Three Separate Pathways for the Initial Oxidation of Limonene, Biphenyl, and Phenol by Rhodococcus sp. Strain T104

  • Rhodococcus sp. strain T104, which is able to grow on either biphenyl or limonene, was found to utilize phenol as sole carbon and energy sources. Furthermore, T104 was positively identified to possess three separate pathways for the degradation of limonene, phenol, and biphenyl. The fact that biphenyl and limonene induced almost the same amount of catechol 1,2-dioxygenase activity indicates that limonene can induce both upper and lower pathways for biphenyl degradation by T104.

  • With respect to a bacterium isolated some of the similarities and differences previously indicated between Thiobacillus thiooxidans and Thiobacillus concretivorus wee confirmed and the bacteria also provided considerable diferences compared to the two species in some repects. Severe precipitation of sulfur occurred in colonies and in liquid media of the organism isolated. The organism isolated utilized nitrate and asparagine as well as ammonium sulfate as a sole nitrogen source and grew well without being nearly inhibited by citrate and malate. This organism also showed the resistance to heats and external physical stimulations. Owing to some characters decribed above and the reports proposed, the organism isolated could be concluded to be Thiobacillus concretivorus and it was suggested that Thiobacillus concretivorus might be an apparently different species from Thiobacillus thiooxidans.

Isolation of a Malonate-utilixing Acinetobacter calcoaceticus from Soil (토양으로부터 Malonate를 이용하는 Acinetobacter calcoaceticus의 분리)

  • A bacterium which can utilize malonate as a sole carbon source was isolated from soil. This strain was identified to be Acinetobacter calcoaceticus by morphological, cultural, phtsiological and biochemical examination. When this microorganism was grown on malonate as a aole carbon source, the enzymes, such as malonyl-CoA synthetase, isocitrate lyase and malate synthase were induced. These results suggest that in this microorganism, malonate is also assimilated through the proposed pathway in Pseudomonas fluorescens: $malonate{\rightarrow}malonyl-CoA{\rightarrow}acetyl-CoA{\rightarrow}glyoxylate\;cycle$.

  • The bacteria, which were capable of producing ${\beta}-1$, 3-glucanase inducibly by utilizing cell wall of Aspergillus fumigatus as a sole carbon source, were isolated from soil in the campus of Kyungpook National University. Among them, the strain which produced the enzyme excellently was selected and identified to be Pseudomonas stutzeri KF 13 by morphological, cultural and physiological examination. The optimal conditions for the enzyme production from Pseudomonas stutzeri KF 13 were investigated. the enzyme production was reached maximum state shen the broth cultured for 72hr at $30^{\circ}C$. And the enzyme showed the highest activity in the medium containing 3.5% cell wall as an inducer, 15% yeast autolysate as a nitrogen source and 0.05% $MnSO_4$ at pH 7.5.

Catabolism of 4-Hydroxybenzoic Acid by Pseudomonas sp. DJ-12

  • A Pseudomonas sp. strain DJ-12 isolated by 4-cholrobiphenyl enrichment culture technique is capable of utilizing 4-hydroxybenzoic acid as a sole source of carbon and energy. The bacterium catabolized 4-hydroxybenzoic acid through the intermediate formation of protocatechuic acid, which was further metabolized. The cell free extracts of pseudomonas sp. DJ-12, grown on 4-hydroxybenzoic acid showed higher activities of 4-hydroxyenzoate 3-hydroxylase and protocatechuate 4,5-dioxygenase, but the activity of catechnol 2,3-dioxygenase was lower. The results suggest that 4-hydroxybenzoic acid is catabolized via protocatechuic acid rather than catechol or gentisic acid in this bacterium and that the protocatechuic acid formed was metabolized through a metacleavage pathway by protocatechuate 4,5-dioxygenase.

  • Poly(l,6-heptadiyne) derivatives with phenoxy and 3,7-dimethyloctyloxyphenoxy substituents were synthesized using metathesis polymerization. Polymerizations using MoCl$\sub$5/ as the sole catalyst gave high yields and the resulting polymers were completely soluble in organic solvents. The polymers' structures and thermal properties were characterized using NMR and UV-Vis spectroscopy, as well as with TGA and DSC thermograms. From the analysis of the high-resolution $\^$13/C-NMR spectra, we was found that these polymers contain six-membered rings. The number-average molecular weights and polydispersities of the polymers were ca. 7.0∼20 ${\times}$ 10$^3$ and 3.1∼5.8, respectively. The glass transition temperatures of the polymers were found to be in the range 91∼159$^{\circ}C$.

Abscisic Acid Binding to Extracts from Normal and Viviparous-1 Mutant Aleurone Layers of Zea mays L.

  • Aleurone layers of normal and vp1 mutant maize kernels were extracted and centrifuged at 100,000g to yield a cytosol fraction. Binding of [3H]ABA cis, trans (+)ABA to a soluble macromolecular components present in the cytosol was demonstrated by Sephadex chromatography and non-denaturing PAGE. The binding component was of high molecular weight and seems to be an aggregate of proteins. A rapid DEAE-cellulose filter method for assaying bound [3H]ABA to a soluble protein was adapted. Binding assays were performed with cytosol that had been preheated or incubated with several enzymes, indicating that heat and protease treatments disrupted the binding. This suggested that binding occurred to proteins. Some properties of the ABA binding proteins were described. The [3H]ABA binding were reduced dramatically when unlabeled ABA was added as a competitor, suggesting a specific binding of [3H]ABA. Gel filtration profiles and autoradiogram of [3H]ABA binding showed no difference in the binding components of Vp1 and vp1/vp1 mutant cytosol, indicating that Vp1 protein is not a sole ABA binding protein.

  • Food irradiation is increasingly recognized as the sole viable alternative to traditional method of food preservation in terms of reducing the overall quantity of spoiled food, reducing energy used in food storage, and reducing reliance on chemicals known to be hazardous but currently required for disinfestation. Irradiation with ionizing radiation can be applied in the fields of sprout inhibition, disinfestation of insects, sterilization, delay of ripening, and improvement of organoleptic properties in food. In order to back up the commercialization of food irradiation in Korea, this review not only dealt with the international background for food irradiation and wholesomeness of irradiated food, but also evaluated economic feasibility of irradiated food, irradiation facilities, and domestic status of food irradiation studies.

  • Edge detection always influenced by noise belong to the original image, therefore need use some methods to sort this issue, mean shift algorithm has the smooth function which suit for the edge detection purpose, so adopted to fade out the unimportant information, and the sensitive noise portions. After this section, use the Canny algorithm to pick up the contour of the objects we focus on, meanwhile select the Soble operator that has the orientation attribute to support the method work well. In final, take experiment and get the perfect result we wanted, make sure this method make sense and better than the sole Edge detection algorithm,

  • Many theorists acknowledge that Modern architecture denied the architect's crafty gesture for the sake of pure formal and compositional status of a building. As well, modern technology in construction has been prevailing as the sole support for modern architecture. But there exists architects who proposed a dialectic development of traditional and modern techniques. This thought was prominent in Germanic circle where technological development was in advance. Throughout $20^{th}$ century, the works of Adolf Loos, Mies Van der Rohe, and Peter Zumthor are representatives in each period. A common point begins from their apprenticeships as craftsmen: Loos and Mies as stonemasons, Zumthor as a cabinetmaker. More than this fact, their craftsmanship is embedded in their works and express creativity of architects. While mass production system raises non-participatory practice that merely require assemblage of products, the adjusted techniques with craftsmanship brings forth a participatory practice that does not limit the creativity of architects.
