비피더스 균주가 가지고 있는 여러 생리 활성을 나타내는 부가가치를 부여한 새로운 고기능성 심치 제품을 개발하기 위해 편성 혐기성 미생물인 비피더스균 가운데 산소에 내성이 있는 Bifidobacterium lactis를 선발하여 김치 제조에 적용하였다. 이 균주의 내산성과 내염성을 각각 알아본 결과 pH 30에서는 별다른 영향을 받지 않았으며 pH 2.5에서도 50% 이상의 생존율을 나타내었다. 그리고 NaCl 농도 3.5%에서도 사멸하지 않는 것으로 나타나 일반적인 김치의 pH와 염농도를 고려할 때 김치에 적용 가능한 것으로 판단되었다. 첨가된 Bifidobacterium lactis가 김치 내에서 생존함을 확인하기 위해서 PCR을 이용하여 확인한 결과 15일 이상 생존하고 있음을 확인할 수 있었다. 그리고 관능평가의 경우 Bifidobacterium lactis 첨가 김치가 전체적인 기호도에서 균주를 첨가하지 않은 대조군 김치와 유사한 특성을 나타냄으로써 Bifidobacterium lactis를 이용한 고기능성 김치의 제조가 가능할 것으로 판단되었다.
Vegetative growth signaling of the opportunistic human pathogenic fungus Aspergillus fumigatus is mediated by GpaA ($G{\alpha}$). FlbA is a regulator of G protein signaling, which attenuates GpaA-mediated growth signaling in this fungus. The flbA deletion (${\Delta}flbA$) and the constitutively active GpaA ($GpaA^{Q204L}$) mutants exhibit enhanced proliferation, precocious autolysis, and reduced asexual sporulation. In this study, we demonstrate that both mutants also show enhanced tolerance against $H_2O_2$ and their radial growth was approximately 1.6 fold higher than that of wild type (WT) in medium with 10 mM $H_2O_2$. We performed quantitative PCR (qRT-PCR) for examination of mRNA levels of three catalase encoding genes (catA, cat1, and cat2) in WT and the two mutants. According to the results, while levels of spore-specific catA mRNA were comparable among the three strains, cat1 and cat2 mRNA levels were significantly higher in the two mutants than in WT. In particular, the ${\Delta}flbA$ mutant showed significantly enhanced and prolonged expression of cat1 and precocious expression of cat2. In accordance with this result, activity of the Cat1 protein in the ${\Delta}flbA$ mutant was higher than that of $gpaA^{Q204L}$ and WT strains. For activity of the Cat2 protein, both mutants began to show enhanced activity at 48 and 72 hr of growth compared to WT. These results lead to the conclusion that GpaA activates expression and activity of cat1 and cat2, whereas FlbA plays an antagonistic role in control of catalases, leading to balanced responses to neutralizing the toxicity of reactive oxygen species.

In this study, we investigated genetic diversity of 22 microsporidian isolates infecting tropical tasar silkworm, Antheraea mylitta collected from various geographical forest locations in the state of Jharkhand, India, using polymerase chain reaction (PCR)-based marker assay: random amplified polymorphic DNA (RAPD). A type species, NIK-1s_mys was used as control for comparison. The shape of mature microsporidians was found to be oval to elongate, measuring 3.80 to $5.10{\mu}m$ in length and 2.56 to $3.30{\mu}m$ in width. Of the 20 RAPD primers screened, 16 primers generated reproducible profiles with 298 polymorphic fragments displaying high degree of polymorphism (97%). A total of 14 RAPD primers produced 45 unique putative genetic markers, which were used to differentiate the microsporidians. Calculation of genetic distance coefficients based on dice coefficient method and clustering with un-weighted pair group method using arithmetic average (UPGMA) analysis was conducted to unravel the genetic diversity of microsporidians infecting tasar silkworm. The similarity coefficients varied from 0.059 to 0.980. UPGMA analysis generated a dendrogram with four microsporidian groups, which appear to be different from each other as well as from NIK-1s_mys. Two-dimensional distribution based on Euclidean distance matrix also revealed considerable variability among different microsporidians identified from the tasar silkworms. Clustering of few microsporidian isolates was in accordance with the geographic origin. The results indicate that the RAPD profiles and specific/unique genetic markers can be used for differentiating as well as to identify different microsporidians with considerable accuracy.
Salmonellosis is a highly contagious bacterial disease that threatens both human and poultry health. Tests that can detect Salmonella in the field are urgently required to facilitate disease control and for epidemiological investigations. Here, we combined loop-mediated isothermal amplification (LAMP) with a chromatographic lateral flow dipstick (LFD) to rapidly and accurately detect Salmonella. LAMP primers were designed to target the Salmonella invA gene. LAMP conditions were optimized by adjusting the ratio of inner to outer primers, $MgSO_4$ concentration, dNTP mix concentration, amplification temperature, and amplification time. We evaluated the specificity of our novel LAMP-LFD method using six Salmonella species and six related non-Salmonella strains. All six of the Salmonella strains, but none of the non-Salmonella strains, were amplified. LAMP-LFD was sensitive enough to detect concentrations of Salmonella enterica subsp. enterica serovar Pullorum genomic DNA as low as $89fg/{\mu}l$, which is 1,000 times more sensitive than conventional PCR. When artificially contaminated feed samples were analyzed, LAMP-LFD was also more sensitive than PCR. Finally, LAMP-LFD gave no false positives across 350 chicken anal swabs. Therefore, our novel LAMP-LFD assay was highly sensitive, specific, convenient, and fast, making it a valuable tool for the early diagnosis and monitoring of Salmonella infection in chickens.
Erwinia amylovora and Erwinia pyrifoliae cause fire blight and black-shoot blight, respectively, in apples and pears. E. pyrifoliae is less pathogenic and has a narrower host range than that of E. amylovora. Fire blight and black-shoot blight exhibit similar symptoms, making it difficult to distinguish one bacterial disease from the other. Molecular tools that differentiate fire blight from black-shoot blight could guide in the implementation of appropriate management strategies to control both diseases. In this study, a primer set was developed to detect and distinguish E. amylovora from E. pyrifoliae by conventional polymerase chain reaction (PCR). The primers produced amplicons of different sizes that were specific to each bacterial species. PCR products from E. amylovora and E. pyrifoliae cells at concentrations of 104 cfu/ml and 107 cfu/ml, respectively, were amplified, which demonstrated sufficient primer detection sensitivity. This primer set provides a simple molecular tool to distinguish between two types of bacterial diseases with similar symptoms.

Tomato yellow leaf curl virus (TYLCV) and tomato spotted wilt virus (TSWV) are well-known examples of the begomovirus and orthotospovirus genera, respectively. These viruses cause significant economic damage to tomato crops worldwide. Weeds play an important role in the ongoing presence and spread of several plant viruses, such as TYLCV and TSWV, and are recognized as reservoirs for these infections. This work applies a comprehensive approach, encompassing field surveys and molecular techniques, to acquire an in-depth understanding of the interactions between viruses and their weed hosts. A total of 60 tomato samples exhibiting typical symptoms of TYLCV and TSWV were collected from a tomato greenhouse farm in Nonsan, South Korea. In addition, 130 samples of 16 different weed species in the immediate surroundings of the greenhouse were collected for viral detection. PCR and reverse transcription-PCR methodologies and specific primers for TYLCV and TSWV were used, which showed that 15 tomato samples were coinfected by both viruses. Interestingly, both viruses were also detected in perennial weeds, such as Rumex crispus, which highlights their function as viral reservoirs. Our study provides significant insights into the co-occurrence of TYLCV and TSWV in weed reservoirs, and their subsequent transmission under tomato greenhouse conditions. This project builds long-term strategies for integrated pest management to prevent and manage simultaneous virus outbreaks, known as twindemics, in agricultural systems.
Aeromonas spp. 감염은 관상어 산업뿐만 아니라 양식업에서도 상당한 경제적 손실을 초래하는 것으로 보고되고 있다. 한편 2022년 12월부터 2023년 1월 사이에 국내에서 사육중인 금붕어(Crassius auratus)에서 Aeromonas병증을 보이는 어체에 대해서 발병 원인균을 규명하기 위해서 조사 하였다. 병증을 보이는 금붕어의 피부와 내부 장기에서 그람 음성균이 분리되었다. Biochemical test와 16S rRNA gene으로는 Aeromonas 속까지 동정할 수 있었으나 종레벨까지는 구분하기 어려웠다. 이에 가장 상동성이 높았던 A.veronii를 대상으로 species-specific PCR을 수행했고 명확히 종 수준으로 동정할 수 있었다. 아울러서 장독소 유전자, 감염 실험 및 항생제 내성 분석을 수행하였다. 금붕어에 대한 균주의 생체 내 병원성 시험 결과, 실험 주사 후 일주일 이내에 감염된 숙주에서 100%의 치사율을 나타냈다. 장독소 유전자인 act 독력 유전자가 검출되었다. 균주의 항균제 감수성 분석 결과 대부분의 항균제에 감수성을 보였으나, Ampicillin, Imipenem, Meropenem 및 Clindamycin 약제에는 내성을 보였다.
흑두청국으로부터 분리된 혈전용해력이 우수한 Bacillus subtilis BB-1(KFCC 11344P)으로부터 혈전용해효소 유전자를 PCR법에 의해 크로닝하였고 이를 BCF-1으로 명명하였다. BCF-1의 DNA 염기서열결정 결과 1,145 bp 크기의 혈전용해 효소로, 일본의 natto로부터 분리된 nattokinase 유전자와 99%의 상동성을 보임을 확인하였다. 혈전용해효소 유전자의 발현을 위하여 Bacillus 발현계인 Bacillus-E. coli의 shuttle vector인 pEB vector에 크로닝 하고 host로서 B. subtilis 168에 형질전환시켜 대량 발현시켰다. 생산된 혈전용해효소의 최 적활성 pH와 온도는 7.0과 $35^{\circ}C$로 확인되었다, 기질에 대한 분해양상을 조사한 결과 fibrin에서만 특이적으로 강한 분해가 일어났으며, skim milk에서 아주 약한 분해능을 보였으나 blood agar, gelatin, casein에서는 전혀 분해능을 보이지 않았다. 특히 blood agar plate에서 분해능이 없는 것으로 보아 혈액 내에서의 적혈구 파괴현상과 같은 부작용에 대한 위험을 배제할 수 있을 것으로 사료된다. BCF-1에 의해 생산된 혈전용해효소는 fibrin 특이적으로 활성을 나타냄을 확인할 수 있으며, 이는 임상적이나 산업적으로 적용하였을 때 부작용에 대한 위험적인 문제는 배제될 수 있으리라 생각된다.
본 논문은 임상검체 이외의 자연계내에서 extended-spectrum $\beta$-lactamase (ESBL) 생성균주의 분리 여부를 확인하고, 분리되었을 경우 이들의 유형을 조사하기 위함이었다. 하수종말처리장, 가물치양식장, 부경대학교 담수어양식장, 공중목욕탕으로부터 분리된 균주를 double disk synergy 검사, 교환접합시험, 등전점 조사 등을 실시하여 하수종말처리장 방류수로붙 extended-spectrum $\beta$-lactamase (ESBL) 생성균주가 26 균주 분리되었다. 균종은 Entero-bacter cloacae와 E. sakazakii, Klebsiella pneumoniae subsp. pneumoniae 및 K. pneumoniae subsp. ozaenae 였다. 이들은 모두 PCR결과 TEM+SHV 복합형이었고 등전점 결과 Klebsiella속의 균들은 pI 5.9, 5.9+5.4 E. cloacae는 $pI{\ge}8.5$, 8.0+5.4, E. sakazakii는 8.0+5.4, 8.0+5.9+5.4였다. 교차접합시험(transconjugation) 결과 피전달 균주인 E. coli RG176 nal$_{r}$에게 K. pneumoniae subsp. pneumoniae (1 균주) K. pneumoniae subsp. ozaenea (5 균주)및 E. cloacae (1 균주)의 $\beta$-lactamase 생성유전자가 전달되었다. 이 결과는 국내에서 임상검체 이외의 자연계에서 분리된 ESBL생성 장내세균의 첫 번째 보고이다.
계통적으로 근연하며 지리적 분포가 유사한 두 종의 Spirastrella 속의 해양 해면, S. panis와 S. abata의 배양 가능한 공생세균 군집구조를 16S rDNA-RFLP 방법에 의해 분석하였다. 공생세균의 배양은 해면추출물 3%를 포함하는 MA 배지를 사용하였다. 증폭된 16S rDNA의 RFLP (restriction fragment length polymorphism) 분석을 위한 제한효소로 HaeIII와 MspI을 이용하였으며, 그 결과 24개의 RFLP type을 구별할 수 있었다. 각 패턴별로 1~5개의 분리균주를 선별하여 부분 염기서열 분석 결과, 알려진 세균 종과 98.4% 이상의 유사도를 나타내었으며 2종의 Spirastrella 해면으로부터 분리된 세균들은 모두 Alphaproteobacteria, Gammaproteobacteria, Firmicutes, Actinobacteria 4개의 강(class)에 포함되었다. Alphaproteobacteria는 S. abata에서 39.3%, S. panis에서 47.6%가 관찰되어 두 해면에서 우점하는 세균 군집이었다. Gammaproteobacteria의 경우 S. abata에서 38.5%로 관찰된 반면 S. panis에서 1.6%의 아주 적은 비율로 관찰되었다. 또한 Bacillus (phylum Firmicutes) 종은 S. abata에서 9.7%를 나타낸 반면, S. panis에서는 44.3%의 분포를 나타내었다. Planococcus maritimus (8.1%, phylum Firmicutes)와 Psychrobacter nivimaris (28.9%, phylum Gammaproteobacteria)는 S. abata에서만 관찰되어 이들은 S. abata에 특이적인 세균 종임을 알 수 있었다. 같은 장소에 서식하는 계통적으로 근연한 두 종의 해면에서 공생세균의 군집 구조는 차이가 큰 것으로 나타났다.
