  • Polyamines are polyvalent cations which are ubiquitously present in pro- and eukaryotic cells. In the present study we investigated the action mechanism of spermine$\^$+4/ (C$\sub$10/), a natural polyamine, on the large-conductance Ca$\^$2+/-activated K (maxi-K) channel using excised patches from the human gastric smooth muscle.(omitted)

Involvement of Polyamines in the Control of Senescence in Lemna gibba G3 Fronds (Lemna gibba G3의 노화조절에 대한 폴리아민의 관여)

  • Exogenously applied spermine, spermidine and putrescine caused a delay of senescence in fronds Lemna gibba G3 under continuous illumination. When the proximal half of a frond containing the meristematic“pockets”was removed, endogenous spermidine level in the distal half (half frond) increased initially to a miximal level, which was followed by a decline during a period of 10 days of incubation in light. No appreciable changes were observed with putrescine or spermine levels. Treatment of fronds with $\alpha$-difluromethylarginine (DFMA) resulted in both reduced level of spermidine and enhancement of chlorophyll loss in half fronds. $\alpha$-difluoromethylornithine (DFMO) was found to be virtually ineffective in either parameter. Results of experiments with ABA and kinetin indicate that there is a close correlation between the progress of senescence and spermidine level in Lemna fronds under illumination. It is suggested that endogenous level of spermidine is associated, at least in part, with frond senescence in this aquatic plant.

Activation of Cabbage Phospholipase D by Polyamines (폴리아민에 의한 양배추 포스포리파제 D의 활성화)

  • The effect of polyamines on the cabbage phospholipase D(PLD) activity was investigated. The PLD activity was determined by pH-stat titration of phosphatidic acid, one of the enzymatic reaction product, using phosphatidyl choline small unilamellar vesicles as a substrate. The cabbage PLD was activated approximately 4 fold by spermine at 1 mM concentration. This spermine effect appears to be similar to the previous report on the PLD activation of rat brain mitochondrial fraction. It was also found that cationic polypetides such as polylysine and polyhistidine exerted a marked enhancement effect on the cabbage PLD. Particularly polyhistidine exerted approximately 5.5 fold enhancement effect at 0.062 mM concentration. The polyamine effect on the cabbage PLD was reexamined in the phosphatidylcholine/sodium dodecyl sulfate mixed micellar system. The relevance of polyamine effect on PLD activity is discussed in relation to the active site of PLD.

Effects of Methylglyoxal bis-(Guanylhydrazone) and Polyamines on Carbohydrate Metabolism during Adventitious Root Formation in Soybean cotyledons (Methylglyoxal bis-(Guanylhydrazone)과 Polymine이 대두 자엽 부정근 형성시 탄수화물 대사에 미치는 영향)

  • In order to study the effects of polyamine inhibitors and polyamines on adventitious root formation, the correlation between adventitious root formation and carbohydrate metabolism was investigated in inoculated soybean (Glycine max L.) cotyledons at the concentration of $10^{-3}\;M$ methylglyoxal bis-(guanylhydrazone)[MGBG], and $10^{-3}\;M$ MGBG plus $10^{-5}\;M$ polyamines (putrescine, spermidine and spermine), respectively, for the adventitious root formation medium. The contents of starch, maltose and sucrose were lower in control and were higher in the treatments containing $10^{-3}\;M$ MGBG, and $10^{-3}\;M$ MGBG plus $10^{-5}\;M$ polyamines during culture. It was shown that the soluble sugar levels, except glucose, were higher than that of control in most of the treatments and the change in glucose contents tended to be similar to that control. The amylase activity increased in control and MGBG treatment, the maltase activity was higher in control and the invertase activity showed less substantial changes during culture.ulture.

Changes in Polyamine and Tyramine Concentrations in Rice (Oryza sativa L.) during Maturation and Preharvest Sprouting (벼 등숙기와 수발아 기간동안 폴리아민과 티라민의 농도변화)

  • The levels of polyamines were measured to investigate the alternative nitrogen metabolism during maturation and sprouting in rice. The rice plants (cv. Ansanbyeo) were cultivated in 20-year-old non-fertilized field. The flag leaves and spikes were collected weekly after the earing stage and the seeds were harvested daily after lodging. Free, bound, and conjugated polyamines were analyzed using reverse phase HPLC. Putrescine, spermidine, spermine, agmatine and tyramine were the major amines found in rice. The level of stress-induced amine, putrescine increased during the preharvest sprouting confirming that the process was a stress to the plants. With all other polyamines, tyramine in free form decreased in flag leaves and panicles during seed maturation. However, agmatine in bound form showed a noticeable increase about 8-fold during 6 weeks period of maturation after which it declined to the bottom level. Among the individual amines, tyramine and spermine in conjugated form showed a marked change during matutation and sprouting. Interestingly, the level of tyramine with all conjugated polyamine decreased in spikes during seed maturation and increased during preharvest sprouting implying that tyrosine decarboxyation and conjugation to phenolic acids may play a key role in preharvest sprouting. Spermine in conjugated form was synthesized only at the early earing stage in the level of $3.4mole\;g^{-1}$ fresh weight, and then decreased to the level of nmole during maturation. Thereafter, it dramatically increased to 2.8 mole during preharvest sprouting. In this study we found the tyramine is a major amine in rice, and it would play a critical role in N-assimilation during seed maturation and sprouting.

In vitro Culture and Acclimatization of Regenerated Plants of Liliem cernum $K_{OMAROV}$ (솔나리 기내배양 및 재분화 식물체의 토양순화)

  • The regenerated-bulblets placed in liquid free media resulted in good formation of roots and bulblets. On 1/4 MS free medium, roots and bulblets were predominantly induced. The 1/4 MS liquid medium supplemented with plant growth regulators was the best suitable condition for elongation of leaves and roots. Somatic embryos were frequently developed from embryogenic callus in liquid media with 2,4-D 1mg/ l . On free liquid media, the viability of callus reduced. As the salt strength of MS media reduces, the viability of callus reduced significantly. However, Leaves were induced from several callus clumps. When leaves, roots and bulb-scale segments were placed on MS media containing NAA 1mg/ l or 2,4-D 1mg/ l and various sucrose concentration, the best result about the differentiation, growth of leaf and the differentiation of leaf was obtained on MS media added 1.5% sucrose and 2,4-D 1mg/ l, 3% sucrose and NAA 1mg/ l, and 1.5% sucrose and NAA 1mg/ l, respectively. Also the better result differentiation, growth of root and differentiation of bulb was obtained on MS media with 6% sucrose and NAA 1mg/ l. Spermidine promoted the growth of leaf and the differentiation of bulb. However, spermine promoted the differentiation of leaf, the differentiation and the growth of root in MS solid media. On the MS liquid media, both spermine and spermidine stimulated organogenesis from bulb-scale segments. Regenerated plantlets were acclimatizated and grown in greenhouse in vermiculite + perlite (1 : 1 by volume) well. The optimal soil condition of rooting for plantlets regenerated was in peat moss.

Polyamine, Cytochrome c and Enzymes Related to the Utilization of Methanol in Methylobacterium extorquens AMI Growing at Different pHs (상이한 수소이온농도에서 성장하는 Methylobacterium extorquens AM1의 메탄올 이용 관련효소와 Cytochrome c 및 폴리아민)

  • The generation time of Methylobacterium extorquens AMI growing on methanol at pH 5.5 and 7.0 was found to be 23 hand 8.3 h. respectively. The bacterium grown at pH 7.0 were found to contain more amounts of spermidine and putrescine than the cell grown at pH 5.5. Cells grown at both conditions exhibited strong methanol dehydrogenase (MDH) activity at the mid-exponential growth phase. The amounts of MDH. however. were found to be almost equal through all gro~1h phases. Cells growing at the stationary phase contained large amounts of cytochrome c. The cytochrome c content was higher in cells growing at pH 7.0 than the cells growing at pH 5.5. Cells growing at pH 5.5 in the presence of putrescine or spermidine contained increased amounts of putrescine. The level of spermine, however. was decreased and that of spermidine was not changed. Spermine added into the medium was found to have no effect on the level of cellular polyamines. Putrescine or spermidine added into the medium stimulated MDH and hydroxypyruvate reductase activities. but did not affect the contents of MDH and cytochrome c. It was found that preincubation of cell-free extracts with polyamines does not stimulate MDH and hydroxypyruvate reductase activities.

Mechanism of Apoptosis Induced by Spermine in MCF-7 Breast Cancer Cells (MCF-7 유방암 세포주에 있어서 spermine에 의해 유도된 세포사멸 기작)

  • In the present work, we show that spermine (spm)-induced cytotoxicity is due to the mitochondrial-dependent pathway triggered by the intracellular $Ca^{2+}$ increase in MCF-7 human breast cancer cells. Spm induced the intracellular $Ca^{2+}$ increase in a dose-dependent manner in the medium containing 1.5 mM $Ca^{2+}$. Even in the $Ca^{2+}$-free medium, spm could induce a minor $Ca^{2+}$ increase in a dose-dependent fashion, suggesting a probable leak from the internal storage. The cytotoxic effect of $Ca^{2+}$ could be further proved by using either BAPTA or ionophore. Spm-induced $Ca^{2+}$ increase led to the release of cytochrome c from mitochondria into the cytosol and the change of mitochondrial membrane potential. In MCF-7 cells, caspase-7 plays a key role in the downstream of apoptosis because caspase-3 is absent. In the cells treated with spm, the cleavage of caspase-7 and -12 was increased almost two-fold. The level of anti-apoptotic Bcl-2 protein decreased to 35% of the control; however, the cells showed increased expression of pro-apoptotic Bax protein about two-fold in response to spm. These results imply that the apoptotic signaling pathway activated by spm is likely to be mediated via the mitochondrial-dependent pathway.

Polyamine Alleviates Inhibition of O2 Evolution by Cd for Spinich (시금치에서 Polyamine에 의한 카드늄의 산소방출억제 경감효과)

  • Polyamine concentrations were clearly enhanced in leaves and chloroplasts by Cd treatment, but not in thylakoid and PSII. It appeared that newly synthesized polyamines by Cd are distributed in stromal space. The accumulated polyamines in stromal space could not be adjacent to thylakoid membranes, suggesting that they are already saturated. The levels of putrescine and spermine were about 36 and 20 fold lower in chloroplast than in whole leaf cells respectively, whereas agmatine level was only 3.7 fold lower. The inhibitory effect of Cd nn $O_2$ evolving process was obviously alleviated by 0.2mM spermine supplement. Polyamines stimulated $O_2$ evolution within the range of 0.5mM in spinach thylakoids. It was also found that stimulating effect of polyamines is about 2 fold higher in dicothyledonous spinach than in monocotyledonous wheat at same concentrations. Furthermore, the enhanced activity of $O_2$ evolution was lowered rather by agmatine treatment than by putrescine treatment in wheat, suggesting a difference between monocot and dicot.
