Early nematode development and subcellular responses in resistant soybean lines PI 88788 and PI 437654 infected with races 3 (R3) and 14 (R14) of soybean cyst nematode (SCN), Heterodera glycines Ichinohe, were compared. SCN R14 nematodes penetrated and developed significantly more than R3 at 5-6 days after inoculation. Both races also penetrated and developed more in PI 88788 than in PI 437654. Syncytia, characterized by cell wall dissolution and cellular hypertrophy, were developed more in PI 88788 than in PI 437654 and more by R14 than R3, for which less necrotic responses occurred in the former than the latter. This suggests that the latter two may be more resistant and less virulent than the former two, respectively. A common structural feature found in each of PI 437654 and PI 88788 in relation to SCN-resistance was the formation of prominent cell wall appositions and nuclear degeneration prior to cytoplasmic degradation in syncytial cells, respectively. Necrosis and cell wall apposition are types of hypersensitive responses occurring at early stages of the nematode infection so that these structural modifications indicate the inhibition of initial syncytial development related to the early nematode development. As soybean cultivars and lines with identical or similar genotypes have the same types of structural features related to SCN-resistance, the structural modifications induced by SCN infection may result from the expression of inheritable resistance genes, of which the information can be used for breeding soybean cultivars and lines specifically resistant to SCN races.
This study was carried out to elucidate the symptom of leaf injury to ozone and to determine varietal difference to ozone injury in rice and soybean plant. Ozone was produced by electrostatic discharge in oxygen and was monitored by UV absorption ozone analyzer. The rice leaves were spotted red and rolling leaf edge, discolored to reddish brown or yellowish white in response to ozone, and the leaves that were severely stressed were withered from the tip of leaf. The soybean ones were also discolored to lemon yellow, yellow or dark brown. The leaf injury in both rice and soybean was clearly appeared at the reverse side of leaf and in lower leaves. Milyang 23 and Nonganbyeo among rice cultivars tested were resistant to ozone, but Chucheongbyeo was resistant to it. The ratio of leaf injury was increased and chlorophyll content was decreased as the extension of ozone exposure from 2 to 8 hours in rice. Keunolkong and Danyeopkong among soybean cultivars tested showed resistant reaction to ozone, but Kwangankong and Muhankong were susceptible to it. It was observed that the soybean plants grown for 45 days after seeding were severely damaged by ozone than those of other growth stages.

The presence of high amounts of seed storage proteins (SSPs) improves the overall quality of soybean seeds. However, these SSPs pose a major limitation due to their high abundance in soybean seeds. Although various technical advancements including mass-spectrometry and bioinformatics resources were reported, only limited information has been derived to date on soybean seeds at proteome level. Here, we applied a tandem mass tags (TMT)-based quantitative proteomic analysis to identify the significantly modulated proteins in the seeds of two soybean cultivars showing varying protein contents. This approach led to the identification of 5,678 proteins of which 13 and 1,133 proteins showed significant changes in Daewon (low-protein content cultivar) and Saedanbaek (high-protein content cultivar) respectively. Functional annotation revealed that proteins with increased abundance in Saedanbaek were mainly associated with the amino acid and protein metabolism involved in protein synthesis, folding, targeting, and degradation. Taken together, the results presented here provide a pipeline for soybean seed proteome analysis and contribute a better understanding of proteomic changes that may lead to alteration in the protein contents in soybean seeds.

Green stem disorder (GSD) of soybean (Glycine max (L.) Merr.) is characterized by delayed senescence of stems with normal pod ripening and seed maturation (Hobbs, 2006). GSD complicates harvesting of soybeans by significantly increasing the difficulty in cutting the affected plants. There is also the potential for moisture in the stems to be scattered on the seed, reducing the grade and storability of the seed. Not only the cause of GSD is yet unknown, but also GSD cannot be evaluated until maturity, therefore the method to evaluate GSD in early growth stage with high sensitivity is necessary. In previous studies, it has been reported that vegetative storage protein (VSP) accumulates and the syndrome of GSD appears in soybean after depod treatment (Fischer, 1999). Soybean VSP is a storage protein which is abundant in young sink leaves and degraded during seed fill (Wittenbach, 1982). Hence, we have established a system to quantify VSP of high sensitivity by using standard protein made by genetically transformed E. coli and specific antibody against VSP, and studied the relationship between VSP and GSD, by depod experiment and drought/excess wet experiments. The result of depod experiment with the cultivar 'Yukihomare' was the same with the previous studies, VSP accumulated much more than control and the syndrome of GSD appeared in soybean in depod treatment. Drought and excess wet had different impact on GSD. Excess wet caused GSD of the cultivar 'Tachinagaha (GSD susceptible)', while drought caused a little syndrome of GSD in the cultivar 'Touhoku 129 (GSD resistant)'. The accumulation of VSP differed between the two cultivars over time. In conclusion, the accumulation of VSP came along with the emergence of GSD. Different cultivars showed different response to drought and excess wet. In the future, it is expected that the dynamics of VSP will be elucidated in detail, leading to the development of early diagnosis technology for green stem disorder and the elucidation of mechanism of soybean GSD.
To analyze their isoflavones contents by HPLC analysis during two years on 60 collected old local traditional soybean varieties from various districts of Korea was conducted. There was a yearly variation in the seed isoflavon concentrations. The total contents on 5 isoflavones were in the range from 16.21mg/g to 25.21 mg/g and from 6.47 mg/g to 15.44 mg/g, in 1997 and 1998, respectively. Collected soybean from Gunsnsi-1(25.21 mg/g) in 1997 and from Gangjingun-3 (16.50 mg/g) in 1998 showed the highest amount of isoflavones as compared with other collected soybean varieties. The highest amount among 5 isoflavones was genistin as 48.45% and 49.73%, in 1997 and 1998, respectively, indicating the genotypic variation in seed isoflavon contents of local soybean cultivar. Our data suggest that it may be feasible for improving soybean variety with higher antioxiadtive activity and substances.
Analysis of breeding gains in grain yield has been intensively conducted in wheat, barley, oat, maize, and soybean. Such information is limited in rice. The objective of this study was to compare the breeding gains and cultural gains contributed to yield gains of Korean rice varieties since early 1900s. Two sets of yield data were used for analysis; the historical yield data of 1908 for old japonica cultivars, and present yield data in the years from 1996 to 1998 for the six cultivars, consisting of previous two old cultivars and four contemporary cultivars. The old cultivars were two native cultivars, Jodongi and Damageum, while contemporary cultivars were two premium quality japonica cultivars, Hwaseongbyeo and Dongjinbyeo, and two Tongil-type cultivars, high yielding cultivars developed from indica/japonica hybridization, Milyang23 and Dasanbyeo. The yield differences of old cultivars between the experiments in 1908 and the experiments from 1996 to 1998 were estimated as cultural gains (1.84 tons $\textrm{ha}^{-1}$) due to the improvement of cultivation technology. Yield differences between the old cultivars and contemporary cultivars were considered total yield gains during the periods. These were 2.51 tons $\textrm{ha}^{-1}$ for japonica cultivars and 3.81 tons $\textrm{ha}^{-1}$ for Tongil-type cultivars. From these data, the genetic gain of 0.67 tons $\textrm{ha}^{-1}$ and 1.97 tons $\textrm{ha}^{-1}$ were estimated for japonica cultivars and Tongil-type cultivars respectively. The ratio between cultural gain and genetic gain appeared to be 2.7:1 for japonica cultivars and 1:1 for Tongil-type cultivars. This analysis clearly showed the higher genetic contribution in Tongil-type cultivars than in japonica cultivars, suggesting a guideline to be used when planning new yield improvement programs. Additional implication has emerged when a better yield response to modem cultivation technology was found in one of the old cultivars, suggesting the combined improvement between breeding and cultural improvement is necessary for attaining the maximum yield capacity of a crop.
In order to evaluate suitability by processing for non-GM soybean cultivars such as Gaechuck#2, Jinyangkong and CJ#1 lacking lipoxygenase (LOX) and kunitz trypsin inhibitor (KTI) protein, physicochemical characteristics and antioxidant activity of Kanjang made from above soybean were compared to Kanjang made from a conventional cultivar (Taekwangkong). Proximate compositions of soybeans were similar for the 4 kinds cultivars. Total phenol and flavonoid contents were significantly higher in cultivars lacking LOX and KTI protein than the Taekwangkong. In Kanjang, contents of total and reducing sugar were higher in Taekwangkong Kanjang than Kanjang from made cultivars lacking LOX and KTI protein. Contents of total and amino type nitrogen were the highest in the Jinyangkong Kanjang. Mineral contents were higher in the Jinyangkong and CJ#1 Kanjangs, amino acid contents were higher in the Kanjang made from 3 cultivars lacking LOX and KTI protein than the Taekwangkong. Taste of the Jinyangkong Kanjang with higher sweety and savory was also found to be superior to that of others in overall acceptability evaluation. Total phenol and flavonoid contents in Kanjang were significantly higher in the Kanjang made from cultivars lacking LOX and KTI protein than the Taekwangkong. Radical scavenging activity of Kanjang was increased in the total phenol contents dependent on. Reducing power by ferric-reducing antioxidant potential (FRAP) was significantly higher the Kanjang made from Gaechuck#2 and CJ#1 than the Taekwangkong. $Fe^{2+}$ chelating activity was higher in Taekwangkong Kanjang than the other cultivars, but its activity was similar to Jinyangkong Kanjang. Therefore, higher nutritional composition, total phenol and flavonoids contents and antioxidant activity in the Kanjang made from soybean cultivars lacking LOX and KTI protein might be provide better benefit for manufacture of another their products.
The present experiments were conducted to investigate the variability of hypocotyl elongation among the major soybean varieties by checking several conditions. The results obtained are summarized as follows. The rate of hypocotyl elongation is the highest during the day from 3.0 to 3.5 after seeding. It follows that it may be reasonable to evaluate the hypocotyl elongation of soybean seeds by comparison of hypocotyl length. And the tested 15 major cultivars could be classified as follow ; long ; Eunhakong, Janggyungkong and Bokwangkong, medium ; Namhekong, Dangyung-kong, Danyubkong, Milyangkong, Dugyukong, Paldalkong, Mangunjoseng, Namchunkong and Seal kong, short ; Gwanggyo, Begunkong and Jangbegkong. The hypocotyl elongation in small seed is longer than large seed. Correlation coefficients(r) for the relationships between 100 seed weight and hypocotyl elongation is -0.2506$^{**}$. As the rising temperature, the hypocotyl length is elongated, and longest at the range of 30 to 35$^{\circ}C$. The effects G $A_3$ hastened the hypocotyl elongation of soybean seed, and ABA, Kinetin and BA inhibit it, and that of those in short hypocotyl cultivars are higher than long hypocotyl cultivars. Hypocotyl length of long hypocotyl cultivars are longer than that of short hypocotyl cultivars under high temperature pre -treatment.
In order to selection of the suitable soybean varieties, the quality characteristics of meju and doenjang on 4 domestic soybean cultivars(Daeweon, Sodam, Jinpeum and Hwangkeum) were evaluated. Crude protein and lipid content was 26.7∼30.5% and 16.89∼19.31%. The content of total free sugar among 4 soybean cultivars were 4.99∼5.26% and composed mainly stachyose(2.59∼3.51%), raffinose and sucrose. The content of total free sugar of meju were the highest in Jinpeum among 2.10∼2.66% and composed mainly glucose. The lightness of meju lump in Hwangkeum was larger decrease than the others, the redness were higher in Daeweon and Hwangkeum, the yellowness was the highest in Sodam. The lightness of meju powder was the highest in Sodam, the redness was the lowest in Sodam, the yellowness was the highest in Hwangkeum. The chemical composition of doenjang were that : was the highest in Jinpeum among the 7.90∼9.71% crude lipid, in Daeweon among 9.67∼13.50% crude protein, in Sodam among 2739∼3825 mg% Formol nitrogen and in Daeweon among 6.88∼8.36% crude saponin, respectively. The content of total free sugar of doenjang were the highest in Hwangkeum among 1.88∼2.22% and composed mainly fructose and glucose. The lightness and yellowness of doenjang was the highest in Sodam, the redness was the highest in Hwangkeum. Doenjang by Hwangkeum had good score for taste, texture and overall quality in sensory evaluation.
Soybeans have various applications, and are a major source of proteins, carbohydrates, and fats. As climate warming has accelerated in recent years, this study was conducted to analyze the effect of different sowing periods on component changes in protein, amino acid, and antioxidant contents of various soybean varieties. The isoflavone content of soybeans that were sown early did not differ with the sowing period. Based on the results, only the total isoflavone content was confirmed in functional soybean cultivars such as Cheongja-5, which had blue-colored seeds; Pungsan and Sowon, which used soybean sprouts; and Jinyang, which lacked a fishy smell. There was no significant difference in the changes in protein and amino acid content in soybean cultivars with respect to the sowing time. The protein and amino acid contents in early-sown soybeans tended to be lower than that in timely sown soybeans. In most varieties, the content of proline was the highest in early-sown soybeans, confirming the presence of stress due to early sowing.
