A total of 191 samples collected from the commercial swine farms located in Chungnam province were investigated by PCR to estimate the prevalence of Lawsonia (L.) intracellularis infection. In the group of the pigs with proliferative enteritis, 14 (93.3%) of 15 intestinal samples and 12 (80.0%) of 15 feces were positive in PCR. In contrast, a relatively low positive rate (18.0%, 29 of 161 samples) was determined in the group of normal healthy pigs. The group of pigs over 120 days showed the highest positive rates (26.8%, 15 of 56 samples). In the comparison of the sequences of 210bp for species specific fragments and 301bp for outer membrane protein, the isolates (L1. L2) showed almost 100% identity with the reference L. intracellularis (L08049, USA). For the sequences of partial 16s rDNA, the homologies among the 5 isolates (L1-L5) were 97.4% to 99.3%, and those of 5 sequences (L1-L5) versus 5 overseas reference strains of L. intracellularis ranged from 98.6% to 99.8%. In the comparison of the nucleotide sequences among 5 isolates and other species in Desulfovibrionales showed 82.4 to 99.5% identities. The 5 isolates shared relatively low identities (76.9% to 84.4%) with the species of alpha-proteobacteria. In phylogenetic analysis based on the 16s rDNA sequences, all of the 5 isolates (L1-L5) were located in the same branch with the strains of L. intracellularis that were previously isolated from the pigs in USA and China. Seven strains of Desulfovibrio sp. were clustered in the neighboring branches, whereas alpha and gamma Proteobacteria showed distant relationship with L. intracellularis strains. The present findings suggest that L. intracellularis infection is endemic in the swine farms in the regions, and that the domestic isolates maintained very limited genetic variation.
P. linteus (Korea Sanghwang, PLKS) and P. baumii (Jangsoo Sangwhang, PBJS) were artificially cultivated under the same conditions. Fruiting bodies were extracted using 70% methanol, 60% ethanol, and hot water. Phenol and flavonoid contents were optimally extracted using 60% ethanol. Antioxidant activities of 60% ethanol extracts from fruiting bodies and mycelia from each Sanghwang mushroom species were measured using DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,2'-azino-bis (3-ethylbisthiazoline-6-sulfonic acid) radical scavenging activities, and FRAP (ferric reducing antioxidant power). The antioxidant activities of fruiting bodies were relatively higher in comparison to those of mycelial samples. In high performance liquid chromatography (HPLC) analysis, styrylpyrone-class poly phenolic compounds, davallialactone, hispidin, hypholomine B, and inoscavin A were detected in fruiting body samples of two Sanghwang mushroom species, but not in their mycelial samples.

The Minke whale, Balaenoptera acutorostrata, is the smallest baleen whale in the suborder Mysticeti. Because this species inhabits coastal areas, it became a main target species of coastal small-type whaling in the North Atlantic and the Northwest Pacific Oceans, and the species' population size dramatically decreased because of over-exploitation. As a result, the International Whaling Commission declared a global moratorium on whaling and launched the development of a management procedure for protecting the whales. Morphological studies, whaling history analysis, and genetic studies conducted mainly by Japanese scientists showed the existence of one unique "E" stock that inhabits the waters around the Korean peninsula and mixes with the "O" stock in the southern part of the Sea of Okhotsk. We used the mitochondrial DNA control region polymorphism of 348 Minke whales bycaught or stranded in Korean waters from 30 October 1998 to 25 June 2005 to assess the whale population structure by year. The frequency of the 10 major haplotypes from the 40 identified haplotypes was not significantly different among groups, suggesting that a subpopulation was not present. A comparison of the genetic distances calculated with Tamura-Nei's method showed that the distances between groups were lower than those within groups, which suggests that there was no genetic difference in the Minke whale populations. The Fst comparison between groups and the phylogenetic tree constructed using the unweighted pair group method with arithmetic mean (UPGMA) and Neighbor Joining (NJ) method also detected no obvious sub-stock structure.
Objective : In this study, we performed quantitative comparison on the content of 10 marker compounds in cinnamon barks from different species and found chemical discrimination between genuine Cinnamomum cassia and other Cinnamomum species (Non C. cassia). Methods : Cinnamon bark samples were extracted using the ultrasonication in 100% methanol for 30 minutes. The samples were analysed using high-performance liquid chromatography with statistical analysis. Results : The analytical method developed in this study met all validation criteria and was applied to the quantification of the 10 marker compounds in cinnamon bark samples. The major chemical discrimination of C. cassia were identified as low content of epicatechin and eugenol, and high contents of benzaldehyde, cinnamaldehyde and cinnamic acid compared to other Non C. cassia samples. Especially, among other compounds, the content of cinnamaldehyde was the highest in the C. cassia and Non C. cassia samples. The result of principal component analysis showed that the samples of C. cassia and Non C. cassia were clearly differentiated via benzaldehyde, cinnamaldehyde, cinnamic acid, eugenol, and epicatechin, which influenced on clustering C. cassia and Non C. cassia samples. Conclusion : C. cassia and Non C. cassia samples were chemically discriminated using the quantitative HPLC analysis. Based on this, it is possible to control the quality of herbal medicines containing Cinnamomi Cortex. It is necessary to further improve the accuracy of discrimination between C. cassia and Non C. cassia species to evaluate cinnamon bark quality.
This study surveyed Gyeongan Stream, a tributary of the Han River Drainage System, from April to October 2017 to investigate the characteristics of fish communities. The survey collected 40 species of 11 families from 48 survey stations using kick nets and cast nets. The dominant and subdominant species were Zacco platypus (48.0%) and Rhynchocypris oxycephalus (17.7%), respectively. The next most abundant species were Pungtungia herzi (6.8%), Z. koreanus (5.0%), Carassius auratus (4.3%), Squalidus gracilis majimae (2.7%), and Rhodeus notatus (2.5%). Among the fish collected, 14 species (35.0%) were Korean endemic species, and the exotic species were Micropterus salmoides, Lepomis macrochirus, and Cyprinus carpio (Israeli type). The land-locked species were Plecoglossus altivelis, Rhinogobius brunneus, and Cottus koreanus, while the species sensitive to climate change was C. koreanus. The community analysis showed that the dominance was higher at the uppermost stream station, whereas diversity and abundance tended to be lower at the upstream station and higher toward the downstream station. The community structure was largely divided into rivers (uppermost stream, upstream, and middle-lower stream) and lake. The river health was mostly good (23 stations, 47.9%) and fair (15 stations, 31.3%). Comparison with past surveys showed that 12 species identified in the past surveys did not appear in this survey; nine species appeared for the first time in this survey; and the ecosystem disturbance species - M. salmoides and L. macrochirus - tended to spread more widely gradually.
Biodiversity holds significant importance within the framework of environmental impact assessment, being utilized in site selection for development, understanding the surrounding environment, and assessing the impact on species due to disturbances. The field of environmental impact assessment has seen substantial research exploring new technologies and models to evaluate and predict biodiversity more accurately. While current assessments rely on data from fieldwork and literature surveys to gauge species richness indices, limitations in spatial and temporal coverage underscore the need for high-resolution biodiversity assessments through species richness mapping. In this study, leveraging data from the 4th National Ecosystem Survey and environmental variables, we developed a species distribution model using Random Forest. This model yielded mapping results of 24 mammalian species' distribution, utilizing the species richness index to generate a 100-meter resolution map of species richness. The research findings exhibited a notably high predictive accuracy, with the species distribution model demonstrating an average AUC value of 0.82. In addition, the comparison with National Ecosystem Survey data reveals that the species richness distribution in the high-resolution species richness mapping results conforms to a normal distribution. Hence, it stands as highly reliable foundational data for environmental impact assessment. Such research and analytical outcomes could serve as pivotal new reference materials for future urban development projects, offering insights for biodiversity assessment and habitat preservation endeavors.

A simple and reliable cryo technique using desiccation and slow freezing of winter dormant buds was employed for 238 core collection of mulberry germplasm collected from diverse geographical regions and maintained under tropical conditions in the ex situ field gene bank to develop long-term biodiversity conservation for ensuring sustainable utilization of these valuable resources. Desiccation and freezing tolerance of bud grafts and excised shoot apices in the axillary buds of different Morus species under in vivo and in vitro condition indicated species-specific variation and most of the wild Morus species were found sensitive. In vitro regeneration and cryopreservation($-196^{\circ}C$) protocols using differentiated bud meristem like axillary winter dormant buds were worked out for a wide range of Morus species, land races, wild and cultivated varieties. Successful cryopreservation of mulberry winter dormant buds of different accessions belonging to M. indica, M. alba, M. latifolia, M. cathayana, M. laevigata, M. nigra, M. australis, M. bombycis, M. sinensis, M multicaulis and M. rotundiloba was achieved. Among wild species Morus tiliaefolia, and M. serrata showed moderate recovery after cryopreservation. Survival rates did not alter after three years of cryopreservation of different Morus species. ISSR markers were used to ascertain the genetic stability of cryopreserved mulberry, which showed no difference detected among the plantlets regenerated from frozen apices in comparison to the non-frozen material.
Two new naturalized species, Amaranthus hybridus and Crepis tectorum, are reported here. Descriptions and illustrations of the species and comparison of morphology with related species are provided. Amaranthus hybridus was first found at Nanjido, Seoul and is widely distributed in the central and southern part of Korean peninsula. New Korean name is given as "Ginteolbireum". It is well distinguished from the related species, A. retroflexus and A. patulus, by its late flowering time (September to October), sparse hair on the leaves, similar length between tepal and fruit, and long inflorescence without division. Crepis tectorum was first found at the entrance of Weoljeongsa Temple in Pyongchang-gun, Gangwon-do and spreads rapidly to other areas. New Korean name is given as "Nadomindulle".
Many species in Gymnodiniales, which are unarmored dinoflagellates, are responsible for marine algal blooms and some of them have potent toxin in the cell. Their taxonomy has so far been well-defined, and several genera (e.g. Akashiwo, Gymnodinium, Karenia) have recently been re-described. In Korea, few works have been carried out on their taxonomical and molecular studies. This study focused on comparison of both morphological and molecular characteristics of five unarmored dinoflagellates on Korean coastal water: Akashiwo sanguinea, Cochlodinium polykrikoides, Gymnodinium catenatum, Gymnodinium impudicum and Karenia aureolum (=K. mikimotoi). Morphological characteristics observed here was in good accordance with the original descriptions of individual species. In addition, none of difference was found in morphological comparisons between the Korean and foreign strains. Furthermore, molecular analysis showed that the SSU rDNA sequences were generally identical according to each species. In some distinct features, A. sanguinea, which has generally the same morphological features, were divided into two groups: one was Korean isolates including European isolates, the other was American isolates. In the two groups, the nucleus was positioned differently: middle of the cells in the Korean isolates (GnSg02, GnSg03), near the epicone in American isolates (CCMP1593, CCMP1837). In addition, this was strongly supported by phylogenetic analysis, inferred from the SSU rDNA sequences. K. aureolum (GrAr01) was corresponded to European G. aureolum (=K. mikimotoi) in shape and position of nucleus, chloroplast, however, which is similar to K. digitata in view of having a finger-like sulcus. This was in good agreement with phylogenetic study of these species. G. catenatum have identical morphology except the ridge location, and their genotype of SSU rDNA was also identical to GenBank data of the same species. From this study, we found that the five Korean unarmored dinoflagellates are identical morphological characteristics and genotype to each species of foreign isolates.
Lichen-forming fungi (LEF) were isolated from 67 Hungarian lichen species from ascospores or thallus fragments. LFF were successfully isolated from 26 species with isolation rate of 38.8%. Of the total number of isolation from ascospores (27 species) and thallus fragments (40 species), 48% and 32.5% of the species were successfully isolated, respectively. Comparison of rDNA sequences of ITS regions between the isolated LFF and the original thallus confirmed that all the isolates originated from the thallus fragments were LEF. The following 14 species of LEF were newly isolated in this study; Acarospora cervina, Bacidia rubella, Cladonia pyxidata, Lasallia pustulata, Lecania hyaline, Lecanora argentata, Parmelina tiliacea, Parmotrema chinense, Physconia distorta, Protoparmeliopsis muralis, Ramalina pollinaria, Sarcogyne regularis, Umbilicaria hirsuta, Xanthoparmelia conspersa and X. stenophylla. Antifungal activity of the Hungarian LFF was evaluated against plant pathogenic fungi of Colletotrichum acutatum, C. coccodes and C. gloeosporioides, causal agent of anthracnose on hot pepper. Among the 26 isolates, 11 LFF showed more than 50% of inhibition rates of mycelial growth of at least one target pathogen. Especially, LFF of Evernia prunastri, Lecania hyalina and Lecanora argentata were remarkably effective in inhibition of mycelial growth of all the tested pathogens with antibiotic mode of action. On the other hands, five isolates of Cladonia furcata, Hypogymnia physodes, Lasallia pustulata, Ramalina fastigiata and Ramalina pollinaria exhibited fungal lytic activity against all the three pathogens. Among the tested fungal pathogens, C. coccodes seemed to be most sensitive to the LFF. The Hungarian LFF firstly isolated in this study can be served as novel bioresources to develop new biofungicides alternative to current fungicides to control hot pepper anthracnose pathogenic fungi.
