Differences in Chemical Composition of Soybean Seeds with Different Usages and Storage Durations (저장기간과 용도가 다른 대두종실의 화학성분 차이)

  • For the comparision of the differences in dry matter weight and the chemical composition of soybean seeds in relation to utilization styles and storage durations, the seed coats and cotyledons including embryo axis separated from the seeds of 3 recommended cultivars stored for 3 years and of 5 recommended cultivars and 2 local varieties with black seed coat and green cotyledons were observed in dry weight, germination percentage, protein, oil, soluble sugar, oilgosaccharide, and starch. In the seeds stored for 3 years, dry weight and germination rate were decreased distinctively. This decrease of seed dry matter resulted from the decrease of the cotyledon weight, and the ratio of seed coat to whole seed was increased. For the change of chemical compositions by the decrement of seed dry weight during the storage, protein, oil, and carbohydrate contents of the seed coat were decreased, and protein content of the cotyledon was markedly decreased in the seeds stored for 3 years. The ratio of seed coat to whole seed was higher in small seed than large seed cultivars. In the local varieties, protein and oil contents were 43.59% and 18.25% in the cotyledons, respectively, which were lower than those of recommended cultivars. But starch and sugar contents were 5.70% and 11.58%, respectively, which were higher than those of recommended cultivars. Sucrose, raffinose and stachyose contents were also higher in the cotyledon of the local varieties.

Survey on Seed-borne Fungi in Some Important Crops of Korea (한국산(韓國産) 주요작물(主要作物)의 종자전염성(種子傳染性) 진균(眞菌)에 관(關)한 조사(調査))

  • One hundred and sixteen seed samples of 16 different crops were tested by routine seed health testing methods. Seeds of rice, soybean and barley were found to be associated with 18, 15 and 7 species of fungi, respectively. Discoloration and spotting of various kinds were observed in rice seeds. Epicoccum purpurascens, Pyricularia oryzae and Trichoconiella pedwickii were higher in these discoloured than in normal seeds of rice. Macrophomina phaseolina, the charcoal-rot fungus is a new record for soybean in Korea. Seeds of sesame were infected by 13 fungi Alternaria sesami, A. sesarmicola and Corynespora cassiicola were detected predominantly. Twenty one species of fungi were detected in seed samples of 12 commercial vegetable crops. Alternaria, Colletotrichum and Fusarium were found predominantly. Some seed samples of commercial vegetable crops were heavily infected with pathogenic fungi and showed heavy seed and seedling rot.

Effect of Methiocarb as a Bird Repellent in Water-Seeding Rice and Soybean Fields (벼 담수직파 및 콩 재배시 Methiocarb 종자 분의에 의한 새 피해 경감 효과)

  • The bird repellent, methiocarb 50% WP, has been used to reduce the bird damage in the crop field in the European countries. The bird damage occaisonally would occur in the wet direct seeded rice and in soybean field, and resulted in decreasing the crop productivity by the reduction of seedling emergence rate. In this experiment, rice seeds, Hwasungbyeo(Oryza sativa), were coated with the different application rate of methiocarb, 5, 10 and 15 per kg seed, and soybean, Taegwangkong(Glycine max), 2.5, 5.0 and 7.5g. In rice, the seeds coated with 10 and 15g of methiocarb were not lost by bird, while those with 5g and control were lost to 37 and 50% of total seeds, respectively. No damage by birds was observed in rice seedling when the coleoptile and radicle of rice were emerged at 7 days after the water seeding. The crop injury of methiocarb reducing the emergence rate of seedlings, shortening the shoot length, and decreasing the leaf number was occurred at the treated of methiocarb 15g per kg seed. In soybean, the loss of the sprouting by birds was lower in the treatment of methiocarb 7.5g per kg seed than that in both the control and the treatments of methiocarb, 2.5 and 5.0g per kg seed. No crop injury by the treatment of methiocarb was observed in all application rate.

Water Uptake and Germination of Soybean Seed as Affected by Soaking Condition (침지조건에 따른 콩 종실의 수분흡수율 및 발아특성)

  • Varietal difference in seed shape and size, water absorption rate, and soybean sprouts was compared. In general, soybean seeds of yellow seed-coat and spherical shape with 100 seeds of 9.0$\pm$2g were evaluated as the best ones; Pungsannamulkong out of tested varieties was considered to be the best one in these respect. Varietal difference in water absorption yale depending on the soaking duration and temperature was recognized; Jungeri and Jillin 3 showed higher water absorption rate at higher temperature but it was completely vice versa for Pungsannamulkong. It took about 15 hours(soaking at 2$0^{\circ}C$) for seed shape of size to grow to 10mm in length. Maximum enlargement in thickness of soybean seeds was made right after the completion of repeated soaking-drying treatment but three hours and nine hours soaking were needed for one time-soaking and non-soaking treatment, respectively. Varietal difference in germination rate was recognized between one hour's soaking at 2$0^{\circ}C$ and two hour's soaking at 15$^{\circ}C$ The growth rate for Jillin 3 was excellent at all soaking methods and temperatures while three hours of soaking was the best for Jungeril and Pungsannamulkong, regardless of soaking temperatures. The growth of hypocotyl length showed somewhat faster in repeated soaking-drying than one time-soaking and non-soaking while the increase of hypocotyl thicknes was better in one-time soaking than repeated soaking-drying.

Tillage practices and fertilization effects on growth and nitrogen efficiency in soybean

  • A field experiment was performed to evaluate the effects of tillage systems and fertilizer management on yield and nutrient uptake in Soybean. The plant height, fresh weight and dry weight of conventional tillage were much higher those observed for no-tillage. Significant differences in plant height were observed under tillage practices combined with fertilizer treatment. However, the greatest plant height (128.47 cm) was observed in conventional tillage with chemical fertilizer, and the lowest (45.4 cm) was observed in the no-tillage with green manure treatment. The highest fresh weight (172.4 g) and dry weight (44.1 g) were observed from the no-tillage chemical treatment in the late flowering stage of soybean. The plant concentration of nitrate was higher (2.29%) in no-tillage with green manure than it was with chemical fertilization. However, nitrogen increased steadily in all treatments, and the highest quantity of total nitrogen (476.7 Kg/ha) was observed in no-tillage with green manure. The N content in the soil decreased gradually just after the vegetative stage. Tillage practices and additional fertilizer application had an adverse effect on the uptake of N, P and K in soybean seeds. The nitrogen concentration in seeds was found to be increased in the no-tillage with green manure treatment. The uptake of more nitrogen induced a better yield. Thus, the no-tillage with green manure treatment had the greatest yield, although no significant difference was observed among foliar-applications and additional fertilization. Additionally, the phosphorus and potassium concentrations in seeds remained the same between the conventional tillage and no-tillage treatments. The results obtained in this study indicate that no-tillage strategies with fertilizers may influence the growth characteristics and mineral uptake in soybean.

TFWT and OBT Concentrations in Soybean Plants Exposed to HTO Vapor at Different Growth Stages (콩의 생육단계별 HTO 증기 피폭에 따른 작물체내 TFWT 및 OBT 농도)

  • Soybean plants were exposed to HTO vapor in an exposure box for 1 hour at different growth stages. Relative concentrations of TFWT at the end of exposure (percent ratios of TFWT concentrations to mean HTO concentrations in air moisture in the box during exposure) decreased on the whole in the order of leaf > shell > seed > stem with the highest values of 40.2% and 6.4% for leaf and stem, respectively. TFWT concentrations reduced by factors of several thousands to several hundred-thousands from the end of exposure till the harvest. The reduction factor decreased in the order of leaf > shell > seed > stem. Relative OBT concentrations at harvest (ratios of the OBT concentration in the dry plant part at harvest to the initial leaf TFWT concentration, ml $g^{-1}$) were in the range of $2.2{\times}10^{-5}{\sim}9.5{\times}10^{-3}$ for seeds being the highest when the exposure was performed at the actively seed-developing stage. The exposure time-dependent variation in the OBT concentration was much greater in seeds and shells than in leaves and stems. It was indicated that OBT would contribute to almost all the radiation dose due to the consumption of soybean seeds in most cases after an acute exposure of growing plants to HTO vapor. Present results are applicable to establishing and validating soybean $^3H$ models for an acute accidental release of HTO.

Soil-to-Plant Transfer Factors of $^{99}Tc$ for Korean Major Upland Crops (우리나라 주요 밭작물에 대한 $^{99}Tc$의 토양-작물체 전이계수)

  • In order to investigate the soil-to-plant transfer factor (TF) of $^{99}Tc$ for Korean major upland crops (soybean, radish and Chinese cabbage), pot experiments were performed in a greenhouse. Soils were collected from four upland fields (two for soybean and two for radish and Chinese cabbage) around Gyeongju radioactive-waste disposal site. Three to four weeks before sowing, dried soils were mixed with a $^{99}Tc$ solution and the mixtures were put into pots and irrigated. TF values were expressed as the ratios of the $^{99}Tc$ concentrations in plants (Bq $kg^{-1}$-dry or fresh) to those in soils (Bq $kg^{-1}$-dry). There was no great difference in the TF value between soils. The TF values for soybean seeds were extremely lower than those for the straws, indicating a very low mobility of $^{99}Tc$ to seeds. As representative TF values of $^{99}Tc$, $1.8{\times}10^{-1}$, $1.2{\times}10^1$, $3.2{\times}10^2$ and $1.3{\times}10^2$ (for dry plants), arithmetic means for two soils, were proposed for soybean seeds, radish roots, radish leaves and Chinese cabbage leaves, respectively. In the case of the vegetables, proposals for fresh plants were also made. The proposed values are not sufficiently representative so successive updates are needed.

Efficient Transformation Method of Soybean Using Meristematic Tissues of Germinating Seeds (발아종자의 분열조직을 이용한 효율적인 콩 형질전환 방법)

  • An efficient transformation method for soybean [Glycine max (L.) Merr.] using meristematic tissues of germinating seeds has been established. The embryonic axes were excised from germinating seeds of Korean soybean cultivar, Iksannamulkong and 0.5-2 cm long segment containing meristematic tissues were prepared by cutting hypocotyl region. The explants were inoculated with Agrobacterium tumefaciens strain LBA4404 harboring a binary vector with the bar gene as a selectable marker gene and a ${\beta}-glucuronidase$ (GUSINT) reporter gene, and then co-cultured for 7 days on co-cultivation medium (CCM). The meristematic tissues were cultured on shoot induction medium (SIMP6) supplemented with 0.4 mg/l $N_6-benzylaminopurine$ (BAP) and 0.1 mg/l indolebutyric acid (IBA) in the presence of 6 mg/l L-phosphinotricin (PPT) for 2 weeks and the surviving explants were transferred to shoot elongation medium (SEMP6). Transformation was confirmed by Southern blot analysis and the transformation efficiencies ranged from 1.48 to 2.07%. The new modified transformation method was successfully implemented for obtaining several transgenic lines with SMV-CP gene. It is expected that this method could efficiently be used for the transformation of recalcitrant soybean cultivars.

Characterization of Isoflavones from Seed of Selected Soybean (Glycine max L.) Resources Using High-Resolution Mass Spectrometry (고해상도 질량 분석을 이용한 대두(Glycine max L.) 우수자원 종자의 이소플라본 특성 평가)

  • In this study, chemical information on a total of 20 individual compounds was constructed to identify isoflavones from the previous reports related with used parts(seeds, leaves, stems, pods) and products of soybean(Glycine max L.). Through constructed library and UPLC-DAD-QToF/MS analysis, a total of 19 individual isoflavones including aglycones, glucosides, acetylglucosides and malonylglucosides as major compounds was identified and quantified from 14 selected soybean seeds. Among them, genistein 7-O-(2"-O-apiosyl)glucoside and genistein 7-O-(6"-O-apiosyl)glucoside(ambocin) were identified tentatively as novel compounds in soybean seeds. Besides, among malonylglucosides, glycitein 4'-O-(6"-O-malonyl)glucoside was estimated for the first time. Total isoflavone contents were distributed from 240.21 to 445.21(mg/100 g, dry matter) and 7-O-6"-O-malonylglucosides were composed of 77.8% on total isoflavone as well as genistein derivatives were confirmed as major class. It was considered importantly that the development of isoflavone-rich varieties was necessary to strengthen their effects such as anti-inflammation, anti-cancer and menopause mitigation. The qualitative and quantitative data presented precisely in this study could be help to select and breed isoflavone-rich varieties. Furthermore, their basic isoflavone profile is expected to be applied to estimate the change of isoflavone conjugates on bioavailability after soy food supplements.

Yield Loss Assessment and Determination of Economic Thresholds Limits against Soybean Anthracnose (콩탄저병의 피해 해석 및 요방제 수준 설정)

  • A field investigation was carried out for two years to analyze yield loss due to soybean anthracnose caused by Colletotrichum truncatum and to determine its economic threshold limit. Anthracnose severity in terms of % diseased pods was negatively correlated with yield, number of normal seeds per plant and number of pods per plant, and positively correlated with % abnormal seeds with correlation coefficients of -0.85, -0.78, -0.64, and 0.80, respectively. A simple linear regression model was obtained as Y=-1.7781X+164.22 with $R^2$=0.8092, when the soybean yields (Y) were predicted using anthracnose severity (X) as an independent variable. The yield levels could be predicted as high as 80.92%. Based on this equation, spray threshold without economic considerations was estimated as 6.9 in % pods infected with anthracnose. Economic threshold limit and economic spray threshold able to compensate the costs of fungicide sprays were determined as 11.9% and 9.5%, respectively.
