Development of an Automated Diffusion Scrubber-Conductometry System for Measuring Atmospheric Ammonia

  • A semi-continuous and automated method for quantifying atmospheric ammonia at the parts per billion level has been developed. The instrument consists of a high efficiency diffusion scrubber, an electrolytic on-line anion exchange device, and a conductivity detector. Water soluble gases in sampled air diffuse through the porous membrane and are absorbed in an absorbing solution. Interferences are eliminated by using an anion exchange devises. The electrical conductivity of the solution is measured without chromatographic separation. The collection efficiency was over 99%. Over the 0-200 ppbv concentration range, the calibration was linear with $r^2$ = 0.99. The lower limit of detection was 0.09 ppbv. A parallel analysis of Seoul air over several days using this method and a diffusion scrubber coupled to an ion chromatography system showed acceptable agreement, $r^2$ = 0.940 (n = 686). This method can be applied for ambient air monitoring of ammonia.

  • Structure H formation experiments were conducted in a semi-batch stirred vessel using methane as the small guest substance and neohexane(NH), tert-butylmethylether(TBME) and methylcyclohexane(MCH) as the large molecule guest substance (LMGS). The results indicate that the rates of gas uptake and induction times are generally dependent on the magnitude of the driving force. When tert-butyl methyl ether is used as the LMGS rapid hydrate formation, much smaller induct ion time and rapid decomposition can be achieved. Liquid-liquid equilibrium (LLE) data for the above LMGS with water have been measured under atmospheric pressure at 275.5, 283.15K, and 298.15K.   • Effects of the plant growth regulator (P. G. R.)on the reaction of proteinase, $\gamma-amylase$ and acid phosphatase were investigated, and also were the conditions of production of P. G. R. by Stroptomyces sp. 445. The P. G. R. had no effect on the act ivities of such enzymes in mung bean seedling. But in germinating seed previously treated with P. G. R. it effected the activity of protease in cotyledon. In the conditions of production of P. G. R., the maxim, activity was appeared in shaking cutlure at $30^{\circ}C$ for 5 days, and by the addition of peptone or casein hydrolysate as nitrogen source, soluble starch as carbon source, and sulfur as metal ion.

무염 원자흡광도법과 흡광광도법에 의한 제제중의 Metoclopramide의 정량 (Determination of Metoclopramide by Flameless Atomic Absorption and Spectrophotometry)

  • Metoclopramide reacts with ammonium cobaltothiocyanate to form a stable ion pair which has an absorption maximum of 625mm. The reaction product was insoluble in water but soluble in most organic solvents. 1,2-Dichlorethane was the best extracting solvent among the several organic solvents tested. Metoclopramide can be determinded not only by visible light spectrophotometry but also indirectly by estimation of cobalt in the organic phase by flameless atomic absorption spectrophotometry. Linear relationship was found between absorbance and concentration in the range of 10$^{-4/}$-10$^{-3}$M by spectrophotometry and 10$^{-5}$-10$^{-4}$M by flameless atomic absorption spectrophotometry. The coefficient of variation by spectrophotometry was 0.9% and that of flameless atomic absorption was 1.8%. There was no interference with excipients, pH, temperature and reaction time. With this method, it is possible to determine accurately metoclopramide and tertiary amines in pharmaceutical preparations.

알카리성 분위기에서 소석고의 수화에 미치는 카르복실산의 영향 (Effect of Carboxylic Acid on the Hydration of Plaster of Paris under Alkalinity)

  • Although various theories have been presented on the mechanism of setting retardation of plaster in addition to organic admixtures. The purpose of this paper is that Hydration studies of plaster of paris in the presence of carboxylic acids under alkalinity are examined in the coordination chemistry. Setting of plaster of paris is retarded by the addition of carboxylic acids except oxalic acid. And setting of plaster of paris contained 5 wt% of Ca(OH)2 is also retarded by the addition of above-mentioned carboxylic acids. The degree of retarding effect under alkalinity is found to be a function of the number of the functional group and the length of carbon chain of carboxyl acids. These reasons are attributed to the soluble complex formation, that is calcium complex formation between calcium ion and carboxylic acids. The author's proposal was confirmed by the results of electrical conductivity measurement. The formation of calcium complex was surpported by IR spectra.

막장의 아미노酸 組成에 關한 硏究 (Studies on the Amino Acids Composition of Makjang)

  • Amino acid composition of Makjang was determined by combined usage of ion exchanged resin and paper chormatography in the following states. a. A fraction soluble in water b. Hydrolysate of the whole Makjang c. Same as a. (p.p.t. formed by tungstic acid or trichloroacetic acid being removed) d. Hydrolysate of c. (T and TCA) By comparing amino acid composition of Makjang with that of its raw material, we found that decomposition of essential amino acids during brewing is slight. From the amino acid composition of a,b,c,d, we discussed the ratio of amino acid liberation during brewing and assumed that Makjang contains peptide-like substances composing of glutamic acid and aspartic acid.

  • The purpose of this study was to analyze the electrophoretic patterns of soluble proteins from ginseng roots and to compare the protein patterns from Korean ginseng and American quinquefolium. The size difference was found in the major protein bands of a molecular weight of about 27,000 between Korean ginseng and American quinquefolium. The protein band of a molecular weight of 22,000 showed a quantitative difference in its amount. The major 27 K proteins appeared to form a complex heterodimer of 66,000 and to have internal bisulfide bonds, from band shifting studies under non-denaturing conditions. Three peaks appeared when the protein extract from root homogenates was purified using gel filtration and DEAE ion exchange chromatography. The examination of physiological activity and further purification of these fractions are underway.

Production of Active Carboxypeptidase Y of Saccharomyces cerevisiae Secreted from Methylotrophic Yeast Pichia pastoris

  • Our previous study showed that the overexpression of carboxypeptidase Y (CPY) of Saccharomyces cerevisiae in Escherichia coli resulted in the formation of insoluble inclusion bodies. To produce soluble CPY, we designed a novel Pichia pastoris expression system, in which the following were inserted into expression vectors: three different signal sequences derived from the mating factor a1 of S. cerevisiae, an inulinase of Kluyveromyces marxianus, and the endogenous signal sequence of CPY. The expression vector pHIL-D2-SSinul-proCPY was the most effective in the production of proCPY among the vectors examined. The purified active CPY was obtained from proCPY by treating with proteinase K, followed by QExcellose ion-exchange column chromatography.

통계분석을 이용한 경산 지역의 초미세먼지(PM2.5) 농도 특성 파악 (Characteristics of PM2.5 in Gyeongsan Using Statistical Analysis)

  • The ambient $PM_{2.5}$ samples were collected by $PM_{2.5}$ sampler from September 2010 to December 2012 at Daegu University, Gyeongsan. A total of 260 samples were collected and 27 species were analyzed by inductively coupled plasma, ion chromatography, and thermal optical reflectance methods. Factor analysis identified four sources such as biomass burning source/secondary aerosol source, soil source, industry source, and incinerator source/mobile source. Also, backward trajectories were calculated using HYSPLIT 4 (Hybrid single-particle lagrangian integrated trajectory) model and PSCF (Potential source contribution function) model was applied to identify the possible source locations of carbonaceous species and water soluble ions species. PSCF results showed that the possible source locations of most chemical constituents measured in Gyeongsan were the industrial areas in the eastern coast of China, northeast regions of China, the Gobi Desert, and east sea of Korea.

Purification of fusion ferritin using silica powder and DEAE chromatography

  • Iron is an essential nutrient for most organisms, which supplied to them in a protein-iron complex known as ferritin. Ferritins are multimeric proteins those are consisted of spherical shell of 24 subunits defining a cavity of about 8nm in diameter. Soluble form of ferritin was separated from disrupted cells, followed by silica powder adsorption. Ferritin was recovered from silica-poweder by distiiled water, which was applied to DEAE anion exchage chromatography. Collected fractions from the DEAE column were assayed to gain the amount and the purity of ferritin by using GF-HPLC.
