^6mL^{-1}$ by diluting with 0.025% PDB, the spore suspension was mixed with each fungicide (1:4, v/v), and $60{\mu}L$ aliquots were dispensed to sterile hole slide glass. Hole slide glasses were placed in a humidified box and incubated for 15 hours at $25^{\circ}C$. Then, spore germination was observed under an optical microscope. At recommended concentration of fungicide prochloraz manganese showed the highest mycelial growth inhibitory effect and dithianon showed the lowest mycelial growth inhibition. The $EC_{50}$ values for the inhibition of spore germination by dithianon and pyraclostrobin were $0.069-0.126{\mu}g/mL$ and $0.37-1.59{\mu}g/mL$, respectively. Although benomyl, prochloraz manganese, azoxystrobin, and tebuconazole did not inhibit the spore germination, they appeared to restrain mycelial growth by abnormal growth of germ tube and mycelium after germination. CONCLUSION: Dithianon seemed to have preventive effect. Prochloraz manganese, azoxystrobin, and tebuconazole were likely to have control effect. Pyraclostrobin is considered to have both preventive and control effect against anthracnose disease of Japanese plum fruit.

The Antidermatophytic Potential of the Marine Isolate of Aspergillus sp. Collected from South Coast of Korea

  • This study was carried out to assess the antidermatophytic potential of the ethyl acetate(EtOAc) extract of the marine isolate of Aspergillus sp.. The fungus was isolated by serial dilution, and was identified Aspergillus sp.. The EtOAc extract of the fungus was examined to evaluate the antidermatophytic efficacy against the fungal pathogens infecting human skin using the disc diffusion and MIC(minimum inhibitory concentration) determination methods. The EtOAc extract($5{\mu}l\;disc^{-1}$) was considered to have the antidermatophytic activity based on the inhibition percentage of the mycelial growth of the fungi tested such as Trichophyton mentagrophytes KCTC 6085, Microsporum canis KCTC 6591, Microsporum canis KCTC 6348, Trichophyton rubrum KCTC 6352, Microsporum canis KCTC 6349 and Trichophyton mentagrophytes KCTC 6316. The percentage of the inhibition ranged from 54% to 81, and the MIC obtained was 62.5, 62.5, 250, 125, 125, and $125{\mu}g\;ml^{-1}$, respectively. The extract had a strong detrimental effect on the spore germination of the tested skin infectious pathogens. These findings strongly support the role of the ethyl acetate extract as a potential antidermatophytic agent.

  • In order to establish an appropriate spray schedule to reduce the infection, 11 kinds of chemicals were tested for their properties on the inhibition of spore germination at the surface of apple fruits and the duration of the inhibitory effect after spray of each chemical was examined from late June to early September with basically 10 day intervals. Actual control efficacy of each chemical by the 8 successive spray and the patterns of waterborne spore dispersals during that periods were also examined. Combining those results with the meteorological observation data, actual control efficacy of each chemical in the given periods could be estimated. It was revealed that folpet, Brodeaux mixture, mancozeb, oxine copper and imminoctadine-triacetate could be used at any time during the possible infection periods. Captan and dithianon could also be used except the rainy season due to the short duration of inhibitory efficacy against spore germination under heavy rain. However, the usefulness of propineb, benomyl and chlorothalonil against the apple white rot could not be demonstrated in this experiment. Thiram, even though has not been used for apple white rot, can also be used before or after the rainy season to control not only white rot but also alternaria blotch.

In Vitro Antifungal Activities of 13 Fungicides against Pepper Anthracnose Fungi (고추 탄저병균에 대한 13종 살균제의 살균 활성)

  • For the effective control of pepper anthracnose, in vitro antifungal activities of 13 commercial fungicides were tested on spore germination, mycelial growth, and sporulation of Colletotrichum gloeosporioides, anthracnose fungus. Among them, captan, chlorothalonil, dithianon, fluazinam and folpet completely inhibited the spore germination of C. gloeosporioides at the concentration of 0.8 ${\mu}g/ml$. They were followed by mancozeb and propineb, showing more than 80% inhibition of spore germination at 20 ${\mu}g/ml$. The mycelial growth of C. gloeosporioides was strongly inhibited by fluazinam and nuarimol. Except for nuarimol, most of the fungicides represented more antifungal activity on the spore germination than the mycelial growth of C. gloeosporioides. Azoxystrobin and metominostrobin, Strobilurin fungicides, were only moderately active against the spore germination and the mycelial growth of C. gloeosporioides, but they were effective antisporulant against C. gloeosporioides. From these results, control of pepper anthracnose will have achieved by preventive spray of the commercial fungicides.

Postharvest Disease Control of Colletotrichum gloeosporioides and Penicillium expansum on Stored Apples by Gamma Irradiation Combined with Fumigation

  • To study the control of postharvest decay caused by Colletotrichum gloeosporioides and Penicillium expansum, gamma irradiation alone or in combination with fumigation was evaluated to extend the shelf life of apples in South Korea. An irradiation dose of 2.0 kGy resulted in the maximum inhibition of C. gloeosporioides and P. expansum spore germination. The gamma irradiation dose required to reduce the spore germination by 90% was 0.22 and 0.35 kGy for C. gloeosporioides and P. expansum, respectively. Microscopic observations revealed that when the fungal spores were treated with gamma irradiation (4.0 kGy), conidial germination was stopped completely resulting in no germ tube formation in C. gloeosporioides. Treatment with the eco-friendly fumigant ethanedinitrile had a greater antifungal activity against C. gloeosporioides and P. expansum in comparison with the non-treated control under in vitro conditions. The in vitro antifungal effects of the gamma irradiation and fumigation treatments allowed us to further study the effects of the combined treatments to control postharvest decay on stored apples. Interestingly, when apples were treated with gamma irradiation in combined with fumigation, disease inhibition increased more at lower (< 0.4 kGy) than at higher doses of irradiation, suggesting that combined treatments reduced the necessary irradiation dose in phytosanitary irradiation processing under storage conditions.

Growth Inhibition Effect of Environment-friendly Farm Materials in Colletotrichum acutatum In Vitro (친환경 유기 농자재의 고추 탄저병(Colletotrichum acutatum) 병원균의 생장 억제 효과)

  • Inhibition effects on spore germination and mycelia growth for pepper anthracnose fungi (Collectricum acutatum) were investigated in vitro using eco-friendly agricultural materials as well as ecofriendly pesticides. The inhibition effect on mycelia growth of anthracnose fungi is the highest when the anthracnose mycelia were treated with a pesticide (commercial name: Koreayeok) that contains a mixture of Bacillus subtilis and Panibacillus polymyxa, resulting in 100% inhibition of the mycelia growth. Meanwhile, the range of 20~40% inhibition effects on the growth of anthracnose mycelia was observed with other commercial agricultural materials. The significant inhibition effects on spore formation of anthracnose fungus were shown in vitro with two water dispersible pesticides containing sulfur [BTB (100%) and SulfurStar (95.1%)], Koreayeok (95.0%), Borstar (99.0%) containing Bordeaux mixture, and Jihabudea-KM containing Psedomonas spp. (96.1%), respectively. Taken from these in vitro results of inhibiting of the spore germination and mycelia growth together, Koreayeok is the most effective on control of pepper anthracnose disease in vitro. In addition, two water dispersible pesticides containing sulfur (BTB and SulfurStar) and Borstar (99.0%) containing Bordeaux mixture are also significantly applicable to prevent pepper plants from anthracnose disease in vitro. It remains to be determined whether the selected eco-friendly agricultural materials in effective control of anthracnose in vitro can be used to control pepper anthracnose in field.

Induction of Phytoalexins by Uptake of Naphthoquinones in Cell Cultures of Petunia (Naphthoquinone류 화합물 흡수에 의한 페튜니아 배양세포내의 Phytoalexin 유도)

  • To induce the phytoalexins in plant cell culture systems, we surveyed the antimicrobial activity following the feeding of five naphthoquinones in cell cultures of petunia. Among naphthoquinones treated, 2,5,7-trihydroxy-3-(5'-hydroxyhexyl)-1,4-naphthoquinone (3-OH NQ ) was most efficiently absorbed into the cells within 48 hr. The crude extracts of cells treated with 3-OH NQ showed a strong inhibition activity on spore germination of Aspergillus candidus $(MIC:\;32\;{\mu}g/ml)$, whereas the untreated cells showed no activity. The two active compounds, 4,2',4',${\beta}$-tetrahydroxychalcone and 4',7-dihydroxyflavone, were isolated from petunia cells treated with 3-OH NQ. The major phytoalexin, 4,2',4',${\beta}$-tetrahydroxychalcone, inhibited strongly the spore germination of A. candidus $(MIC:\;16\;{\mu}g/ml)$.
