Identification of Quantitative Trait Loci Associated with Isoflavone Contents in Soybean Seed

  • Soybean seeds contain high amounts of isoflavones that display biological effects and isoflavone content of soybean seed can vary by year, environment, and genotype. Objective of this study was to identify quantitative trait loci that underlie isoflavone content in soybean seeds. The study involved 85 $F_2$ populations derived from Korean soybean cultivar 'Kwangkyo' and wild type soybean 'IT182305' for QTL analysis associated with isoflavone content. Isoflavone content of seeds was determined by HPLC. The genetic map of 33 linkage groups with 207 markers was constructed. The linkage map spanned 2,607.5 cM across all 33 linkage groups. The average linkage distance between pair of markers among all linkage groups was 12.6 cM in Kosambi map units. Isoflavone content in $F_2$ generations varied in a fashion that suggested a continuous, polygenic inheritance. Eleven markers (4 RAPD, 3 SSR, 4 AFLP) were significantly associated with isoflavone content. Only two markers, Satt419 and CTCGAG3 had F-tests that were significant at P<0.01 in $F_2$ generation for isoflavone content. Interval mapping using the $F_2$ data revealed only two putative QTLs for isoflavone content. The peak QTL region on linkage group 3, which was near OPAG03c, explained $14\%$ variation for isoflavone content. The peak QTL region on linkage group 5, which was located near OPN14 accounted for $35.3\%$ variation for isoflavone content. Using both Map-Maker-QTL $(LOD{\geq}2.0)$ and single-factor analysis $(P{\leq}0.05)$, one marker, CTCGAG3 in linkage group 3 was associated with QTLs for isoflavone content.   • The present study was carried out to determine the effect of a radionuclide, cesium-137, in soybean, which is an element released usually from nuclear facilities. Soybean plants were grown on the pots treated with cesium-137 $0.5{\sim}60{\mu}Ci/1kg$ soil and the uptake, translocation and accumulation of the radiocesium in the plant parts were measured at different growth stage. The results are summarized as follows: 1) Visual toxic symptoms on the plants due to treatment of radioactive cesium were not observed up to $60{\mu}Ci/10Kg$ soil in a pot. 2) The uptake of cesium-137 in soybean plant was increased with increment of concentration applied, while the uptake of potassium was proportionally decreased, indicating to have an ion antagonistic relationship between them. 3) The absolute amounts of cesium-137 in the plants were gradually increased by the pod setting stage, but rather reduced at harvesting stage. The accumulation occurred more in the leaves and stems than the soybean seeds. 4) The rate of uptake was ranged from 0.069 to 0.005 with proportional decrease by increasing concentration applied and the rate of Cs-137 translocation from plants to seeds was averaged 38.6% in soybean plant. The concentration coefficient was 0.04 in the soybean seeds from the pots treated with $20{\mu}Ci$ of cesium-137 and decreased with increment of cesium-137 applied.

Lipoxygenase Inhibitor from Defatted Nutmeg Seed

  • Lipoxygenase inhibitory acitivity of the methanolic extract of 60 different kinds of plant seeds was determined by a spectrophotometric method using a soybean lipoxigenase(SLO) and linolenic acid. Among the extracts examined, the methanolic extract of nutmeg(Myristical fragrans)seed showed the most potent SLO inhibitory activity. To isolate SLO inhibitor, hence, the defatted methanol extract was further partitioned with ether, ehtylacetate , and n-butanol , stepwise. The ether souble fraction was successively chromatographed on silica gel, Sephadex LH-20 and preparative TLC. Three phenolic compounds were isolated , and one of them showing a strong SLO inhibition activity was identified as a 2,6-dihydroxy-9-(3', 4', -dihydroxyphenyl)nonylphenone (IC50a=0.39$\mu\textrm{g}$/ml) by 1H-& 13C0NMR, IR, and MS spectroscopy.

Effects of Leaf and Pod Removal on Assimilate Translocation in Soybean Plants (적엽 및 제협처리가 콩의 동화물질 전류에 미치는 영향)

  • Effects of leaf and pod removal on changes in dry weight and on the contents of soluble sugar, starch, protein and oil in leaves and seeds of soybean [Glycine max(L.) Merr.] cultivar ‘Hwangkeumkong’ were measured at the research farm of Korea University in 1992. The upper 40% and lower 60% of leaves and pods were subjected to treatments at the growth stage of beginning pod(R3). Upper leaf-lower pod removal showed the highest leaf and the lowest seed dry weights. Soluble sugar content was no different among treatments in leaves and seeds. The highest starch content was found in leaves of upper leaf-lower pod removal. Protein content was higher in lower leaves than upper leaves and the lowest in seeds of lower leaf-upper pod removal which had the highest oil content in leaves and seeds. These results apparently indicated that photoassimilates were mobilized from upper leaves to lower seeds, and protein sources were moved from lower to upper parts but weak in remobilization from leaves for the long distance translocation during the reproductive growth period.

  • Collected 38 varieties of colored-soybean seed cultivated under a given cultural conditions were tested in the various chemical compositions and rheological properties in seeds. As it were, protein, fat, carbohydrate, ash and water contents as a chemical contents were studied not only on their statistical variations, but also on interrelationships with their collaborated rheological characteristics; hardness, cohesiveness, elasticity, gumminess and chewiness. For the rheological analysis of the materials, the General Foods Texturometer and typical texturometer curve were used.

Seed Quality of Soybean Produced from Upland and Drained-Paddy Field

  • This study was carried out to evaluate the seed quality of soybeans produced from upland and drained-paddy fields. Soybeans from drained-paddy field showed significantly higher in the 100 seeds weight and greater in the size of seed length than those from upland fields. However, there are no significant differences in seed width and thickness between upland and paddy fields. In case of Hunter's color value, the lightness (L) was significantly higher in the upland soybeans, but the a (redness) and b (yellowness) values were higher in the drained-paddy field soybeans. Seed appearance of drained-paddy field was poor than that of upland field. Soybeans produced from the drained-paddy field showed higher protein content, whereas, lipid and ash contents were higher in the upland field. Soybeans from upland field had lower contents of total amino acids compared to drained-paddy fields. No statistical differences were found in palmitic, linoleic, and linolenic, but stearic, oleic, saturated fatty acids (SFA), and unsaturated fatty acids (USFA) showed significant differences between soybean seeds from upland and drained-paddy fields. Genistein content was higher in the drained-paddy fields, while daidzein and glycitein contents were higher in the upland fields. This result suggested that the soil condition of drained-paddy field is more favorable to synthesis genistein than daidzein and glycitein.

Identification of Soybean Glycinin Precursor In Vitro (대두 세포내에서 Glycinin 전구체의 존재 확인)

  • Glycinin is the major storage protein in soybean. It has been known that a molecule of glycinin is composed of 6 subunits, each of which consists of two different kinds of polypeptides, acidic (A) and basic (B) one (NW 39K and 19K, respectively). To study the molecular origin and the relationship of glycinin subunit polypeptides, antibodies against A-and B-polypeptide were obtained by immunizing rabbits with either of the antigens purified by gel filtration and preparative electrophoresis. Each antibody was not only specific for its own antigen polypeptide in soybeans but also recoginzed the precursor which was synthesized in vivo and in vitro. The polyadenylated mRNAs were isolated from immature seeds and leaves and were translated in vitro using wheat germ extract. One of the seed-specific translation products. MW 60K, was identified to be the precursor of glycinin subunit by immunoprecipitation with antibodies against glycinin A- and B-polypeptide. Mature A- and B-polypeptides were not detected in the translte in vitro. These results suggest that the precursor polypeptide is synthesized from the mRNA and is cleaved to yield A- and B-polypeptides which from a glycinin subunit in the cell. Glycinin genes were expressed with the maturation of soybean seeds in a tissue-specific and developmental stage-specific manner.

Plant Protective and Growth Promoting Effects of Seed Endophytes in Soybean Plants

  • Seed-borne diseases reduce not only the seed germination and seedling growth but also seed quality, resulting in the significant yield loss in crop production. Plant seed harbors diverse microbes termed endophytes other than pathogens inside it. However, their roles and application to agricultures were rarely understood and explored to date. Recently, we had isolated from soybean seeds culturable endophytes exhibiting in-vitro antagonistic activities against common bacterial and fungal seed-borne pathogens. In this study, we evaluated effects of seed treatment with endophytes on plant growth and protection against the common seed-borne pathogens: four fungal pathogens (Cercospora sojina, C. kikuchii, Septoria glycines, Diaporthe eres) and two bacterial pathogens (Xanthomonas axonopodis pv. glycines, Pseudomonas syringae pv. tabaci). Our experiments showed that treatment of soybean seeds with seed endophytes clearly offer protection against seed-borne pathogens. We also found that some of the endophytes promote plant growth in addition to the disease suppression. Taken together, our results demonstrate agricultural potential of seed endophytes in crop protection.
