Real-time PCR Quantification of White Spot Syndrome Virus (WSSV) and Hepatopancreatic Parvovirus (HPV) Loads in Shrimp and Seawaters of Shrimp Ponds on the West Coast of South Korea

  • Viral diseases are major emerging problems of shrimp that have affected the production, and even complete losses for shrimp farms. In this study, we developed a sensitive TaqMan real-time PCR method to quantify white spot syndrome virus (WSSV) and hepatopancreatic parvovirus (HPV) in the shrimp and pond water in which fleshy shrimp, Fenneropenaeus chinensis, and Pacific white shrimp, Litopenaeus vannamei, are reared. WSSV and HPV in pond seawaters ranged from $1.65{\times}10^3$ to $2.43{\times}10^9$ and from 0 to $4.43{\times}10^5$ copies/L of seawater, respectively. Of 20 ponds analyzed, all pond water and shrimp were positive for WSSv. L. vannamei showed higher susceptibility to WSSV than F chinensis. HPV was detected only in the pond water for F chinensis. In shrimp tissue, however, HPV was found in both species, with 23-times higher infection rate in F chinensis than L. vannamei. The total bacterial counts in the pond water ranged from $2.23{\times}l0^3$ to $1.98{\times}l0^5\;CFU/mL$. The variations in total bacterial count for each pond appeared to correlate to the variations of the WSSV load. Statistical analysis indicated that there was no significant difference (P>0.05) between the WSSV load in pond water and shrimp, and there was no relationship between total bacterial load and viral load in the pond water. However, a significant difference (P<0.01) was found between HPV load and L. vannamei and F chinensis pond water.

Quaternary Sea Levels Estimated from River Terraces of the Ungcheon River, Midwestern Coast of South Korea (態川川流域의 河成段丘로부터 推定되는 舊汀線高度와 그 意義, 韓國 西海岸의 第四紀 環境變化 究明에 있어서 臨海山岳地域 小河川 河成段丘 硏究의 重要性 考察)

  • River terraces of glacial and interglacial periods are most developed in the Ungcheon River, midwestern coastal region of south Korea. Among these terraces, interglacial river terraces correspond to the thalassostatic terraces of eastern coastal region of Korea. Thus the former shoreline altitudes of the coastal region around Ungcheon River can be estimated by using relative heights of these interglacial thalassostatic terraces of Ungcheon River The former shoreline altitudes estimated from interglacial thalassostatic terraces of Ungcheon River are 80m, 50${\sim}$60m, 40${\sim}$45m, 30m, 25m(?), 15${\sim}$20m, and 10m. These estimates are almost identical with those of Quaternary sea levels of eastern coastal region. Among the above estimates of Ungcheon River, the former shoreline altituded of 15~20m and 10m correspond to the ancient sea levels of $\pm$18m and $\pm$10m of eastern coastal region which were injudged as the last interglacial culmination period and late warmer period of the last interglacia(5e and 5a substages of oxygen isotope stage), respectively. Therefore there is a possibility that the rest of the above former shoreline altitudes of the coastal region aroune Jngcheon River also correspond to those of eastern coastal region. On the basis of the above possibility it can be proposed that the eastern and western coastal region of Korean Peninsula have undergone tectonic uplift of equall amount since the middle Quaternary Period.

Annual Reproductive Cycle of Wild Female Yellow Croaker, Larimichthys polyactis (자연산 암컷 참조기, Larimichthys polyactis의 생식년주기)

  • Annual reproductive cycle of small yellow croaker, Larimichthys polyactis were histologically investigated based on samples captured nearby the coast around Chujado, Youngkwang and Kanghwa, South Korea from February $2004{\sim}February$,2005. By histological survey, the annual reproductive cycle was classified into the following successive stages: The primary growth stage from September to October, the second growth stage from October to December, the mature stage from January to April, the final mature, ovulation and spawning stage from May to June, the degeneration stage from June to July and the rest stage from August to September. Throughout anatomical observation of the ovary, it is found that the small yellow croaker, L. polyactis belongs to an asynchronous and multiple spawner from late spring to early summer.

Distribution of indicator species of copepods and chaetognaths in the middle East Sea of Korea and their relationships to the characteristics of water masses (한국 동해 중부 해역의 지표성 요각류 및 모악류의 분포와 수괴 특성)

  • Zooplankton samples were collected vertically from different layers with a closing net at 14 stations in the middle East Sea of Korea in February, August and September to study distribution of biological indicators for analysis of water masses. Horizontal and vertical distributions of important species of copepods and chaetognathas known as indicator species were closely related to distributions of different water masses and oceanic fronts. Pleuromamma gracilis, Calanus tenuicornis, Sagitta enflata and Sagitta minima were found to be reliable indicator species to determine warm water mass with warm core, and Calanus cristatus, Calanus tonsus and Sagitta elegans could be used as cold water species for evaluating the movement of cold current from North Korea, and Gaetanus armiger was deep sea water species. Therefore, it was found that North Korean Cold Current down to the south along the coast appeared to be significant in the surface around Chumunjin area, and from here towards the south the cold water containing S. elegans submerged under warm water with S. enflata which were about $2{\~}4^{\circ}C$ higher than that of the vicinity and reappeared near Chukpeon area in surface layer. In the layer between loom and 300m depths, distribution of Pleuromamma gracilis and Sagitta bedoti indicated that warm water mass and front zone influenced by the different water systems were formed in northwestern area off Ulreung-do. In $300{\~}500m$ layer, the proper cold water could be estimated to be present in the northwestern area off Ulreung-do throughout the survey period by the high abundance of Gaetanus armiger. In August, distributions of S. bedoti, S. enflata and S. minima were valuable index to find oceanic fronts and warm core.

Comparisons of the Pan and Penman Evaporation Trends in South Korea (우리나라 증발접시 증발량과 Penman 증발량 추세 비교분석)

  • The effects of geographical and climatic factors on annual and monthly pan and Penman evaporation were analyzed. 52 climatological stations were selected and trend analyses were performed. Furthermore, cluster analysis and multiple linear regression analysis were performed to understand the effects of geographical and climatic factors on pan and Penman evaporation. Based on stepwise multiple linear regression analysis, annual pan evaporation is proved to be mainly controlled by urbanization as geographical factor, and annual pan evaporation is also controlled by temperature, relative humidity, wind speed, and solar radiation as climatic factor. Especially wind speed is considered to be most significant climatic factor which affects pan evaporation. Meanwhile, Penman evaporation is not affected by geographical factors but it is affected by climate factors such as temperature, relative humidity, wind speed, and solar radiation except precipitation. Furthermore, the study results show that only proximity to coast affects pan evaporation trend on July; however, geographical and climatic factors do not affect pan evaporation trends in annual basis and monthly basis (January, April, and October). On the other hand, Penman evaporation trends were not affected by geographical factors in annual and monthly basises.

Landscape Changes of Coastal Lagoons during the 20th Century in the Middle East Coast, South Korea (한국 중부 동해안 석호의 20세기 경관 변화)

  • Coastal lagoon has experienced a natural geomorphic development process which has been aggraded after the climax of transgression in Holocene. This study estimates superiority on landscape conservation of lagoons and degree of landscape changes during the 20th century as conservation ratios of area and shore length, and analyzes the causes of the changes and classifies the lagoons based on the data by case study of the major 7 coastal lagoons(Hwajinpo, Songjiho, Gwangpoho, Yeongrangho, Maeho, Hyangho, and Gyeongpoho) in the middle East Coast of the Korean Peninsular. Based on the conservation ratios of area and shore length, the areal change is in order of Songjiho($0.56km^2$, 92%), Hwajinpo($2.06km^2$, 90%), Yeongrangho($0.96km^2$, 86%), Hyangho($0.32km^2$, 76%), Gyeongpoho($0.90km^2$, 52%), Maeho($0.14km^2$, 50%), and Gwangpoho($0.07km^2$, 32%), and the shore length change is in order of Hwajinpo(11.90km, 100%), Hyangho(3.34km, 90%), Yeongnangho(7.21km, 89%), Gyeongpoho(7.11km, 79%), Songjiho(5.56km, 79%), Gwangpoho(1.16km, 62%), and Maeho(2.16km, 58%). Therefore, the characteristics of landscape changes of the lagoons in the study area can be represented in order of Hwajinpo(Al), Hyangho(A2), Yeongrangho(A3), Songjiho(A4), Gyeongpoho(B4), Gwangpoho(B5), Maeho(B6). Serious process for land use and industrial development has changed landscape around lagoons decreasing the area of coastal plains dramatically up to this century. Because small lagoons such as Maeho and Gwangpoho have experienced severe transformation and destruction, and Gyeongpoho was transformed into artificial lake for urbanization and tourism, they show dramatic landscape change.

Distribution of Total Mercury in Korean Coastal Sediments (한반도 연안역 표층퇴적물 내 총 수은 분포 특성)

  • To determine the distribution of mercury (Hg) in the coastal surface sediments around the Korean peninsula, the baseline concentration of Hg was estimated, the extent of contamination was assessed, and the factors controlling the distribution were discussed. The concentrations of Hg in surface sediments were significantly high in Jinhae-Masan Bay in the South Sea, Ulsan-Onsan Bay and Yeongil Bay in the East Sea, but Hg in other sediments showed a similar distribution to Cs and relatively very low concentration between 0.21 and $39.5{\mu}g/kg$ ($13.6{\pm}7.80{\mu}g/kg$). Compared to the sediment quality guidelines in Korea, 8 % of the surface sediments (n=282) analyzed in this study exceeded the values of the threshold effects level (TEL), and six sediments collected around Onsan Port were higher than the value of the probable effects level (PEL). The contamination levels of Hg were assessed by the enrichment factors using the baseline concentration (2.06Cs+1.75) based on the residual analysis from the linear regression line for Cs, and further, factors controlling the distribution of Hg were discussed by the comparison with geochemical substances depending upon the Hg enrichment level. Hg concentrations were correlated well with Cs concentration in the range of less than 1.69 of EF implying grain size control, while in the range of 1.69 and 4.03 Hg concentrations were correlated well with Fe oxyhyroxide and organic carbon contents, which indicates Hg was enriched by superior sorption capability. On the meanwhile, samples with higher EFs (4.03 to 74.9) showed fairly positive correlations with other metals (Cu, Zn, Pb) rather than geochemical substances. For samples in Youngil Bay and Ulsan-Onsan Bay (n=30), Hg concentrations were correlated only with other metals rather than geochemical substances implying simultaneous supply of metal particles from metal refineries. But samples at Gosung, Sokcho and Uljin coast were correlated well with organic carbon even though they had high EFs. In addition, samples in Jinhae-Masan Bay with high contents of S were enriched by relatively high sulfide formation.

Numerical Simulation of Residual Currents and tow Salinity Dispersions by Changjiang Discharge in the Yellow Sea and the East China Sea (황해 및 동중국해에서 양쯔강의 담수유입량 변동에 따른 잔차류 및 저염분 확산 수치모의)

  • A three-dimensional hydrodynamic model with the fine grid is applied to simulate the barotropic tides, tidal currents, residual currents and salinity dispersions in the Yellow Sea and the East China Sea. Data inputs include seasonal hydrography, mean wind and river input, and oceanic tides. Computed tidal distributions of four major tides($M_2,\;S_2,\;K_1$ and $O_1$) are presented and results are in good agreement with the observations in the domain. The model reproduces well the tidal charts. The tidal residual current is relatively strong around west coast of Korea including the Cheju Island and southern coast of China. The current by $M_2$ has a maximum speed of 10 cm/s in the vicinity of Cheju Island with a anti-clockwise circulation in the Yellow Sea. General tendency of the current, however, is to flow eastward in the South Sea. Surface residual current simulated with $M_2$ and with $M_2+S_2+K_1+O_1$ tidal forcing shows slightly different patterns in the East China Sea. The model shows that the southerly wind reduces the southward current created by freshwater discharge. In summer during high runoff(mean discharge about $50,000\;m^3/s$ of Yangtze), low salinity plume-like structure(with S < 30.0 psu) extending some 160 km toward the northeast and Changjiang Diluted Water(CDW), below salinity 26 psu, was found within about 95 km. The offshore dispersion of the Changjiang outflow water is enhanced by the prevailing southerly wind. It is estimated that the inertia of the river discharge cannot exclusively reach the around sea of Cheju Island. It is noted that spatial and temporal distribution of salinity and the other materials are controlled by mixture of Changjiang discharge, prevailing wind, advection by flowing warm current and tidal current.

Wintertime Extreme Storm Waves in the East Sea: Estimation of Extreme Storm Waves and Wave-Structure Interaction Study in the Fushiki Port, Toyama Bay (동해의 동계 극한 폭풍파랑: 토야마만 후시키항의 극한 폭풍파랑 추산 및 파랑 · 구조물 상호작용 연구)

  • In February 2008, high storm waves due to a developed atmospheric low pressure system propagating from the west off Hokkaido, Japan, to the south and southwest throughout the East Sea (ES) caused extensive damages along the central coast of Japan and along the east coast of Korea. This study consists of two parts. In the first part, we estimate extreme storm wave characteristics in the Toyama Bay where heavy coastal damages occurred, using a non-hydrostatic meteorological model and a spectral wave model by considering the extreme conditions for two factors for wind wave growth, such as wind intensity and duration. The estimated extreme significant wave height and corresponding wave period were 6.78 m and 18.28 sec, respectively, at the Fushiki Toyama. In the second part, we perform numerical experiments on wave-structure interaction in the Fushiki Port, Toyama Bay, where the long North-Breakwater was heavily damaged by the storm waves in February 2008. The experiments are conducted using a non-linear shallow-water equation model with adaptive mesh refinement (AMR) and wet-dry scheme. The estimated extreme storm waves of 6.78 m and 18.28 sec are used for incident wave profile. The results show that the Fushiki Port would be overtopped and flooded by extreme storm waves if the North-Breakwater does not function properly after being damaged. Also the storm waves would overtop seawalls and sidewalls of the Manyou Pier behind the North-Breakwater. The results also depict that refined meshes by AMR method with wet-dry scheme applied capture the coastline and coastal structure well while keeping the computational load efficiently.

Melanogenesis regulatory constituents from Premna serratifolia wood collected in Myanmar

  • Melanin is a mixture of pigmented biopolymers synthesized by epidermal melanocytes that determine the skin, eye, and hair colors. Melanocytes produce two different kinds of melanin, eumelanin (dark brown/black insoluble pigments found in dark skin and dark hair and pheomelanin (lighter red/yellow). The biological role of melanin is to prevent skin damage by ultraviolet (UV) radiation. However, the overproduction or deficiency of melanin synthesis could lead to serious dermatological problems, which include melasma, melanoderma, lentigo, and vitiligo. Therefore, regulating melanin production is important to prevent the pigmentation disorders. Myanmar has a rich in natural resources. However, the chemical constituents of these natural resources in Myanmar have not been fully investigated. In the effort to search for compounds with anti-melanin deposition activity from Myanmar natural resources, five plants were collected in Myanmar. Extracts of these collected five plants were tested for anti-melanin deposition activity against a mouse melanoma cell line (B16-F10) induced with ${\alpha}$-melanocyte-stimulating hormone (${\alpha}$-MSH) and 3-isobutyl-1-methylxanthine (IBMX), and their anti-melanin deposition activities were compared with the positive control, arbutin. Among the tested extracts, the CHCl3 extracts of the Premna serratifolia (syn: P. integrifolia) wood showed anti-melanin deposition activities with IC50 values of $81.3{\mu}g/mL$. Hence, this study aims to identify secondary metabolites with anti-melanin deposition activity from P. serratifolia wood of Myanmar. P. serratifolia belongs to the Verbenaceae family and is widely distributed in near western sea coast from South Asia to South East Asia, which include India, Malaysia, Vietnam, Cambodia, and Sri Lanka. People in Tanintharyi region located in the southern part of Myanmar utilize the P. serratifolia, Sperethusa crenulata, Naringi crenulata, and Limonia acidissima as Thanaka, traditional cosmetics in Myanmar. Thanaka is applied in the form of paste onto skins to make it smooth and clear, as well as to prevent wrinkles, skin aging, excessive facial oil, pimples, blackheads, and whiteheads. However, the chemical constituents responsible for their cosmetic properties are yet to be identified. Moreover, the chemical constituents of P. serratifolia was almost uncharacterized. Investigation of the P. serratifolia chemical constituents is thus an attractive endeavor to discover new anti-melanin deposition active compounds. The investigation of the chemical constituents of the active CHCl3 extract of P. serratifolia led to isolation of four new lignoids, premnan A (1), premnan B (2), taungtangyiol C (3), and 7,9-dihydroxydolichanthin B (4), together with premnan C (5) (assumed to be an artifact), one natural newlignoid,(3R,4S)-4-(1,3-benzodioxol-5-ylcarbonyl)-3-[(R)-1-(1,3-benzo dioxol-5-yl)-1-hydroxy methyl]tetrahydro-2-furanone (6), and five known compounds (7-11)1,2). The structures of all isolated compounds were determined on the basis of their spectroscopic data and by comparison with the reported literatures. The absolute configurations of 1-3 and 5 were also determined by optical rotation and circular dichroism (CD) data analyses1). The anti-melanin deposition activities of all the isolated compounds were evaluated against B16-F10 cell line. 7,9-Dihydroxydolichanthin B (4) and ($2{\alpha},3{\alpha}$)-olean-12-en-28-oic acid (11) showed strong anti-melanin deposition activities with IC50 values of 18.4 and $11.2{\mu}M$, respectively, without cytotoxicity2). On the other hand, compounds 1-3, 5, and 7 showed melanogenesis enhancing activities1). To better understand their anti-melanin deposition mechanism, the effects of 4 and 11 on tyrosinase activities were investigated. The assay indicated that compounds 4 and 11 did not inhibit tyrosinase. Furthermore, we also examined the mRNA expression of microphthalmia-associated transcription factor (MITF), tyrosinase (TYR), tyrosinase-related protein-1 (TRP-1), and tyrosinase-related protein-2 (TRP-2). Compounds 4 and 11 down-regulated the expression of Tyr and Mitf mRNAs, respectively. Although the P. serratifolia wood has been used as traditional cosmetics in Myanmar for centuries, there are no scientific evidences to support its effectiveness as cosmetics. Investigation of the anti-melanin deposition activity of the chemical constituents of P. serratifolia thus provided insight into the effectiveness of the P. serratifolia wood as a cosmetic agent.
