Endoparasites of Small Mammals in Edo State, Nigeria: Public Health Implications

  • Some small mammals occur as household pests and harbour a number of parasites that could be of public health importance. This study profiled the helminth and protozoan parasites in trapped small mammals within and around human dwelling places (houses) located across 4 major towns (Auchi, Benin, Ekpoma, and Uromi) and environs in Edo state, Nigeria. Six genera (Apodemus sp., Crocidura sp., Mastomys natalensis, Mus musculus, Rattus sp., and Sorex sp.) were identified from 502 trapped small mammals. Overall, M. musculus (71.9%) and Rattus rattus (20.1%) were the most frequently trapped. In total, on examination of blood, gastrointestinal contents, and brain tissues, 12 helminth taxa (Angiostrongylus sp., Aspicularis sp., Capillaria sp., Gongylonema sp., Heterakis spumosa, Hymenolepis diminuta, Hymenolepis nana, Mastophorus muris, Moniliformis moniliformis, Nippostrongylus brasiliensis, Strongyloides sp., Trichosomoides sp., and Trichuris sp.), and 6 protozoan parasites (Babesia sp., Trypanosoma lewisi, Plasmodium sp., Eimeria sp., Isospora sp., and Toxoplasma gondii) were isolated. Most prevalent helminths with relatively heavy mean intensity were Strongyloides sp. and Heterakis spumosa, while Plasmodium, Eimeria, and Isospora were the most prevalent protozoan parasites. Generally, intrinsic factors like sex and age had marginal influence on the rate and burden of infection in M. musculus and R. rattus. Although the infection rate and prevalence of zoonotic parasites were low, they were largely recovered in rodents from Ekpoma. This study elucidates the public health implication of the presence of zoonotic parasites in these small mammals.

Siphonostomatoid Copepoda (Crustacea) Associated with Invertebrates from Tropical Waters

  • Siphonostomatoid copepods associated with marine invertebrates are described from tropical waters of the West Indies and Madagascar. They belong to the families Asterocheridae (7 new genera and 39 new species), Dinopontiidae (one new species), and Nanaspididae (one new species). New taxa of the Asterocheridae are 14 species of Asterocheres (Asterocheres unioviger n. sp., A. trisetatus n. sp., A. bahamensis n. sp., A. tricuspis n. sp., A. plumosus n. sp., A. peniculatus n. sp., A. oricurvus n. sp., A. planus n. sp., A. sensilis n. sp., A. indivisus n. sp., A. nudicoxus n. sp., A. tenuipes n. sp., A. galeatus n. sp., and A. fastigatus n. sp.); 3 species of Hetairosynella n. gen. (Hetairosynella bifurcata n. sp.; H. angulata n. sp.; H. aculeata n. sp.); 15 species of Asteropontius (Asteropontius capillatus n. sp., A. membranulatus n. sp., A. plumatus n. sp., A. parvipes n. sp., A. humesi n. sp., A. angulatus n. sp., A. latioriger n. sp., A. magnisetiger n. sp., A. pinnatus n. sp., A. trifilis n. sp., A. orcafer n. sp., A. bilinguis n. sp., A. dentatus n. sp., A. minutisetiger n. sp., and A. bispinifer n. sp.); Collocherides minutus n. sp.; Cyclocheres sensilis n. gen. n. sp.; Stenomyzon edentatum n. gen. n. sp.; Cephalocheres flagellatus n. gen. n. sp.; Humesimyzon pusillum n. gen. n. sp.; Thermocheres validus n. gen. n. sp.; and Gascardama longisiphonata n. gen. n. sp. New taxa of the remaining two families are Stenopontius spinulatus n. sp. in the Dinopontiidae and Stephopontius ahni n. sp. in the Nanaspididae. Asterocheres crinoidicola Humes, Asteropontius genodon Stock, and Asteropontius ungellatus Stock are redescribed. Asteropontius gonioporae Kim is reported as a new record from Madagascar. A key to species of Asteropontius is provided.

  • For the production of glucose-1-phosphate from sucrose, bacteria having sucrose phosphorylase were isolated from Kimchi. Among them, JS-05, newly isolated strain having high activity of sucrose phosphorylase was selected and identified as Leuconostoc sp. The specific activity of sucrose phosphorylase of Leuconostoc sp. JS-05 was the highest when the strain was cultured at 25C for 20 hrs in the medium (pH 7.5) containing 10 g sucrose, 5g corn steep liquor, and 2.5g yeast extract per liter.

Characteristics of Biosurfactant Producing Pseudomonas sp. Z1 (생물 계면활성제를 생산하는 Pseudomonas sp. Z1의 특성)

  • One hundred forty five bacterial colonies which were able to degrade crude oil were isolated from soil samples that were contaminated with oil in the Daejon area. Among these colonies, one bacterial strain was selected for this study based on its low surface tension ability, and this selected bacterial strain was identified as Pseudomonas sp. Z1 through physiological-biochemical tests and analysis of its 16S rRNA sequence. Pseudomonas sp. Z1 showed a high resistance to antibiotics such as chloramphenicol and ampicillin, as well as heavy metals such as lithium, manganese, and barium. It was found that the optimal pH and temperature for biosurfactant production of Pseudomonas sp Z1 were pH 6.0-7.0 and $30^{\circ}C$, respectively. After ten hours of inoculation, the biosurfactant activity of the culture broth decreased rapidly, and had maximum surface tension (28 dyne/cm) after twenty-one hours incubation. The biosurfactant activity of the culture broth was also decreased up to 2% NaCl concentration.

  • The hydrolysis of biochemical kinetic resolutions for 2-[1-(acetoxy-2, 2, 2-trifluoroethyl)]-5-trimethylsilylfuran(1) using kinds of Lifase(talpase, paratase, M1000L, SP398, SP435) was investigated. In the results SP435 of Novo Nordish comp any was confirmed to show the capacity of biochemical kinetic resolutions, and 2-(1-(2, 2, 2-trifluoro-1-hydroxyethyl)]-5-trimethylsilylfuran(2) to have the optical purity expressed as 92.4% enantiomeric excess(ee) was produced in this studies.

Physicochemical properties of granular formulation using Paenibacillus sp. AC-1 as a microbial fungicide (미생물 살균제 Paenibacillus sp. AC-1 입제의 물리화학적 특성)

  • In order to commercialize Paenibacillus sp. AC-1 and to minimize its harmful side effects, four granular formulations were prepared using AC-1 powder, adjuvant, and carrier and then their physicochemical properties of the formulations were investigated. Out of the carriers tested, the best one was talc for the formulation. Viable cells was stabilized during the formulating process. Viable cells in the granules formulated with Paenibacillus sp. AC-1 powder were stabilized at storage temperature range ($4{\sim}50^{\circ}C$) after 12 weeks. The release rate of viable cells from granules into water under a static condition were eluted over 90% in 7 hours and breakdown rates of particle were 100% in 1 day. Among the tested formulations, granular formulation comprising of 20% of Paenibacillus sp. AC-1 powder, 7% of polycarboxylate as surface active agent, 1% of sodium polyacrylate as adjuvant, the rest as carrier showed to be best.

Association between the Human Surfactant Protein-A(SP-A) Gene Locus and Chronic Obstructive Pulmonary Disease in Korean Population (한국인에서 만성폐쇄성폐질환과 인체 폐 표면 활성제 단백-A 유전자 대립형질의 상관관계)

  • Backgrounds: This study investigated whether or not a polymorphism in the gene encoding the surfactant protein A(SP-A) has any bearing on the individual susceptibility to the development of chronic obstructive pulmonary disease(COPD) in a genetically homogenous Korean population. Methods: The genotypes of 19 COPD patients and 20 healthy neonates as controls were tested using a polymerase chain reaction followed by restriction fragment length polymorphism analysis for the SP-A gene. Results: The specific frequencies of the 6A2 and 6A18 alleles of SP-A1 and the 1A2 allele of SP-A2 were much higher in the COPD group than control group (p<0.05). However, the frequencies of the 6A3 and 6A4 alleles of SP-A1 and the 1A0 allele of SP-A2 in the COPD group were significantly lower than the control group. In the COPD group, the frequencies of the +50 locus genotypes GG of SP-A1 and the +9 locus genotypes CC of SP-A2 were 85.0% and 60.6%, respectively, and 19.7% and 24.8% in the control group, respectively. The frequencies of the polymorphic genotypes or alleles showed a statistically significant difference between the COPD group and the control group (P<0.05). Conclusion: A genetic polymorphism in SP-A is associated with the development of COPD in the Korean population.

Effect of Different Biosynthetic Precursors on the Production of Nargenicin $A_1$ from Metabolically Engineered Nocardia sp. CS682

  • Nargenicin $A_1$ is a 28-membered polyketide macrolide, with antibacterial activity against methicillin-resistant Staphylococcus aureus, produced by Nocardia sp. CS682. In this study, the production of nargenicin $A_1$ was improved by enhancing the supply of different biosynthetic precursors. In Nocardia sp. CS682 (KCTC11297BP), this improvement was ~4.62-fold with the supplementation of 30 mM methyl oleate, 4.25-fold with supplementation of 15mM sodium propionate, and 2.81-fold with supplementation of 15 mM sodium acetate. In Nocardia sp. metK18 and Nocardia sp. CS682 expressing S-adenosylmethionine synthetase (MetK), the production of nargenicin $A_1$ was improved by ~5.57-fold by supplementation with 30 mM methyl oleate, 5.01-fold by supplementation with 15 mM sodium propionate, and 3.64-fold by supplementation with 15 mM sodium acetate. Furthermore, supplementing the culture broth of Nocardia sp. ACC18 and Nocardia sp. CS682 expressing the acetyl-CoA carboxylase complex (AccA2 and AccBE) with 30 mM methyl oleate, 15 mM sodium propionate, or 15 mM sodium acetate resulted in ~6.99-, 6.46-, and 5.58-fold increases, respectively, in nargenicin $A_1$ production. Our overall results showed that among the supplements, methyl oleate was the most effective precursor supporting the highest titers of nargenicin $A_1$ in Nocardia sp. CS682, Nocardia sp. metK18, and Nocardia sp. ACC18.

Biocontrol of Anthracnose of Chili Pepper by Bacillus sp. NAAS-1 (Bacillus sp. NAAS-1을 이용한 고추 탄저병 생물학적 방제)

  • Bacillus sp. NAAS-1 isolated from the field of Chili pepper was tested for biocontrol activity against anthracnose pathogen of Chili pepper caused by Colletotrichum acutatum. The antifungal activity of Bacillus sp. NAAS-1 culture broth was compared with synthetic fungicide containing carbendazim (40%) and kasugamycin (3.45%). Bacillus sp. NAAS-1 showed a similar fungicidal activity against the anthracnose pathogen at the concentration of 50 ${\mu}L/mL$ in comparison to the fungicide containing carbendazim (40%) and kasugamycin (3.45%) using a cup method. Bacillus sp. NAAS-1 also exhibited its potent fungicidal activity against the anthracnose in vivo test at the concentration of 50 ${\mu}L/mL$ when compared to the fungicide containing carbendazim (40%) and kasugamycin (3.45%).

Human selenium binding protein-1 (hSP56) is a negative regulator of HIF-1α and suppresses the malignant characteristics of prostate cancer cells

  • In the present study, we demonstrate that ectopic expression of 56-kDa human selenium binding protein-1 (hSP56) in PC-3 cells that do not normally express hSP56 results in a marked inhibition of cell growth in vitro and in vivo. Down-regulation of hSP56 in LNCaP cells that normally express hSP56 results in enhanced anchorage-independent growth. PC-3 cells expressing hSP56 exhibit a significant reduction of hypoxia inducible protein (HIF)-$1{\alpha}$ protein levels under hypoxic conditions without altering HIF-$1{\alpha}$ mRNA (HIF1A) levels. Taken together, our findings strongly suggest that hSP56 plays a critical role in prostate cells by mechanisms including negative regulation of HIF-$1{\alpha}$, thus identifying hSP56 as a candidate anti-oncogene product.
