Changes in Quality Characteristics and Chemical Components of Sweet Potatoes Cultivated using Different Methods (재배방법에 따른 고구마의 품질특성 및 화학성분 변화)

  • Effects of cultivation methods on quality characteristics, pasting characteristics, chemical components, and antioxidant activities of sweet potatoes (Ipomoea batatas (L.) Lam) were determined. The Brix degree, hunter color value, pasting characteristics, moisture, protein, and mineral contents of the sweet potatoes showed significant differences from cultivation methods. The total polyphenol and flavonoid contents of the methanolic extracts of the sweet potato's pericarp were higher than sweet potato's sarcocarp. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity of the sweet potato's pericarp on the conventional culture and successful cropped hairy vetch culture was 776.38 and 715.20 mg TE/100 g sample. The 2,2'-azino-bis-3-ethylbenzo-thiazoline-6-sulfonic acid (ABTS) radical scavenging activity of the sweet potato's pericarp on the conventional culture and successful cropped hairy vetch culture was 708.03 and 708.58 mg TE/100 g sample. Generally, there was a difference in antioxidant compound content and radical scavenging activity on the methanolic extract of sweet potato with cultivation methods.

Proteolytic Yeasts Isolated from Mackerel (Scomber japonicus) (고등어에서 분리된 부패성 효모)

  • Microbiological spoilage of marine fish is complex process occurring by bacteria, yeasts and molds. There have been rare study for saprophytic yeasts although having enormous numbers of bacteriological studies on the spoilage of marine fish. The 14 genera of yeasts isolated from mackerel (Scomber japonicus) with high frequency of occurrence were Candida sp., Rhodotorula sp., Torulopsis sp., Cryptotoccus sp. and Tricosporon sp. Among these ones Candida lipolytica was identified as the strongest proteolytic yeast, then named Candida lipolytica FM5 (C. lipolytica FM5). C. lipolytica FM5 showed optimum growth at $25^{\circ}C$, pH 7.0 and could grow at $5^{\circ}C$ and in medium containing $10\%$ sodium chloride, To evaluate the saprophytic activity of the selected strain, C, lipolytica FM5 and Pseudomonas fluorescens ATCC 17571 which is one of representative spoilage bacteria were individually inoculated into the sterilized fish muscle homogenates, and then pH changes and volatile basic nitrogen (VBN) values were checked during the storage at various temperatures. According to the experimental results, the productions of VBN by C. lipolytica FM5 in the fish muscle homogenates were 50 mg-N/100 g at $5^{\circ}C$, 152 mg-N/100 g at $15^{\circ}C$ and 379 mg-N/100 g at $25^{\circ}C$ for 1 week storage, respectively. Above results were nearly same as in case of Ps. fluorescens ATCC 17571 inoculation. It suggest that sapyophytic yeasts also have important role in spoilage of marine fish.

Infection status with helminthes in feral cats pur-chased from a market in Busan, Republic of Korea

  • The present study was performed to investigate the infection status with helminth in a group of feral cats in Korea. More than 29 helminth species including adults or eggs were detected in visceral and fecal samples of the examined cats. Among these were a host of nematodes, including toxocarids, Ancylostoma sp. and the larva of Anisakis simplex; trematodes, including Clonorchis sinensis, Paragonimus westermani, Eurytrema pancreaticum, Pharyngostomum cordatum, Metagonimus spp., Heterophyes nocens, Pygidiopsis summa, Heterophyopsis continua, Stictodora fuscata, Stictodora lari, Acanthotrema felis, Stellantchasmus falcatus, Centrocestus armatus, Procerovum varium, Cryptocotyle sp., Echinostoma revolutum, Echinostoma hortense, Echinochasmus japonicus, Stephanoprora sp., Plagiorchis muris, Neodiplostomum sp. and diplostomulum. We also detected a variety of cestodes, including Spirometra erinacei, Taenia taeniaeformis and unidentified species of tapeworm. We also found examples of the acanthocephalan, Bolbosoma sp. In our assessment of the stools, we detected at least 12 species of helminth eggs. These findings confirmed that feral cats in Korea are infected with a variety of helminth parasite species. Furthermore, among the helminths detected, E. pancreaticum, S. fuscata, S. lari, A. felis, S. falcatus, C. armatus, P. varium, Cryptocotyle sp., E. revolutum, E. japonicus, Stephanoprora sp., P. muris, Neodiplostomum sp. and Bolbosoma sp. represent helminth fauna which have not been reported previously in feral cats in the Republic of Korea.

Analysis of the Bacteria in Nuclear Medicine (핵의학 검사실내 세균 분석)

  • In this study, the bacterial contamination level of equipments and devices in the nuclear medicine department of a university hospital was investigated. CNS was detected from the sample collected from the door opening button of the nuclear medicine department. Bacillus sp. was detected from the table and CNS with Bacillus sp. were detected from the control button at the PET-CT room no.1. Also, CNS was detected from the table and the control button at the PET-CT room no.2. In the distribution room no.1, CNS and Bacillus sp. were detected while CNS being detected from the distribution room no.2 and CNS with Bacillus sp. being detected from the distribution room no.3. In the injection room, Enterrococcus faecium and Pontoea sp. were detected. On the table of the ecsomatics room, Pontoea sp. was detected. Bacillus sp. was detected from the inside of the syringe Pb shield and CNS was detected from the outside. Enterrococcus faecium was detected from the Gamma camera table and Bacillus sp. was detected from the door grip. On the chair at the patient waiting room, Pseudomonas aeruginosa abd Bacillus sp. were detected. Therefore, it was understood that infection should be prevented by securely sterilizing examination devices after each examination, maintaining cleanliness by regular sterilization of waiting chairs and such objects with a number of direct contacts with patients, and infection education for the features of nuclear department.

Isolation of Agarivorans sp. JS-1 and Characterization of Its β-Agarase (한천분해세균 Agarivorans sp. JS-1의 분리 및 β-아가라제의 특성 규명)

  • This report looks at an agar-degrading marine bacterium and characterization of its agarase. Agar-degrading marine bacterium JS-1 was isolated with Marine agar 2216 media from seawater from the seashore of Sojuk-do, Changwon in Gyeongnam Province, Korea. The agar-degrading bacterium was named as Agarivorans sp. JS-1 by phylogenetic analysis based on 16S rRNA gene sequencing. The extracellular agarase was prepared from the culture media of Agarivorans sp. JS-1 and used for characterization. Relative activities at 20℃, 30℃, 35℃, 40℃, 45℃, 50℃, 55℃, and 60℃ were 70%, 74%, 78%, 83%, 87%, 100%, 74%, and 66%, respectively. Relative activities at pH 5, 6, 7, and 8 were 91%, 100%, 90%, and 89%, respectively. Its extracellular agarase showed maximum activity (207 units/l) at pH 6.0 and 50℃ in 20 mM Tris-HCl buffer. The residual activity after heat treatment at 20℃, 30℃, and 50℃ for 30 minutes was 90%, 70%, and 50% or more, respectively. After a 2-hour heat treatment at 20℃, 30℃, 35℃, 40℃, and 45℃, the residual activity was 80%, 68%, 65%, 63%, and 57%, respectively. At 50℃ and above, after heat treatment for 30 minutes, the residual activity was below 60%. Thin layer chromatography analysis suggested that Agarivorans sp. JS-1 produces extracellular β-agarases as they hydrolyze agarose to produce neoagarooligosaccharides such as neoagarohexaose (20.6%), neoagarotetraose (58.5%), and neoagarobiose (20.9%). Agarivorans sp. JS-1 and its thermotolerant β-agarase would be useful in the production of neoagarooligosaccharides, showing functional activity such as inhibition of bacterial growth and delay of starch degradation.

Pathological Characteristics of Saprolegnia sp. to Snakehead and Effect of Essential Oils to the Growth of the Water Mold (양식 가물치(Channa argus)에 대한 Saprolegnia sp.의 병리학적 특성과 물곰팡이의 생장을 제어하는 정유의 영향)

  • A Saprolegnia sp. was isolated from cultured snakehead, Channa argus, and its physiological characteristics were investigated. The optimum temperature, pH and NaCl concentration for mycelial growth of Saprolegnia sp. were $25^{\circ}C$, 6.0 and 0%, respectively. The mycelial growth was increased with the addition of 10 mM phosphate and 10mg/L casamino acid. The essential oils extracted from three plants, Artemisia princeps var. orientalis, Thuja orientalis and Chamaecyparis obtusa have been tested to know whether they inhibit the growth of Saprolegnia sp. at six different oil concentrations(10, 100, 500, 1,000, 1,500 and 2,000 ppm). Essential oil from A. princeps var. orientalis began to inhibite the mycelial growth of Saprolegnia sp. at the concentration more than 10ppm. Using other essential oils from T. orientalis and C. obtusa, those initially inhibited the mycelial growth of Saprolegnia sp. at the concentration over 10 ppm and complate inhibition of mycelial growth was observed at over 500 ppm. The histopathological features of Snakehead infected by Saprolegnia sp. were studied. A club shape of gill lamella epithelial cells was observed in the gill. The mycelial cells were penetrated into muscular tissue, and the accumulation of the ceroid was observed in the liver, spleen and kideny tissue in common. The necrosis of tubular epithelial cells was seen in the liver tissue, parenchymal tissue in the spleen and tubular epithelial cells in the kidney.

Phylogenetic Analysis on Wild Cordyceps Collected from Miryang Region of South Korea (밀양근교에서 채집한 야생 동충하초 계통의 PCR 산물에 근거한 계통 유전학적 연구)

  • The phylogenetic relationships among thirty-two strains (P1~P32; including Cordyceps sp., Paecilomyces sp., Beauveria sp., Aranthomyces sp., Isaria sp. and Himenostilbe sp.) in Miryang region located in the southern part of Korea, were investigated based on internal transcribed spacer (ITS) sequences of ribosomal DNA. A fragment of ITS region was amplified by polymerase chain reaction (PCR) using the specific primer pairs ITS1 and ITS4. After obtained same size of PCR products from various strains, we cloned them into a pGEM-T easy vector to determine their sequences. BLAST analyses of the nucleotide sequence ITS1, 5.8S and ITS2 gene fragments revealed the identity and their phylogenetic relationship. Among 32 strains isolated from Miryang region, Cordyceps militaris was shared 100% sequences with Genbank (AY49191, EU825999, AY491992), while some species are not shared perfectly with reported sequences. For example, strain P17 (P. tenuipes in Ulju-gun Gaji Mountain) has some differences among the other strains of P. tenuipes (Miryang-si Jocheon-eup, Miryang-si Gaji Mountain) and those of gene bank. We conclude that ITS analyses with strains in the suburbs of Miryang in this study can be effectively used as a tool for classification, evaluation and collection of the natural eco-type genetic resources.

Endoparasites of Small Mammals in Edo State, Nigeria: Public Health Implications

  • Some small mammals occur as household pests and harbour a number of parasites that could be of public health importance. This study profiled the helminth and protozoan parasites in trapped small mammals within and around human dwelling places (houses) located across 4 major towns (Auchi, Benin, Ekpoma, and Uromi) and environs in Edo state, Nigeria. Six genera (Apodemus sp., Crocidura sp., Mastomys natalensis, Mus musculus, Rattus sp., and Sorex sp.) were identified from 502 trapped small mammals. Overall, M. musculus (71.9%) and Rattus rattus (20.1%) were the most frequently trapped. In total, on examination of blood, gastrointestinal contents, and brain tissues, 12 helminth taxa (Angiostrongylus sp., Aspicularis sp., Capillaria sp., Gongylonema sp., Heterakis spumosa, Hymenolepis diminuta, Hymenolepis nana, Mastophorus muris, Moniliformis moniliformis, Nippostrongylus brasiliensis, Strongyloides sp., Trichosomoides sp., and Trichuris sp.), and 6 protozoan parasites (Babesia sp., Trypanosoma lewisi, Plasmodium sp., Eimeria sp., Isospora sp., and Toxoplasma gondii) were isolated. Most prevalent helminths with relatively heavy mean intensity were Strongyloides sp. and Heterakis spumosa, while Plasmodium, Eimeria, and Isospora were the most prevalent protozoan parasites. Generally, intrinsic factors like sex and age had marginal influence on the rate and burden of infection in M. musculus and R. rattus. Although the infection rate and prevalence of zoonotic parasites were low, they were largely recovered in rodents from Ekpoma. This study elucidates the public health implication of the presence of zoonotic parasites in these small mammals.

Effect of Glucose Concentration on the Production of Erythritol by Trichosporon sp.

  • The effect of glucose concentration on the production of erythritol by Trichosporon sp. was mainly studied. The specific growth rate and production rate of erythritol gave the highest values of $0.23 h^{-1}\; and\; 4.2 g/\ell/h,\; respectively,\; on\; 100 g \; glucose/\ell$ of medium. The conversion yield of erythritol during the exponential phase and the stationary phase was constantly maintained at 19% and 51 %, respectively, while the glucose concentration in the medium varied from 100 g/$\ell$ to 400 g/$\ell$. The maximum overall erythritol conversion yield of 47% was obtained when the glucose concentration in the medium was 400 g/$\ell$. It corresponded to a 74% increase compared with the 100 g/$\ell$ glucose medium. The diauxy growth of this microbe was also observed. It grew exponentially consuming glucose, then after the second lag phase, biomass slowly increased using glycerol and erythritol.

  • In this paper, a fault diagnosis and control integrated system (FDCIS) was developed to control the thermoelectric (TE) power in the SP-100 space reactor. The objectives of the proposed model predictive control were to minimize both the difference between the predicted TE power and the desired power, and the variation of control drum angle that adjusts the control reactivity. Also, the objectives were subject to maximum and minimum control drum angle and maximum drum angle variation speed. A genetic algorithm was used to optimize the model predictive controller. The model predictive controller was integrated with a fault detection and diagnostics algorithm so that the controller can work properly even under input and output measurement faults. With the presence of faults, the control law was reconfigured using online estimates of the measurements. Simulation results of the proposed controller showed that the TE generator power level controlled by the proposed controller could track the target power level effectively even under measurement faults, satisfying all control constraints.
