  • To study the function and structure of Golgi apparatus in the spermiogenesis of long-fingered bat (Miniopterus schreibersi fuliginosus), the testis obtained from adult bat was treated with the prolonged osmification or fixed with ferrocyanide reduced osmium. golgi apparatus was oval shape in early Golgi phase, and was composed of cortex and medullar enclosing acrosome in mid Golgi phase. The vesicles of crescent shape Golgi apparatus were closed or fused with small or large vesicles at the periphery of acrosome. Golgi apparatus moved behing the acrosome face in cap phase, but the Golgi apparatus was still active. According to this, Golgi apparatus appears to be involved in the formation of acrosome and sperm tail. Transfer of materials from Golgi to acrosme seems to be carried out not only by fusion of large vesicles with acrosomal vesicles but also by detachment of coated vesicle from various cisternae of Golgi fusing with acrosomal vesicle.

  • In order to investigate the spermiogenesis of the Korean Native Jin-do dog, the tissue sampled from the testis and the epididymis of matured dog were conventionally prepared as thin sections. And then these sections were observed under a transmission electron microscope. Based on the development of various permanent organelle and certain transient structures, the spermiogenesis of the dog could be divided into four major phases: Golgi, cap, acrosome and maturation; and these phases had been futher subdivided into seven stages. In the maturatin phase, the karyoplasm has been condensed completely and the nucleus eleongated as the movement of the unclear ring. The equatorial segment has notable waist-form at the late acrosomal stage. In the cross section of the tail, it shows a typicl 9+9+2 pattern of the axial fiber bundle condisting of the dense fibers, the inner fibers, and a central pair. The dense fiber number 1, 5, 6, and 9 were larger in diameter than the rest of the dense fibers.

  • Testes from the drake and the gander have been examined by the electron microscopy in thin sections in order to examine the spermiogenesis and the structure of spermatozoa. The spermiogenesis can be divided into three stages: early spermatid, nuclear elongation, and matured spermatid. In the early spermatid of the drake, there are thread-like material in the nucleus, a prominent nuclear envelope around the nucleus, and big lumens in the cytoplasm. The shape of the gander's mitochondria in the early spermatid is slender compared to that of the drake, and the inner membrane of the mitochondria is thicker than the outer membrane. The distal centriole of the drake and the gander in the early spermatid is a long hollow cylinder form. In the nuclear elongation stage, elongated nucleus forms two or three cross sections in one spermatid cell and it is surrounded by the amorphous sheath. The nucleus of the matured spermatid is compact and its apical end is covered with acrosome cap and acrosome spine. The axoneme is surrounded by the amorphous material.

  • In order to investigate the spermiogenesis of Korean cattle spermatozoa, the tissue obtained from the testis and the epididymis of six matured bull were conventionary prepared as thin sections and these sections were observed under a transmission electron microscope. Based on the development of the various permanent organelles and certain transient structures, the spermiogenesis of the Korean cattle could be divided into four main phases: Golgi, cap, acrosome and maturation, and these phases had been further subdivided into a total of nine stages. In the maturation phase, the condensation of the nuclear karyoplasm had progressed from the u, pp.r side to the lower side of the nucleus as the movement of the nuclear ring which would be disa, pp.ared after maturation.

  • Testes from the allied rock wallaby(Petrogale assimilus) have been examined by the electron microscopy in thin sections in order to examine spermiogenesis and structure of spermatozoa. The spermiogenesis can be divided into nine stages: early Golgi, late Golgi, collapsing, nuclear protrusion, condensation and flattening, nuclear shaping, rotation, nuclear ring contraction, and maturation. The acrosome has been abruptly formed following the collapse of expanded acrosomal vesicle without the cap stage described in the eutheria. The flatly condensed nucleus rotates obliquely to the axis of the axial filament complex and the folded acrosome covers the anterior third of the dorsal nuclear surface forming a wide subacrosormal space as the nuclear ring has contracted. The Sertoli cell reaction and spur are prominent during the nuclear protrusion and rotation stages. A mature spermatozoon has S-shape head which has an extended part reaching to the vicinity of the middle piece.

  • 성숙한 한국산 집고양이(Felis domesticn)의 정자변태 과정을 알아보기 위하여 정소 및 부정소의 조직을 채취하여 광학현미경과 전자현미경으로 관찰한 결과는 다음과 같았다. 1 세포 구조의 차이에 따라 한국산 집고양이의 정자변태과정을 출지, 두모. 첨체. 성숙 그리고 이탈단계로 구분하였는데. 즉 골지, 첨체 및 이탈단계를 각각 전 ·후기로, 두모단계는 전 ·중 ·후기로 성숙단계는 1단계로 하여 전체를 10기로 나눌 수 있었다. 2 정자의 꼬리는 두모중기부터 형성하기 시작한다. 3 염색질 과립은 골지단계에서 응축되기 시작하여 정체후기에서 구형의 형태로 진행되고 성숙단계에서 소멸된다.

  • Spermatid differentiations during spermiogenesis and sperm ultrastructures in male Mercenaria stimpsoni were investigated by transmission electron microscopic observations. In the early stage of the spermatid during spermiogenesis, a few granules and a proacrosomal granule, which is formed by the Golgi complex, become a proacrosomal vesicle. Consequently, it becomes an acrosome by way of the process of acrosome formation. The morphologies of the sperm nucleus type and the acrosome of this species have a curved cylindrical type and cap shape, respectively. The spermatozoon is approximately $48-51{\mu}m$ in length including a curved cylinderical sperm nucleus (about $4.18{\mu}m$ long), an acrosome (about $0.52{\mu}m$ in length) and tail flagellum ($42-45{\mu}m$ long). As some ultrastructural characteristics of the acrosomal vesicle, the peripheral parts of two basal rings show electron opaque part (region), while the apex part of the acrosome shows electron lucent part (region). These charateristics of the sperm belong to the family Veneridae in the subclass Heterodonta, unlike a characteristic of the subclass Pteriomorphia showing all part of the acrosome being composed of electron opaque part (region). Therefore, it is easy to distinguish the families or the subclasses by the acrosome structures. Exceptionally, In particular, a cylinder-like nucleus of the sperm is curved (the angle of the nucleus is about $80^{\circ}$), as seen in some species of Veneridae (range from $0^{\circ}$ to $80^{\circ}$). The number of mitochondria in the midpiece of the sperm of this species are four, as one of common characteristics appeared in most species except for a few species in Veneridae in the subclass Heterodonta. Cross-sectioned axoneme of the sperm tail flagellum shows a 9+2 structure.

  • The morphological characteristics of spermatogenic cells during the spermiogenesis of Paradoxornis webbiana were studied by transmission electron microscope. Spermiogenesis of P. webbiana was divided into ten phase. The chromatin granules became fibrous granules at the Golgi phase, gradually condensed at the cap phases, condensed as a stick at the acrosomal phase, and finally, a perfect nucleus was formed at the maturation phase. The formation of sperm tail began at the early Golgi phase, and completed at the late maturation phase. In particular, the dense materials existed in the sperm neck, which is wedged between the tip of segmented columns and the first mitochondria of the middle piece. The axone in the neck were surrounded by the dense materials. The axonema in spermatozoon contains a 9+2 arrangement of microtubules: 9 doublets, and 2 central single microtubules. Mitochondrial bundles of middle piece were composed of a pair of arms, which surrounded the axone of the middle piece by the $15^{\circ}$ angled-helical structure. The outer membrane of mitochondria were surrounded by microtubules in plasma membrane of the sperm. The undulating membrane had a helical structure, and the sperm plasma membrane was surrounded by undulating membrane.

  • Spermiogenesis in the Korean squirrel, Tamias sibiricus, was investigated by transmission electron microscopy. Spermiogenesis was divided into Golgi, cap, acrosome, maturation and spermiation phases based on the characteristics of acrosomal changes and nuclear shape. Beside, the Golgi, cap and acrosomal steps were subdivided into three phases of early, middle and late phase respectively, the maturation step was divided into two phases of early and late phase, and spermiation step has only one phase. Thus, the spermiognesis of T. sibiricus was divided into a total of twelve phases. In Golgi phase (steps 1-3), a well developed Golgi complex was located close to the vesicles, the acrosomal vesicle fixd to a recess of nuclear membrane at step 3. During cap phase (steps 4-6), the acrosomal vesicle spred over the nuclear surface to cover a third of the nucleus, and the acrosomal granule was not yet flattened. At acrosomal phase (steps 7-9), the nucleus and acrosome were elongated but nucleoplasm was not condensed. During maturation phase (steps 10-11), the nucleoplasm was more condensed, and the mitochondria completely arranged the center of axoneme. The spatulate-sperm head was completely formed at spermiation phase (step 12).

Spermiogenesis in the Korean Striped Fleid Mouse Apodcmus agrarius coreae (한국산 등줄쥐(Apodemus agrarius coreae)에서의 정자변태)

  • In order to study the process of spermiogenesis of the korean striped field mouse Apodemus agrorfus coreae, the cell differentiation of seminiferous epithelium and morphological features of mature sperm in cauda epididymis was examined and the results are as follows: Spermiogenesis was divided, according to the features of cell structure; Golgi, cap, acrosome and spermiation phases were further subdivided into two steps of early and late phases respectively, and maturation phase has only one step. Hence, the spermiogenesis consists of nine phases. in the changes of the chromatin in nucleus, the chromatin granules began to be condensed in the cap phase and the condensation proceeded to form a globular of nucleus at the acrsome phase. Finally, the chromatin regularization was completed and perfect nucleus of sperm was formed at the maturation phase. Sperm head had the falciform, and the outer dence fibers of middle piece were arranged in a horseshoe fashion. The outer dence fiber number 1, 5, 6 and 9 was larger than other fiber number 2, 3, 4, 7, 8.
