Extraction Characteristics and Antioxidative Activity of Lycium chinense Extracts (구기자 추출물의 추출특성 및 항산화 효과)

  • The present study was conducted to investigate extraction characteristics and antioxidative activity of Lycium chinense extracts. Lycium chinense were extracted by reflux extraction(RE) under different extraction conditions including solvent. The solid yield, turbidity, color value, titratable acidity, free sugar contents, electron donating ability(EDA) and superoxide dismutase(SOD)-like ability of Lycium chinense extracts were determined. The highest solid yield value were obtained with water of 10 fold. No significant difference in turbidity and color value were found among the extracts prepared with various extraction solvents, 75$\%$ ethanol, 50$\%$ ethanol and water. The highest titratable acidity were obtained with water of Lycium chinense. The free sugar contents of Lycium chinense extracted with water showed the highest value. Lycium chinense extracts with water included higher contents of free sugar compared with those of the other solvent extracts, 50$\%$ ethanol and 75$\%$ ethanol extracts. The total polyphenol compounds content of Lycium chinense extracted with 50% ethanol showed the highest value. Lycium chinense extracts with 50$\%$ ethanol included higher contents of total polyphenol compound compared with those of the other solvent extracts, water and 75$\%$ ethanol extracts.

The total polyphenol compound content of Schiznadra chinensis extracted with 50% ethanol showed the highest value. Schiznadra chinensis extracts with 50% ethanol included higher contents of total polyphenol compound compared with those of the other solvent extracts, water and 75% ethanol extracts. The electron donating ability of extracts were 60.87% in water, 57.24% in 50% ethanol, and 55.61% in 75% ethanol.

Optimization of extraction conditions of flavonoid compounds from Thyme (Thymus vulgaris Libiatae) (타임으로부터의 플라보노이드 화합물의 추출 조건 최적화)

  • Total polyphenol contents, flavonoid contents, 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activities of ethanol extracts of 20 herbal plants (sage, turmeric, mace, bayleaf, fenugreek, oregano, blackpepper, whitepepper, clove, marjoram, cinnamon, coriander, basil, dillseed, mustard, cadamon, thyme, celery, rosemary, cumin) were analyzed for the screening of high flavonoid-containing plant resource. Thyme extract, showing the highest flavonoid content and a high degree of antioxidant activity, was selected as a bioactive cosmetic material. The total polyphenols and flavonoids contents of thyme extracts were measured as 6.90 mg chlorogenic acid equivalent (CAE)/100 g and 1.71 mg naringin equivalent (NE)/100 g, respectively, and DPPH radical scavenging activities was 90%. Among the tested organic solvents, hexane gave the highest extraction yield. Thus hexane was selected as the most suitable solvent for the extraction of thyme. Response surface method was used to obtain optimized extraction conditions for thyme: reaction temperature of 35.9 ℃, raw material to hexane ratio of 1.63:25 (w/v), and reaction time of 192 min. These predicted extraction conditions was validated by a total flavonoid extraction experiment showing a value equivalent to 96.3% of the predicted total flavonoid content. It is expected that the optimized solvent extraction conditions could be used for the production of flavonoid using thyme.

Optimal Extraction Conditions to Produce Rosemary Extracts with Higher Phenolic Content and Antioxidant Activity (폴리페놀 함량과 항산화력에 따른 로즈마리의 최적 추출조건 확립)

  • We evaluated the effects of temperature, time, solvent type, and solvent concentration on the phenolic content and antioxidant activity of rosemary extracts. The antioxidant capacity of rosemary extracts was demonstrated by their ability to scavenge stable free radicals, and the phenolic content of the extracts was determined using the Folin Ciocalteu method. The highest values were obtained by water-based extraction at $90^{\circ}C$ for 30 min, a 75% methanol solution at $60^{\circ}C$ for 30 min, and a 50% ethanol solution at $70^{\circ}C$ for 10 min. The aqueous solution of 75% methanol, extraction temperature of $60^{\circ}C$, and extraction time of 30 min were the most efficient parameters for the extraction of polyphenols from dry rosemary.

  • Poly (styrene-b-methyl methacrylate) (PS-b-PMMA) was synthesized by free radical telomerization: the telomerization of styrene with $CCI_4$ by using AIBN as initiator followed by a second telomerization of methyl methacrylate using $CCI_3$ end group of the resulting polymer as the macrotelogen, with AIBN initiation, gave the styrene-methyl methacrylate block copolymer. The effects of the concentration of the macrotelogen, the concentration of monomer, the molecular weight of the macrotelogen, the reaction temperature and the concentration of the solvent on the formation of the block copolymer were investigated. Block copolymers containing up to 10 weight percent PMMA were obtained by adjusting the reaction conditions.

  • Sulphuric acid leaching was conducted to extract the metal values from spent refinery catalyst. More than 95% of Ni and V and 30% of Mo could be leached out in 1 M sulphuric acid and 1 hr of leaching time. The decrease in Mo leaching was due to typical characteristic of Mo matrix. The activation energies of the leaching reactions showed the dissolution process follows a diffusion control mechanism. In order to leach out all Mo, further the leaching experiments were conducted with sulfur free spent refinery catalyst. For sulfur free spent refinery catalyst, a two step process of leaching with 1 M sulphuric acid followed by sodium carbonate washing showed better leaching than a two step leaching process with sodium carbonate followed by sulphuric acid washing, with almost 99% leaching of Ni, Mo and V. Solvent extraction using LIX 841 were conducted for a leach liquor containing Ni, 2 g/L; V, 9 g/L, Mo, 0.6 g/L. More than 98% of Mo was extracted from the leach liquor at A:O ratio of 5:2 in a 2 stage process. Similarly V was extracted at A:O ratio of 5:3 in a 2 stage process with 82% of total V extraction.

Free Radical Scavenging Effect of Seed and Skin Extracts of Black Olympia Grape (Vitis labruscana L.) (거봉 포도 종자 및 과피 추출물의 항산화 활성)

  • Optimal extraction condition and free radical scavenging effect of ethanol extracts of Olympia grape (Vitis labruscana L.) seeds and skins were determined. Extraction yield of grape seeds increased with increasing extraction temperature increased, but was not affected by ethanol concentration in grape skins. Highest extraction yields obtained with 100% ethanol concentration at $78^{\circ}C$ were 36 and 49.5% in seeds and skins, respectively. Ethanol extract of grape seeds obtained at $78^{\circ}C$ showed stronger $RC_{50}\;(409.7\;{\mu}g/mL)$ than that of grape skins. Among organic solvent fractions, ethylacetate fraction obtained from ethanol extract of grape seed showed the strongest $RC_{50}\;(136.7\;{\mu}g/mL)$. Overall, ethanol extracts of grape seeds and their organic fractions showed significantly stronger free radical scavenging effect than those of skin extracts.

Preparation and Release Characterization of Biodegradable Poly($\varepsilon$-caprolactone) Microcapsules Containing Tocopherol (토코페롤을 함유하는 생분해성 폴리($\varepsilon$-카프로락톤) 마이크로캡슐의 제조 및 방출 특성)

  • The biodegradable poly($\varepsilon$-caprolactone) (PCL) microcapsules containing tocopherol were prepared by oil-in-water emulsion solvent evaporation method. The features of the microcapsules were investigated in the manufacturing conditions and degradation behaviors. The form and structural feature of the microcapsules were measured by scanning electron microscope and X-ray diffraction, respectively. The surface free energy of the microcapsules was executed using contact angle measurement. As a result, the microcapsules were more stable and spherical with poly(vinyl alcohol) given in a surfactant. The surface free energy and crystallinity of microcapsules were decreased with increasing the core concentration, and degradation of PCL was occurred after 21 days. The release behaviors were examined by Uv/vis. spectrophotometer. It was found that the release rate of the microcapsules was increased with increasing the stirring rate, due to the increased interface between microcapsules and release media.

Investigation of Antioxidative Activity and Stability of Ethanol Extracts of Licorice Root (Glycyrrhiza glabra) (감초의 에탄올 추출물의 항산화 활성 및 안정성 조사)

  • This study was carried out to optimize the conditions for the extraction of antioxidative materials from licorice root, Glycyrrhiz glabra. Chipped licorice roots were extracted with several solvents and their antioxidative activities were tested to determine the optimal extraction solvent. Among the solvents tested, 95% ethanol gave the highest free radical scavenging activity, and was therefore chosen as the optimal extracting solvent. The optimum extraction temperature and time were $20^{\circ}C$ and 12 hr, respectively. Next, the free radical scavenging activity of the ethanol extract was compared with that of other known antioxidants such as ${\alpha}-tocopherol$, butylated hydroxytoluene (BHT) and butylated hydroxyanisole (BHA). Ethanol extract of licorice root had greater antioxidative activity than ${\alpha}-tocopherol$ and a similar level to that of the two synthetic antioxidants (BHA and BHT). Moreover, the antioxidative activity of the ethanol extract was inhibited neither by heat treatment at $180^{\circ}C$ for 30 min nor by treatment at extreme pH. These findings suggest that ethanol extract of G. glabra may be useful as a natural antioxidant.
