Effect of Sophora Subprostrata Fractions on Focal Ischemic Brain Damage Induced by Middle Cerebral Artery Occlusion in Rats(II) (광두근 분획물이 중대뇌동맥폐쇄에 의한 뇌허혈손상에 미치는 효과(II) -조직화학적 평가를 기준으로-)

  • This research was peformed to investigate protective effect of Sophora Subprostrata fractions against focal ischemic brain damage after middle cerebral artery(MCA) occlusion using intraluminal suture. Rats were divided into six groups: MCA-occluded group(Control): each administered groups with Sophora Subprostrata total phase(Total), Sophora Subprostrata Aqueous phase (Aqueous), Sophora Subprostrata BuOH phase(BuOH), and Sophora Subprostrata Alkaloid phase(Alkaloid) after MCA-occlusion; sham-operated group(Sham). The right MCA was occluded by A poly-L-lysine coated 4-0 nylon suture thread through the internal carotid artery permanently. Sophora Subprostrata and fractions were administered orally(Smg/ml) for 7 days after MCA-occlusion. The Drain tissue was stained with $2\%$ triphenyl tetrazolium chloride on ischemic brain tissue(2mm section). The results showed that 1) Sophora Subprostrata total phase reduced infarct size and total infarct volume compared to the control group at 24 hours after MCA-occlusion, 2) Sophora Subprostrata Aqueous phase reduced infarct size and total infarct volume compared to the control group at 24 hours after MCA-occlusion, 3) Sophora Subprostrata Alkaloid phase reduced infarct size compared to the control group at 24 hours after MCA-occlusion, but 4) at 7 days after MCA-occlusion, Sophora Subprostrata did not show effective recovery compared with control group. Sophora Subprostrata has protective effects against brain damage at the early stage of focal cerebral ischemia. Sophora Subprostrata total and Aqueous phase produced more pronounced protective effect against focal ischemic brain damage.

  • This research was performed to investigate protective effect of Sophora Subprostrata fractions against focal ischemic brain damage after middle cerebral artery(MCA) occlusion. Rats were divided into six groups: MCA-occluded group(Control); each administered groups with Sophora Subprostrata total phase(Total), Sophora Subprostrata Aqueous phase (Aqueous), Sophora Subprostrata BuOH phase(BuOH), and Sophora Subprostrata Alkaloid phase(Alkaloid) after MCA-occlusion; sham-operated group(Sham). The right MCA was occluded by A poly-L-lysine coated 4-0 nylon suture thread through the internal carotid artery permanently. Sophora Subprostrata and fractions were administered orally(5mg/ml) for 7 days after MCA-occlusion. The behavior of ischemic rats were examined at 24 hours, 3, 5 and 7 days after MCA-occlusion from the views of 4 different aspects: posture & balance tests(4 subtests), reflex tests(6 subtests), muscle-tone tests(3 subtests), and foot-fault test. The results showed that 1) in muscle tone test, Sophora Subprostrata total phase only increased reduced muscle tone function from 3 to 7 days, 2) in reflex test, Sophora Subprostrata total and Aqueous phase increased fast recovery from 24 hours and 3 days, 3) in posture & balance test, Sophora Subprostrata total and Aqueous phase increased fast recovery from 24 hours, and Sophora Subprostrata BuOH and Alkaloid phase increased posture & balance function from 3 days, but 4) in motor function test, Sophora Subprostrata did not show effective recovery compared with control group. In conclusion, Sophora Subprostrata has protective effects against brain damage at the early stage of focal cerebral ischemia. Sophora Subprostrata total and Aqueous phase produced more pronounced protective effect against focal ischemic brain damage.

Effects of Sophora Subprostrata against Focal Cerebral Ischemic Damage by Middle Cerebral Artery Occlusion in Rats (광두근이 백서 중대뇌동맥 폐쇄에 의한 국소뇌허혈손상에 미치는 효과)

  • Objective : This research was performed to investigate protective effects of Sophora subprostrata, against ischemic brain damage after a middle cerebral artery(MCA) occlusion. The effect was estimated using histological test, neurobehavioural test, and biochemical test. Methods : Rats(Sprague-Dawley) were divided into four groups: Sham operated group, MCA occluded group, Sophora subprostrata administrated group after MCA occlusion, and Normal group. The MCA was occluded by intraluminal method. Sophora subprostrata was administrated orally twice(l and 4 hours) after middle cerebral artery occlusion. The neurobeavioural test was performed at 3 hours, 6 hours, 9 hours and 24 hours after the surgery by posture reflex test and swimming behavioural test. All groups were sacrificed at 24 hours after the surgery. The brain tissue was stained with 2% triphenyl tetrazolium chioride(TTC) or 1 % cresyl violet solution, to examine effect of Sophora subprostrata on ischemic brain tissue. The blood samples were obtained from the heart of rats. Tumor necrosis factor-a level was measured from sera using Enzyme-Linked Immunoabsorbent Assay(ELISA). Results : The results showed that (1) Sophora subprostrata reduced infarct size and total infarct volume by 54.8% compared to the control group, (2) that neuronal death, which was shown by decrease in cell number and size, was attenuated significantly in the boundary area of the infarction, (3) that serum $TNF-{\alpha}$ㆍlevel was reduced significantly, and finally, there was significant recovery of motor deficit at 3 hours after MCA occluded by Swimming behavioural test. Conclusions :In conclusion, Sophora subprostrata has protective effects against ischemic brain damage at the early stage of ischemia.

Neuroprotecticve Effect of Sophora Subprostrata on Transient Global Ischemia in Gerbil (광두근(廣豆根)의 Gerbil 전뇌(全腦)허혈에 대한 신경손상방어효능 연구)

  • Objectives : This research was performed to investigate protective effect of Sophora Subprostrata against transient global ischemic damage after 5-min two vessel occlusion. Methods : Gerbils were divided into three groups: Normal group, 5-min two vessel occlusion (2VO) group, Sophora Subprostrata administrated group after 2VO. The CCAs were occluded by microclip for 5min. Sophora Subprostrata was administrated orally(12mg/ml) for 7 days after 2VO. The histological and immunohistochemistrical analysis was performed at 72 hours and 7 days after the surgery each. For histological analysis, the brain tissue was stained with 1% cresyl violet solution and Immunohistochemistry for BAX and Bcl-2 was carried out to examine effect of Sophora Subprostrata on ischemic brain tissue. Results : The results showed that (1) Sophora Subprostrata has the protective effect against ischemia in CA1 area of the gerbil hippocampus 7 days after 5-minute occlusion, (2) the treatment of Sophora Subprostrata inhibits the expression of Bax relatively after 2VO-induced ischemia. That protective effect of the Sophora Subprostrata seems to be performed by regulating the proportion of Bax and Bcl-2 protein, (3) in hypoxia/reperfusion model using PC12 cell, the Sophora Subprostrata extract has the protective effect against ischemia in the dose of $2{\mu}/m{\ell}$ and $20{\mu}/m{\ell}$.This study suggests that Sophora Subprostrata has neuroprotective effect against neuronal damage following cerebral ischemia in vivo with a widely used experimental model of cerebral ischemia in Mongolian gerbils and that Sophora Subprostrata regulates the proportion of Bax and Bcl-2 protein following ischemia. And, Sophora Subprostrata extract has protective effects also on a hypoxia/reperfusion cell culture model using PC12 cell. Conclusions : Sophora Subprostrata has protective effects against ischemic brain damage at the early stage of ischemia.

Pollen Morphology of the Woody Fabaceae in Korea (한국에서 생육하는 콩과 목본식물의 화분형태)

  • The pollen morphology of 31 taxa, representing all woody species from 12 genera of Fabaceae in Korea, was examined by light microscope and scanning electron microscope. The pollen grains were monad and prolate spheroidal or rarely subprolate in equatorial view. The pollen grain aperture was trizonocolporate or rarely trizonooperculate, which does not occur in the other families of Fabales in Korea. The exine showed a great variety of sculpture pattern, i.e. psilate, microperforate, densely microperforate, perforate, polygonal perforate, macroperforate, macroperforate with granules in holes, polygonal macroperforate, verrucate with pitted perforate striae, verrucate-perforate, granulate, rugulate, reticulate or refined reticulate. The pollen morphological data supported the idea that the genera Echinosophora and Sophora were congeneric. Caragana (Galegeae) was quite close to the Sophora group (Maackia, Sophora and Echinosophora) of the Sophoreae in each measured pollen parameter and exine sculpture pattern. Eleven genera of Fabaceae were recognized based on differences in aperture type, pollen grain shape and exine pattern, and a generic pollen key of the woody Fabaceae in Korea was developed. A key to pollen types of Lespedeza was also suggested. The examined taxa of Coronilla and Wisteria were recognized by their exine patterns.

The Study of Preservative System in Cosmetics using Botanical Antimicrobial (식물유래 천연항균제를 이용한 화장품에서의 방부시스템에 관한 연구)

  • Anti-microbial activities of Scutellariae Radix Extracts and Sophora flavecens Extracts was tested by formulation, such as skinsoftener, emulsion for 4 weeks. Control contained no preservative, test group contained 1.00% Scutellariae Radix extracts and 1.00% Sophora flavecens extracts and positive control contained parabens and imidazolidinyl urea. To determine the anti-microbial activity of these extract, the 4 germs such as Escherichia coil, Pseudomonas aeruginosa, Staphylococcus aures, Canida albicans were used. The test groups showed significant anti-microbial activities against the 4 germs at 2 and 3 weeks as compared with control. Anti-microbial activities of these extracts were similar to positive control. Considering that the Scutellariae Radix Extracts and the Sophora flavecens Extracts have a significant anti-microbial activities against 4 germs, it is possible as natural preservative in cosmetics.

Effect of Disease Resistance on Oral Administration of Lightyellow Sophora Extract in Olive Flounder (생약제 고삼 뿌리 열수추출물의 넙치 투여시 질병 저항성에 미치는 영향)

  • The effects of lightyellow sophora, Sophora flavescens extract were tested on non-specific immune response and a disease resistance of olive flounder, Paralichthys olivaceus. For feeding trial, weight gain of fish fed different treated groups were not significant differences. Serum analysis of treated and control group did not demonstrated any differences. Lysozyme activity in the kidney and spleen of the 0.05% treated group on $4^{th}$ and $8^{th}$ week showed significant increase, respectively. In addition, phagocytic activity of the 0.05% treated group on $4^{th}$ and 8th week showed significant increase compared to the control group. Histopathology of the liver and kidney tissue of the administrated group for $4^{th}$, $8^{th}$ and $12^{th}$ week showed no particular signs of tissue degeneration. The treated group was higher than control group by analyzing the relative percent survival (RPS) of the experimental challenge of Edwardsiella tarda. Therefore, the lightyellow sophora showed effective in preventing fish-pathogenic edwardsiellosis for a certain period of time.

  • Extract of drugs in commerce was administered to mice for 3 days and its fatigue effects were compared with those of P. ginseng extract and A. sessiliflorum extract. Mice were used for these experiments divided into ethyl alcohol and water extract of P. cocos, S. japonica, P. multiflorum, L. chinense, A. cochinchinensis, S. indicum, N. nucifera were given ordinary dose orally. For antifatigue test, swimming performance were determined after administration of the extract The group of P. cocos gave high improvement of the swimming performance in ethyl alcohol extract group. And the group of S. japonica an improvement was also observed in water extract group.

Study on the Change of Antioxidant Activity by Enzymatic Hydrolysis in Sophora japonica Linne, Houttuynia cordata Thunberg, Leonurus japonicus Houttuyn (괴화, 어성초, 익모초에서 효소 분해에 의한 항산화 활성 변화 연구)

  • Objectives: The enzymatic hydrolysis is one of the processing methods that improve its effectiveness on medicinal herbs. In this research, changes in ingredients and activity by enzymatic hydrolysis were studied. Methods: For this study, a carbohydrate hydrolase such as viscozyme, which converts glycosides to aglycone, was applied to induce constituent changes in Sophora japonica Linne, Houttuynia cordata Thunberg and Leonurus japonicus Houttuyn. Changes in antioxidant activity were measured using the 1,1-diphenyl-2-picrylhydrazl (DPPH) method, and changes in ingredients were analyzed by high performance liquid chromatography. Results: As a result of enzymatic hydrolysis, the content of quercetin was increased from 1.26 mg/g to 29.66 mg/g in Sophora japonica Linne, from 0 mg/g to 0.66 mg/g in Houttuynia cordata Thunberg and from 0.43 mg/g to 0.71 mg/g in Leonurus japonicus Houttuyn. As a result of the antioxidant experimentation, the IC50 of Sophora japonica Linne decreased from 5 ug/ml (MeOH extract) and 9.1 ug/ml (EtOAc fraction) to 3.0 ug/ml, Houttuynia cordata Thunberg decreased from 15.6 ug/ml (MeOH extract) and 13.6 ug/ml (EtOAc fraction) to 11.2 ug/ml, and Leonurus japonicus Houttuyn decreased from 14.4 ug/ml (MeOH extract) and 12.6 ug/ml (EtOAc fraction) to 10.2 ug/ml. Conclusion: In conclusion, it was confirmed that glycoside rutin contained in the three medicinal herbs was changed to quercetin which is the aglycone, by the enzymatic hydrolysis using viscozyme. In terms of antioxidant activity, Sophora japonica Linne showed a significant antioxidant activity value that closes to the control group butylated hydroxyanisole. Houttuynia cordata Thunberg and Leonurus japonicus Houttuyn showed a minor increase in antioxidant activity.

  • The linkage pattem of two side chains i.e., a isopentenyl and a lavandulyl group in kushenol M(I), a flavonoid from Sophora flavescens was established by the aid of 2-D NMR techniques, especially DEPT, ${13}C-^1H$ COSY and COLOC experiments. Thus, I was unequivocally determined as (2R, 3R)-5, 7, 2', 4'-tetrahydroxy-6-isopentenyl-8-lavandulyflavanonol.
