Assessing persistence of cruciferous crops in the field

  • We assessed the persistence of eight major cruciferous crops-leaf mustard, oilseed rape, cabbage, broccoli, cauliflower, Chinese cabbage, turnip, and radish-growing in the field. In the first part of our experiment, we tested the viability of seeds that had been buried at two different soil depths for up to 16 months. We then broadcast seeds over the soil surface and left them undisturbed to investigate the survivorship of the resultant plants over two years. Seed viability was significantly affected by plant taxa and burial depth, but not substantially affected by the duration of burial. Although seeds of leaf mustard had the greatest viability among all crops examined here, the viability rates were significantly lower at 2 cm depth than at 15 cm. Seeds of leaf mustard, oilseed rape, broccoli, turnip, and Chinese cabbage remained viable throughout the 16-month period. A study of plant demography revealed that only leaf mustard and oilseed rape succeeded in producing seeds and overwintering in the undisturbed field. However, neither of those species competed well with other plants long-term and their overall growth and survival rates declined during the evaluation period. In addition, insect herbivory severely decreased the growth of all of these crops. Our results suggest that populations of leaf mustard and oilseed rape do not tend to persist in the field for more than a few years without disturbance and external seed inputs.

Lactobacillus Persisters Formation and Resuscitation

  • Lactobacillus is a commonly used probiotic, and many researchers have focused on its stress response to improve its functionality and survival. However, studies on persister cells, dormant cells that aid bacteria in surviving general stress, have focused on pathogenic bacteria that cause infection, not Lactobacillus. Thus, understanding Lactobacillus persister cells will provide essential clues for understanding how Lactobacillus survives and maintains its function under various environmental conditions. We treated Lactobacillus strains with various antibiotics to determine the conditions required for persister formation using kill curves and transmission electron microscopy. In addition, we observed the resuscitation patterns of persister cells using single-cell analysis. Our results show that Lactobacillus creates a small population of persister cells (0.0001-1% of the bacterial population) in response to beta-lactam antibiotics such as ampicillin and amoxicillin. Moreover, only around 0.5-1% of persister cells are heterogeneously resuscitated by adding fresh media; the characteristics are typical of persister cells. This study provides a method for forming and verifying the persistence of Lactobacillus and demonstrates that antibiotic-induced Lactobacillus persister cells show characteristics of dormancy, sensitivity of antibiotics, same as exponential cells, multi-drug tolerance, and resuscitation, which are characteristics of general persister cells. This study suggests that the mechanisms of formation and resuscitation may vary depending on the characteristics, such as the membrane structure of the bacterial species.

Allozyme Diversity and Population Genetic Structure in Korean Endemic Plant Species : II. Hosta yingeri (Liliaceae)

  • Levels of genetic diversity, population genetic structure, and gene flow in Hosta yingeri, a herbaceous perennial endemic to Taehuksan, Sohuksan, and Hong Islands, were investigated. Starch gel electrophoresis was conducted on leaves for 101 plants collected from three populations. Although the distribution of thespecies is restricted in the islands, it maintains high levels of genetic variatin; 64% of polymorphic loci in at least one population (Ps), the mean number of alleles per locus (Ap) of 1.92, and the mean effective number of alleles per locus (Aep) of 1.52. Overall, mean genetic diversity (Hep=0.250) was substantially higher than mean estimate for species with very similarlife history traits (0.102). Large populaton size, the persistence of multiple generations within populations, high fecundity, predominantly outcrossing breeding system, large size of pollinator visitation areas may be explanatory factors contributing the higher level of genetic diversity maintained within populations. Analysis of fixation indices showed an overall slight excess of heterozygotes (mean FIS=-0.066) relative to Hardy-Weinberg expectations, which may in part be due to the near self-incompatible breeding system in the species. Significant differences in allele frequencies among populaitns were found for 14 out of 16 polymorphic loci (P<0.05). Slightly more than 80% of the total variation in the species was common to all populations (GST=0.198). As expected, indirect estimate of the number of migrants per generation (Nm=0.45, calculated from mean GST) and nine private alleles found in the three populations indicate that gene movement among three isolated island populations was low.

Geranium purpureum Vill.: A new casual alien plant in Korea

  • Naturalization centers are hotspots where accidently introduced seeds first germinate and grow. Railways play important roles as corridors for movement, having positive effects on species dispersal and persistence, though they can also be invasion routes for undesirable plants. Geranium purpureum Vill., a new alien species in Korea, was found at the Bumil Railway Station, Busan, Gyeongsangnam-do, Korea. Most native Geranium species in Korea belong to the subgenus Geranium and section Geranium, but G. purpureum belongs to the subgenus Robertium and section Ruberta, with the "carpel-projection" method of seed dispersal. In other countries, G. purpureum is often compared with G. robertianum L., which is an ornamental plant in Korea. We assume that G. purpureum is a casual alien plant and that it is not likely to spread to other provinces, as it scarcely survives when the mean temperature in January is below $4^{\circ}C$. Moreover, it has not been found further north than Busan. We provide a key of allied taxa, information about the habitat, a description of the morphological characters, illustrations, and photographs.

Valuation of Forest Habitat Functions of Endangered Mammals Using Species Distribution Model

  • It is estimated that there is a total of approximately 100,000 species in Korea. However, the number is currently about 30,000 and only 16,027 species are listed in the 'Species Korea' (as of December, 2014). Of the listed species, 51 species are designated as the Endangered Species Class I while 195 species are in the Class II, totaling 246 endangered species including 20 mammals. Under the circumstances that development (e.g., roads) is increasingly threatening the persistence of endangered mammals, it is significant to identify and preserve suitable habitats for them. In this context, evaluating the values of the suitable habitat environment would serve as essential information for development decision making. This study estimated the values of endangered mammals' forest habitats through spatialization of habitat services. In doing so, a species distribution model, Maximum Entropy Model (MaxEnt) was utilized for a group of endangered mammals including, mountain goat, wildcat, marten cat, and flying squirrel. To calculate the values per unit area, a benefit transfer method was used based on the point-estimate technique with the best available values estimated previously. The range of discount rate of 3.0 to 5.5 percent was applied taking the notion of social discount rate into account. As a result, the province with the highest values for endangered mammal habitats appeared to be Gangwon, followed by Gyeongbuk and Gyeongnam. The monetary values of the endangered mammal habitats were estimated to be 330 billion to 421 billion won per year.

Susceptibility of Laboratory Rodents to Trichinella papuae

  • Members of the genus Trichinella are small nematodes that can infect a wide range of animal hosts. However, their infectivity varies depending on the parasite and host species combination. In this study, we examined the susceptibility of 4 species of laboratory rodents, i.e., mice, rats, hamsters, and gerbils to Trichinella papuae, an emerging non-encapsulated Trichinella species. Trichinella spiralis and Trichinella pseudospiralis were also included in this study for comparison. Fifteen animals of each rodent species were infected orally with 100 muscle larvae of each Trichinella species. Intestinal worm burden was determined at day 6 and 10 post-inoculation (PI). The numbers of muscle larvae were examined at day 45 PI. The reproductive capacity index (RCI) of the 3 Trichinella species in different rodent hosts was determined. By day 6 PI, 33.2-69.6% of the inoculated larvae of the 3 Trichinella species became adult worms in the small intestines of the host animals. However, in rats, more than 96% of adult worms of all 3 Trichinella species were expelled from the gut by day 10 PI. In gerbils, only 4.8-18.1% of adult worms were expelled by day 10 PI. In accordance with the intestinal worm burden and the persistence of adults, the RCI was the highest in gerbils with values of $241.5{\pm}41.0$ for T. papuae, $432.6{\pm}48$ for T. pseudospiralis, and $528.6{\pm}20.6$ for T. spiralis. Hamsters ranked second and mice ranked third in susceptibility in terms of the RCI, Rats yielded the lowest parasite RCI for all 3 Trichinella species. Gerbils may be an alternative laboratory animal for isolation and maintenance of Trichinella spp.

Primary Production System in the Southern Waters of the East Sea, Korea III. Vertical Distribution of the Phytoplankton in Relation to Chlorophyll Maximum Layer

  • Vertical profiles of the chlorophyll ${\alpha}$, phytoplankton abundance, nutrients and sigma-t were compared with the vertical distribution of phytoplankton species in conjunction with $^{14}$C primary production in the Southern Waters of the East Sea, Korea. In the upper mixed layer the water column was only weakly stratified and ambient nitrogenic nutrient concentrations were markedly depleted. Dissolved silicate seemed to be another limiting nutrient in the surface layer. The occupation of different water depths by several dominant diatom species was well explained by the degree of silicification of each cell and the silicate concentration of ambient seawater. Subsurface chlorophyll maxima were continuously observed in the lower parts of the euphotic layer and the depth coincided with nutricline, supporting our view that chlorophyll maximum was sustained partially by enhancement of in situ growth of phytoplankton and partially by increase of cellular chlorophyll content. The persistence of chlorophyll maximum layer was attributed to the physiological adaptation of the phytoplankters to low light intensities and to the utilization of regenerated nutrients. Integrated water column production of organic matter by photosynthesis appeared to be better related to phytoplankton cell division than to the cell growth in terms of biosynthesis of pigments and other intracellular components.

국내 농업용수의 유해중금속 수질권고기준 도출 (Derivation of Agricultural Water Quality Guidelines for Heavy Metals in Korea)

  • Korean water quality guidelines for agricultural were derived to protect agricultural water uses according to the Canadian methodology. The adverse effect of heavy metals in agricultural water was of concern due to its persistence, bioaccumulation and ecotoxicity to crop plants. The ecotoxicity data of cadmium, copper, lead, and zinc were collected for the crops grown in Korea, and used to estimate the species maximum acceptable toxicant concentration (SMATC), which corresponds to the water quality guideline. Values of irrigation per year and soil bulk density were revised to reflect the Korean situation. The estimated guideline values for cadmium, copper, lead, and zinc were 0.01, 0.5, 0.1, and 1.0 mg/L, respectively. These values are in agreement with the agricultural water quality guidelines of foreign countries and current Korean water quality standard for the protection of public health. Current water quality standards in agricultural uses were for management of public water resource, and was not prepared to protect crop plants from contaminants. The results of this study will be a basis for the designation of Korean water quality guidelines for the protection of agricultural water uses in the future.

Comparison of the cultivation performance between Korean (Sugwawon No. 301) and Chinese strains (Huangguan No. 1) of kelp Saccharina japonica in an aquaculture farm in Korea

  • Saccharina japonica was introduced to both Korea and China from Hokkaido, Japan, and it has become an economically important species in both nations. After a long period of cultivation, several varieties of S. japonica have been developed in Korea and China. In this study, we conducted aquacultural research on the persistence of thalli from two kelp cultivars, one from China (Huangguan No. 1) and one from Korea (Sugwawon No. 301), between December 2015 and November 2016 in Haenam, Korea. The maximum length was $247.8{\pm}13.0$ and $227.5{\pm}42.0cm$, respectively, which were significantly longer in Sugwawon No. 301 than in Huangguan No. 1. The maximum width was $29.9{\pm}5.4$ and $23.2{\pm}1.9cm$, respectively, which were significantly wider in Huangguan No. 1 than in Sugwawon No. 301. The mean biomass obtained from the culture ropes was for Sugwawon No. 301 was $3.5{\pm}0.3kg\;wet\;wt\;m^{-1}$ and for while Huangguan No. 1 was $3.1{\pm}1.0kg\;wet\;wt\;m^{-1}$ of culture rope. After August, the persistence of the thalli of Sugwawon No. 301 was two months longer than that of Huangguan No. 1. We found that the Sugwawon No. 301 performed as well as the Huangguan No. 1 in Korean waters possibly due to increased flexibility as a result of the different cell arrangements of the two cultivars. Overall, the use of the Sugwawon No. 301 cultivar rather than the Huangguan No. 1 cultivar of S. japonica appears the best alternative to help to ensure a stable year round algal feed supply for the Korean abalone industry.

ppGpp: Stringent Response and Survival

  • Adaptation to any undesirable change in the environment dictates the survivability of many microorganisms, with such changes generating a quick and suitable response, which guides the physiology of bacteria. During nutritional deprivation, bacteria show a stringent response, as characterized by the accumulation of (p)ppGpp, resulting in the repression of stable RNA species, such as rRNA and tRNA, with a concomitant change in colony morphology. However, genes involved in amino acid biosynthesis become over-expressed to help bacteria survive under such conditions. The survivability of pathogenic bacteria inside a host cell also depends upon the stringent response demonstrated. Therefore, an understanding of the physiology of stringent conditions becomes very interesting in regulation of the growth and persistence of such invading pathogens.
