STUDIES OF GRAVITY WAVES USING MICHELSON INTERFEROMETER MEASUREMENTS OF OH(3-1)BANDS

  • As part of a long-term program for polar upper atmospheric studies, temperatures and intensities of the OH(3-1) bands were derived from spectrometric observations of airglow emissions over King Sejong station($62.22^{circ}S,\;301.25^{circ}E$). These measurements were made with a Michelson interferometer to cover wavelength regions between 1000nm and 2000 nm. A spectral analysis was performed to individual nights of data to acquire information on the waves in the upper mesosphere/lower thermosphere. It is assumed that the measured fluctuations in the intensity and temperature of the OH (3-1) airglow were caused by gravity waves propagating through the emission layer. Correlation of intensity and temperature variation revealed oscillations with periods ranging from 2 to 9 hours. We also calculated Krassovsky’s parameter and compared with published values.

Evaluation of proteomic strategies for analyzing ubiquitinated proteins

  • Ubiquitin is an essential, highly-conserved small regulatory protein in eukaryotic cells. It covalently modifies a wide variety of targeted proteins in the forms of monomer and polymers, altering the conformation and binding properties of the proteins and thus regulating proteasomal delivery, protein activities and localization. Mass spectrometry has emerged as an indispensable tool for in-depth characterization of protein ubiquitination. Ubiquitinated proteins in cell lysates are usually enriched by affinity chromatography and subsequently analyzed by mass spectrometry for identification and quantification. Ubiquitin-conjugated amino acid residues can be determined by unique mass shift caused by the modification. Moreover, the complex structure of polyubiquitin chains on substrates can be dissected by bottom-up and middle-down mass spectrometric approaches, revealing potential novel functions of polyubiquitin linkages. Here I review the advances and caveats of these strategies, emphasizing caution in the validation of ubiquitinated proteins and in the interpretation of raw data.

  • Two derivatives of star shaped compounds based on naphthylamine with symmetric trisubstituted benzene as core, methoxy and ethoxy as end substitutions are synthesized. The synthesized compounds are characterized by UV-visible, FT-IR and NMR spectrometric techniques. The electronic and thermal properties of the compounds are studied using cyclic voltametry (CV) and differential scanning calorimetry (DSC) respectively. The data's obtained have similarity with the arylamines that have been already used in optoelectronic devices. So these compounds are interesting materials for applications in such devices.

Xyloglucan in the Differentiating Xylem of the Populus deltoides M.

  • The chemical composition of the differentiating xylem of Populus deltoides M. was investigated and compared with that of sapwood. The cell wall polysaccharides were extracted sequentially from adifferentiating xylem and fractionated with gel chromatography. The sugar composition of each fraction was analyzed with G..C and H.P.L.C. The cell wall of the differentiating xylem is rich with the pectin substance and hemicellulose compared with that of sapwood. The water-extracted polysaccharides from the differentiating xylem were composed mainly of xylose and glucose residues. The sugar composition of some of the fractions in the gel filtration of purified $H_{2}O$ polysaccharide suggest that xyloglucan was extracted with $H_{2}O$ from differentiating xylem. Also, we can supposed that the purified $H_{2}O$ polysaccharide might be xyloglucan from the spectrometric data(IR and NMR) of purified $H_{2}O$ polysaccharide.

Continuous On-line Estimation of Cell Growth and Substrate Consumption Using a Computer-coupled Mass Spectrometer (Computer-coupled Mass Sepctrometer를 이용한 세포증식과 기질소모의 연속적 On-line추정)

  • From the on-line mass spectrometric analyese of the exhaust gaseous composition of fermentor and the material balance equations for oxygen and carbon dioxide, oxygen uptake rate (OUR) and carbon dioxide evolution rate (CER) were calculate using a personal computer (IBM PC-AT) interfaced to a quadrupole mass spectromter. The calculate OUR and CER were used for the indirect estimation of cell and substrate concentrations during the batch culture of Candida utilis. For the estimation of sustrate concentration, the yield model of Pirt was applied. It was found that the cell and substrate (glucose) concentration could be ssatisfactorily estimataed and the results showed the more accurate estimations of both fermentation state variables when OUR data were used than CER data.

  • Continuous on-line monitoring of cell concentration and growth rate in aerobic batch fermentation process was carried out by analyzing the exhaust gas composition of tormentor with a quadrupole mass spectrometer. From the mass spectrometric analyses of major gaseous components, i.e. $N_2$, $O_2$, $CO_2$ and $H_2O$, and the material balance equations for oxygen and carbon dioxide, oxygen uptake rate (OUR) rind carbon dioxide evolution rate (CER) were instantaneously calculated using a computer (16-bit IBM PC-AT) interfaced to a quadrupole mass spectrometer. The calculated OUR and CER data were used for the estimation of cell concentration and growth rate of Candida utilis during batch culture. It was found that the cell concentration could be satisfactorily estimated from the data of OUR arid CER during the culture and this method could be successfully und for the continuous monitoring of cell growth and growth rate.

Physicochemical Characteristics of Korean Folk Sojues (전통민속소주의 물리화학적 특성)

  • In order to provide a quality index of Korean folk sojues, physicochemical properties of Korean folk sojues (Andong soju, Moonbaesul, Leekangju, Jindo Hongju, Chebiwon soju, Yethyang (rice) and Yethyang (barley)), Paekrosul, Chinese kaoliangchiews (Moutaichiew, Ergoutoutiu, Chuyehchingchiew, Zhikukaoliangchiew and Paigal), Japanese Senbatanuki shochu and two whiskies were determined. The pH of sojues ($3.43{\sim}5.85$) were mainly influenced by total acidity which was described as acetic acid. The conductivities of Paekrosul and Leekangju showed relatively high value of $246\;and\;122.7{\mu}S/cm$, respectively. Korean folk sojues and Paekrosul showed maximum absorption at 274 or $278{\sim}280nm$ in the spectrometric study. We performed principal components analysis (PCA) of physicochemical properties and spectrometric data to extract representative characteristics and to compare the similarity or the dissimilarity. The PCA plot showed the distinguished cluster of Korean folk sojues comparing with Chinese kaoliangchiew or Paekrosul etc.

  • Chemical investigation of the plant Tristemma hirtum P. Beauv (Melastomataceae) resulted to the isolation of a new flavonol glycoside named quercetin-7-O-${\alpha}$-D-arabinofuranoside (1), together with nine known compounds including 3'-hexadecanoyl-2'-(9aZ)-tetradecanoyl-glycerol 1'-O-[${\beta}$-D-galactopyranosyl-(1'' ${\rightarrow}$ 6'')-${\alpha}$-D-galactopyranoside] (2), arjunolic acid (3), ${\beta}$-sitosterol-3-O-${\beta}$-D-glucopyranoside (4), terminolic acid (5), quercetin (6), asiatic acid (7), maslinic acid (8), $1{\beta}$-O-galloylpedunculagin (9) and 6-hydroxyapigenin 7-O-${\beta}$-D-glucopyranoside (10) from the methanol extract using normal and reversed phase column chromatography. The structures of these compounds were determined by comprehensive interpretation of their spectral data mainly including 1D- 2D-NMR ($^1H-^1H$ COSY, HSQC, and HMBC) spectroscopic and ESI-TOF-MS mass spectrometric analysis.

Characterization and Cytotoxic Activities of Nonadecanoic Acid Produced by Streptomyces scabiei subsp. chosunensis M0137 (KCTC 9927)

  • The substance 0116p, which exhibits cytotoxicity against human macrophage cell line THP-1, was isolated from a mycelial extract of Streptomyces scabiei subsp. chosunensis M0137. The cytotoxic substance was purified by Diaion-HP2O adsorption, solvent extraction, Sephadex LH-20 column chromatography, and silica-gel column chromatography. The molecular formula is C$\_$19/H$\_$38/O$\_$2/ (MW301.10) based on elemental and spectrometric analysis. It was identified as nonadecanoic acid by NMR spectral data. It exhibits cytotoxic activities in various human cancer cell lines, including A549, SK-OV-3, SK-MEL-2 and HCT-15. In addition, 0116p also inhibits IL-12 production in lipopolysaccharide-activated macrophages.

A Study on the Component Analysis of Sappan Wood Extracts (소목 추출물의 구조분석)

  • Colorants were extracted from the heartwood of sappan lin. with MeOH under reflux, and the concentrate or the powder of dye was prepared by low pressure concentration method using suitable organic solvent. Various components were isolated from sappan wood, and the chemical structure and mechanism of compound having the excellent antibacterial and deodorization properties were analyzed. The results obtained are as follows ; The seventeen components of sappan wood were seperated by HPLC chromatography, and the five components among them were existed more than 6% and the other components were existed lower than 0.6%. The resolving powers of the non-polar solvent and polar solvent systems were evaluated by their ability to resolve the samples. It showed that chloroform-methanol-water(800:150:10) system has the best resolving power. Although the seperation rate is very slow, polyamide C-100 column chromatography gives a clear seperation of sappan wood. On the basis of the spectrometric data such as IR, UV, $GC-Mass,\;^1H-NMR,\;^{13}C-NMR\;and\;^1H-^{ 13}C-NMR$, the chemical structure of compound haying the excellent antibacterial and deodorization properties was established as brazilin containing the functional groups such as two quaternary carbon, one benzyl carbon, methylene contiguous to oxygen and methylene caused by oxygen atom.
