Introduction of Maize Transposable Elements, Ac and Ds into the Genome of a Diploid Potato Species (옥수수 전위유전자 Ac 및 Ds의 2배체종 감자 Genome 내로의 도입)

  • Two maize transposable elements, immobilized Ac (iAc) and Ds, have been introduced into the genome of a diploid potato clone (Solanum tuberosum Group Phureja clone 1.22). The iAc is a modified Ac that is supposed to be unable to transpose but is expected to trans-activate the transposition of a Ds that is unable to transpose by itself. When the leaf and stem explants of in vitro shoots of the clone 1.22 were inoculated with Agrobacterium tumefaciens strains harboring binary vectors containing the iAc and the Ds, calli were formed from the explants on media containing 50 mg/L of kanamycin, and shoots were regenerated from the calli. The regenerated shoots formed roots when cultured on media containing 100 mg/L of kanamycin, whereas untransformed shoots did not form roots on the same media. The PCR amplification of the DNA's from the transgenic plants confirmed that the iAc and the Ds elements were introduced into the potato genome of 1.22.

Study on Antioxidative Activity of Plant Extracts in Fish Oil (식물 추출물이 어유의 산화 안정성에 미치는 영향)

  • Extract from forty four species of natural plants in Korea were obtained. Each extinct (1,000 ppm) was added into fish oil and their antioxidative activities were screened by rancimat, From the results, seven species including Brassica juncea, Solanum tuberosum, Capsella bursa, Ulums davidiana, Broussonetia kazinoki, Oenanthe javanica, and Rhus chinensis were selected due to their relatively prolonged induction periods. When the concentration (100 - 1,000 ppm) of the selected extract was considered, the most prolonged induction period was observed from Solanum tuberosum throughout the studied concentrations, suggesting that its extinct showed the strongest antioxidative activity against fish oil. In addition, peroxide value (POV) and thiobarbituric acid reactive substance (TBARS) value were considered at $60^{\circ}C$ for 5 days. Among the selected extract, extract from Solanum tuberosum showed the lout POV while the extract from Brassica juncea var showed the least TBARS values after 5 days. Also, the free radical scavenging effect against DPPH radical were studied with the selected extract, showing that the extract from Rhus chinensis was found to be the most effective.

  • By high performance liquid chromatography, separation and quantification of glucoalkaloids $({\alpha}-chaconine$and${\alpha}-solanine)$ from potato (Solanum tuberosum, var. May Queen) was established using periderm and cortex. $Nucleosil-NH_2\;(10{\mu}m)$ was packed in two stainless steel columns ($4.0\;ID{\times}15\;cm$ and $4.0\;ID{\times}25\;cm$) which were connected in sequence and eluted with the mixture of tetrahydrofuran, phosphoric acid buffer and acetonitrile (50 : 25 : 25, v/v/v) at the flow rate of 1ml/min. and the absorbance were read at 208 nm. Retention time was 6.92 min. for ${\alpha}-chaconine$ and 10.96 min. for ${\alpha}-solanine$ with complete separation. This method took 12 min. per sample and seemed best. ${\alpha}-Chaconine$ and ${\alpha}-solanine$ were found much in periderm than in cortex.
