Microbial Modification of Extracellular Polysaccharides

  • Some trials to alter the structure of extracellular polysaccharides by means of biotransformation and microbial modification have been reported. Seaweed alginate was acetylated by intact and resting cells of Pseudomonas syringae ATCC 19304. Glucose analogs such as 3-O-methyl-D-glucose used as sole carbon sources was directly incorporated into curdlan by agrobacterium sp. ATCC 31749. The 2-amino-2-deoxy-D-glucose (glucosamine)and 2-acetamido-2-deoxy-D-glucose (N-acetylglucosamine) were incorporated into microbial cellulose by Acetobacter xylinum ATCC 10245. The changed monomeric composition in pullulan by Aureobasidium pullulans ATCC 42023 as well as zooglan by Zoogoea ramigera ATCC 25935 was another effect of glucose analogs used a carbon source. There was no effect of glucose analogs found in polysacharide-7 (PS-7) produced by Beijerinckia indica. ATCC 21423.

  • A vendor supplies a product to a sole/major buyer on a lot-for-lot basis under deterministic inventory control conditions. The basic premise is that the setup cost reduction technologies are available to both the buyer and the vendor, and that the vendor's inventory and setup reduction investment costs differ from the buyer's. Therefore, an individually designed ordering and setup cost reduction policy will likely cause mismatches between the vendor's and the buyer's optimal cycle times. For this situation, we show that a joint optimal setup cost reduction and ordering policy, together with an appropriate side payment(quantity discount or premium price) schedule, can be designed in a spirit in a spirit of coordination to eliminate mismatches in individual optimal cycle times.

Conversion of Citron (Citrus junos) Peel Oil by Enterobacter agglomerans

  • Citron peel oil was extracted from citron (Citrus funas) fruit by steam distillation, and was used as starting material for microbial conversion to synthesize attractive flavor compounds by using Enterobacter agglomerans 6L. E. agglomerans was isolated from citron peel and was able to metabolize the citron peel oil and grew well ($A_{600}:\;3.0$) on the citron peel oil as the sole carbon source. Multiple terpene metabolites were produced by E. agglomerans 6L on M9 salt media with citron oil vapor. The identified bioconversion products from the citron peel oil included trans-2-decenal, octanol, $\delta$­valerolactone, $\gamma$-valerolactone, cryptone, hydroxycitronellol, cuminol, and $\gamma$-dodecalactone.

  • A bacterial strain A-6 producing the high level of an extracellular inulin fructotransfe rase(depolymerizing)(EC 2.4.1.93) which converts inulin into di-D-fructofuranose dianhydride III (DFAIII) was isolated from soil. The isolated strain could be classified as a species belonging to the genus Arthrobacter based on its morphological and physiological characteristics identified in this work. Production of the enzyme was induced by inulin, and the highest activity was detected in the slightly acidic medium supplemented with 2.5% inulin and 0.1% trypton as a sole carbon and a nitrogen source, respectively. Under the optimal conditions, the enzyme activity in the culture supernatant reached approximately 60 uints/ml after 96 hours of cultivation. The optimum pH and temperature for the crude enzyme preparation from Arthrobacter sp. A-6 were pH 5.0 and 60$\circ$C , respectively. The DFA produced by the action of the inulin fructotransferase was confirmed to be DFAIII by paper chromatography, HPLC and $^{13}$C-NMR spectroscopy.

Two Different Pathways (a Chlorocatechol and a Hydroquinone Pathway) for the 4-Chlorophenol Degradation in Two Isolated Bacterial Strains

  • Two isolated strains, Comamonas testosteroni CPW301 and Arthrobacter ureafaciens CPR706, were able to use 4-chlorophenol (4-CP) as a sole carbon and energy source. CPW301 was found to degrade 4-CP via a meta-cleavage pathway in which the chloro-substituent was eliminated even when 4-chlorocatechol was cleaved by the catechol 2, 3-dioxygenase. In contrast, CPR706 removed chloride from 4-CP prior to the ring-fission reaction, producing hydroquinone as a transient intermediate during 4-CP degradation. CPR706 exhibited much higher tolerance for 4-CP than CPW301, which was indicated by the maximum degradable concentration and degradation rate.

치코리뿌리 성분(成分)에 관(關)한 연구(硏究) (Studies on the Constituents of the Chicory Root)

  • Proximate composition, minerals and fatty acid in dried chicory root (moisture content 7.0%) are analyzed and subsequent results are as follows: Crude protein, crude fat, crude fiber, total sugar and ash content in chicory root are 8.6%, 1.6 %, 6.9%, 58.5% and 4.2%, respectively. Mineral content of Ca, P, Fe, Mg and Si in the root are 1,560, 180, 10,600 and 180 mg%, respectively. Other minerals such as K, Na, Al, Zn, Ag, Cu and Ti are also determined. Unsaturated fatty acid content in total fat of the root is 65.4%, Particularly high in linoleic acid. Uridine-5'-diphospho-glucose, as sole nucleotide-sugar in the root, was detected.

  • International Factoring transaction in Korea is different from that of financially advanced countries in terms of legal system and commercial and financial practices. As for the domestic factoring, Korean factors are only involved in advances often on a with recourse basis. With regard to the international factoring, Korean factors do not accommodate whole account receivables from clients (suppliers) but handle on a selective basis. Among Korean banks, KEXIM (Export and Import Bank of Korea) is sole factor for international transactions. Currently KEXIM and several foreign banks handle factoring provide factoring services with limitation to invoice discounting which is largely extended to large corporate names. Therefore this is far different from factoring in Europe and Americas designed for small exporters with non recourse advances. In respect of legal environment, receivable assignment is subject to debtor' acknowledge or approval of such assignment according to Civil Law Act. To remove the legal obstacles, Korean government have prepared new law which allows factor's own notification of assignment (and thereby reimbursement right) to debtor with some evidences.

Plasmid- and Chromosome-Mediated Assimilation of Phenol and Cyanide in Pseudomonas sp. Strain PhCN

  • Pseudomonas sp. PhCN strain, which has the potential to utilize phenol and cyanide as a sole carbon and nitrogen source, was isolated. A comparison of the effect of cyanide on phenol degradation and vice versa by strain PhCN showed that the degradation time was significantly delayed by an increase in either phenol or cyanide concentration, and the greatest activities were obtained in basal medium containing a low concentration of cyanide and phenol. This strain contained two plasmids of approximately 120 kb (pPhCN-1) and 110 kb (pPhCN-2). Plasmid curing experiments produced a plasmid-free strain as well as strains containing either the 120- or the 110 kb plasmid. The strains were tested for their ability to utilize phenol and KCN. The results demonstrated that the ability to utilize phenol was encoded by the 120 kb plasmid, whereas the ability to utilize cyanide appeared to be encoded by the chromosome.

Optical Resolution of DL-Pipecolic Acid by Fermentation Using Pseudomonas sp. PA09

  • Pseudomonas sp. PA09 was isolated from farm soil and used for the optical resolution of D-pipecolic acid from DL-popecolic acid. The strain PA09 consumed L-pipecolic acid preferentially as the sole carbon and energy source, thus accumulating D-pipecolic acid in the culture broth. Optimization to improve the enantiomeric excess and yield was performed. The time course experiment showed that the strain OP09 consumed L-pipecolic acid almost to completion after 35h of cultivation, and the enantiomeric excess and the yield (% of residual D-pipecolic acid) were 99.8 and 96.0%, respectively.

  • In recent years, through the rapid development in personal computer technology, it has become possible to make a radar simulator based on the personal computer. The advantage of the personal Computer aidd radar simulator lies in its cost effectiveness, when comparing with that using the real radar. Although there have been studies carried out to develop radar simulator using PC and the products of thi kind is in the commercial market already, they are all using the mockup of the real radar, and therefore, the price of the simulator is still rather expensive. In this respect, this thesis aims to develop a ARPA radar simulator which is running on the sole PC, so that the students of the maritime educational institution may get ARPA radar training easily and cheaply. The simulator developed in this thesis using Visual Basic is found to run successfully on the 486PC, and it is expected that this new simulator system designed first time in Korea would be used as an easily accessible ARPA radar training equipment.
