An infectious virus isolated from soybeans (대두위축병원 바이러스에 관한 연구)

  • Soybean stunt virus (SSV) was newly isolated in Korea from naturally infected soybeans (Glycine max). The main symptoms caused by this virus on soybean cultivars are crinkling, mild mottling and reduction in plant size. This virus induced local lesion on the inoculated leaves of Chenopodium amaranticolor, C quinoa and Vigna sinensis, and mosaic symptoms on Nicotiana tabacum (Bright yellow, KY-57). The virus was inactivated at 60C, and was infectious at dilution of $10^3$. Extract juice became infective 3 days later at room temperature. The virus was transmitted by green peach apid (Myzus persicae). This virus closely is related serologically to cucumber mosaic virus. The virus particles observed in the electron microscopy were spherical types of 30mm in diameter.

Effect of Defoliation on the Grain Yield of Two Soybean Cultivars Grown under Different Population Densities (적엽처리가 재식밀도를 달리한 대두품종의 수량 및 수량구성요소에 미치는 영향)

  • Defoliation effect of two soybean cultivars on the grain yield was studied under different planting densities planted at two different planting date. Leaf removal caused a linear decrease of grain yield with increased amounts of defoliation in the case of lower population densities, while in the case of higher population densities over 40 plants per sqare meter, the slight removal of lower leaves indicated increased grain yields by 13% to 35% compared with control depending on the cultivar and planting time. The pod number per plant was the most effective factor influencing grain yield. The grain weight and the percent of matured grain become more important components for higher yield with decreased population densities depending on cultivar.

Sensitive and Pathovar-Specific Detection of Xanthormonas campestris pv. glycines by DNA Hybridization and Polymerase Chain Reaction Analysis

  • Xanthomonas campestris pv. glycines causes bacterial pustule disease on susceptible soybean leaves and produces a bacteriocin, named glycinecinA, against most xanthomonads including Xanthomonas campestris pv. vesicatoria. One of the 5 isolated DNA regions responsible for bacteriocin production, a 1.7 kb DNA region for the glycinecinA gene, was used as a probe to detect the presence of the homolog DNA in other bacterial strains. Among 55 bacterial strains tested, only X. campestris pv. glycines showed the positive signal with glycinecinA DNA. Two oligomers, heu2 and heu4, derived from a glycinecinA DNA were used to carry out the polymerase chain reaction (PCR) analysis with chromosomal DNA from 55 different bacterial strains including 24 different strains of X. campestris pv. glycines, 9 different pathovars of xanthomonads, and other 22 bacterial strains of different genus and species. By separation of the PCR products on agarose gel, a 0.86 kb DNA fragment was specifically detected when X. campestris pv. glycines was present in the amplification assay. The 0.86 kb fragment was not amplified when DNA from other bacteria was used for the assay. Southern analysis with glycinecinA DNA showed that the PCR signal was obtained with X. campestris pv. glycines isolates from various geographic regions and soybean cultivars. Therefore, the 1.7 kb DNA region for the glycinecinA gene can be used for the pathovar-specific probe for the DNA hybridization and the primers heu2 and heu4 can be used for the pathovar-specific primers for the PCR analysis to detect X. campestris pv. glycines.

Variation of Major Characters in Soybean Varieties II. Effects of Day Length (대두품종의 주요특성 변이 II. 일장조건의 영향)

  • These experiments were conducted to investigate the effect of day length on some chracteristics and seed weight in the soybean cultivars. Those were classified into three type's of seed weight with delaying seeding date in the previous paper. Flowering was delayed as the longer day length was employed. When the length became longer. the number of leaves at flowering greatly length of main stem and the number of nodes at maturity greatly increased. The seed weight decreased when the day length was longer than 14 hours. Negative correlation was obtained between seed weight and day length. There was negative correlation between seed weight and the number of days required to flowering, and length of main stem and the number of nodes at maturity. Type I cultivars decreased in seed weight when the day length was longer than 16 hours, but this trend was found to be clear with type II cultivars. Type III cultivars decreased in seed weight with increasing the day length.

Effect of Leaf and Pod Removal on Nodal Sink Characters in Soybean (잎과 꼬투리 제거가 콩의 절위별 Sink 형질 변이에 미치는 영향)

  • The effects of leaf and pod removal on variation of nodal sink components in determinate soybean [Glycine max (L.) Merr.] cultivar 'Danyeobkong' were measured at the experiment field of Chonbuk Provincial Rural Development Administration in 1991. Contrary to the conventional numbering system, node order in this experiment was numbered from top to bottom node. The leaves and pods of main stem from terminal to 5th node, below 6th node, or branches were removed at the growth stage of beginning pod(R3). In the leaf removal treatment, number and weight of pod and seed were highly decreased in upper part leaf removal, especially in removed part. In the pod removal treatment, number and weight of pod and seed were slightly increased in the other part. Cracked seed coat ratios were also high in the leaf removal treatment compared with control but low in the pod removal treatment.

An Investigation of Prepenetration structure by the soybean anthracnose fungus, Glomerella glycines (콩 탄저병균(Glomerella glycines)의 침입 전 구조에 관한 연구)

  • In order to find out the effect of contactor to form appressorial structure of Glomerella glycines, this experiment was carried out by using several contactors such as cover glass. cellophane, vinyl and oil paper respectively. In the case of cover glass placed on a drop of conidial suspension of the fungus which is incubated on water agar for 12 hours, 67.7 percent of appressoria were resulted, whereas no appressorial structure was found in the control. Vinyl was known best physical contactor and cellophane, cover glass and oil paper were fairly good in that order. Effects of temperature, time and relative humidity on the formation of appressoria of G. glycines were similar to each of optimum growth range of temperature and relative humidity $25^{\circ}-30^{\circ}C$ and 70-100 Percent, respectively. In addition, maximum appressorial formation was resulted in the conidial suspension incubated on soybean leaves for 36 hours. No appressorium was found at above $35^{\circ}C$, below R.H. 70 percent and 5 hours incubation.

Inhibitory Effect of Green-Yellow Vegetables on the Mutagenicity in Salmonella Assay System and on the Growth of AZ-521 Human Gastric Cancer Cells (녹황색 채소류의 돌연변이유발 억제 및 AZ-521 위암세포의 성장 저해효과)

  • The antimutagenic effect of green-ye1low vegetables on the mutagenicities induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) and N-nitrosodimethylamine (NDMA) in Salmonella assay system and also their inhibitory effect on AZ-521 human gastric cancer cells were studied. Twenty-four items from twenty six kinds of vegetables(92%) revealed antimutagenic activity toward MNNG (p< 0.0l, 0.05). Perilla leaf, Korean cabbage, cauliflower, lettuce, mustard leaf, water dropwort, small water dropwort, carrot and burdock inhibited the mutagenicity more than 80%. The methanol extracts of the vegetables also showed the antimutagenic activity toward NDMA (p< 0.01, 0.05). Especially, perilla leaf, kale, soybean sprout and onion inhibited more than 80% of the NDMA induced mutagenicity in S. typhimurium TA100. Small water dropwort and perilla leaf exhibited the strong inhibitory effect (97~100%) on the growth of the AZ-521 human gastric cancer cells. Soybean sprout, water dropwort, broccoli, crown daisy, green red pepper, red pepper leaves, spinach, cabbage and sweet potato also inhibited growth of the cancer cells (p < 0.001~0.05).

Survey of the Food Preferences and Nutrient Intakes of Elderly People Hospitalized in a Nursing Hospital in Daegu (대구지역 요양병원 입원노인의 식품선호도 및 영양소 섭취 실태 조사)

  • The number of elderly in care facilities is increasing in parallel with a rapid increase in the elderly population in Korea. We analyzed the meal satisfaction ratings, food preferences, and nutritional intakes of 74 patients aged ≥65 years living in a nursing hospital in Daegu. Food preferences were high for cooked rice, rice gruel, meat soup, soybean paste stew, beef, frozen pollack, squid, egg, spinach, sea mustard, cabbage kimchi, apple, peanut, drinking yogurt, pickled perilla leaves, and salted squid. Preferences for duck (P<0.01) and vegetable (P<0.01) cooking method were significantly different for men and women, and preference for soybean paste stew increased with age (P<0.05). Regarding nutrient intakes, men had higher energy (P<0.01), carbohydrates (P<0.05), and zinc (P<0.05) intakes than women, while women had higher vitamin C (P<0.05) intake than men. Furthermore, energy (P<0.001), carbohydrates (P<0.001), protein (P<0.05), dietary fiber (P<0.01), vitamin B6 (P<0.01), phosphorus (P<0.05), and zinc (P<0.001) intakes significantly decreased with age. Men had a significantly higher nutrient adequacy ratios (NARs) for carbohydrates (P<0.05) and cal- cium (P<0.01) than women, while women had a significantly higher NARs for protein (P<0.05), vitamin A (P<0.01), vitamin C (P<0.05), thiamine (P<0.01), riboflavin (P<0.001), iron (P<0.001), and zinc (P<0.01). Mean adequacy ratios were 0.69 for men and 0.75 for women. In particular, NARs for carbohydrates (P<0.01), vitamin B6 (P<0.01), and calcium (P<0.05) decreased significantly with age. In summary, the study shows that supplementing vitamin C, vitamin B6, calcium, and iron, reducing sodium, and providing meals that reflect preferences will improve the nutritional statuses of elderly residents in a nursing hospital.

Identification of hrcC, hrpF, and maA Genes of Xanthomonas campestris pv. glycines 8ra: Roles in Pathogenicity and Inducing Hypersensitive Response on Nonhost Plants

  • Nonpathogenic mutants of Xanthomonas campestris pv. glycines were generated with Omegon-Kim to isolate genes essential for pathogenicity and inducing hypersensitive response (HR). Three nonpathogenic multants and two mutants showing slow symptom development were isolated among 1,000 colonies tested. From two nonpathogenic mutants, 8-13 and 26-13, genes homologous to hrcC and hrpF of X. campestris pv. vesicatoria were identified. The nonpathogenic mutant 8-13 had a mutation in a gene homologous to hrpF of X. campestris pv. vesicatoria and failed to cause HR on pepper plants but still induced HR on tomato leaves. The nonpathogenic mutant 26-13 had an insertional mutation in a gene homologous to hrcC of X. campestris pv. vesicatoria and lost the ability to induce HR on pepper leaves but still caused HR on tomato plants. Unlike other phytopathogenic bacteria, the parent strain and these two mutants of X. campestris pv. glycines did not cause HR on tobacco plants. a cosmid clone, pBL1, that complemented the phenotypes of 8-13 was isolated. From the analysis of restriction enzyme mapping and deletion analyses of pBL1, a 9.0-kb Eco RI fragment restored the phenotypes of 8-13. pBL1 failed to complement the phenotypes of 26-13, indicating that the hrcC gene resides outside of the insert DNA of pBL1. One nonpathogenic mutant, 13-33, had a mutation in a gene homologous to a miaA gene encoding tRNA delta (2)-isopentenylpyrophosphate transferase of Escherichia coli. This indicated that tRNA modifications in X. campestris pv. glycines may be required for expression of genes necessary for pathogenicity. The mutant 13-33 multiplied as well as the parent strain did in the culture medium and in planta, indicating that loss of pathogenicity is not due to the inability of multiplication in vivo.

Environmental contamination and geochemical behaviour of heavy metals around the abandoned Songcheon Au-Ag mine, Korea

  • The objective of this study is to investigate the contamination levels and dispersion patterns of arsenic and heavy metals and to estimate the bioaccessible fraction of the metals in soil and plant samples in the vicinity of the abandoned Songcheon Au-Ag mine. Tailings, soils, plants (Chinese cabbage, red pepper, soybean, radish, sesame leaves, green onion, lettuce, potato leaves, angelica and groundsel) and waters were collected around the mine site. After appropriate preparation, all samples were analyzed for As, Cd, Cu, Pb and Zn by ICP-AES and ICP-MS. Elevated levels of As and heavy metals were found in tailings. Mean concentrations of As in agricultural soils were higher than the permissible level. Especially, maximum level of As in farmland soil was 513 mg/kg. The highest concentrations of As and Zn were found in Chinese cabbage (6.7 mg/kg and 359 mg/kg, respectively). Concentrations of As, Cd, and Zn in most stream waters which are used for drinking water around this mine area were higher than the permissible levels regulated in Korea. Maximum levels of As, Cd and Zn in stream waters were 0.78 mg/L, 0.19 mg/L and 5.4 mg/L, respectively. These results indicate that mine tailings can be the main contamination sources of As and heavy metals in the soil-water system in the mine area. The average of estimated bioaccessible fraction of As in farmland soils were $3.7\%$ (in simulated stomach) and $10.8\%$ (in simulated small intestine). The highest value of bioaccessible fraction of metal in farmland soils was $46.5\%$ for Cd.
