Component Content, Antioxidant and ACE Inhibitory Activity of Ramie (Leaf Boehmeria nivea M.) according to Breeding Lines (모싯잎의 육성 계통별 성분 함량, 항산화 및 ACE 저해 활성)

  • Ramie (Boehmeria nivea M.) has been used for fiber materials in Korea traditionally, but in recent years, the concern with ramie leaves for the food industry such as tteok (a kind of Korean rice cake) industry has been increasing, so a study for eatable ramie is required for the expansion of ramie consumption. Moreover, the ramie varieties for the food industry are not established, so the natural species are cultivated in general; therefore, it is very important to select the ramie varieties for the food industry such as rice cakes, tea, beverage and so on. This study was undertaken to compare the physiochemical properties among 9 ramie lines selected in the Yeonggwang-gun Agricultural Technology Center to select the eatable ramie varieties for the food industry. The contents of the protein among 9 ramie lines was 6.21~7.56% and had the highest content in the YG55. The folic acid (folate) and vitamin C content had varying differences among the 9 lines; the content of folate showed $771.52{\sim}1,978.84{\mu}g%$, that of vitamin C showed 149.42~275.34 mg%. The ACE inhibitory activity appeared to be the highest in YG88 (21.5%) among the 9 ramie varieties tested.

Difference in Macrobenthic Community Structures at Thermal Effluent Discharge Areas of Two Nuclear Power Plants in Korea (원자력발전소의 온배수 배출해역에서 대형 저서동물 군집구조의 차이)

  • This study investigated and compared sediment properties and macrobenthic community structures within heated effluent plumes at the discharge areas of Kori Nuclear Power Plant (KNPP) and Sinkori Nuclear Power Plant (SNPP) in Korea, which have different thermal effluent discharge systems. There were significant differences in sand, clay and organic carbon contents between sediments at the two discharge areas. Species richness and abundance of macrobenthos were higher at the SNPP discharge area than at that of the KNPP, although the values at both areas were comparable to previous studies in coastal areas of eastern Korea.

  • Cloud droplet activation process is well described by $K{\ddot{o}}hler$ theory and several parameterizations based on $K{\ddot{o}}hler$ theory are used in a wide range of models to represent this process. Here, we test the two different method of calculating the solute effect in the $K{\ddot{o}}hler$ equation, i.e., osmotic coefficient method (OSM) and ${\kappa}-K{\ddot{o}}hler$ method (KK). To do that, each method is implemented in the cloud droplet activation parameterization module of WRF-CHEM (Weather Research and Forecasting model coupled with Chemistry) model. It is assumed that aerosols are composed of five major components (i.e., sulfate, organic matter, black carbon, mineral dust, and sea salt). Both methods calculate similar representative hygroscopicity parameter values of 0.2~0.3 over the land, and 0.6~0.7 over the ocean, which are close to estimated values in previous studies. Simulated precipitation, and meteorological variables (i.e., specific heat and temperature) show good agreement with reanalysis. Spatial patterns of precipitation and liquid water path from model results and satellite data show similarity in general, but on regional scale spatial patterns and intensity show some discrepancy. However, meteorological variables, precipitation, and liquid water path do not show significant differences between OSM and KK simulations. So we suggest that the relatively simple KK method can be a good alternative to the OSM method that requires various information of density, molecular weight and dissociation number of each individual species in calculating the solute effect.

Population Genetic Structure of Potentilla discolor Bunge, Rosaceae in Korea (한국내 솜양지꽃의 집단 유전 구조)

  • The genetic diversity and population structure of fifteen Potentilla discolor Bunge populations in Korea were determined using genetic variations at 19 allozyme loci. Fourteen of the 19 loci (73.7%) showed detectable polymorphism. Genetic diversity at the species level and at the population level was high ($H_{ES}\;=\;0.215$, $H_{EP}\;=\;0.196$, respectively), whereas the extent of the population divergence was relatively low $(G_{ST}\;=\;0.069)$. Total genetic diversity values $(H_T)$ varied between 0.0 and 0.656, giving an average overall polymorphic loci of 0.292. The interlocus variation of genetic diversity within populations $(H_S)$ was high (0.274). On a per locus basis, the proportion of total genetic variation due to differences among populations $(G_{ST})$ ranged from 0.010 for Pgm-2 to 0.261 for Pgd-2 with a mean of 0.069, indicating that about 6.9% of the total allozyme variation was among populations. Wide geographic ranges, perennial herbaceous nature and the persistence of multiple generations are associated with the high level of genetic variation in P. discolor. The estimate of gene flow based on $G_{ST}$ was high among Korean populations of P. discolor (Nm = 3.36).

Identification and Phylogenetic Relationship at Cytochrome Oxidase Subunit I (COI) Gene among Korean Terrestrial Planarian Taxa (한국 내 육지플라나리아 간 치토크롬 산화효소의 동정과 계통유전학적 관계)

  • Sequence data of Cytochrome Oxidase Subunit I (COI) gene of mitochondria were used to elucidate the taxonomy and phylogenetic relationships of the terrestrial planarian taxa in Korea. Published COI gene sequences from Family Bipaliidae in GenBank were also included in the phylogenetic analysis. The aligned data sets for Terricola ranged from 387 to 444 nucleotides (bp) as a result of differences in insert nucleotides. The phylogeny based on COI analysis was not congruenced with the morphological traits. Bipalium nobile included the remainder taxa (Bipalium adventitium, Bipalium venosum, Bipalium kewense, and Bipalium multilineatum). Internal nodes were strongly supported (>91%). The phylogenetic tree on COI analysis showed that most identified species were well separated from each other. The main phylogenetic analysis formed monophyletic groups. COI gene of mitochondria could have the resolving power for taxonomy information for the terrestrial planarian taxa in Korea.

Analysis of Rutin Contents in Buckwheat and Buckwheat Foods (메밀 및 메밀식품에서의 루틴함량의 분석)

  • In this study, the rutin contents in buckwheat and buckwheat foods were determined. Rutin in buckwheat and buckwheat foods was extracted with methanol and separated by High Performance Liquid Chromatography(HPLC) equipped with Lambda-Max Model 481 detector set at 355 nm using a ${\mu}$ Bondapak $C_{18}$ column and a 2.5% acetic acid:methanol:acetonitrile (35:5:10, v:v:v) solvent. There were differences in the rutin contents among the different species of buckwheat. The rutin contents in buckwheat groats were ranged from 8.84 mg to 24.77 mg/100g. The rutin contents in commercial buckwheat groats and flours were ranged from 15.04 mg to 20.92 mg/100g. The rutin contents in commercial dried buckwheat noodles, steamed buckwheat noodles, and buckwheat cookies were ranged from 1.76 mg to 10.84 mg/100g.

Contents of Free-Sugars & Free-Sugaralcohols in Pleurotus ostreatus, Lentinus edods & Agaricus bisporus (느타리버섯, 표고버섯 및 양송이의 유리당(遊離糖)과 당(糖)알코올 조성)

  • Compositions of free-sugars and sugar-alcohols in three cultivated mushrooms, Pleurotus ostreatus, Lentinus edodes and Agaricus bisporus were analyzed by gas-liquid chromatography and compositional differences of these components depending on species, sizes of fruiting body and portions(pileus and stipe) were investigated. Glucose, fructose, trehalose, glycerol, arabitol and mannitol were identified in each of the three mushrooms. The major components were mannitol and arabitol in Lentinus edodes, mannitol in Agaricus bisporus, trehalose and mannitol in Pleurotus ostreatus. The total free-sugars and sugar-alcohols content were in the range of 12.7-18.9% in Lentinus edodes, 9.2-16.1% in Agaricus bisporus and 8.2-10.6% in Pleurotus ostreatus. Total contents of these components in Pleurotus ostreatus and Agaricus bisporus were higher in pileus than those in stipe and it was decreased as pileus size development in all of three edible mushrooms.

Reanalysis of Ohno's hypothesis on conservation of the size of the X chromosome in mammals

  • In 1964, Susumu Ohno, an evolutionary biologist, hypothesized that the size of X chromosome was conserved in mammalian evolution, and that this was based on chromosomal length. Today, unlike Ohno's method which was based on estimated lengths, we know the exact lengths of some mammalian sequences. The aim of this study was to reanalyze Ohno's hypothesis. In mammalian species, variation in the length of the X chromosome is greater than in the autosomes; however, this variation is not statistically significant. This means that differences in chromosomal length occur equally in the X chromosome and in the autosomes. Interspersed nuclear elements and genetic rearrangements were analyzed to maintain the same variance between the length of the X chromosome and the autosomes. The X chromosome contained fewer short interspersed elements (SINEs) (0.90 on average); however, it did contain more long interspersed elements (LINEs) than did autosomes (1.56 on average). An overall correlation of LINEs and SINEs with genetic rearrangements was observed; however, synteny breaks were more closely associated with LINEs in the autosomes, and with SINEs in the X chromosome. These results suggest that the chromosome-specific activities of LINEs and SINEs result in the same variance between the lengths of the X chromosome and the autosomes. This is based on the function of interspersed nuclear elements, such as LINEs, which can inactivate the X chromosome and the reliance of non-autonomous SINEs on LINEs for transposition.

The Effects of Mycoplasma Infection on Semen Parameters (in Normal Human Semen) (남성에서 Mycoplasma 감염이 정액검사소견에 미치는 영향에 관한 연구)

  • Mycoplasmas have long been suspected of contributing to involuntary infertility in couples. However considerable disagreement exits concerning the role of genital mycoplasma infection in human infertility. Several investigators have noted abnormalities in the semen analysis of men with positive mycoplasma cultures, and early epidemiologic studies indicated that Ureaplasma urealyticum was linked to human reproductive failure on the basis of higher frequencies of isolation from infertile versus fertile couples and successful pregnancies in infertile couples after doxycycline therapy. However, subsequent investigators have questioned these findings because there are many studies in which treatment for mycoplasma in the male or female did not demonstrate an improved pregnancy rate, and semen samples from unexplained infertile men containing ureaplasmas have not revealed poorer motility, fewer spermatozoa and more aberrant forms. The objective of this study were to investigate the incidence rate of mycoplasma in semen and to investigate whether the presence of mycoplasma in semen makes significant difference to the semen volume, sperm motility and sperm counts. The results were that the rate of isolation of mycoplasma species was 70.3%. Semen volume is $2.84{\pm}1.01ml$ for culture negative and $3.15{\pm}1.42ml$ for culture positive group. Sperm motility is $46.23{\pm}15.80%$ for culture negative and $50.09{\pm}15.69%$ for culture positive group, and sperm count is $95.47{\pm}47.14({\times}(P)10^6/ml)$ for culture negative and $86.73{\pm}47.59({\times}10^6/ml)$ for culture positive group. In conclusion, we suggest that the presence of mycoplasma in semen makes no significant differences to the sperm parameters.

Yeasts Associated with Fruits and Blossoms Collected from Hanbat Arboretum, Daejeon, Korea (한밭 수목원의 과일과 꽃으로부터 효모의 분리)

  • Yeasts are common inhabitants of the phyllosphere, but our knowledge of their diversity in various fruits and blossoms are limited. We collected different kinds of blossoms and unripened fruits from Hanbat arboretum, Daejeon, Korea at the year of 2013. Yeasts were isolated by plating of suspensions prepared for collected samples onto YPD medium containing antibiotics. BLAST searches were subsequently performed for the comparison of the partially determined sequences of D1/D2 domain of 26S rDNA. As a result, we isolated 57 yeast strains of 31 species from 29 different kinds of blossoms and 6 kinds of fruits samples. We found huge differences in yeast flora depending on the sample collection season.
