Monitoring of Pathogenic Bacteria, Heavy Metals, and Pesticide Residues in Commercial Edible Dry Flowers (시판 23종 꽃차의 유해세균, 중금속 및 잔류농약 평가)

  • Some flowers have a high sensual appeal owing to their unique shape, color, smell, and taste and have been used as functional food and oriental medicine. Recently, edible dry flowers (EDFs) have attracted social attention as noble sources of functional teas. In this study, for the risk assessment of EDFs, pathogenic bacteria, heavy metals, and pesticide residues were monitored in 23 types of commercial EDF. No Enterobacteria spp. and Listeria spp. were found in all EDF products. However, common aerobic bacteria (3.24~3.85 Log CFU/g) were found in EDF, namely, Pueraria lobata, Chamaemelum nobile, Acacia decurrens, Rhododendron mucronulatum Turcz, Oenothera lamarckiana, Brassica napus, and Prunus serrulata. Staphylococcus aureus was found in 11 and Salmonella sp. was found in 8 of the 23 EDFs. Considering the cold extraction of EDF for tea and beverages, the regulation of pathogenic bacteria in EDFs is necessary. No heavy metals such as Pb, Cd, Co, Cr, Cu, Ni, and As were found in all EDFs, except the dry flower of Hemerocallis fulva, which contained Pb at 0.08 ppm. Different pesticides and fungicides were found in EDFs, but their concentrations were very low (0.01~0.08 ppm) and below the maximal residue level. Only the dry flower of Chrysanthemum morifolium had a high content of chlorpyrifos (0.215 ppm), which is long-lasting pesticide. Our results suggest that the establishment of EDF regulations for pesticide residue, culture separation between edible and garden flowers, and guidelines for preventing pathogenic microbial contamination are necessary.

Heterologous Expression of Interferon α-2b in Lactococcus lactis and its Biological Activity against Colorectal Cancer Cells

  • Type I Interferons (IFNα) are known for their role as biological anticancer agents owing to their cell-apoptosis inducing properties. Development of an appropriate, cost-effective host expression system is crucial for meeting the increasing demand for proteins. Therefore, this study aims to develop codon-optimized IFNα-2b in L. lactis NZ3900. These cells express extracellular protein using the NICE system and Usp45 signal peptide. To validate the mature form of the expressed protein, the recombinant IFNα-2b was screened in a human colorectal cancer cell line using the cytotoxicity assay. The IFNα-2b was successfully cloned into the pNZ8148 vector, thereby generating recombinant L. lactis pNZ8148-SPUsp45-IFNα-2b. The computational analysis of codon-optimized IFNα-2b revealed no mutation and amino acid changes; additionally, the codon-optimized IFNα-2b showed 100% similarity with native human IFNα-2b, in the BLAST analysis. The partial size exclusion chromatography (SEC) of extracellular protein yielded a 19 kDa protein, which was further confirmed by its positive binding to anti-IFNα-2b in the western blot analysis. The crude protein and SEC-purified partial fraction showed IC50 values of 33.22 ㎍/ml and 127.2 ㎍/ml, respectively, which indicated better activity than the metabolites of L. lactis NZ3900 (231.8 ㎍/ml). These values were also comparable with those of the regular anticancer drug tamoxifen (105.5 ㎍/ml). These results demonstrated L. lactis as a promising host system that functions by utilizing the pNZ8148 NICE system. Meanwhile, codon-optimized usage of the inserted gene increased the optimal protein expression levels, which could be beneficial for its large-scale production. Taken together, the recombinant L. lactis IFNα-2b is a potential alternative treatment for colorectal cancer. Furthermore, its activity was analyzed in the WiDr cell line, to assess its colorectal anticancer activities in vivo.

Effects of Ground Vegetation and Pyrethroid Spray on the Population Dynamics of Panonychus citri (Acari: Tetranychidae) and Natural Enemies in Citrus Orchard: A Short-term Effect (감귤원에서 초생관리와 합성피레스로이드계 조합처리가 귤응애와 천적의 발생양상에 미치는 단기효과)

  • This study was conducted to examine the effects of grass vegetation (W: manual weeding, NW: herbicide sprays) and pyrethroid spray (P: pyrethroid spray, NP: no pyrethroid spray) on the population dynamics of Panonychus citri and natural enemies in citrus orchards. Two essential hypothesis were made to test the population dynamics: 1) weed planting promotes natural enemies by offering habitat and alternative food sources, resulting in the reduction of P. citri populations, and 2) pyrethroid spray removes natural enemies by its non-selective toxicity, resulting in the increasement of P. citri populations. The observed natural enemy populations (mainly Phytoseiids and Agistemus sp.) were not different largely from the expected values in the hypothesis, which assumes more abundant natural enemies in weeds and no pyrethroid plots. Although some discrepancy was occurred in NW+NP and W+NP plots in 2011, the observed values were almost same with expected values in 2012. In overall, pesticide effect was strongly significant and pyrthroids removed largely natural enemies. Although habitat (weeds) effect showed a conflict result, natural enemy population increased in plots allowing weed growth, when considering the increased autumn population relatively compared to that of spring-summer population. The decreased abnormal P. citri populations in pyrethroid plots could be explained under the assumption of a strong repellent behavior of P. citri to the pyrethroids.

MHY2251, a New SIRT1 Inhibitor, Induces Apoptosis via JNK/p53 Pathway in HCT116 Human Colorectal Cancer Cells

  • Sirtuins (SIRTs) belong to the nicotinamide adenine dinucleotide (NAD+)-dependent class III histone deacetylase family. They are key regulators of cellular and physiological processes, such as cell survival, senescence, differentiation, DNA damage and stress response, cellular metabolism, and aging. SIRTs also influence carcinogenesis, making them potential targets for anticancer therapeutic strategies. In this study, we investigated the anticancer properties and underlying molecular mechanisms of a novel SIRT1 inhibitor, MHY2251, in human colorectal cancer (CRC) cells. MHY2251 reduced the viability of various human CRC cell lines, especially those with wild-type TP53. MHY2251 inhibited SIRT1 activity and SIRT1/2 protein expression, while promoting p53 acetylation, which is a target of SIRT1 in HCT116 cells. MHY2251 treatment triggered apoptosis in HCT116 cells. It increased the percentage of late apoptotic cells and the sub-G1 fraction (as detected by flow cytometric analysis) and induced DNA fragmentation. In addition, MHY2251 upregulated the expression of FasL and Fas, altered the ratio of Bax/Bcl-2, downregulated the levels of pro-caspase-8, -9, and -3 proteins, and induced subsequent poly(ADP-ribose) polymerase cleavage. The induction of apoptosis by MHY2251 was related to the activation of the caspase cascade, which was significantly attenuated by pre-treatment with Z-VAD-FMK, a pan-caspase inhibitor. Furthermore, MHY2251 stimulated the phosphorylation of c-Jun N-terminal kinase (JNK), and MHY2251-triggered apoptosis was blocked by pre-treatment with SP600125, a JNK inhibitor. This finding indicated the specific involvement of JNK in MHY2251-induced apoptosis. MHY2251 shows considerable potential as a therapeutic agent for targeting human CRC via the inhibition of SIRT1 and activation of JNK/p53 pathway.

A Case of Psychogenic Tremor Improved by Complex Korean Medicine Treatment Including Cheongshimondam-tang-gamibang and Acupuncture Treatment - A Case Report (청심온담탕가미방과 침구치료를 포함한 복합한의치료로 호전된 심인성 떨림 환자 1례 - 증례보고)

  • Objectives: A case study about the effectiveness of integrative Korean medicine treatment in a psychogenic tremor patient. Case presentation: A 36-year-old male diagnosed with psychogenic tremor with low back pain underwent 29 days of inpatient treatment and 4 weeks of outpatient treatment. His pattern identification was a psychogenic tremor of the weakness of the heart and gall bladder (SimDamHeoGup) type. The treatments were herbal medicine and acupuncture. He took Cheongshimondam-tang for 29 days and Chunwangbosim-dan for 6 weeks when he had symptoms. He also received acupuncture for 20 minutes twice a day at GB20 (Pungji), L14 (Hapgok), HT4 (Yeongdo), PC6 (Naegwan), HT7 (Sinmun), GB34 (Yangleungcheon), ST36 (Joksamni), SP6 (Sameumgyo), and LR3 (Taechung). After 8 weeks of treatment, the Fahn Tolosa Marin rating (FTM) scale of his tremor, at rest, dropped from Grade 4 on both upper extremities, trunk, and both lower extremities at the time of admission to Grade 2 in both upper extremities and trunk, and Grade 0 in both lower extremities at the time of discharge. His Beck Anxiety Inventory (BAI) score decreased from 38 to 7, and his numerical rating scale (NRS) decreased from 8 to 1 for low back pain and from 9 to 2 for tremor. A follow-up visit to the hospital 2 months after the end of treatment confirmed continued symptom improvements and no significant side effects. Conclusions: This study suggests the possibility of treating psychogenic tremor using only Korean medicine treatments. Further studies with control groups and long-term follow-up are needed.

Inhibitory Effect of Dendrobium moniliforme on Degranulation and Histidine Decarboxylase Expression in RBL-2H3 Cells (RBL-2H3 세포에서 탈과립과 histidine decarboxylase 발현에 미치는 석곡(Dendrobium monilifrme)의 효과)

  • The stems of Dendrobium moniliforme are used in traditional Oriental medicine as a Yin tonic to nourish the stomach, promote the production of body fluid, and reduce fever. This study investigated the effects of the aqueous extract of D. moniliforme stems (DME) on mast cell degranulation and the expression of tumor necrosis factor-α (TNF-α), interleukin-4 (IL-4), and histamine-synthesizing enzyme histidine decarboxylase (HDC). We used rat mast cell line RBL-2H3 cells and stimulated them with PMA plus calcium ionophore (PMACI). Pretreatment with DME significantly inhibited PMACI-induced β-hexosaminidase release and the expression of TNF-α, IL-4, and HDC. Furthermore, DME suppressed PMACI-induced nuclear translocation of the nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) and activator protein 1 (AP-1). In addition, HDC expression was inhibited by SP600125 (JNK inhibitor), PD98059 (ERK inhibitor), and SB203580 (p38 kinase inhibitor). Finally, the phosphorylation of p38 kinase, extracellular signal-regulated kinase 1/2 (ERK1/2), and c-Jun N-terminal kinase (JNK) was inhibited by pretreatment with DME. These results suggest that DME has inhibitory effects against degranulation, cytokine (TNF-α and IL-4) and HDC expression, and that HDC expression is mediated by MAPK signaling. These findings suggest that DME may have therapeutic potential in the treatment of hypersensitive and inflammatory diseases.

Selecting and evaluating microorganism strains to prepare low-salt doenjang for flavoring via the fermentation of rice and soybeans (쌀 및 콩 발효 균주선발과 이를 적용한 조미용 저염된장의 평가)

  • This study investigated the strains and fermentation characteristics of used to ferment a mixture of rice and soybeans to manufacture low-salt doenjang for flavoring. The soybean and rice mixture was fermented using three selected strains of Aapergillus oryzae and Bacillus sp. The changes in quality of the fermented products were found to be dependent on the aging period. Therefore, the strain and a suitable aging period were seleted based on the increases in AN, total sugar, and reducing sugar. The fermented products were prepared and mixed, using the selected or commercially available strains (the sample and control, respectively), to create low-salt doenjang. Following this, their characteristics were compared. The sample had a higher content of taste-related ingredients(free amino acid, nucleic acid-related substances) than the control. Using the selected strain to ferment a rice and soybean mixture will thus be expected to enhance the flavor of industrially produced seasoned doenjang.

Diversity and Succession of the Bacterial Community during the Initial Fermentation Period in Modernized Soy Sauce (Ganjang) (개량식 간장의 발효 초기 단계에서의 미생물 다양성 및 천이에 관한 연구)

  • The taste and quality of soy sauce, a fermented liquid condiment, is greatly influenced by microbial metabolism during fermentation. To investigate the microbiological characteristics of ganjang during the initial fermentation process, we prepared meju (fermented soybean) blocks fermented with starter cultures and solar salts and analyzed the microbial community quantitively using 16S rRNA gene profiling from ganjang that had been fermented over a five-week period. The ganjang samples were collected and analyzed after soaking for week one (1W), three (3W), and five (5W) weeks. We found that Halomonadaceae was significantly higher in the 1W group (89.83%) than the 3W and 5W groups (14.46%, and 13.78%, respectively). At a species level, Chromohalobacter beijerinckii and Chromohalobacter canadensis were the dominant species in the 1W group but several taxa such as Bacillus subtilis, Pediococcus acidilactici, and Enterococcus faecalis were more abundant in the 3W and 5W groups. Pearson correlation analysis of the relative abundance of the bacteria showed a negative correlation between Chromohalobacter and two bacterial genera Bacillus and Enterococcus. Beta-diversity showed a statistical distinction between the 1W and the 3W and 5W groups, while no significance was evident between the 3W and 5W groups. Linear discriminant effect size analysis was used to identify biomarkers and significant differences in the relative abundance of several halophilic bacteria, Bacillus sp. and lactic acid bacteria at 1W, 3W, and 5W, recpectively, which indicates the important role of the bacterial community at these time points.

Analyzing the Relationship between Environmental Consciousness and Railway Choice Behavior (환경의식과 철도이용행동의 관련성 분석)

  • The purpose of this research is to clarify the relation between environmental consciousness and railway usage behavior. Author would locate this research on position of basic survey to promote railway use according to Low Carbon Green Growth policy in Korea. In this research, we would perform descriptive analysis using data of research on the actual condition of railway use in 2008, Daegu, and describe its relationship. In addition, we would suggest some idea about policy which can promote railway use. The order of railway choice behavior noticed in clustering of environmental consciousness was cooperative behavior type, middle type and non-cooperative behavior type. It suggests that environmental consciousness has effect on transportation choice behavior. Specially, railway improvement isn't enough to promote railway use. And, it is advisable to carry out the improvement in such a way that it may encourage the nation to move from the current environmental consciousness stage to cooperative behavior. Moreover, we assumed Binary Probit(BP) model using SP data of time or condition of transportation expense compared with passenger car and bus. As the results, modified likelihood ratio of two BP models is favorable variables. And it occurred that mode was transferred from passenger car to railway when it showed higher social environment consciousness and low selfish environment consciousness, because t-statistic which represents selfish environment consciousness showed significance in 95% confidence level. That is, it can be described that environment consciousness affect on the intention of railway use.

Taxonomic Characteristics of Chironomids Larvae from the Hangang River at the Genus Level. (한강 수계 내 서식하는 깔따구류 유충의 속 수준에서의 분류 형질)

  • The Hangang River* is necessary to manage the water environment of severe pollution due to the high density of residential areas, parks, and agriculture and the large population concentrated there. Benthic macroinvertebrates, such as chironomids larvae, are bioindicator species that reflect environmental changes and are crucial for water quality monitoring. In this study, we investigated morphological characteristics and molecular analysis of the chironomids larvae inhabiting the Hangang River area for water environment surveys. For this research, 20 rivers, lakes, and urban area in the Hangang River basin were selected. Chironomids larvae were collected from July to September 2022, and their appearance and characteristics were identified through morphological identification. In addition, phylogenetic analysis was performed based on the mtCOI gene sequences of the collected chironomids larvae, and identification at the genus level was confirmed. As a result, 32 species and 18 genera of 3 subfamilies of Chironomidae larvae were identified, and Stictochironomus sp. dominated most sites(6 sites). The morphological characteristics of the identified chironomids larvae, such as the mentum, ventromental plate, and antenna, were organized into table and pictorial keys, and a Bayesian inference molecular phylogeny was presented. These results provide basic morphological information for genus-level identification and can be used as fundamental information for water quality management.
