Copolyester of 3-Hydroxybutyrate and 3-Hydroxyvalerate Produced by Methylobacterium sp. GL-10 (Methylobacterium sp. GL-10이 생산하는 3-Hydroxybutyrate와 3-Hydroxyvalerate의 Copolyester)

  • - The further study for the identification of the previously reported pink-pigmented facultative methylotrophic bacterium (PPFM) GL-10 was carried out. The PPFM GL-10 was Gram nagative, rod, and motile by a single polarly inserted flagellum. The colonies were smooth, pink, circular, along with convex with entire margin. The isolate could utilize C1 compounds and a variety of multicarbon substrates as sole carbon and energy source. The isolate was obligately aerobic, and exhibited both catalase and oxidase activities. The deoxyribonucleic acid base composition was 65-67 mol% guanine plus cytosine. The isolate was mostly identical with Methylobacterium extorquens and named Methylobacterium sp. strain GL-10. Methylobacterium GL-10 accumulated a copolyester of 3-hydroxybutyrate and 3-hydroxyvalerate (poly-3HB/3 HV) when grown in nitrogen-free culture media containing sodium propionate as substrate at the second polyester accumulation stage. The composition of copolyester, as determined from $^1h$ NMR spectra, was 23 mol% of 3-hydroxyvalerate (3HV).

Production, Purification, and Characterization of Taxol and 10-DABIII from a new Endophytic Fungus Gliocladium sp. Isolated from the Indian Yew Tree, Taxus baccata

  • We have isolated endophytic fungi from the Indian yew tree, Taxus baccata, and then screened for taxol production. Out of the 40 fungal cultures screened, one fungus Gliocladium sp. was found to produce taxol and 10-DABIII (10-deacetyl baccatin III). These compounds were purified by TLC and HPLC and characterized using UV-spectroscopy, ESI-MS, MS/MS, and proton NMR. One liter of Gliocladium sp. culture yielded $10\;{\mu}g$ of taxol and $65\;{\mu}g$ of 10-DABIII. The purified taxol from the fungus showed cytotoxicity towards cancer lines HL-60 (leukemia), A431 (epidermal carcinoma), and MCF-7 (breast cancer).

  • Acetobacter strains are bacteria that can synthesize cellulose when grown on an undefined medium containing glucose. Several culture conditions affecting cellulose production by Acetobacter sp. A9 were examined by cultivating cells under shaking cultures. The addition of buffering agents, such as 3-(N-morpholino) propanesulfonic acid (MOPS) and CaCO$_3$, increased cellulose production. It suggests that pH of culture medium is important to an economical mass cellulose production. Addition of bead (Ф10 w) to culture medium stimulated 'disintegrated bacterial cellulose' production.

  • A strain of non-sulfur photosynthetic bacteria possessing large amount of ubiquinone-10 in the cell mass, was isolated from waste water collected from the Nakdong River. It was identified as Rhodobacter sp. N-2 by a series of studies on its morphological. cultural and physiological characteristics. The organism appeared to got higher cell mass under circumstances of anaerobic growth conditions than under photosynthetic aerobic cultures. Biotin and thiamin and niacin could be used as good growth factors.

Effects of substance P on mineralization markers and heme oxygenase-1 Expression in human immortalized periodontal ligament cells

  • Substance P (SP) is known to be expressed in the nerve fibers of dental pulp and periodontal tissues. It was recently reported that SP expression increased in response to orthodontic force. In the present study, we investigated the effect of SP on expression of mineralization markers and heme oxygenase-1 (HO-1) in human immortalized periodontal ligament (IPDL) cells. Cell viability was measured using a 3,4,5-dimethylthiazol-2-yl-2,5-diphenyl tetrazolium bromide (MTT) assay. The expression of mineralization markers, including alkaline phosphatase (ALP), osteonectin (ON) and bone sialoprotein (BSP), and heme oxygenase-1 (HO-1) was assessed by reverse transcription-polymerase chain reaction (RT-PCR) and Western blot analysis. SP did not significantly change human IPDL cell viability, with the exception of the 24 hour treatment group. Treatment of human IPDL cells with $10^{-10}$ to $10^{-4}M$ SP upregulated mineralization marker and HO-1 expression in a time- and concentration-dependent manner. Our results suggest that SP may modulate osteoblastic cell differentiation of human IPDL cells through a mechanism involving HO-1 expression.

  • A bacterium which produce hyaluronic acid complex was isolated from soil, and identified as Klebsiella sp. L-10. The maximal hyaluronic acid complex production was obtained when the strain was cultured at $37^{\circ}C$ for 18hrs with shaking in the optimal medium containing 0.1% yeast ext., 3% tryptone, 5% glucose, 10mM $K_2HPO_4$and $KH_2PO_4$, respectively and initial pH6.5 and the final hyaluronic acid complex production under the above condition was 1,400mg per liter of cultures.

The Distribution of Fish Larvae in the Korea Strait (대한해협의 자치어 분포상)

  • Larvae of fishes were collected at 26 stations in the Korea Strait, by horizontal towings using the fish larval net (Norpac) in the surface layer and by oblique towings using Isaac-Kid mid-water trawl in the mid-water layer in summer (July) and in autumn (November), in 1983. The towing speed was 1.5 knots. Of the larval fish examined, 3,529 individuals were identified to generic and specific levels. They belong to 27 families, consisting of 38 species in 37 genera. Unidentified were 114 specimens. The most abundant species was Engraulis japonica which occupied $88.5\%$ of the whole 3,645 individuals. Diaphus sp. took the second place with 50 individuals ($l.3\%$). Seasonally abundant species were in the order of Engraulis japonica, a Gobid, Trichiurus lepturus, and Diaphus sp. in summer, and in the order of Diaphus sp., Engraxlis japonica, Bothus sp., and a Ophichthid in autumn. Of these larvae, Engraulis japonica and Diaphus sp. continuously appeared from summer to Autumn. Champsodon sp., Diaphus sp. and Synagrops philippinensis were unrecorded species in Korea.

Application of SP Monitoring in the Pohang Geothermal Field (포항 지열 개발지역에서의 SP 장기 관측)

  • To delineate geothermal water movement at the Pohang geothermal development site, Self-Potential (SP) survey and monitoring were carried out during pumping tests. Before drilling, background SP data have been gathered to figure out overall potential distribution of the site. The pumping test was performed in two separate periods: 24 hours in December 2003 and 72 hours in March 2004. SP monitoring started several days before the pumping tests with a 128-channel automatic recording system. The background SP survey showed a clear positive anomaly at the northern part of the boreholes, which may be interpreted as an up-flow Bone of the deep geothermal water due to electrokinetic potential generated by hydrothermal circulation. The first and second SP monitoring during the pumping tests performed to figure out the fluid flow in the geothermal reservoir but it was not easy to see clear variations of SP due to pumping and pumping stop. Since the area is covered by some 360 m-thick tertiary sediments with very low electrical resistivity (less than 10 ohm-m), the electrokinetic potential due to deep groundwater flow resulted in being seriously attenuated on the surface. However, when we compared the variation of SP with that of groundwater level and temperature of pumping water, we could identify some areas responsible to the pumping. Dominant SP changes are observed in the south-west part of the boreholes during both the preliminary and long-term pumping periods, where 3-D magnetotelluric survey showed low-resistivity anomaly at the depth of $600m\~1,000m$. Overall analysis suggests that there exist hydraulic connection through the southwestern part to the pumping well.

  • Fugi are eukaryotic, nonphotosynthetic, filamentous or unicellular organisms, most of which grow on nonliving materials as saphrophytes. The majority are therefore opportunistic pathogens and predisposing factors often contribute to the establishment of fungal infection. These include an alteration in the normal flora of the host by prolonged administration of antibiotics, immunosup-pression, concurrent infections, damage to the skin or mucous membranes, constantly moist areas of skin or the exposure to a large infective dose, and as with fungal spores. Fungi may cause a variety of diseases which may be due directly to fungal invasion of tissue or more often to the ingestion of toxins produces by fungi in growing, standing or stored grains and other animals feeds. In this experiment, contaminated fugi were isolated and identified from animal feedstuffs such as Korean cattle, milking cows, pigs and chickens. Twelve genues were isolated from animal feeds, they are 9 from Korean cattle and milking cows feeds, 6 from pigs feeds, and 10 from chickens feeds. Among them, most frequently encountered species was Yeast(56 strains), followed by Fusarium sp(41 strains), Aspergillus sp(20 strains), each of Micorsporum sp and Trichophyton sp(17 strains), Penicilium sp(12 strains), in order. And also minority was isolated as Candide sp(4 strains), Trichoderma sp(3 strains), each of Epidermophytom sp and Absida sp(2 strains), and each of Sporothrix sp and Maduromyces sp(1 strain). Among the Aspergillus sp 20 isolates, A flavus(5 strains), A nidulans(4 strains), A fumigatus(3 strains), A glucans(3 strains), A niger(3 strains) and A terreus(2 strains) were identified.

A Biological Study on the Methanol-Utilizing Bacteria (Methanol 자화세균에 관한 생물학적 연구)

  • By the successive enrichment culture, more than 250 methanol-utilizing bacteria were isolated from various samples such as soil, waste water and sewage. Two strains of which were selected and tentatively identified as Acinetobacter sp. and Pseudomonas sp. experiments were carried out to determine the growth conditions for the higher biomass yield and to demonstrate the difference to protein composition dependent upon carbon sources of these two species. the results were as follows ; 1. the optimum pH was determined as 8 in the both species. The optimum temperature in Acinetobacter sp. was $25^{\circ}C{\sim}30^{\circ}C$ and pseudomonas sp. was $30^{\circ}C-35^{\circ}C$. The optimum initial concentration of mthanol was determined as 1-2% in Acinetobacter sp. and 2-3% in pseudomonas sp. 2. The optimum concnetrations of nitrogen source, micro-elements, and vitamins such as biotin and thiamine-HCl in Acnetobactar sp. were 1g $(NH_4)_3SO4,\;1{\sim}3mg\;Mn^{++},\;4mg\;Fe^{++},\;10{\mu}g\;biotin,\;and\;100{\mu}g$ thiamine-HCl per liter medium. In the Pseudomonas sp., 2g $(NH_4)_3SO4,\;1mg\;Mn^{++},\;trace\;amounts\;of\;Fe^{++},\;5{\mu}g\;biotin,\;and\;100{\mu}g$ thiamine HCl per liter were effective. Maximum biomass yield was 2.5g/l in Acinetobacter sp. and 4.8g/l in Pseudomonas sp. 3. Protein composition of the two strains exhibited that alkai-labile protein was higher than alkali-stable protein. In Pseudomonas sp., the contents of acid soluble fraction and alkali-stable protein of the cells grown in the methanol medium were higher than in sucrose medium. On the other hand, in Acinetobacter sp., alkalilabile protein of the cells grown in sucrose medium was higher than in methanol medium.
