Sixty-five molds isolated from clinical specimens were included in this study. All the isolates were molds that could be identified morphologically, strains that are difficult to identify because of morphological similarities, and strains that require species-level identification. PCR and direct sequencing were performed to target the internal transcribed spacer (ITS) region, the D1/D2 region, and the β-tubulin gene. Comparative sequence analysis using the GenBank database was performed using the basic local alignment search tool (BLAST) algorithm. The fungi identified morphologically to the genus level were 67%. Sequencing analysis was performed on 62 genera and species level of the 65 strains. Discrepancies were 14 (21.5%) of the 65 strains between the results of phenotypic and molecular identification. B. dermatitidis, T. marneffei, and G. argillacea were identified for the first time in Korea using the DNA sequencing method. Morphological identification is a very useful method in terms of the reporting time and costs in cases of frequently isolated and rapid growth, such as Aspergillus. When molecular methods are employed, the cost and clinical significance should be considered. On the other hand, the molecular identification of molds can provide fast and accurate results.

We perform an experimental study of concrete beam with pultruded fiber reinforced polymer(FRP) plank using as a permanent formwork and the tensile reinforcement. A satisfactory bond at the interface between the smooth surface of the pultruded plank and the concrete must be developed for the FRP plank and the concrete to act as a composite structural member. Two kinds of aggregate were bonded to the FRP plank using a commercially available epoxy. No additional flexural or shear reinforcement was provided in the beams. For comparison we test two types of control specimen. One control did not have any aggregate bonded to the FRP plank and the other control had infernal steel reinforcing bars instead of the FRP plank. The beams were loaded by central patch load to their ultimate capacity. The experimental results were compared to current ACI 318 (2005) and ACI 440 (2006) code predictions. This study demonstrates that the FRP plank has the potential to serve as formwork and reinforcing for concrete structures.

The National Environmental Specimen Bank (NESB) has set up a plan to develop reference materials in the facility for assuring analytical quality and validating analytical methods for its monitoring samples. Some of the crucial characteristics that reference materials must consist of are homogeneity and stability of both intra and inter-bottles. In this study, we examined the homogeneity and stability of cryogenically-milled muscle samples, from Common Carp (Cyprinus carpio) for total mercury. Homogeneity was tested using ANOVA analysis and regression analysis was used to test short-term stability. The variations of total mercury concentration did not significantly differ between the intra and the inter-bottle (F=0.8, p=0.37). Additionally, relative standard deviation of the total mercury concentration showed low values (2.28%). For the short-term stability test, total mercury variations were not statistically significant as demonstrated by the result of the regression analysis (F ratio = 3.11, p = 0.18). This suggests that the cryogenic-milling process has statistically proven the degree of homogeneity and short-term stability for samples of carp muscles in the chemical analysis for total mercury.

The National Environmental Specimen Bank (NESB) has been sampling and cryogenically preserving various wildlife specimens to monitor bio-accumulations of chemical pollutants since 2010. Recently, the NESB set up a plan to develop reference materials at their facility to assure the analytical quality of and validate the analytical methods for their monitoring samples. One of the crucial characteristics of reference materials is intra- and inter-bottle homogeneity. In this study, we used ANOVA for total mercury concentrations in some samples to validate their homogeneities after milling and homogenization. We examined the intra- and inter-bottle homogeneities of two cryogenically-milled samples (Korean mussel (Mytilus coruscus) and black-tailed gull's egg (Larus crassirostris). The variations in the total mercury concentrations were not significantly different intra- and inter-bottle (mussel: F=0.74, p=0.67; gull egg: F=1.96, p=0.10). Additionally, the relative standard deviations of the total mercury concentrations showed low values (mussel: 2.02%, gull egg: 1.78%). Therefore, the cryogenic-milling process statistically proves the homogeneity of the materials of mussels and sea gull eggs for chemical analysis for total mercury.
This study was carried out to apply the taxonomic characters to identify and to provide the habitat circumstances for forty one un-introduced environmentally harmful plants in the Korea which have been designated by Ministry of Environment in 2016. We investigated the dried plant specimen from several herbaria and performed field habitat survey in western coast area of United States including California, Oregon and Washington. We suggest the first result for ecological risk discussion, weed risk assessment, taxonomic characters and classification keys for 8 species with the related species - Carduus acanthoides L. (Asteraceae), Carduus tenuiflorus W. Curtis (Asteraceae), Onopordum acanthium L. (Asteraceae), Chromolaena odorata (L.) R.M. King & H. Rob. (Asteraceae), Hydrocotyle ranunculoides L.f. (Apiaceae), Oenanthe pimpinelloides L. (Apiaceae), Ehrharta erecta Lam. (Poaceae), and Paspalum conjugatum P.J. Bergius (Poaceae).
Morphological and molecular characterization of clinostomid metacercariae (CMc) was performed with the specimens collected in fish from Korea and Myanmar. Total 6 batches of clinostomid specimens by the fish species and geographical localities, 5 Korean and 1 Myanmar isolates, were analyzed with morphological (light microscopy and SEM) and molecular methods (the cytochrome c oxidase 1 gene and internal transcribed spacer 1/5.8S rRNA sequence). There were some morphological variations among CMc specimens from Korea. However, some morphometrics, i.e., the size of worm body and each organ, ratio of body length to body width, and morphology of cecal lumens, were considerably different between the specimens from Korea and Myanmar. The surface ultrastructures were somewhat different between the specimens from Korea and Myanmar. The CO1 sequences of 5 Korean specimens ranging 728-736 bp showed 99.6-100% identity with Clinostomum complanatum (GenBank no. KM923964). They also showed 99.9-100% identity with C. complanatum (FJ609420) in the ITS1 sequences ranging 692-698 bp. Meanwhile, the ITS1 sequences of Myanmar specimen showed 99.9% identity with Euclinostomum heterostomum (KY312847). Five sequences from Korean specimens clustered with the C. complanatum genes, but not clustered with Myanmar specimens. Conclusively, it was confirmed that CMc from Korea were morphologically and molecularly identical with C. complanatum and those from Myanmar were E. heterostomum.
Five years have passed since the first set of environmental samples was taken in 2011 to represent various ecosystems which would help future generations lead back to the past environment. Those samples have been preserved cryogenically in the National Environmental Specimen Bank(NESB) at the National Institute of Environmental Research. Even though there is a strict regulation (SOP, standard operating procedure) that rules over the whole sampling procedure to ensure each sample to represent the sampling area, it has not been put to the test for the validation. The question needs to be answered to clear any doubts on the representativeness and the quality of the samples. In order to address the question and ensure the sampling practice set in the SOP, many steps to the measurement of the sample, that is, from sampling in the field and the chemical analysis in the lab are broken down to evaluate the uncertainty at each level. Of the 8 species currently taken for the cryogenic preservation in the NESB, pine tree samples from two different sites were selected for this study. Duplicate samples were taken from each site according to the sampling protocol followed by the duplicate analyses which were carried out for each discrete sample. The uncertainties were evaluated by Robust ANOVA; two levels of uncertainty, one is the uncertainty from the sampling practice, and the other from the analytical process, were then compiled to give the measurement uncertainty on a measured concentration of the measurand. As a result, it was confirmed that it is the sampling practice not the analytical process that accounts for the most of the measurement uncertainty. Based on the top-down approach for the measurement uncertainty, the efficient way to ensure the representativeness of the sample was to increase the quantity of each discrete sample for the making of a composite sample, than to increase the number of the discrete samples across the site. Furthermore, the cost-effective approach to enhance the confidence level on the measurement can be expected from the efforts to lower the sampling uncertainty, not the analytical uncertainty. To test the representativeness of a composite sample of a sampling area, the variance within the site should be less than the difference from duplicate sampling. For that, a criterion, ${i.e.s^2}_{geochem}$(across the site variance) <${s^2}_{samp}$(variance at the sampling location) was proposed. In light of the criterion, the two representative samples for the two study areas passed the requirement. In contrast, whenever the variance of among the sampling locations (i.e. across the site) is larger than the sampling variance, more sampling increments need to be added within the sampling area until the requirement for the representativeness is achieved.
The National Environmental Specimen Bank (NESB) has been collecting broad leave samples to monitor environmental pollution from five different designated sampling areas. In order to ensure the reproducibility and comparability of the results, all the procedures from selecting trees and pooling leaves to make the representative sample are defined in the standard operation procedures(the SOP). The representative samples were subjected to the chemical analyses for some heavy minerals and Polycyclic Aromatic Hydrocarbons(PAHs). The uncertainty levels involved in each step of the SOP, that is, the sampling and the chemical analysis, were derived using the Robust ANOVA, which enables the relative comparison among the different levels of pollutants concentrations with confidence. Furthermore, the effect of the varying degrees of precipitation on the pollutants concentration of the leaves was also examined. Overall, the biological difference estimated from the duplicate samples was found to exceed the variation across the site, implying even aerial deposition over site. Samples from Gwanak Mt. showed highest heavy metal concentrations than the other sites. Washing off effect of the pollutants adhering in the form of particles on the leaf surface was found to be affected by the cumulative precipitation.

For the effective control of Syphilis, many investigators have developed a more rapid, simple and economical screening serological test which is adequately sensitive and specific. To fulfill the requirements of a more rapid serologic test for syphlis, a substitute for the conventional serum specimen was needed since considerable time and labor are involved in the processing of blood to serum. Burdon suggested the use of plasma in the serologic tests for syphilis as a substitute for serum. He noticed that plasma was more sensitive than serum in the Kline and Kahn tests, and attributed this to the presence of more antibody-like substance, "reagin" in plasma than in serum. However, to make plasma sufficiently sensitive, it was necessary to inactivate plasma by heating at a temperature of $56^{\circ}C$ for about 30 minutes. Heating of plasma resulted in the precipitation of fibrinogen which made centrifugation necessary to obtain dear plasma. Since the chief disadvantage to the use of unheated plasma(or serum) was a reduction in sensitivity of results-which probably was due to a labile factor such as complement-Portnoy et al began to consider rapid chemical methods of inactivation of plasma(or serum). They experienced that choline chloirde was shown to be anticomplementary which suggested its use as an inactivating agent for unheated plasma(or serum). In 1959 Portnoy et al reported the Rapid Plasma Reagin(RPR) Test for syphilis which is a more rapid, economical and simple. But still this test has many disadvantages as a rapid performing, field and office procedure, because it requires the usual laboratory equipments such as centrifuge, rotating machine, microscope etc. To substitute these disadvantages of the RPR test, in 1962, Portnoy et al developed the Rapid Plasma Reagin(RPR) card test for syphilis, which has the following advantages: a) Simplicity and rapidity of performance, b) Requires no laboratory equipments, c) Stable antigen suspension, d) Adequate sensitivity and specificity. This RPR card test can be used as a rapidly performing and screening test in field investigation, outpatient clinics, small laboratories and hospitals doing limited syphilis serology, and predonor in blood bank. Private clinic which has limited laboratory equipment and technic for syphilis serology can also use this RPR card test as a tool in the rapid diagnosis of syphilis. It was thought that this RPR card test is a useful tool in Korea for private physician and mass survey for syphilis diagnosis. But Portnoy patented the reagents needed for the performing the RPR card test. Therefore authors developed newly the reagents and according to Portnoy's method evaluated the newly developed. RPR card test compared with the VDRL, Kolmer CF, and RPCF tests. The RPR card and VDRL tests were performed plasma and serum from the total 1,132 cases. Among these 1,131 cases, 521 were syphilis suspected laboratory specimens, and 611 were syphilis unsuspected healthy young men. After screening with these two tests, the RPR card and VDRL tests, reactive specimens to the above one or both tests were retested by the Kolmer CF and RPCF tests.

Strength anisotropy is a typical feature of thin-bedded rock masses and their strength will be degraded subjected to water immersion effect. Such effect is crucial for the operation of hydropower plant because the impoundment lifts the water level of upstream reservoir and causes the rock mass of nearby slopes saturated. So far, researches regarding mechanical property of natural thin-bedded rock masses and their strength variation under water immersion based on field test method are rarely reported. This paper focuses on a thin-bedded stratified rock mass and carries out field test to investigate the mechanical property and strength variation characteristics. The field test is highlighted by samples which have a large shear dimension of 0.5 m*0.5 m, representing a more realistic in-situ situation than small size specimen. The test results confirm the anisotropic nature of the concerned rock mass, whose shear strength of host rocks is significantly larger than that of bedding planes. Further, the comparison of shear strength parameters of the thin-bedded rock mass under natural and saturated conditions show that for both host rocks and bedding planes, the decreasing extent of cohesion values are larger than friction values. The quantitative results are then adopted to analyze the influence of reservoir impoundment of a hydropower plant on the surrounding rock mass stability of diversion tunnels which are located in the nearby slope bank. It is evaluated that after reservoir impoundment, the strength degradation induced incremental deformations of surrounding rock mass of diversion tunnels are small and the stresses in lining structure are acceptable. It is therefore concluded that the influences of impoundment are small and the stability of diversion tunnels can be still achieved. The finings regarding field test method and its results, as well as the numerical evaluation conclusions are hoped to provide references for rock projects with similar concerns.
