Isolation of Protoplasts from Cultured Cells of Potato (Solanum tubersoum L.) Tuber Tissue (감자(Solanum tuberosum L.) 괴경의 배양세포로부터 원형질체의 분리)

  • Protopasts were isolated from cultured cells of potato (Solanum tuberosum L.) tuber tissue. The ability of callus formation from the culture cells was higher in cultivars Dejima and Superior than in Shimabara and Irish Cobbler on Lam's medium. Therefore, the former was used as sources for protoplast isolation. Friable calli were transferred to liquid media and cells in exponential phase were used for protoplast isolation. In both of Dejima and Superior, the yield of protoplasts was high in the enzyme solution of 2% Onozuka cellulase and 1% macerozyme. Also, viability of isolated protoplasts was very good. Thus, it seems that these protoplasts would be applicable to various aims of research.

Effect of Solution Culture System on Growth and Mini-tuber Yield of Hydroponically Grown Potato(Solanum tuberosum L. cv. Dejima) (양액재배방식이 '대지' 감자(Solanum tuberosum L.)의 생육 형 소괴경 형성에 미치는 영향)

  • This study was conducted to investigate the effect of solution culture systems on growth and mini-tuber yields of hydroponlcally grown potato (Solanum tuberosun L. cv. Dejima). The growth of stem and foliage at 40 and 60 days after planting were better in aeroponics system than the other systems, and stem length and number of stems per plant at 90 days after planting were the greatest as of 65.4 cm, 7.3 in aeroponics and the lowest as of 49.5 cm, 3.2 in scoria media system. Stolon length was the longest as of 30.4 cm in aeroponics at 90 days after planting, and number of stolons was the greatest as of 10.5 in NFT. Number of mini-tubers per plant at 90 days after planting was 67.1, 62. 5, 20.1 and 18.0 in aeroponics, NFT, perlite and scoria media, respectively. The perlite media system made the fewest enlargement of lenticels of mini-tubers. The results indicate that aeroponic system can be used effectively for mini-tuber potato production.

Detection of Genetic Variation and Gene Introgression in Potato Dihaploids Using Randomly Amplified Polymorphic DNA (RAPD) Markers

  • Randomly amplified polymorphic DNAs were employed to study the genetic variation and gene introgression in potato dihaploids (2n=24) which were generated after interspecific pollination of tetraploid cultivars (2n=4X=48, Solanum tuberosum cv Irish Cobbler, Superior and Dejima) by haploid inducer clones (2n=2X=24, Solanum phureja 1.22, Hes-5 and Hes-6). Genetic variation and DNA marker segregation among dihaploids were observed. Most dihaploids contain S. tuberosum specific RAPD markers but haploid inducer-specific RAPD markers were also found in some dihaploids. Of six different arbitrary 10-mer oligonucletide primers which showed polymorphism betwen tetraploid cultivars and haploid inducers used, three generated amplification products which seemed to be derived from the S. phureja parent. Our results indicate that chromosomes of dihaploids may not be pure S. tuberosum and the dihaploids may not be produced by parthenogenesis.

  • To observe the stimulating effect of low dose ${\gamma}$ radiation on the dormancy breaking and growth, microtubers of two potato cultivars (Solanum tuberosum L. cv. Dejima and cv. Superior) were irradiated at the dose of 0.5-30 Gy. Though it varied with cultivars and storage duration, sprouting rate, plant growth and tuber yield were promoted by 2-8 Gy irradiation in microtuber of 'Dejima' stored at low temperature. On the other hand, in microtuber of 'Superior', sprouting rate was promoted by 2 and 4 Gy irradiation, and the growth and tuber yield by 4 Gy irradiation. These results suggest that low dose of ${\gamma}$ radiation could have stimulating effects on the dormancy breaking of microtuber and potato growth.

  • To observe the stimulating effects of low dose ${\gamma}$ - radiation on the dormancy breaking and physiological activity, potato (Solanum tuberosum L. cv. Dejima) were irradiated at the dose of $0.5{\sim}30$ Gy. Low dose ${\gamma}$ - radiation exhibited promoting effects on the sprouting rate within the range of optimum dose of 1 Gy and 2 Gy and on the sprout length within the range of optimum dose of 2 Gy and 4 Gy. Regardless of storage duration and irradiation doses, the number of sprouts was enhanced. The number of sprouts, which were reserved for 15 days after production (DAP), were significantly increased under 4, 8 and 16 Gy irradiation. The growth of 45 DAP sprouts was extremely stimulated under 4 Gy irradiation accompanying the increase of peroxidase activity in the plantlet. In this study, it was also suggested that the activities of antioxidative enzymes of potato plantlets were not related to the irradiation dose during the plantlet development with the exception of decrease in catalase activity.

  • To observe the stimulating effect of low dose ${\gamma}$ radiation on the dormancy breaking and growth, microtubers induced in vitro of two potato (Solanum tuberosum L. cv. Dejima and cv. Superior) cultivars with different storage duration were irradiated with ${\gamma}$ radiation at the dose of $0.5{\sim}30$ Gy. Sprouting rate, growth and tuber yield of ‘Dejima’ microtuber were increased by ${\gamma}$ radiation in the range of $2{\sim}16$ Gy. In the microtuber of ‘Superior’, the sprouting rate was promoted by 2 Gy and 4 Gy irradiation, and the growth and tuber yield by 0.5 Gy and 4 Gy irradiation. There were not that much difference in chlorophyll content of potato plantlet by the low dose ${\gamma}$ irradiation. These results suggested that low dose ${\gamma}$ radiation stimulated the dormancy breaking and potato growth.

  • In order to compare salt tolerance among potato cultivars (Solanum tuberosum L.), in vitro plants of each cultivar were cultured on the Murashige and Skoog's medium containing different levels of NaCl (0, 75, 150, and 225 mM). The suitable level of NaCl for a comparison of the shoot and root growth between potato cultivars was 75mM. Shoot length and weight were better in 'Dejima', 'Superior', 'Jayoung', and 'Haryeong' than in other cultivars such as 'Goun' and 'Atlantic'. Normal root growth was observed in 'Seohong', 'Superior', and 'Haryeong', while 'Goun', 'Atlantic', 'Dejima', 'Jowon', and 'Chuback' showed no root. Proline contents in all the cultivars increased as affected by NaCl levels, then the increase amount was fewer in the cultivars showed a better growth in both shoot and root than in others showed a poor growth. As a result of this study, 'Superior' and 'Haryeong' were determined as salt tolerant cultivars and therefore using these cultivars it would be possible to potato production at salt arable land such as reclaimed tidal.

Effects of Culture Type and Inoculation Quantity in Bioreactor on Production of Potato Plantlets

  • Potato (Solamum tuberosum 'Dejima') plantlets were investigated on culture type and initial quantity of inoculation in bioreactor and survival rate by hydroponics for mass production. rode stems (1 to 1.5cm in length) of potato plantlets multiplied in vitro were grown for 3 weeks in liquid Murashige and Skoog (MS) medium with sucrose $30 g\; L^{-1}$. When plantlets (80-node inoculation) were raised in 10L balloon type bubble (BB) bioreactor, the healthiest growth of plantlets was obtained from explants cultured in ebb & flow culture with medium supplied periodically 12 times per day. The suitable inoculation quantity of 20L BB bioreactor was 120 pieces of stem segments (mean 2.2g fresh weight) in ebb & flow culture. Number of nodal shoot was eight on the average. In controlled culture room, survival rate of plantlets at 7 days after stem cutting was above $70\%$ when they were acclimatized by hydroponics grown in deep flow and solid medium culture. The highest survival rate of the stem cutting plantlets was in nutrient solution adjusted to EC $1.4dS{\cdot}m^{-1}$. Stem cutting plantlets through one culture could be obtained $670\~900$, when plantlets were grown in ebb & flow culture during 3 weeks using a 20L bioreactor with initial 120 pieces of nodal segments. 11 is possible In do mass production of seedlings cultured in bioreactor and hydroponics.

  • Bacterial specs isolated from common scab lesion on potato (Solanum tuberosum L. c.. Dejima) tuber was identified as Streptomyces turgidiscabies. This organism had flexuous spore chains and grey spore mass color, produced melanin pigment on ISP 7, but did not produce on ISP 6. S. turgidiscabies grew on agar media at pH 4.5, used L-arabinose, D-fructose, D-glucose, D-mannitol, raffinose, rhamnose, sucrose, D-xylose and meso-inositol as carbon sources, and was susceptible to 7% NaCl, thallium acetate (10 $\mu\textrm{g}$/ml, 100$\mu\textrm{g}$/ml), crystal violet (0.5 $\mu\textrm{g}$/ml), phenol (0.1%, wt/vol), oleandomycin (100 $\mu\textrm{g}$/ml), and streptomycin (20 $\mu\textrm{g}$/ml).
