  • The purpose of this study was to develop molecular identification method for medical mushrooms and their preparations based on the nucleotide sequences of nuclear large subunit (LSD) rDNA. Four specimens were collected of each of the three representative medicinal mushrooms used in Korea: Ganoderma Incidum, Coriolus versicolor, and Fomes fomentarius. Fungal material used in these experiments included two different mycelial cultures and two different fruiting bodies from wild or cultivated mushrooms. The genomic DNA of mushrooms were extracted and 3 nuclear LSU rDNA fragments were amplified: set 1 for the 1.1-kb DNA fragment in the upstream region, set 2 for the 1.2-kb fragment in the middle, and set 3 for the 1.3-kb fragment downstream. The amplified gene products of nuclear large subunit rDNA from 3 different mushrooms were cloned into E. coli vector and subjected to nucleotide sequence determination. The sequence thus determined revealed that the gene sequences of the same medicinal mushroom species were more than $99.48\%$ homologous, and the consensus sequences of 3 different medicinal mushrooms were more than $97.80\%$ homologous. Restriction analysis revealed no useful restriction sites for 6-bp recognition enzymes for distinguishing the 3 sequences from one another, but some distinctive restriction patterns were recognized by the 4-bp recognition enzymes AccII and HhaI. This analysis was also confirmed by PCR-RFLP experiments on medicinal mushrooms.

  • Trichoderma species/isolates exhibited varied degree of agglutination on sclerotial (Sc) and hyphal (Hy) surface of Macrophomina phaseolina. The agglutination efficiencies on Sc and Hy ranged from $11\;to\;57\%$. Isolates of T. harzianum (Th) and T. viride (Tv) showed greater agglutination on Sc ($23-57\%$) and Hy ($16-47\%$). Different enzymes (trypsin, pepsin, proteinase k, a-chymotrypsin, lyticase and glucosidase) and inhibitors (tunicamycin, cycloheximide, brefeldin A, sodium azide, dithiothreitol and SDS) reduced the agglutination potential of conidia of Th-23/98 and Tv-25/98; however, the extent of response varied greatly in different treatments. Different fractions of Th-23/98 and Tv-25/98 exhibited haemagglutinating reaction with human blood group A, B, AB and O. Haemagglutinating activity was inhibited by different sugars and glycoproteins tested. Crude haemagglutinating protein from outer cell wall protein fraction of Th-23/98 and Tv-25/98 were eluted on Sephadex G-100 column. Initially Th-23/98 and Tv-25/98 exhibited two peaks showing no agglutination activity; however, lectin activity was detected in the third peak. Similar to crude lectin, the purified lectin also exhibited haemagglutinating activity with different erythrocyte source. SDS-PAGE analysis of partially purified lectin revealed single band with an estimated molecular mass of 55 and 52 kDa in Th-23/98 and Tv-25/98, respectively. Trypsin, chymotrypsin and b-1,3-glucanase totally inhibited lectin activity. Similarly, various pH also affected the haemagglutinating activity of Th-23/98 and Tv-25/98. From the present observations, it can be concluded that the recognition/attachment of mycoparasite (T. harzianum and T. viride) to the host surface (M. phaseolina) may be most likely due to lectin-carbohydrate interaction.

  • Based on its geophysical feature, the island has long been recognized as a separate and self-sustaining space independent of neighboring continent or other islands. Literary tradition has used the island as a metaphor for a utopian alternative to mundane human society with its various kinds of wrongdoings. Recent nature writings have taken up this island metaphor to emphasize the wholeness of the ecosystem in specifically designated natural community or landscapes such as national parks or wilderness preservation areas. Human-nature relations as border-divided area is also recognized as the island. Modern island biogeography, however, has disproved such a concept of islands as autonomous, revealing the contrasting fact that the richness of species on an undisturbed island is determined largely by species immigration from and emigration to a source of colonists. This scientific finding has posited the island as the interconnected nature, but the public and metaphoric use of it still resorts to the old concept of it as isolated and autonomous nature, because this image has been ingrained deeply in our consciousness and culture. Considering the negative consequences from the recognition of nature and nature-humans as isolated space, we need a new nature metaphor that embodies interconnectedness in nature and of human-nature relations. Such feature of interconnectedness is best embedded in the concept of ecotone. Some ecotones are created and maintained through human participation in nature, and this human induced nature of ecotone denotes the possibilities of a constructive relation between them. The substitution of the island with the ecotone as the concept of nature and the image of human-nature relations is expected to correct ecocritical practices of reading of nature writing, which has been predominantly interpreted within the orientation of nature itself and nature-human relations as an isolated and self-autonomous island. Adopting the ecotone in literary study enables ecocriticism to dig out cultural elements embedded in nature writing and reveal socio-political, ideological factors hidden behind the writers' portrayal of nature as islands.

  • This paper proposes a decision method of fuzzy weights by utilizing degrees of contribution in order to classify insect footprint patterns having difficulties to classify species clearly. Insect footprints revealed delicately in the form of scattered spots since they are very small. Therefore it is not easy to define shape of footprints unlike other species, and there are lots of noises in the footprint patterns so that it is difficult to distinguish those from correct data. For these reasons, the extracted feature set has obvious feature values with some uncertain feature values, so we estimate weights according to degrees of contribution. If the one of feature values has distinct difference enough to decide a class among other classes, high weight is assigned to make classification. A calculated weight determines the membership values by fuzzy functions and objects are classified into the class having a superior value.atu present experimental resultseighrontribution. • We conducted the study on the relationship between the distribution of coastal sand dune plants and edaphic factors from the shoreline to inland in sand dune ecosystem. The application of TWINSPAN classification based on 10 species, led to the recognition of three vegetative groups (A-C), which associated with their habitats (foredune, hummuck in semistable zone and stable zone). The associations were separated along soil gradient far from the seashore. The relationships between species composition and environmental gradients were explained by canonical correspondence analysis (CCA). Distance from the shoreline was an important indicator to determine soil properties (pH, total ion contents, sand particle sizes, organic matters and nitrogen contents) from the seaward area to inland area and distribution pattern of coastal sand dune plants. Group A is foredune zone, characterized by Calystegia soldanella; group included typical foredune species such as Elymus mollis, Carex kobomugi, Ixeris repens, C. soldanella and Glehnia littoralis. Group B on semi-stabilized zone was characterized by Vitex rotundifolia, a perennial woody shrub. This group was associated the proportion of fine sand size (100 to 250 μm). The results on the proportion of soil particle size showed a transition in sand composition, particularly with respect to the proportion of fine sand size that occurred from the foredune ridge at 32.5 m to the Vitex rotundifolia community at 57.5 m from the shoreline. Group C on stabilized zone was characterized by Zoysia macrostachya, Lathyrus japonicus and Cynodon dactylon and were associated soil organic matter and nitrogen contents. The spatial distribution of plants in the Goraebul coastal sand dune system may result from the interactions between the plant species and environmental heterogeneity.

  • We propose a PfSGA(parameter-free species genetic algorithm) to learn the topological structure of MLP classifiers being adequate to given applications. The PfSGA is a combinational method of SGA(species genetic algorithm) and PfGA(parameter-free genetic algorithm). In SGA, we divide the total search space into several subspaces(species) according to the number of hidden units, and reduce the unnecessary search by eliminating the low promising species from the evolutionary process. However the performances of SGA classifiers are readily affected by the values of parameters such as mutation ratio and crossover ratio. In this paper, therefore, we combine SGA with PfGA, for which it is not necessary to determine the learning parameters. Experimental results on benchmark data and sign language words show that PfSGA can reduce the learning time of SGA and is not affected by the selection parameter values on structural learning. The results also show that PfSGA is more efficient than the exisiting methods in the aspect of misclassification ratio, learning rate, and complexity of MLP structure.

  • To determine whether pattern recognition based on metabolite fingerprinting for whole cell extracts can be used to discriminate cultivars metabolically, leaves of nine commercial Orostachys plants were subjected to Fourier transform infrared spectroscopy (FT-IR). FT-IR spectral data from leaves were analyzed by principal component analysis (PCA) and Partial least square discriminant analysis (PLS-DA). The dendrogram based on hierarchical clustering analysis of these PLS-DA data separated the nine Orostachys species into five major groups. The first group consisted of O. iwarenge 'Yimge', 'Jeju', 'Jeongsun' and O. margaritifolius 'Jinju' whereas in the second group, 'Sacheon' was clustered with 'Busan,' both of which belong to O. malacophylla species. However, 'Samchuk', belong to O. malacophylla was not clustered with the other O. malacophylla species. In addition, O. minuta and O. japonica were separated to the other Orostachys plants. Thus we suggested that the hierarchical dendrogram based on PLS-DA of FT-IR spectral data from leaves represented the most probable chemotaxonomical relationship between commercial Orostachys plants. Furthermore these metabolic discrimination systems could be applied for reestablishment of precise taxonomic classification of commercial Orostachys plants.

  • The hornet species are so similar in shape that they are difficult for non-experts to classify, and because the size of the objects is small and move fast, it is more difficult to detect and classify the species in real time. In this paper, we developed a system that classifies hornets species in real time based on a deep learning algorithm using a boundary box. In order to minimize the background area included in the bounding box when labeling the training image, we propose a method of selecting only the head and body of the hornet. It also experimentally compares existing boundary box-based object recognition algorithms to find the best algorithms that can detect wasps in real time and classify their species. As a result of the experiment, when the mish function was applied as the activation function of the convolution layer and the hornet images were tested using the YOLOv4 model with the Spatial Attention Module (SAM) applied before the object detection block, the average precision was 97.89% and the average recall was 98.69%.

  • Hypoxia-inducible factor $1{\alpha}\;(HIF1{\alpha})$ is a transcription factor that plays a key role in the adaptation of cells to low oxygen stress and oxygen homeostasis. The oxygen-dependent degradation (ODD) domain of $HIF1{\alpha}$ is responsible for the negative regulation of $HIF1{\alpha}$ in normoxia. The interactions of the $HIF1{\alpha}$ ODD domain with partner proteins such as von Hippel-Lindau tumor suppressor (pVHL) and p53 are mediated by two sequence motifs, the N- and C-terminal ODD(NODD and CODD). Multiple sequence alignment with $HIF1{\alpha}$ homologs from human, monkey, pig, rat, mouse, chicken, frog, and zebrafish has demonstrated that the NODD and CODD motifs have noticeably high conservation of the primary sequence across different species and isoforms. In this study, we carried out molecular dynamics simulation of the structure of the $HIF1{\alpha}$ CODD motif in complex with the p53 DNA-binding domain (DBD). The structure reveals specific functional roles of highly conserved residues in the CODD sequence motif of $HIF1{\alpha}$ for the recognition of p53.

  • We investigated the occurrence of the dinoflagellate genus Alexandrium in the nineteen Korean coastal sites from July to October 2013. Alexandrium-like planktonic cells were microscopically observed only in four out of the 19 sampling sites. From the samples containing Alexandrium-like cells 22 clonal cultures of Alexandrium species were established by single cell or single chain isolation method. Taxonomic identity of the 4 different strains ascertained by the robust analyses of morphological and molecular genetic characteristics were confirmed to be A. catenella, A. affine, A. fraterculus and an unidentified Alexandrium sp. for which strain WEB-Alex-01 was assigned. It was ascertained that in spite of hot summer diverse Alexandrium species attaining up to four were distributed in the study area, in contrast with the long empirical recognition that the emergence of Alexandrium species is restricted to cooler seasons like spring or autumn in Korean coastal waters. Morphology and genetic characteristics of Alexandrium sp. strain WEB-Alex-01 are different from any other previously reported Alexandrium species from Korean seas, which implies that further studies on taxonomic, physiological, ecological and toxicological properties of the newly recorded Alexandrium species are needed.
