Different Expression of Extracellular Matrix Genes: Primary vs. Recurrent Disc Herniation

  • Objective: Recurrent lumbar disc herniation has been reported to occur in 5% to 15% of surgically treated primary lumbar disc herniation cases. We investigated the molecular biologic characteristics of primary herniated discs and recurrent discs to see whether the recurrent discs has the similar biological features with primary herniated discs. Methods: Primary hemiated disc and recurrent disc cells were obtained by discectomy of lumbar disc patients and cells were isolated and then taken through monolayer cultures. We compared chondrogenic and osteogenic mRNA gene expression, and western blot between the two groups. Results: The mRNA gene expression of recurrent disc cells were increased 1.47* times for aggrecan, 1.38 times for type I collagen, 2.04 times for type II collagen, 1.22 times for both Sox-9 and osteocalcin, and 1.31 times for alkaline phosphatase, respectively, compared with the primary herniated lumbar disc cells (*indicates p < 0.05). Westem blot results for each aggrecan, type I collagen, type II collagen, Sox-9, osteocalcin, and alkaline phosphatase were similar between the primary herniated disc cells and recurrent disc cells. Conclusion: These results indicate that the recurrent disc cells have similar chondrogenic and osteogenic gene expression compared to primary herniated disc cells. Therefore, we assumed that the regeneration of remaining discs could fill the previous discectomy space and also it could be one of the factors for disc recurrence especially in the molecular biologic field.

Effect of Pahyeolsandong-tang (Poxiesanteng-tang) in Tibia Fracture-induced Mice (경골 파혈산동탕(破血散疼湯)이 골절 생쥐의 골 유합에 미치는 영향)

  • Objectives The main purpose of this study was to evaluate the bone healing effect of Pahyeolsandong-tang (PHT)(Poxiesanteng-tang) extract in tibia fracture-induced mice. Methods PHT was extracted using a solution of 35% ethanol in 60℃ for 8 hours. Mice were randomly divided into 4 groups (normal, control, PHT 50 and PHT 100). Mice of experimental groups were medicated with PHT 50 or 100 mg/kg for 7 to 21 days. To clarify the effect of bone fracture healing, relative messenger RNA (mRNA) expressions of osteocalcin (OCN), runt-related transcription factor 2 (Runx2), osterix (OSX), Sox9, collagen type II alpha 1 chain (Col2a1), receptor activator of nuclear factor kappa-B ligand (RANKL), osteoprotegerin (OPG) were examined. Results In in vitro experiment, relative mRNA expression of OCN, Runx2, Col2a1 was significantly increased in PHT treated group to compare with control differentiation group. In in vivo experiment, relative mRNA expression of OCN, Runx2, OSX, Sox9, Col2a1, RANKL, OPG was significantly increased in PHT treated group. Conclusions This study showed that PHT accelerates bone fracture healing through the activation of osteoclasts and osteoblasts. It was showed that PHT significantly promotes osteoblasts differentiation by osteoblast differentiation markers such as OCN, Runx2, Col1a2. Also it was investigated that PHT had stimulatory effect on osteoblasts function through enhancing OCN, Runx2, OSX, Sox9, Col2a1 and, osteoclasts function through enhancing RANKL and OPG markers. PHT effectively promotes bone fracture healing process through activation of osteoblasts and osteoclasts.

  • The main purpose of this study was to characterize the air pollutants emission factors in electric power plant (EPP) using fossil fuels. The electric power plant is a major air pollution source, thus knowing the emission characteristics of electric power plant is very important to develop a control strategy. The major air pollutants of concern from EPP slacks are particulate matter (PM), sulfur oxides (SOx), nitrogen oxides (NOx), carbon monoxide (CO) and heavy metals. Throughout the study, the following results are estimated - PM : 8.671E-05 ∼ 8.724E+01 PM emission (kg) per fuel burned (ton) - SOx : 4.149E-04∼7.877E+01 SOx emission (kg) per fuel burned (ton) - NOx 1.578E-02∼9.857E+00 NOx emission (kg) per fuel burned (ton) - CO : 3.800E-04∼1.291E+00 CO emission (kg) per fuel burned (ton) - Hg : 1.220E+01∼3.108E+02 Hg emission (mg) per fuel burned(ton) From the statistical analysis by Wilcoxon signed ranks test between the emission factors of ours and U.S. EPA's, we can yielded that : p 〉0.05.

miR-133a-3p and miR-145-5p co-promote goat hair follicle stem cell differentiation by regulating NANOG and SOX9 expression

  • Objective: Hair follicle stem cells (HFSCs) differentiation is a critical physiological progress in skin hair follicle (HF) formation. Goat HFSCs differentiation is one of the essential processes of superior-quality brush hair (SQBH) synthesis. However, knowledge regarding the functions and roles of miR-133a-3p and miR-145-5p in differentiated goat HFSCs is limited. Methods: To examine the significance of chi-miR-133a-3p and chi-miR-145-5p in differentiated HFSCs, overexpression and knockdown experiments of miR-133a-3p and miR-145-5p (Mimics and Inhibitors) separately or combined were performed. NANOG, SOX9, and stem cell differentiated markers (β-catenin, C-myc, Keratin 6 [KRT6]) expression levels were detected and analyzed by using real-time quantitative polymerase chain reaction, western blotting, and immunofluorescence assays in differentiated goat HFSCs. Results: miR-133a-3p and miR-145-5p inhibit NANOG (a gene recognized in keeping and maintaining the totipotency of embryonic stem cells) expression and promote SOX9 (an important stem cell transcription factor) expression in differentiated stem cells. Functional studies showed that miR-133a-3p and miR-145-5p individually or together overexpression can facilitate goat HFSCs differentiation, whereas suppressing miR-133a-3p and miR-145-5p or both inhibiting can inhibit goat HFSCs differentiation. Conclusion: These findings could more completely explain the modulatory function of miR-133a-3p and miR-145-5p in goat HFSCs growth, which also provide more understandings for further investigating goat hair follicle development.

  • With the significant growth of the number of vehicles environmental problems is raised. NOx SOx, and PM emissions in diesel powered vehicles are larger than that in gasoline because the development of pollutants reduction techniques has not been yet achieved. So it is need to develop after-tratment or to convert into alternative fuel to satisfy emission regula-tion. Among the after-treatment systems to reduce the diesel emissions studies with diesel oxidation catalyst(DOC) are done greatly. In this study using DOC reduction efficiency with the change of temperature and catalyst loading was calculated through measurements of CO, HC, PM. and SOX.

A Study on Recycling of Waste Tire (폐타이어 재 자원화를 위한 연구)

  • Compared to other waste, waste tire has much discharge quantity and calorie. When we use waste heat from waste tire, it can be definitely better substitute energy than coal and anthracite in high oil price age. To use as a basic data for providing low cost and highly effective heating system, following conclusion was founded. Annual waste tire production was 19,596 million in 1999, Recycling ratio was almost 55% and more than 8.78 million was stored. Waste tire has lower than 1.5% sulfur contain ratio which is resource of an pollution, So it is a waste fuel which can be combustion based on current exhaust standard value without any extra SOx exclusion materials. Waste tire has 9,256Kcal/kg calorific value and it is higher than waste rubber, waste rubber, waste energy as same as B-C oil. When primary and second air quantity was 1.6, 8.0 Nm$^3$/min, dry gas production time was 270min and total combustion time was 360 min. In the SOx, NOx, HC of air pollution material density were lower than exhaust standard value at the back of cyclone and dusty than exhaust standard value without dust collector.

  • In this study, CALPUFF, a three-dimensional atmospheric diffusion model, was used to predict the degree of influence of pollutants generated during clean center operation on surrounding areas. To drive the CALPUFF model, CALMET, a weather field calculation model, was used. Due to the influence of the wind field, air pollutants from the Clean Center diffused in the southeast direction, increasing the distribution area. SOx satisfies atmospheric environmental standards with an annual average value of 0.02 ppm or less NOx satisfies atmospheric environmental standards with an annual average value of 0.03 ppm or less. Dust (PM-10) satisfies atmospheric environmental standards with an annual average value of 50㎍/m3 or less and 24 hours average value of 100㎍/m3 or less. CO satisfies atmospheric environmental standards with an 8 hours average value of 9 ppm or less and an 1 hour average value of 25 ppm or less.

A Study of Emission Volume of Air Pollutants in Suwon City (수원시 대기오염발생량 조사에 관한 연구 -자동차에 의한 발생 중심으로-)

  • This study was carried out to investigate the emissions of the air pollutants from the automobile in Suwon city. To estimate emissions due to by automobile the data of express highway and the national road used an observation traffic volume, and the other roads used a method by Vehicle kilometer traveled(VKT). In the emissions due to by automobile from Suwon city, CO was highest 36,290.4 ton/year, NOx at 19,392.1 tons, HC 5,095.4 tons and PM 2,788.7 tons was highly order. SOx emissions in the whole Suwon city by fuel types was investigated with 178ton/year from the Diesel motorcar, 26.9 ton/year and 6.2 ton/year from the gasolines and LPG automobiles, respectively. VOC emissions from the automobile was investigated with 366.4 ton/year (29.22%) from Gwonseon-gu, 329.2 ton/year (26.25%) Yeongtong-gu, 319.9 ton/year (25.51%) Jangan-gu, 238.6 ton/year (19.03%) Paldal-gu.

Isolation and In vitro Culture of Pig Spermatogonial Stem Cell

  • The present study identified the favorable conditions for isolation, enrichment and in vitro culture of highly purified, undifferentiated pig spermatogonial stem cell (SSC) lines that proliferate for long periods of time in culture. The colonies displayed morphology similar to miceSSC and were positive for markers of SSC (PGP9.5), proliferating germ cell (PigVASA), pre-meiotic germ cell (DAZL) and pluripotency (OCT4, SSEA-1, NANOG, and SOX2) based on immuno-cytochemistry and RT-PCR. The purity of these colonies was confirmed by negative expression of markers for sertoli cell (GATA4 and SOX9), peritubular myoid cell (${\alpha}$-SMA), differentiating spermatogonial and germ cells (c-KIT). The colonies could be maintained with undifferentiated morphology for more than two months and passaged more than 8 times with doubling time between 6-7 days. Taken together, we conclude that pigSSC could be successfully isolated and cultured in vitro and they possess characteristics similar to miceSSC.

This study was carried out lo rcmove the stlfur and mineral mancrs in the samplw using wet msg~xiic separatol ant1 oil agglomeration apparatus. The rcsults for the wet magnetic separalion showed that the total sulfur removal from Kangnung coal sample was 60.8% with 82.6% combustible recovery. For the results of oil agglomeration testa, combustible recavety, ash nod sulfur rcmovcl horn Maro coal sample were 98.0, 70.9 and 95.7 percent, respectively.
