Volatile Flavor Composition of White-flowered Lotus by Solid-phase Microextraction (Solid-Phase Microextraction에 의한 백련의 휘발성 향기 성분 분석)

  • This study investigated the chemical composition of headspace gas from white-flowered lotus (Nelumbo nucifera Gaertner). Volatile flavor compositions of headspace from white-flowered lotus (floral leaf, stamen, flower stalk, stem) were investigated through the solid-phase microextraction method using polydimethylsiloxane-divinylbenzene fiber. The headspace was directly transferred to a gas chromatography-mass spectrometry. Sixty-three volatile flavor constituents were detected in the headspace of lotus floral leaves, and undecanoic acid (7.81%) was the most abundant component. Fifty-three volatile flavor constituents were detected in the headspace of lotus stamina, and isobutylidene phthalide (7.94%) was the most abundant component. Forty-four volatile flavor constituents were detected in the headspace of lotus flower stalks, and 3-butyl dihydrophthalide (11.23%) was the most abundant component. Fifty-nine volatile flavor constituents were detected in the headspace of lotus stems, and ligustilide (16.15%) was the most abundant component. The content of phthalides was higher in the headspace of flower stalks and stems, while alcohols and acids were the predominant compounds in lotus floral leaves.

Removal Characteristics of Cu(II) ion in Aqueous Solution by Solid-Phase Extractant Immobilized D2EHPA and TBP in PVC (D2EHPA와 TBP를 PVC에 고정화한 고체상 추출제를 사용한 수용액 중의 Cu(II) 이온 제거특성)

  • Removal characteristics of Cu(II) ions by solid-phase extractant immobilized D2EHPA and TBP in PVC were investigated. Cu(II) ion concentrations in the solution and removal capacity of Cu(II) ion according to operation time were compared. The lower the initial concentration of Cu(II) ion in aqueous solution was, the removal capacity of Cu(II) ion by solid-phase extractant was increased relatively. The bigger the initial concentration of Cu(II) ion was, the removal capacity of Cu(II) ion was increased relatively. The pseudo-second-order kinetics according to operation time was showed more satisfying results than the pseudo-first-order kinetics for the removal velocity of Cu(II) ion. The removal capacity of Cu(II) ion was 0.025 mg/g in aqueous solution of pH 2, but the removal capacity of Cu(II) ion was increased to 0.33 mg/g mg/g in aqueous solution of pH 4 according to increasing pH.

  • Adefovir dipivoxil which was originally developed by Gilead Sciences has been used as treatments of HIV and HBV, especially a therapeutics for HBeAg positive and negative chronic patients. We developed highly efficient purification method using reverse phase column chromatography for mass production and a stable amorphous Adefovir dipivoxil using solid dispersion method. Reverse phase column chromatography led to highly pure product, more than 99.7% by HPLC and can be used for mass production compared with normal column chromatography. Solid dispersion method containing watersoluble polymer and Isomalt showed improved stability of amorphous Adefovir dipivoxil against heat and moisture.

Simple and Highly Efficient Synthesis of [$^{11}C$]methionine Using Solid-Phase Extraction Method (고정상 추출법을 이용한 효율적인 [$^{11}C$]methionine의 합성)

  • We developed simple and highly efficient synthesis method for [$^{11}C$]methionine using solid-phase extraction method. For synthesis, we used C18 cartridge. [$^{11}C$]methionine was synthesized on C18 cartridge according to the solid-phase [$^{11}C$]methylation of precursor L-homocysteine thiolactone hydrochloride. The radiochemical yields of [$^{11}C$]methionine was $48.9{\pm}7.93%$ decay corrected (results of 30 syntheses, mean$\pm$SD), with average production higher than 180 mCi. This procedure showed high yield and simple synthesis of [$^{11}C$]methionine.

Synthesis of Zn4Sb3 by Solid State Reaction and Hot Pressing, and Their Thermoeletric Properties (고상 합성과 진공 압축성형에 의한 Zn4Sb3의 제조 및 열전특성)

  • Thermoelectric $Zn_4Sb_3$ alloys were synthesized by a conventional powder metallurgy process consisting of solid state reaction and hot pressing. Single phase $Zn_4Sb_3$ was successfully produced by the annealing of cold compact starting with the mixed elemental powders, and subsequent hot pressing yielded single phase bulk specimens without microcracks. Phase transformations in this alloy system during synthesis were investigated using XRD, SEM and EDS. Thermoelectric properties as a function of temperature were investigated from room temperature to 600 K and compared with results of analogue studies. Transport properties at room temperature were also evaluated. Thermoelectric properties of single phase $Zn_4Sb_3$ materials produced by this process are comparable to the published data. Synthesis by solid state reaction and hot pressing offers a potential processing route to produce a bulk $Zn_4Sb_3$

Synthesis and characterization of α-mangostin imprinted polymers and its application for solid phase extraction

  • α-mangostin imprinted polymers have been synthesized by a non-covalent imprinting approach with α-mangostin as a template molecule. The α-mangostin molecularly imprinted polymers (MIPs) prepared by radical polymerization using methacrylic acid, ethlylene glycol dimethacrylate, benzoyl peroxide, and acetonitrile, as a monomer, crosslinker, initiator, and porogen, respectively. The template was removed by using methanol:acetic acid 90:10 (v/v). The physical characteristics of the polymers were investigated by Fourier Transform Infrared spectroscopy (FTIR), scanning electron microscopy (SEM), and thermogravimetric analysis (TGA). The rebinding studies were carried out by batch methods. The results exhibited that the MIPs was able to adsorb the α-mangostin at pH 2 and the contact time of 180 min. The kinetic adsorption data of α-mangostin performed the pseudo-second order model and followed the Langmuir isotherm model with the adsorption capacity of 16.19 mg·g-1. MIPs applied as a sorbent material in solid-phase extraction, namely molecularly imprinted solid-phase extraction (MISPE) and it shows the ability for enrichment and clean-up of α-mangostin from the complex matrix in medicinal herbal product and crude extract of mangosteen (Garcinia mangostana L.) pericarp. Both samples, respectively, which were spiked with α-mangostin gives recovery more than 90% after through by MISPE in all concentration ranges.

Utility of solid phase extraction for colorimetric determination of lead in waters, vegetables, biological and soil samples

  • A highly sensitive, selective and rapid method for the determination of lead based on the reaction of lead (II) with 5-(4'-chlorophenylazo)-6-hydroxypyrimidine-2,4-dione (CPAHPD) and the solid phase extraction of the Pb(II)-CPAHPD complex with Amberlite XAD-2000 was developed, in the presence of pH 5.6 buffer solution and Triton X-114 medium. CPAHPD reacts with lead to form a violet complex with a molar ratio of 2:1 (CPAHPD to lead). This complex was enriched by the solid phase extraction with Amberlite XAD-2000. An enrichment factor of 500 was obtained by elution of the complex from the resin with a minimal amount of isopentyl alcohol(0.2 mL). In isopentyl alcohol medium,the molar absorptivity of the complex is $1.13{\times}10^6L\;mol^{-1}cm^{-1}$ at 647 nm. Beer's law is obeyed in the range of $5.0-160ng\;mL^{-1}$ in the measured solution. The relative standard deviation for 10 replicate samples of $50ng\;mL^{-1}$ level is 1.26%. The detection and quantification limits reaches 1.5 and $4.7ng\;mL^{-1}$ in the original samples. The presented procedure was successfully applied for determination of lead content in real samples such as vegetables, waters, biological and soil samples with satisfactory results.

Effects of Chemical Inhomogeneity on Phase Coexistence in Pb(Zr, Ti))$O_3$ Ceramics at Morphotropic Tetragonal and Rhombohedral Phase Boundary (정방정상과 능면체상의 경계조성 Pb(Zr, Ti)$O_3$ 세라믹스에서 화학조성의 불균일성이 상공존에 미치는 영향)

  • In order to identify the origin of phase coexistence at morphotropic tetragonal and rhombohedral boundary in PZT ceramics, the effect of chemical inhomogeneity on phase coexistence region was investigated. Two kinds of PZT ceramics with different chemical homogeneity were prepared by conventional solid state reaction and co-precipitation method. There was coexistence of tetragonal and rhombohedral phase over a wide composition range in PZT ceramics calcined by solid state reaction, and there was also phase coexistence of which region was reduced considerably in sintered samples. And phase coexistence region was not observed in co-precipitated PZT ceramics(within 1 mole%). Therefore compositional fluctuation is considered to be major origin of the phase coexistence at morphotropic phae boundary in PZT ceramics.

  • Prediction performances by Einstein's equation of diffusivity, Peskin's model, Three-Equation model, Four-Equation model and Algebraic Stress Model, have been compared by analyzing twophase (air-solid) turbulent jet flow. Turbulent kinetic energy equation of dispersed phase was solved to investigate effects of turbulent kinetic energy on turbulent diffusivity. Turbulent kinetic energy dissipation rate of particles has been considered by solving turbulent kinetic energy dissipation rate equation of dispesed phase and applying it to turbulent diffusivity of dispersed phase. Results show that turbulent diffusivity of dispersed phase can be expressed by turbulent kinetic energy ratio between phases and prediction of turbulent kinetic energy was improved by considering turbulent kinetic energy dissipation rate of dispersed phase for modelling turbulent diffusivity. This investigation also show that Algebraic Stress Model is the most promising method in analyzing gas-solid two phaes turbulent flow.

Reverse Micellar Extraction of Fungal Glucoamylase Produced in Solid-State Fermentation Culture

  • Partial purification of glucoamylase from solid-state fermentation culture was, firstly, investigated by reverse micellar extraction (RME). To avoid back extraction problems, the glucoamylase was kept in the original aqueous phase, while the other undesired proteins/enzymes were moved to the reverse micellar organic phase. The individual and interaction effects of main factors (i.e., pH and NaCl concentration in the aqueous phase, and concentration of sodium bis-2-ethyl-hexyl-sulfosuccinate (AOT) in the organic phase) were studied using response surface methodology. The optimum conditions for the maximum recovery of the enzyme were pH 2.75, 100 mM NaCl, and 200 mM AOT. Furthermore, the optimum organic to aqueous volume ratio ($V_{org}/V_{aq}$) and appropriate number of sequential extraction stages were 2 and 3, respectively. Finally, 60% of the undesired enzymes including proteases and xylanases were removed from the aqueous phase, while 140% of glucoamylase activity was recovered in the aqueous phase and the purification factor of glucoamylase was found to be 3.0-fold.
