An Ecological Corridor Plan in an Urban Neighborhood Park - A Case Study of Noryangjin Neighborhood Park in Dongjak-gu, Seoul - (도심지역 산지형 근린공원내 도로에 의한 단절지역 생물이동통로 조성계획 연구 - 동작구 노량진근린공원을 대상으로 -)

  • This study was carried out to design a bridge-type ecological corridor plan in a forested neighborhood park affected by road construction in Dongjak-gu, Seoul. In order to study the site conditions, we analyzed topography, ecosystem structure, and user behavior and trail use. Existing vegetation was classified into 12 types. Based on a vegetation analysis, the Populus albaglandulosa and Robinia pseudoacacia communities, where planted species are dominant, were distributed extensively in the southern forest area. Planted areas with a single-layer structure of Korean landscape woody plants and Robinia pseudoacacia communities with a single-layer structure were distributed extensively in the northern forest and water-supply area. Based on a study of 28 quadrats, the similarity index between the multi-layer plant communities distributed in the southern forest and the single-layer planted areas was low. Twenty-four species of wild birds(355 individuals) were found in the survey area, including nine interior species and three urban species. The study of user behavior and numbers showed most users were walkers and few users were observed in the southern forest while most users were observed in the northern forest and water supply area. We selected some wild birds as model species to represent migrating species believed to use this park as an ecological corridor during migration. We suggested the new park plan include the following: improvement of vegetation structure for wildbird migration and habitat, connection of park trails for users and presentation of a landscape linked to nature.

  • For the gene tagging of Armoracia rusticana, maize controlling element Ac and Ds were introduced into A.rusticana via Agrobacterium-mediated transformation method. We established an efficient in via regeneration and transformation system for gene transfer in A. rusticana. The optimum in via regeneration condition has been obtained from leaf, petiole and root organs on modified MS medium supplemented with NAA 0.1 mg/L plus BA 1.0 mg/L for direct shooting and with free growth regulators for root induction for transformation, the leaf, petiole and root explants of A. rusticana were concultivated with Agrobacterium tumefaciens, LBA4404 which carries a binary vector pEND4K containing maize controlling element Ac or Ds, respectively: Selections were performed in the shoot induction medium supplemented with 100 mg/L kanamycin, and 500 mg/L carbenicillin transformation frequency showed about 8 to 10% in case of leaf disks. PCR md Southern blot analyses showed that the Ac and the Ds elements were integrated into the chromosome of donor plants.

Transformation of Rice (Oryza sativa L.) with Phosphate Transporter cDNA from Tobacco (Nicotiana tabacum L.) (담배 인산수송자 유전자를 이용한 벼의 형질전환)

  • In order to improve phosphate use efficiency of rice using phosphate transporter (PT), transgenic rice plants containing a tobacco PT gene were developed. Calli from Dongjinbyeo (Oryza sativa L.) were cocultured with A. tumefaciens LBA 4404 harboring PT gene. Multiplied calli were transferred to MS medium supplemented with 50 mg/L hygromycin, 500 mg/L carbenicillin, 2 mg/L kinetin, 0.1 mg/L NAA. After 2 weeks, hygromycin resistant shoots were obtained from the calli on the selection medium. The putative transgenic shoots were transferred to rooting MS medium supplemented with 250 mg/L cabenicillin. Plant regeneration rate from the calli was about 52%. Stable incorporation of the tobacco PT gene into rice genomic DNA was confirmed by PCR and Southern blot analysis.

  • This study was carried out to develop a model system using selection method for disease resistant plant breeding programs using a herbicide bialaphos-resistant patII gene as a gene-based marker. Spraying bialaphos could eliminate the susceptible plants from the segregating populations such as ${F_2}^{\prime}s$ and thereafter. Tomato cv. Momotaro-yoke was transformed with patII gene 60 independent transformants were acquired. Total 42 transformants were analyzed in transgene copy numbers by Southern blotting and the segregation ratios for the bialaphos resistance. Statistical analysis revealed that the transgene copy numbers and the segregation ratios were not always coincided, especially having the tendency of underestimating the real numbers of the transgenes in the multicopy lines. A two-stepwise screening method was applied to select $T_1$ tomato plants which linked the transgenic patII to a disease resistance gene (I2 and Ve). Based on the resistant to susceptible ratios, T-20 plant was finally selected due to the estimated linkage 12-13 cM between the patII gene to the I2 gene on chromosome 11. This newly developed system could be applied to any economical crop in breeding programs.

The Distribution and Habitation Characteristics of Zostera marina L. along the Southern Coast of Korea (남해안에서 자생하는 거머리말(Zostera marina L.)식물의 분포와 생육지 환경)

  • An ecological study was conducted to determine the geographic distribution, community structure, and habitat characteristics of eelgrass, Zostera marina L. beds along the southern coast of Korea. Plants and sediment samples were collected during June 2000 and December 2000 on twenty-eight locations, including two Cheju Island stations, which were used to compare morphological characteristics with habitat types. Z. marina populations existed from the intertidal to subtidal zone, mainly in the bays along the coast and the island, the barrier reef, and the estuary where the water depth was 0.5${\sim}$8.0m. Salinity range in Z. marina beds ranged 18.2 to 34.5%$_o$. Sediments of Z. marina beds contained 49.7${\sim}$99.1% of sand and were classified into sand, muddy sand, and sandy mud. Mean grain size varied from 1.5 to 4.4 phi. Height of vegetation shoots varied from 54.7 to 171.4 cm, depending on water depth, location, substrata and habitat types. quantitative morphological features that enabled recognition of the two phonetic groups were short-narrow leaf type and long-broad leaf type. Statistical analysis indicated that biomass of individual plants and their quantitative morphological characteristics were significantly correlated.

Flora of Mt. Balbaek (Gyeongbuk, Gyeongsan) (발백산(경북, 경산)의 관속식물상)

  • This study was conducted to study the vascular plants of Balbaeksan, located on the border between Gyeongsan-si and Cheongdo-gun, Gyeongsangbuk-do. It was performed 13 times from September 2020 to November 2021, and the investigated vascular plants were identified as a total of 93 families, 302 genera, 485 species, 10 subspecies, 40 varieties, 13 varieties, and 545 taxa. Endemic plants were investigated in 18 taxa, 9 taxa of rare plants; Vulnerable (VU) 5 taxa, Least concerned (LC) 3 taxa, Data Deficient (DD) 1 taxa, Red List plants 5 taxa; 3 taxa of least concerned (LC), 2 taxa of Not Evaluated (NE), 183 taxa of biological resources subject to export approval, 78 taxa of specific taxonomy; There were 9 taxa of grade IV, 20 taxa of grade III, 20 taxa of grade II, and 28 taxa of grade I. 4 taxa of plants subject to climate change adaptation; 2 taxa of endemic plants, 1 taxa of southern plants, 1 taxa of northern plants, 4 taxa of climate change biomarker species, 2 taxa of candidate species of climate change biomarker, 65 taxa of exotic plants, and 3 taxa of ecosystem disturbance species. Plants 235 taxa (43.1%), medicinal plants 257 taxa (47.2%), ornamental plants 148 taxa (27.2%), woody plants 54 taxa (9.9%), herbaceous plants 37 taxa (6.8%), industrial plants 15 Taxa (2.8%), 11 taxa (2.0%) of fiber-bearing plants, and plants of unknown use were identified as 73 taxa (13.4%). The flora of Mt. Balbaek was investigated for the first time, and plants to be preserved in order to secure useful plant resources and biological sovereignty were identified. So continuous monitoring is necessary to prevent the introduction of foreign plants that may be caused by the recent increase in the number of people coming and going.

A Routine System for Generation of Fertile Transgenic Rice Plants Using Biolistic Method

  • A routine system based on particle bombardment of embryogenic callus for recovery of fertile transgenic rice (Oryza sativa L.) plants was developed. Embryogenic callus was established within 2-3 months from calli derived from mature seeds of Korean rice cultivar, Nagdongbyeo. The callus was bombarded with the plasmid pRQ6 containing the $\beta$-glucuronidase gene (gusA) and hygromycin phosphotransferase gene (hph, conferring resistance to hygromycin B), both driven by CaMV 35S promoter. Placement of cells on an osmoticum-containing medium (0.2 M sorbitol and 0.2 M mannitol) 4 hrs prior to and 16 hrs after bombardment resulted in a statistically significant increase with 3.2-fold in transient expression frequency gusA. In five independent experiments, the average frequency of transformation showing GUS activities was $8.86\%$. A large number of morphologically normal, fertile transgenic rice plants were obtained. Integration of foreign gene into the genome of $R_0$ transgenic plants was confirmed by Southern blot analysis. GUS and HPT were detected in $R_1$ progeny and Mendelian segregation of these genes was observed in $R_1$ progeny.

Vetch Effects for the Low-input No-till Direct-Seeding Rice-Vetch Cropping System

  • A field experiment was carried out to observe the weed control effects of vetch and to evaluate vetch characteristics on clay loam soil in no-till direct-sown rice-vetch cropping systems. The effects of weed control, forage productivity, and N content of vetch plants were investigated. With the progress of plant growth, density of Chinese milk vetch (Astragalus sinicus L.) gradually decreased, but densities of foxtail and other weeds decreased steeply due to the depression by the over-shaded vetch canopy in a no-till direct-sown rice-vetch cropping paddy field. The vetch density in tillage systems was lower than in notillage cropping systems. Lower vetch density occurred with an increase in foxtail density and other weeds. Weed control effect increased by the progress of vetch growth, which indicated that the vetch canopy over-shaded the weeds. Vetch straw was degraded rapidly submergence after with water at the time of wet sowing of rice. Early harvesting of vetch seed resulted in lower seed germination. To acquire enough seedlings without re-sowing, the harvesting of seed should be delayed at least 28 days after the flowering stage in order to ensure the vetch population is sustainable in a no-till direct-sown rice-vetch cropping system. In order to improve the survival of vetch plants, vetch seeds should germinate from the heading .stage to before the full-ripening stage of rice plants. To enhance the percent of over-wintering survival, vetch seeds should germinate no later than the end of October in southern Korea. The dry weight of vetch plants increased with the progress of vetch growth until the flowering stage but N content decreased for 30 days from before the flowering stage (2.9%) until the ripening stage (1.8%). We concluded that Chinese milk vetch could have an effect on weed control before the flowering stage, sustainability without re-sowing of seed annually, and effective green manure for rice pre-crop in no-till direct-sown rice-vetch relay cropping systems.

Isolation of Antifungal Bacterial Strain Bacillus sp. against Gray Mold infected in Kiwi Fruits and its Disease Control (참다래 잿빛곰팡이 병원균에 대한 길항균 Bacillus sp. 분리와 병해 억제 작용)

  • This study was carried out to identity the effects of antifungal bacteria isolated from the soil grown kiwi fruit plants on the growth inhibition of Botrytis cinerea causing gray mold in kiwi fruit plants in the southern districts of Jeonnam. Two hundred and fifty antagonistic microorganisms were isolated and examined into the antifungal activity against Botrytis cinerea. We screened and isolated four bacterial strains which strongly inhibited Botrytis cinerea from the soil grown kiwi fruit plants. And the best antifungal bacterial strain which called CHO 163 was finally selected. Antagonistic microorganism CHO 163 was identified to be the genus Bacillus sp. based on the morphological and biochemical characterization. Bacillus sp. CHO 163 showed 86.9% of antifungal activity against Botrytis cinerea. By the bacterialization of culture broth and heated filtrates of culture broth, Bacillus sp. CHO 163 showed almost all of antagonistic activity against Botrytis cinerea. And we also confirmed that in vitro the treatment of Bacillus sp. CHO 163 cultured by SD+B+P broth efficiently controled the growth of Botrytis cinerea causing gray mold in kiwi fruit plants.

Inoculation with Indole-3-Acetic Acid-Producing Rhizospheric Rhodobacter sphaeroides KE149 Augments Growth of Adzuki Bean Plants Under Water Stress

  • The use of plant growth-promoting rhizobacteria is economically viable and environmentally safe for mitigating various plant stresses. Abiotic stresses such as flood and drought are a serious threat to modern agriculture. In the present study, the indole-3-acetic acid-producing rhizobacterium R. sphaeroides KE149 was selected, and its effects on the growth of adzuki bean plants under flood stress (FS) and drought stress (DS) were investigated. IAA quantification of bacterial pure culture revealed that KE149 produced a significant amount of IAA. Moreover, KE149 inoculation notably decreased stress-responsive endogenous abscisic acid and jasmonic acid and increased salicylic acid in plants under DS and FS. KE149 inoculation also increased proline under DS and methionine under FS. In addition, KE149 inoculation significantly increased the levels of calcium (Ca), magnesium (Mg), and potassium (K) while lowering the sodium (Na) content in the plant shoot under stress. KE149-treated plants had markedly greater root length, shoot length, stem diameter, biomass, and higher chlorophyll content under both normal and stressed conditions. These results suggest that KE149 could be an efficient biofertilizer for mitigating water stress.
