An infectious full-length clone of Soybean mosaic virus (SMV) strain G5H was constructed under the control of the cauliflower mosaic virus 35S promoter. The cloned SMV G5H established infections upon simple rub-inoculation of soybean leaves with intact plasmid DNA. We demonstrated that this SMV G5H infectious DNA clone caused typical characteristic symptoms and virulence of SMV strain G5H in twelve tested soybean cultivars. Soybean cultivars Lee74, Somyungkong and Sowonkong developed systemic mosaic symptom while Kwanggyo, Taekwangkong, Hwangkeumkong and Geumjeongkong-l showed systemic necrosis. In contrast, Geumjeongkong-2, Jinpumkong-2, L29, V94-5152 and Ogden showed resistant response against SMV-G5H infection. We also determined full-length sequence of cloned SMV-G5H. The phyogenetic analyses reveal that SMV-G5H is most closely related to SMV-G5, and support that SMV-G5H might be derived from SMV-G5 by recombination rather than mutation.
The soybean cyst nematode (Heterodera glycines Inchinoe; SCN) is a devastating pest of soybean and is responsible for significant losses in yield. The use of resistant cultivars is the effective method to reduce or eliminate SCN damage. The objective of this research is to identify AFLP markers linked to the SCN resistant genes. Bulked genomic DNA was made from resistant and susceptible genotypes to SCN and a total of 19 primer combinations were used. About 31 fragments were detected per primer combination. The banding patterns were readily distinguished in resistant and susceptible bulked genotypes. Polymorphic fragments were detected between resistant and susceptible bulked genotypes in the primer combination of CGT/GGC, CAG/GTG and CTC/GAG. In primer combinations of CGT/GGC and CAG/GTG, bulked resistant genotype produced a polymorphic bands. However, in primer of CTC/GAG, bulked susceptible genotype produced a polymorphic fragments. Three AFLP markers identified as a polymorphic fragments between bulked genomic DNA were mapped in 85 F2 population. Among them, only two markers, CGT/GGC and CTC/GAG, was linked and was mapped. Broad application of AFLP marker would be possible for improving resistant cultivars to SCN.
A species of Corynespora was isolated from red-brown spots developed on leaves of soybean. The fungus was identified as Corynespora cassiicola and proved to be pathogenic to soybean plants. Growth of this fungus in vitro was optimal at $27^{\circ}C$, and poor at lower than 19 and higher than $35^{\circ}C$. Sporulation was abundant on the potato dextrose malt agar and moderate on V-8 juice agar and potato dextrose agar, The relative susceptibility of several soybean cultivars to Corynespora cassiicola was evaluated in the greenhouse. Cultivar Hwangkum of soybeans appered to be resistant to C cassiicola and other cultivars were susceptible or moderately resistant.
Soybean [Glycine max (L.) Merrill] crops, grown in a rice soybean rotation, can suffer when grown in soil with excessive moisture. The objective of this work were to determine the reduction in growth and yield, responses of vegetative and reproductive growth of soybean to excessive soil moisture achieved by prolonged irrigation. Responses of different cultivars were determined at growth stages from V6 to R8 to clarify the sensitive growth stages or characteristics to excessive soil moisture. Cultivar differences in response to excessive soil moisture condition were conspicuous in seed dry weight and harvest index (HI) but not in the response of seed number or pod number per plant. The timing of irrigation causing the condition of excessive soil moisture influenced the vegetative or reproductive traits. Soybean plants were more affected by irrigation commencing at the pre-flowering than at the post-flowering stage. Post-flowering irrigation did not reduce growth of vegetative organs significantly; in fact the growth of stems and leaves was facilitated by the prolonged irrigation commencing at flowering. Differences between cultivar response to prolonged irrigation were assumed to relate to the reduced amount of assimilates translocated to the reproductive organ.

Fusarium root rot is an increasingly severe problem in soybean cultivation. Although several Fusarium species have been reported to infect soybean roots in Heilongjiang province, their frequency and aggressiveness have not been systematically quantified in the region. This study aimed to investigate the diversity and distribution of Fusarium species that cause soybean root rot in Heilongjiang province over two years. A total of 485 isolates belonging to nine Fusarium species were identified, with F. oxysporum and F. solani being the most prevalent. Pot experiments were conducted to examine the relative aggressiveness of different Fusarium species on soybean roots, revealing that F. oxysporum and F. solani were the most aggressive pathogens, causing the most severe root rot symptoms. The study also assessed the susceptibility of different soybean cultivars to Fusarium root rot caused by F. oxysporum and F. solani. The results indicated that the soybean cultivar DN51 exhibited the most resistance to both pathogens, indicating that it may possess genetic traits that make it less susceptible to Fusarium root rot. These findings provide valuable insights into the diversity and distribution of Fusarium species that cause soybean root rot and could facilitate the development of effective management strategies for this disease.
Soybean seeds deteriorate even before harvest, especially under high temperature and high relative humidity conditions. This study was conducted to determine the effect of harvest date on seed quality in early maturing soybean cultivars. Soybean cultivars used in the experiment were Damyang-native, having small seed with yellow color, and Yuhsuzumi, having large seed with green color. The length of physiological maturity to grain harvest was 24 days in Yuhsuzumi and 8 days in Damyang-native. The moisture contents of seed were 25.3 % in Yuhsuzumi and 14.5 % in Damyang-native cultivar when pod reached its mature pod color. The seed color of Yuhsuzumi started to turn yellow, and its germination rate was greatly decreased when harvest was delayed after maturity, However, Damyang-native cultivars produced seed of high quality regardless of delayed harvest.
We examined changes in the amino acid composition of soybean sprouts of four different Korean soybean cultivars (Yutae, Jinunee, Subaktae, and Baktae). Total amino acids levels in all samples 1 day after cultivation were lower than those in soybeans but increased at cultivation times greater then 3 days. Glutamic and aspartic acids, the most abundant amino acids in all soybean cultivars studied at day 0 of sprouting, constituted more than 30% of all amino acids. Aspartic acid content of hypocotyls markedly increased, but glutamic acid levels decreased 5-day-old sprouts of Jinunee and 3-day-old sprouts of the other cultivars. Moreover, compositions of aspartic acid decreased in the order hypocotyl > root > cotyledon and Yutae > Baktae > Jinunee > Subaktae.
Wilted soybean plants were collected from soybeans cultivation fields in Korea from 2014 to 2016. Fusarium spp., Colletotrichum spp., Rhizoctonia spp., Macrophomina sp., Phytophthora spp., and Calonectria ilicicola were obtained from the infected samples. Out of these, Fusarium spp. were the dominant species (79.1%). In total, 53 isolates were identified as F. solani species complex, F. oxysporum species complex, F. graminearum species complex, and F. fujikuroi species complex based on mycological characteristics. Sequence typing analysis was conducted using translation elongation factor 1 alpha (TEF) to confirm the identification of isolates. All isolates were identified as F. solani, F. oxysporum, F. commune, F. asiaticum, and F. fujikuroi based on phylogenetic analysis of TEF sequences. Pathogenicity of 44 isolates was tested on three cultivars of soybean using the root dip inoculation method. Out of 5 Fusarium species, only F. asiaticum could not cause the symptoms or be weak. Ten isolates were selected based on pathogenic characters and species identification to investigate the host range and screen soybean cultivars for resistance. Fusarium solani, F. oxysporum, and F. commune were aggressive only to soybean, and F. fujikuroi was aggressive to kidney bean, yellow cowpea, black cowpea, adzuki bean as well as soybean. All 13 Korean soybean cultivars were susceptible to F. commune and F. fujikuroi. Out of 13 cultivars, cv. Janggi, cv. Poongsannamul, and cv. Socheongja were resistant to Fusarium wilt, while cv. Hwanggeumol and Chamol were susceptible to Fusarium wilt.
Three soybean cultivars, Hwangkeum, Tanyeob and Enrei were planted in the same pot under glasshouse conditions to investigate the influence of the different soil water content such as pF 1.4(wet), 2.1(control) and 3.6(dry) on the transpiration rate, dry matter production and water requirement. The transpiration rate remained the high constant rates under the wet soil condition and the control than the dry condition, and showed a linear correlation between transpiration rate and solar radiation under the all condition of soil water. The transpiration rate highly increased in the morning, but dramatically decreased in the other time in a day. The dry matter production was higher under the conditions of wet soil and the control than that under the dry condition. Also, the dry matter production Tanyeob was higher than other cultivars under all soil water content. The water requirement was higher for Enrei and lower Tanyeob than the control.

The purpose of this study was to investigate the effects of shifts in sowing time on the ecological responses, growth and yields of major soybean cultivars in a southern region of South Korea. Experiments were carried out in Naju, Jeonnam Province (latitude $35^{\circ}$ 04'N, longitude $126^{\circ}$ 54'E) for three years from 2008 to 2010. The test cultivars included Saeol-kong and Tawon-kong of the summer type, Taekwang-kong and Pungsannamul-kong, and Cheongja3 of the autumn type. Sowing took place on May 15 and 30, June 15 and 30, and July 15 of each year. Of the summer type soybean cultivars, Saeol-kong showed a smaller curtailment in days from sowing to flowering, days from flowering to maturity, and days from sowing to maturity according to sowing time postponement than Tawon-kong. Of the autumn type soybean cultivars, Taekwang-kong exhibited a lower photoperiodic response in reproductive growth period than Pungsannamul-kong and Cheongja3, both of which recorded higher level photoperiodic responses in vegetative growth stages and reproductive growth periods than other test cultivars, with the former exhibiting higher levels than the latter. Most of the test cultivars tended to decrease in stem length, node numbers of the main stem, and stem diameter according to postponed sowing time, but there were no significant differences in stem length and node numbers of the main stem of the Saeol-kong cultivar. Differences in sowing times did not affect the first setting pod node order of Saeol-kong and Tawon-kong summer type cultivars of internode lengths of all of the test cultivars. All of the test cultivars tended to show decreases in pod number per plant due to postponement of sowing time except for the Saeol-kong cultivar. The variation was more prominent in small grain cultivars such as Tawon-kong and Pungsannamul-kong with the latter autumn type cultivar showing especially large variation. Yields were the greatest for the Tawon-kong, Taekwang-kong, and Pungsannamul-kong cultivars sown on May 30 and Cheongja3 sown on May 30 and June 15. There were no significant differences in the yields of Saeol-kong for different sowing times from May 30 to July 15, with the yields lowest for the batch sown on May 15.
