  • 토마토 뿌리에서 분리한 Bacillus velezensis T20E-257 균주는 식물촉진효과가 있었고, 본 연구에서 T20E-257 균주의 유전체 서열을 해독하였다. 유전체 초안에서 포함된 2개 contig는 총 염기서열이 3,900,066 bp고, G + C content가 46.7%이었다. 유전체에서 단백질 유전자 3,708개, rRNA 유전자 27개, tRNA 유전자 86개를 확인하였다. 항균활성을 가지는 2차 대사산물 생합성 관련 유전자군과 식물생육촉진에 관여하는 IAA와 2,3-butadiol 생합성 관련 유전자를 T20E- 257 균주 유전체에서 확인하였다.

  • 토마토 뿌리에서 분리한 Chryseobacterium sp. T16E-39 균주는 식물생육촉진과 역병, 시들음병에 대한 억제효과가 있었다. 이 균주의 유전체 염기서열은 4,873,888 염기쌍이었으며 G + C 함량은 35.22%이었다. 이 유전체는 4,289개 단백질 유전자, 15개 rRNA 유전자, 71개 tRNA 유전자를 포함하였다. T16E-39 균주의 유전체에서 인산가용화, 식물호르몬 조절, 항산화 활성, 키틴 분해, 제9형 분비시스템에 관여하는 유전자를 확인하였으며, 이들 유전자는 식물의 생육을 촉진하고 병발생을 억제하는 기작과 관련되어 있을 것으로 판단된다.

감자 '하령' 품종의 친환경재배 가능성 평가 (Evaluation of the Potential for Environment-Friendly Cultivation of Potato cv. 'Haryeong')

  • 본 시험은 2006년 고령지농업연구소에서 육성한 '하령' 감자를 실제 농가에서 재배함으로써 다양한 병원균에 대한 포장저항성을 평가하여 친환경 재배 가능성을 검토하기 위하여 수행되었다. 품종별 평균 AUDPC는 '수미' 품종이 1377.7, '하령' 품종이 590.3으로서 '수미' 품종보다 '하령' 품종이 매우 강한 포장저항성을 나타내었으나, 시험처리 간에는 뚜렷한 경향을 나타내지 않았다. 수량은 모든 처리구에서 '하령' 품종이 많았으며, 단위면적(10a) 당 평균 수량은 '하령' 품종이 2,870 kg, '수미' 품종이 2,301 kg으로 '하령' 품종이 25% 증수를 보였다. 두 품종의 평균 건물률은 '수미' 품종이 16.1%, '하령' 품종이 18.8%로서 '하령' 품종이 2.7% 높았으나 처리 간 건물률의 차이는 나타나지 않았다. 본 시험을 통하여 '하령' 품종은 친환경 감자 재배를 위한 품종으로 매우 적합할 것으로 확인되었으며, 시비수준, 병해관리 등에 대한 추가적인 연구가 진행되어야 할 것으로 판단되었다.

A New Potato Cultivar "Early Valley", with High Yield and Early Maturity

  • "Early Valley", is an early maturing potato cultivar with high yield potential. "Early Valley" is a clonal selection resulting from the cross between 'Suncrisp' and 'A87109-10'. It has medium plant height and light green foliage. "Early Valley" has medium flowering habit and white flowers. Tubers are smooth, yellow skin, light yellow flesh, round tuber shape, medium eye depth, and medium dormancy and good keeping quality. It has stable yield under wide range of climatic conditions. "Early Valley" is resistance to late blight, but moderately susceptible to common scab and hollow heart. This cultivar is also resistant to potato rotting at harvesting during the raining season. "Early Valley" has high level of antioxidant activity (about three times higher) and vitamin C (higher by 40%) than the 'Superior'. This cultivar has high level of tuber uniformity and capable of yielding 36.56 t/ha which is 17.07% higher than the control potato cultivar 'Superior' under optimum agronomical practices.

A New Distinct Clade for Iranian Tomato spotted wilt virus Isolates Based on the Polymerase, Nucleocapsid, and Non-structural Genes

  • Tomato spotted wilt virus (TSWV; Genus Orthotospovirus: Family Tospoviridae) is one of the most destructive viruses affecting a wide range of horticultural crops on a worldwide basis. In 2015 and 2016, 171 leaf and fruit samples from tomato (Solanum lycopersicum) plants with viral symptoms were collected from the fields in various regions of Iran. ELISA test revealed that the samples were infected by TSWV. The results of RT-PCR showed that the expected DNA fragments of about 819 bp in length were amplified using a pair of universal primer corresponding to the RNA polymerase gene and DNA fragments of ca 777 bp and 724 bp in length were amplified using specific primers that have been designed based on the nucleocapsid (N) and non-structural (NSs) genes, respectively. The amplified fragments were cloned into pTG19-T and sequenced. Sequence comparisons with those available in the GenBank showed that the sequences belong to TSWV. The high nucleotide identity and similarities of new sequences based on the L, N, and NSs genes showed that minor evolutionary differences exist amongst the isolates. The phylogenetic tree grouped all isolates six clades based on N and NSs genes. Phylogenetic analysis showed that the Iranian isolates were composed a new distinct clade based on a part of polymerase, N and NSs genes. To our knowledge, this is the first detailed study on molecular characterization and genetic diversity of TSWV isolates from tomato in Iran that could be known as new clade of TSWV isolates.

Screening of Stat3 inhibitory effects of Korean herbal medicines in the A549 human lung cancer cell line

  • Background: The transcription factor signal transducer and activator of transcription 3 (Stat3)is constitutively activated in many human cancers. It promotes tumor cell proliferation,inhibits apoptosis, induces angiogenesis and metastasis, and suppresses antitumor hostimmune responses. Therefore, Stat3 has emerged as a promising molecular target for cancertherapies. In this study, we evaluated the Stat3-suppressive activity of 38 herbal medicinestraditionally used in Korea.Methods: Medicinal herb extracts in 70% ethanol were screened for their ability to suppressStat3 in the A549 human lung cancer cell line. A Stat3-responsive reporter assay system wasused to detect intracellular Stat3 activity in extract-treated cells, and Western blot analyseswere performed to measure the expression profiles of Stat3-regulated proteins.Results: Fifty percent of the 38 extracts possessed at least mild Stat3-suppressive activities(i.e., activity less than 75% of the vehicle control). Ethanol extracts of Bupleurum falcatumL., Taraxacum officinale Weber, Solanum nigrum L., Ulmus macrocarpa Hance, Euonymus alatusSieb., Artemisia capillaris Thunb., and Saururus chinensis (Lour.) Baill inhibited up to 75% of thevehicle control Stat3 activity level. A549 cells treated with these extracts also had reducedBcl-xL, Survivin, c-Myc, and Mcl-1 expression.Conclusion: Many medicinal herbs traditionally used in Korea contain Stat3 activity-suppressing substances. Because of the therapeutic impact of Stat3 inhibition, these resultscould be useful when developing novel cancer therapeutics from medicinal herbs.

Morphometric Characterisation of Root-Knot Nematode Populations from Three Regions in Ghana

  • Tomato (Solanum lycopersicum) production in Ghana is limited by the root-knot nematode (Meloidogyne incognita, and yield losses over 70% have been experienced in farmer fields. Major management strategies of the root-knot nematode (RKN), such as rotation and nematicide application, and crop rotation are either little efficient and harmful to environments, with high control cost, respectively. Therefore, this study aims to examine morphometric variations of RKN populations in Ghana, using principal component analysis (PCA), of which the information can be utilized for the development of tomato cultivars resistant to RKN. Ninety (90) second-stage juveniles (J2) and 16 adult males of M. incognita were morphometrically characterized. Six and five morphometric variables were measured for adult males and second-stage juveniles (J2) respectively. Morphological measurements showed differences among the adult males and second-stage juveniles (J2). A plot of PC1 and PC2 for M. incognita male populations showed clustering into three main groups. Populations from Asuosu and Afrancho (Group I) were more closely related compared to populations from Tuobodom and Vea (Group II). There was however a single nematode from Afrancho (AF4) that fell into Group III. Biplots for male populations indicate, body length, DEGO, greatest body width, and gubernaculum length serving as variables distinguishing Group 1 and Group 2 populations. These same groupings from the PCA were reflected in the dendogram generated using Agglomerative Hierarchical Clustering (AHC). This study provides the first report on morphometric characterisation of M. incognita male and juvenile populations in Ghana showing significant morphological variation.

  • The purpose of this study is to present the basic data for establishing the management plan of forest ecosystem in Honam-Jeongmeak and Geumnamhonam-Jeongmaek by surveying and analysing the vascular plants distributed in Jangansan(A-site), Palgongsan(B-site), Mandeoksan(C-site), Chuwolsan (D-site), Cheonunsan(E-site) and Jeamsan(F-site). The numbers of flora were 813 taxa including 115 families, 406 genera, 700 species, 4 subspecies, 94 varieties and 15 forms. There were 500 taxa in A-site, 427 taxa in B-site, 402 taxa in C-site, 449 taxa in D-site, 345 taxa in E-site and 524 taxa in F-site. The rare plants were 12 taxa including Paeonia japonica, Iris minutoaurea, Chloranthus fortunei and so forth. In IUCN Red List categories, there were 1 taxon of EN, 2 taxa of VU, 8 taxa of LC and 1 taxon of DD. The Korean endemic plants were 20 taxa including Salix koriyanagi, Thalictrum actaefolium var. brevistylum, Saussurea pseudogracilis and so forth. The specific plants by floristic region were 108 taxa including 58 taxa of grade I, 30 taxa of grade II, 16 taxa of grade III, 2 taxa of grade IV and grade V, respectively. The plants adaptable to climate change were 18 taxa including 11 taxa of southern plant and 7 taxa of northern plants. The naturalized plants were 49 taxa including Rumex nipponicus, Brassica juncea, Trifolium repens and so forth. The invasive alien plants were 4 taxa including Rumex acetocella, Solanum carolinense, Ambrosia artemisiifolia and Aster pilosus.

Identification and Expression Analysis of Genes Induced in Response to Tomato chlorosis virus Infection in Tomato

  • Tomato (Solanum lycopersicum) is one of the most widely grown and economically important vegetable crops in the world. Tomato chlorosis virus (ToCV) is one of the recently emerged viruses of tomato distributed worldwide. ToCV-tomato interaction was investigated at the molecular level for determining changes in the expression of tomato genes in response to ToCV infection in this study. A cDNA library enriched with genes induced in response to ToCV infection were constructed and 240 cDNAs were sequenced from this library. The macroarray analysis of 108 cDNAs revealed that the expression of 92 non-redundant tomato genes was induced by 1.5-fold or greater in response to ToCV infection. The majority of ToCV-induced genes identified in this study were associated with a variety of cellular functions including transcription, defense and defense signaling, metabolism, energy, transport facilitation, protein synthesis and fate and cellular biogenesis. Twenty ToCV-induced genes from different functional groups were selected and induction of 19 of these genes in response to ToCV infection was validated by RT-qPCR assay. Finally, the expression of 6 selected genes was analyzed in different stages of ToCV infection from 0 to 45 dpi. While the expression of three of these genes was only induced by ToCV infection, others were induced both by ToCV infection and wounding. The result showed that ToCV induced the basic defense response and activated the defense signaling in tomato plants at different stages of the infection. Functions of these defense related genes and their potential roles in disease development and resistance to ToCV are also discussed.

Agrobacterium을 이용한 토마토 떡잎에서 CRISPR-Cas9 시스템의 임시발현 시 토마토 떡잎 발달 단계에 따른 유전자교정 효율 변화 (Observation of Gene Edition by the Transient Expression of CRISPR-Cas9 System During the Development of Tomato Cotyledon)

  • BACKGROUND: Before generating transgenic plant using the CRISPR-Cas9 system, the efficiency test of sgRNAs is recommended to reduce the time and effort for plant transformation and regeneration process. The efficiency of the sgRNA can be measured through the transient expression of sgRNA and Cas9 gene in tomato cotyledon; however, we found that the calculated efficiency showed a large variation. It is necessary to increase the precision of the experiment to obtain reliable sgRNA efficiency data from transient expression. METHODS AND RESULTS: The cotyledon of 11th, 15th, 19th, and 23rd-day-old tomato (Solanum lycopersicum cv. Micro-Tom) were used for expressing CRISPR-Cas9 transiently. The agrobacterium harboring sgRNA for targeting ALS2 gene of tomato was injected through the stomata of leaf adaxial side and the genomic DNA was extracted in 5 days after injection. The target gene edition was identified by amplifying DNA fragment of target region and analyzing with Illumina sequencing method. The target gene editing efficiency was calculated by counting base deletion and insertion events from total target sequence read. CONCLUSION: The CRISPR-Cas9 editing efficiency varied with tomato cotyledon age. The highest efficiency was observed at the 19-day-old cotyledons. Both the median and mean were the highest at this stage and the sample variability was also minimized. We found that the transgene of CRISPR-Cas9 system was strongly correlated with plant leaf development and suggested the optimum cotyledon leaf age for Agrobacterium-mediated transfection in tomato.
