Turnover of Phosphate Compounds in Chlorella cells in a P-free medium (인산결핍배지에 있어서의 Chlorella 세포내의 인산화합물의 전환)

  • Using the Chlorella cells which had been uniformly labeled with $^{32}P$, the distribution of phosphorus in various fractions of cell material was investigated. Uniformly $^{32}P$-labeled Chlorella cells were further grown in a P-free medium, and some protions of the cells were taken out at intervals during the culture, and subjected to analyze the contents of $^{32}P$ in various fractins of the cell constituents. 2. Analysis of the $^{32}P$-labeled Chlorella cells showed that the highest in P-content was the fraction of RNA followed by those of lipid, RNA-polyphosphate complex, acid-insoluble polyphosphate, acid-soluble polyphosphate, DNA and protein. 3. During the culture of $^{32}P$-labeled Chlorella cells in a P-free medium, amounts of phosphate in DNA, protein and lipid fractions increased, while the P-contents in the fraction of RNA-polyphosphate complex decreased as well as those of acid-insoluble polyphosphate and acid-soluble polyphosphate fractions. 4. It was inferred that phosphorus used in the syntheses of DNA and protein was taken from polyphosphates of the cells, and RNA-polyphosphate complex would play an important role as a phosphate pool.

Ureide Distribution in Nitrogen Fixing Soybean Plant under External Phosphorus Limitation (인산결핍 조건하에서 질소고정식물체내의 Ureide 분배)

  • Soybean plants inoculated Bradrhizobium japonicum MN 110 were grown in outdoor perlite pots with nitrogen free nutrient solution containing 1.0 mM-P(control) and 0.05 mM-P(stress) and harvested at 28, 35, 42 and 49 days after transplanting (DAT) to examine the effect of phosphorus deficiency on ureide concentration of and distribution to diffrent plant organ in nitrogen fixing soybean plant during the vegetative growth. Total dry mass of control plants increased 8.9 fold and that of phosphorus deficient plant increased 2.7 fold during the experimental period. Phosphorus deficiency reduced total phosphorus and nitrogen accumulation by 80%,40% respectively, at 28 DAT and 93%, 84%, respectively, at 49 DAT. Nitrogen concentration was reduced by phosphorus deficiency in all tissues with leaf and stem tissues affected to a greater degree than nodule and root tissues at every sampling date. Phosphorus deficiency significantly reduced soluble reduced-N and ureide-N concentration in leaf and stem but did not affect those in root. The proportion of soluble reduced-N in leaf was reduced from 60% to 50% but increased from 10% to 20% in the roots. The proportion of ureide-N in leaf of control plants was higher than that in phosphorus deficient plants, whereas, roots of phosphorus deficient plants contained a higher propotion of ureide-N than those of control plants. These indicated that phosphorus deficiency not only inhibit nitrogen fixation of nodules but also restrict the translocation of fixed nitrogen out of the root system into the xylem.(Received April 4, 1997; accepted May 2, 1997)

Effects of Organic Acids on Availability of Phosphate and Growth of Corn in Phosphate and Salts Accumulated Soil

  • Accumulated Phosphate can be released by ligand exchange reaction of organic acids. The objective of this study was to evaluate effects of the organic acids on the availability of phosphate and the growth of crop in phosphate and salts accumulated soil. Soil samples were collected from farmer's plastic film house. Available phosphate and electrical conductivity of soil were $3,005mg\;kg^{-1}$ and $16.63mg\;kg^{-1}$ which were 6 and 8 times higher than the optimum range of soil for crop growth, respectively. Corns were cultivated in pots for 2 months. Treatments were no treatment (control), phosphate fertilizer (P), citric acid (CA) 1, 5, 10 mM, and oxalic acid (OA) 1, 5, 10 mM. Water soluble phosphorus, available phosphate, corn growth and uptake were determined after cultivation. Results showed that organic acids increased water soluble phosphorus and available phosphate. For the level of 10 mM, the order of effectiveness of organic acids for water soluble P was citric acid (44%) > oxalic acid (32%). Height and dry weight of corns were increased significantly by the treatment of citric acid 1 and 5 mM. Also, corn absorbed more phosphorus, nitrogen, potassium, calcium and magnesium in the treatment of citric acid 1 mM than these of other treatments. Even though phosphate availability of soil was enhanced by addition of citric acid 10 mM, the growth of corns decreased because high concentration of citric acid caused salt damage by increasement of electrical conductivity. Thus, the citric acid of 1 mM has the potential to improve the availability of phosphate and the healthy growth of corns.

Reaction Characteristics of Piggery Wastewater for Biological Nutrient Removal (생물학적 영양염류 제거를 위한 돈사폐수의 반응 특성)

  • This study was performed to investigate the reaction characteristics of piggery wastewater for biological nutrient removal. The reaction characteristics were discussed the fraction of organics, the behavior of nitrogen, nitrification, denitrification, and the behavior of phosphorus. The fraction of readily biodegradable soluble COD was 11-12 percent. The ammonia nitrogen was removed via stripping, nitrification, autotrophic cell synthesis, and heterotrophic cell synthesis. The removal percents by each step were 12.1%, 68.9%, 15.0%, and 4.0%, respectively. Nitrification inhibition of piggery wastewater was found to occur at an influent volumetric loading rate over 0.2 NH$_{3}$-N kg/m$^{3}$/d. Denitrification rates were the highest in the raw wastewater and the lowest in the anaerobic effluent. The denitritation of piggery wastewater came out to be possible, and the rate of organic carbon consumption decreased about 10 percent. The phosphorus removed was released in the form of ortho-p in the aerobic fixed biofilm reactor, it was caused by autooxidation. The synthesis and release of phosphorus were related to the ORP and the boundary value for the phase change was about 170mV. In the synthesis phase, the phosphorus removal rate per COD removed was 0.023mgP$_{syn}$/mgCOD$_{rem}$. The phosphorus contents of the microorganism were 4.3-6.0% on a dry weight basis.

  • On the seventh day of mounting male and feamale cocoons are distinguished. A part of those male and female pupae is irradiated for 40 min. with 1,000r-ray irradiation. Those eggs obtained from the irradiated pupae are divided into six groups of 5, 15, 45, 90, 720 and 2,400 minutes after laying. The cahanges of accid soluble phosphorus in the each group of eggs are as follows. 1. In the control group of the maximum quantity in acid soluble phosphorus was 1,456r/100mg D.P. in the 2,400 minutes group, while the minimum quantity was 1,233r/100mg D.P. in the 720 minutes group. 2. In the mated group of *irradiated female moths and control male moths, the quantity of acid soluble phoshorus was small as compared with the control group in the five minute group of 1,288r/100mg D.P. and in the 15 minutes group of 1,299r/100mg D.P. while the quantity increased to 1,484r/100mg D.P. in the 45 minutes group. The quantity began to decrease again to 1,400r/100mg D.P. in the 2,400 minutes group which was smaller than that of the control group. The quantity was slightly small in the silkworm eggs laid in the early stage when compared with that of mated group of *irradiated male moths and control female moths. Both groups showed the larger quantity in the 720 minutes group than that of the control group. The quantity of acid soluble phosphorus in the mated group of control female moths .and *irradiated male moths was larger in variation than in the mated group of control male moths and *irradiated female moths. 3. In the mated group of *irradiated male and female moths the largerst quantity was 1,760r/100ms D.P. in 2,400 minutes group and second was 1,736r/100mg D.P. in 90 minutes group, and third was 1,288r/100mg D. P., in 5 minutes group. Generally the quantity was small in silkworm eggs laid by the irradiated one in the early stage than the control group, but in the later period of egg the quantity began to increase more than that of control group.

  • Immobilization is seen as a promising technology for lead remediation. In a laboratory experiment, immobilization of lead with soluble P was tested as a function of reaction time and P concentration. The P treated with an acidic solution to enhance heavy metal immobilization was worked into the soil, and within 7 days, lead was stabilized. Different molar ratios of soluble phosphates (super-phosphate and KH$_2$PO$_4$) would be considerably effective to accelerate the formation of highly insoluble minerals due to the lack of leachable Pb in the contaminated soil. Although it was demonstrated that the addition of soluble phosphates with an acidic solution significantly reduced available lead in soil up to over 95%, remaining phosphorus in soil matrix might cause a possible groundwater eutrophication in the near future.

Impoundments Increase Potential for Phosphorus Retention and Remobilization in an Urban Stream

  • Weirs are conventional structures that control water level and velocity in streams to facilitate water resource management. Despite many weirs built in streams, there is little information how weirs change hydrology regime and how that translates to sediment and phosphorus (P) responses. This study evaluated the influence of weirs on P retention and mobilization in an urban tributary of the Han River in Korea. Total P concentrations in sediments upstream of weirs were higher than the downstream site, mainly due to the increase of potentially available fractions (labile P and aluminum- and iron-bound P) (p < 0.05). Equilibrium phosphorus concentrations ($EPC_o$) were lower than soluble reactive phosphorus (SRP) concentrations of stream waters, but there was an increasing trend of sediment $EPC_o$ upstream of weirs compared to the downstream site (p < 0.001) indicating a greater potential for P release upstream of weirs. Sediment core incubation showed that SRP release rates upstream of weirs were higher than the downstream site under anoxic conditions of the water column (p < 0.01), but not under oxic conditions. SRP release rates under anoxic conditions were greater than that measured under oxic conditions. Un-neutral pH and increased temperature could also enhance SRP release rates upstream of weirs. We conclude that weirs can increase P retention within stream sediments and potentially promote significant P releases into waters, which in turn cause eutrophication.

Effects of Nitrogen Sources on RNA, DNA and other Phosphorus Fractions of Soybean Cultivars Different in Phosphorus Sensitivity (인산감수성(燐酸感受性)이 다른 대두품종(大豆品種)의 RNA, DNA 및 기타 인산형태(燐酸形態)에 대(對)한 질소원(窒素源)의 영향(影響)에 관한 연구(硏究))

  • RNA, DNA and other phosphorus fractions were determined in the leaf and root of soybean plants different in phosphorus sensitivity grown in $NH_4-N,\;NO_3-N$ and urea medium. The phosphorus sensitive cultivars contained higher ASIP (acid soluble inorganic phosphorus) than the tolerant cultivars with all nitrogen sources. ASIP was highest in the urea treated plants and lowest in the nitrate treated plants. Total phosphorus content was mostly affected with increase in ASIP. When ASIP increased, acid solsuble organic phosphorus(ASOP), phospholipids (L-P), RNA-P, residual phosphorus (R-P) tended to increase, while DNA-P showed little change. The percent RNA-P or DNA-P of total phosphorus in the nitrate treated plant was twice that in the ammonium treated plant, which were also higher in tolerant cultivars regardless of nitrogen sources. The percent ASOP in total acid soluble phosphorus $(ASOP/ASP{\times}100)$ decreased as phosphorus sensitivity decreased. Indications are that phosphorus sensitivity depends on the relative sizes of phosphorus metabolic pools. Total dry matter yield was negatively correlated with total phosphorus (r=0.84 significant at 0.01P), ASIP (0.84 significant at 0.01P) and residual phosphorus (0.69 significant at 0.05P). ASOP showed positive correlation with L-P, RNA-P and DNA-P but negative with R-P. RNA-P was significanly correlated only with L-P (0.63 at P=0.01). There was significant interaction (0.01) among nitrogen sources, cultivars and phosphorus metabolic pools. Phosphorus sensitivity and ammonium toxicity appear to be same in view of energy metabolism, that is, the former inhibits the conversion of ATP to ADP (energy releasing) through phosphate potential while the latter inhibits ATP formation (energy storing).

Effects of Phosphorus and Iron on the Phytotoxicity of Lettuce (Lactuca sativa L.) in Arsenic-contaminated Soil

  • We examined the effect of simultaneous application of phosphorus (P) and iron (Fe) on the phytotoxicity of lettuce in arsenic (As) contaminated soil using response surface methodology (RSM). To stabilize As and supply nutrient into soil, Fe and P were treated, respectively. Water soluble As and P was decreased by Fe application but increased by P application. Through phytotoxicity test, the result showed that only the addition of P affected lettuce root elongation even though both P and Fe were added. The correlation coefficients between root elongation and other indices indicated that the As content in the roots seemed to be the main reason that root growth was impeded. We could verify that the former result was not a passing phenomenon and Fe was necessarily needed to protect secondary pollution by exclusive usage of P fertilizer.

Monitoring and Analysis of Nutrients in Sediments in the Riverbed (하천 퇴적물의 영양염류 모니터링)

  • Characterization of sediment in the riverbed is of importance for effective water quality management, yet have not been monitored sufficiently. This paper reports monitoring results of nutrient concentrations of sediments. Surface waters and sediments were sampled four times during rainy season at five monitoring points. Organics of overlying water were increased after high flow condition followed by decreasing tendencies. Soluble phosphorus fraction among total phosphorus was increased after high flow condition while total phosphorus was in decreasing tendencies. Monitoring result suggested that more extended monitoring scheme for flow rate, scouring velocity, and suspended material is required for analyzing relationship between water quality and sediment.
