A report on the Myxobolus sp. (Myxosporea : Bivalvulida), found from the gills of the pale chub, Zacco platypus (한국산 담수어류 피라미의 아기미에 기생하는 Myxobolus sp.에 관하여)

  • The mature spores of present Myxobolus sp. was ovoid in front view with no distinct redges of folds, lemon-shaped in side view with a straight sutural ridge. Spore valves showed symmetrical and smooth. Spores were 9 to $12{\mu}m$ (Mean=$10.4{\pm}0.7$, n=50)in length, 6 to $9{\mu}m$ (Mean=$7.7{\pm}0.6$, n=50) in width and 5.0 to $7.5{\mu}m$ (Mean=6.2, n=7) in thickness. Two polar capsules of spore were pyriform in shape, equal or mearly equal in size, 3 to $6{\mu}m$(Mean=$4.6{\pm}0.6$, n=50) in length, 2 to $3{\mu}m$(Mean=$2.2{\pm}0.3$, n=50) in width, Polar filaments of spore were composed with six to seven coils within capsules. Extended polar filaments were 55 to $135{\mu}m$ (Mean=78.7, n=50)in length. The shape and measurements of the present Mysobolus sp. spores were very similar with the spore of M. cyprinicola Reuss, 1906.

  • nif (nitrogen fixation)-H.D, K genes of Frankia sp. SNU 014201. a symbiotic strain isolated from root nodule of Alnus hirsura, were found to be located in the genome on 13.5 kb of EcoRI, 18.0 kb of BamHI, 10.5 kb of BglII and 4.5 kb of KpnI fragments. Using EMBL-3 BamHI arms of bacteriophage lambda. the genomic library was constructed. from which fourteen recombinant phage nif-clones were selected. Among them, Ahnif-I2 had insert DNA of 18 kb, in which 7.9 kb of BamHl fragment contained nif-H, D, K and 3.6 kb of HindlIl/KpnI had nif-H and partial -D. Therefore, the 7.9 kb and 3.6 kb fragments were subcloned and partial restriction maps were constructed. As the results, nif-F1, D.K genes were found to be located continuously on the 6.5 kb of HindII/BamHI and 5.2 kb of SalIIBamHI fragment in the genome of Frankia sp. SNU 014201.

Biosorption of Lead and Cobalt by Absidia coerulea and Thraustochitrium sp. (Absidia coerulea와 Thraustochitrium Sp. 에 의한 납과 코발트의 생물흡착)

  • Nonliving Absidia coerulea and Thraustochitrium sp. were used as biosorbents to remove lead and cobalt that are one of representative pollutant in wastewater and radioactive liquid waste. The optimum pH range for maximum lead and cobalt removal was increased 6.5~11.4 and 8.6~12.0 for Absidia coerulea and 4.2~10.5 and 8.9~11.6 for Thraustochitrium sp. to compared to biosorbent-free control, pH of 8.4~11.2 and 10.5~11.5, respectively. With 1 g biosorbent/L at initial solution pH 5.0. Absidia coerulea and Thraustochitrium sp. took up lead from aqueous solutions to the extent of 104 and 125 mg/g biomass, respectively, whereas Absidia coerulea and Thraustochitrium sp. at initial pH 6.0 took up only 2 and 20 mg/g biomass of cobalt, respectively. For initial 500 mg Pb/L at initial pH 5.0. optimum amount of biosorbent for maximum lead uptake was 0.2 g/L for Absidia coerulea and Thraustochitrium sp., whereas optimum 3.0 g biosorbent/L was needed for initial 200 mg Co/L at initial pH 6.0. Absidia coerulea and Thraustochitrium sp. had higher adsorption capacity for lead than that of cobalt.

Design of a S-Band Transfer-Type SP4T Using PIN Diode (PIN 다이오드를 이용한 S-대역 고출력 경로선택형 SP4T 설계)

  • In this paper, the design of a PIN diode S-band transfer-type SP4T including its driver circuit is presented. Each path of the SP4T is composed of the cascade connection of series-shunt PIN diodes to improve the isolation performance. The SP4T is implemented using chip type PIN diodes and a 20 mil AIN substrate fabricated using thin film technology. The driver circuit for the SP4T is designed using a multiplexer and four NMOS-PMOS push-pull pair. From on-wafer measurement, the fabriacted SP4T shows a maximum insertion loss of 1.1 dB and a minimum isolation of 41 dB. The time performance of the driver circuit is evaluated using the packaged PIN diodes with the identical PIN diode chip, and the transition time for on-off and off-on are below 100 nsec. For an input power level of 150 W, the measured insertion loss and isolation are close to those of the on-wafer measurement taking into consideration of the coaxial package mismatch and insertion loss.

주정증류 폐액을 이용한 Actinobacillus sp. EL-9로부터 Poly-$\beta$-Hydroxybutyrate의 생산 및 폐약의 처리

  • Alcoholic distillery wastes are utilized as dual purposes to produce PHB in lower production cost and to reduce the amount of waste to be treated. In this study, various attempts were made to increase PHB production under various conditions by Actinobacillus sp. EL-9 in a shaker culture. The addition of glucose, NH$_{4}$NO$_{3}$ to alcoholic distillery wastes slightly promoted cell mass and PHB production. Enzyme hydrolysis of alcoholic distillery wastes increased the production of PHB than that of untreated waste and acid hydrolysis treatment. The PHB weight in alcoholic distillery wastes was 1.91 g/l. Fermentation process of PHB production reduced the amount of COD value up to 54%, which reduced organic loading rate and capacity of activated sludge system.

  • Sphingomyelinase (SMase EC:3.l.4.l2) has been suggested to play important roles in the cell cycle, differentiation, apoptosis, inflammation, and the regulation of eukaryotic stress responses. SMase inhibitors may be a powerful tool to elucidate and regulate these cellular responses in which SMase involves. We first isolated an SMase inhibitor, named sphinin, from a strain of soil actinomycetes, F50970. Sphinin inhibited Mg/sup 2+/ -dependent neutral SMase from chicken embryo at 1.2 ㎍/㎖ of IC/sub 50/ Sphinin also inhibited acidic SMase, but it had no inhibitory activity on PI-PLC and PC-PLC, suggesting that sphinin is a specific inhibitor of SMase. The strain F50970 was identified as a Streptomyces sp. by its spiral spore chain, LL-diaminopimelic acid, menaquinone patterns of MK-9 (H'6) and MK-9 (H'8), FA-2c type of fatty acid pattern, and other morphological, physiological, and cultural characteristics.

Evaluation of Intrinsic Bioremediation of Methyl Tert-butyl Ether (MTBE) Contaminated Groundwater

  • This paper reported the use of real-time polymerase chain reaction (PCR), denaturing gradient gel electrophoresis (DGGE), and the culture-based method in the intrinsic bioremediation study at a petroleum contaminated site. The study showed that phenol hydroxylase gene was detected in groundwater contaminated with benzene, toluene, ethylbenzene, xylene isomers (BTEX) and methyl tert-butyl ether (MTBE). This indicated that intrinsic bioremediation occurred at the site. DGGE analyses revealed that the petroleum-hydrocarbon plume caused the variation in microbial communities. MTBE degraders including Pseudomonas sp. NKNU01, Bacillus sp. NKNU01, Klebsiella sp. NKNU01, Enterobacter sp. NKNU01, and Enterobacter sp. NKNU02 were isolated from the contaminated groundwater using the cultured-based method. Among these five strains, Enterobacter sp. NKNU02 is the most effective stain at degrading MTBE without the addition of pentane. The MTBE biodegradation experiment indicated that the isolated bacteria were affected by propane. Biodegradation of MTBE was decreased but not totally inhibited in the mixtures of BTEX. Enterobacter sp. NKNU02 degraded about 60% of MTBE in the bioreactor study. Tert-butyl alcohol (TBA), acetic acid, 2-propanol, and propenoic acid were detected using gas chromatography/mass spectrometry during MTBE degraded by the rest cells of Enterobacter sp. NKNU02. The effectiveness of bioremediation of MTBE was assessed for potential field-scale application.

재래누룩에서 분리한 곰팡이를 이용한 탁주의 성분분석

  • 쌀과 같은 곡류는 전분질을 당분으로 전환시켜 술을 제조하여야 하므로 미생물이 생성하는 효소가 필요한데 그 효소원이 누룩이며 누룩은 주류의 품질이나 생산량에 영향을 미치는 가장 중요한 요소라고 할 수 있다. 본 연구는 전국44개 지역의 누룩에서 순수 분리된 89종의 곰팡이 중 효소역가와 당화력이 비교적 뛰어난 10종(Aspergillus sp. SH-412 Heunghae, Aspergillus sp. SH-422 Ulsan, Rhizopis sp. SH-606 Imdeok, Aspergillus sp. SH-607 Yhesan, Aspergilus sp. SH-613 Wolseong, Rhizopus sp. SH-654 Uncheon, Aspergillus sp. SH-660 Jeonkok, Aspergillus sp. SH-667 Dongseong, Aspergillus sp. SH-669 Uncheon, Aspergillus sp. SH-696 Daecheon)과 대조균주로 Aspergillus kawachii CF1002 그리고 효모 Saccharomyces cerevisiae를 사용하셨으며 탁주제조 중 균주별 술덧의 산도, 환원당, 아미노산도와 제조된 완성주의 유기산, 유리아미노산, 유리당, 휴젤유 및 색도를 HPLC, GC Mass, 색차계로 측정한 결과 다음과 같은 결론을 얻었다. 균주별 술덧의 주정도는 모든 실험구에서 Aspergillus kawachii보다 높았으며 특히 Aspergillus sp. SH-422는 14.9%로 가장 수율이 좋았다. 환원당은 Aspergillus sp. SH-613이 0.49%로 가장 높았고 Aspergillus sp. SH-422는 0.37%로 가장 낮았으며 산도는 시간의 경과에 따라 감소했으며 술덧 발효중 아미노산도는 시간의 경과에 따라 약간 상승 후 다소 낮아지는 경향을 보였다. 각 균주별 술덧의 유기산은 tataric acid, malic acid, succinic acid, lactic acid, acetic acid가 검출되었으며 lactic acid 함량이 2.0∼3.2g/100ml로 가장 많이 검출되었다. 유기산은 Aspergillus sp. SH-669가 가장 높게 확인되었으며 Aspergillus sp. SH-607이 가장 낮게 분석되었다. 술덧의 주 아미노산은 histidine, alanine, glutamic acid, leucine, tryptophan순으로 검출되었으며 특히 alanine은 주류에 단맛을 주는 성분으로 모든 실험구에서 많은 양이 검출되었다. 각 균주별 술덧의 유리당은 glucose가 가장 많이 검출되었으며 Fusel oil은 iso-butyl alcohol과 iso-pentyl alcohol 이 가장 많이 검출되었다. 한편 균주별 술덧의 색차를 측정한 결과 L값은 63.33∼41.98, a값은 0.09∼-3.47, b값은 17.41∼4.90으로 나타났다.

Turfgrass Probiotics Reduce Population of Large Patch Pathogen and Improve Growth of Zoysiagrass (유용미생물 처리에 따른 들잔디 재배지의 갈색퍼짐병 병원균 감소와 잔디생육 촉진 효과)

  • To prevent large patch disease, caused by Rhizoctonia solani AG-2-2, in zoysiagrass a fungicide, Tebuconazole and three microbial agents Streptomyces sp. Burkholderia sp. and Streptomyces sp. S8 were applied in commercial turfgrass cultivation field in Sanchung, Gyeongnam, Korea. All treatments showed 50% reduced the pathogen population in thatch layer throughout the yearly cultivation period. Not only reduced the pathogen population, Tebuconazole, Streptomyces sp. Burkholderia sp. and Streptomyces sp. S8 treatment also enhanced turfgrass growth, chlorophyll and proline content. Malondialdehyde contents in each treatment was reduced from 6.2~28.9% when compared with the control. Taken together, reduction of pathogen population in soil lowered the disease incidence or severity, and allowed the turfgrass developed as normal condition. The results suggested that the selected microbial agents may use as biological control and growth promotion agents for the Zoysia turfgrass.

Inhibition of the Jun Aminoterminal Kinase by SP600125 Blocks PMA-Induced 92kDa Type IV Collagenase Secretion (PMA 유도 제4형 단백분해효소 분비를 차단하는 SP600125에 의한 Jun Aminoterminal Kinase의 억제)

  • 제 4형 콜라젠을 분해하는 MMP-9은 조직 재생에 중요한 역할을 하기 때문에 주목받아 왔는데, 이전의 문헌들에서 PMA에 의해 이 유전자 발현이 강하게 상승발현된다고 알려져 있다. 비록 MMP-9 발현을 조절하는 PMA의 기전이 잘 밝혀지지는 않았지만, 다른 유전자발현에서의 이 phorbol ester의 효과는 c-raf-1-ERK 신호전달통로의 활성에 관해 연구되어 오고 있다. 하지만 이번 연구에서 저자는 MMP-9 발현에서 PMA의 상승효과에는 대개는 스트레스성 자극과 관련된 JNK1 의존성 신호전달과정이 또한 필요하다는 다른 가능성을 조사하였다. 그 결과 JNK 억제재중 하나인 SP 600125가 UM-SCC-1세포주에서 PMA에 의해 유도된 MMP-9 상승발현을 용량의존적으로 억제하는 것으로 나타났고 72시간동안 전처치를 한 경우에 그의 억제효과가 최대이었다. phorbol ester로 처리한 세포에 GAL4 luciferase reporter와 vector를 주입해서 조사한 결과 PMA가 c-Jun transacting 활성을 상승시켰다. 그뿐 아니라 PMA에 의한 MMP-9 촉진자 활성에는 AP-1 motif가 필요하며 이 motif에 c-Jun이 결합하는 것을 EMSA를 통해 확인하였다. 결론적으로 UM-SCC-1 세포주에서 PMA에 의한 MMP9의 증가된 발현은 이미 밝혀진 ERK 경로뿐 아니라 JNK 경로를 통한 결과임이 밝혀져 이 경로를 차단하는 방법이 또하나의 암치료 방향을 제시해주고 있다.
