Development of Oriental Melon Harvesting Robot in Greenhouse Cultivation (시설재배 참외 수확 로봇 개발)

  • Oriental melon (Cucumis melo var. makuwa) should be cultivated on the soil and be harvested. It is difficult to find because it is covered with leaves, and furthermore, it is very hard to grip it due to its climbing stems. This study developed and tested oriental melon harvesting robots such as an end-effector, manipulator and identification device. The end effector is divided into a gripper for harvest and a cutter for stems. In addition, it was designed to control the gripping and cutting forces so that the gripper could move four fingers at the same time and the cutter could move back and forth. The manipulator was designed to realize a 4-axis manipulator structure to combine orthogonal coordinate-type and shuttle-type manipulators with L-R type model to rotate based on the central axis. With regard to the identification device, oriental melon was identified using the primary identification global view camera device and secondary identification local view camera device and selected in the prediction of the sugar content or maturity. As a result of the performance test using this device, the average harvest time was 18.2 sec/ea, average pick-up rate was 91.4%, average damage rate was 8.2% and average sorting rate was 72.6%.

Comparative Genetic Characterization of Plasmids of Agrobacterium Species Isolated in Korea (한국산 Agrobacterium plasmid의 유전학적 성상에 관한 연구)

  • The soil bacterium Agrobacterium tumefaciens is a plant pathogen that cause3 crown gall tumors by infecting the wounded dicotyledonous plants and subsequent integration of bacterial DNA into plant nuclear DNA. Virulent A. tumefaciens strains harbor a large Ti (tumor-inducing) plasmid that carries genes essential for tumorigenesis. In the present study, 13 strains (Malus pumila Mill; $A_{1-3}$, Populus monilifera; $W_{1-6}$, Populus tomentiglandlosa; $P_{1-3}$ and Rosa species; $R_1$) of Agrobacterium isolated in korean crown gall tumors and plasmids were observed in 6 strains ($W_2$, $W_3$, $W_6$, $P_1$, $P_3$ and $A_2$). The test for crown gall tumor formation was resulted only in ATCC15955 and $KW_2$ strains inoculated into the stem of sun flower and the development was observed for 4 and 6 weeks after inoculation. Above two Ti plasmids (pTi) were purified by cesium chloride-ethidium bromide density gradient centrifugation and digested with restriction enzyme and fragments of pTiATCC 15955 and $pTiKW_2$ observed by EcoR I ; 25&27, Hind III; 23&21, BamH I ; each 20 and Hpa I ; 12&27, and sizes of pTiATCC15955 and and $pTiKW_2$ calculated as 200 and 87 kbases. Octopine was isolated from tumor tissue ($W_{1-6}$ and $P_{1-3}$) and these strains confirmed as octopine type.

Simultaneous traveltime inversion of surface and borehole seismic data in Pungam basin (풍암분지 시험시추공 주변에서의 지표 및 시추공 초동주시 동시역산)

  • Velocity structures were defined in the vicinty of the 140-m deep test borehole in the pungam basin through simultaneous inversion of surface seismic refraction and far-ofset VSP traveltime data. Seismicenergy generated at the surface by a seisgun was recorded both at 42 surface locations at 3-m intervalsalong the profiles in the N20E and its orthogonal directions and at 71 m depth in the borehole. Forthe ofset VSP study, seismic energy was generated by a 5 kg sledgehamer at the surface in the horizontal ofset range of -19.5∼+19.5 m from the borehole. The seismic signals were detected at 9∼99 m depths with 1∼2 m intervals and recorded for 204 ms per shot. After shot static corrections,first-arrival times picked from both the surface refraction and borehole records were simultaneouslyinverted to yield velocity tomograms. The tomograms indicate that a 1.5 m thick soil layer with velocities les than 500 m/s overlies basements having a velocity range of 3,067 ∼5,717 m/s. Within the basements,∼4 m and deeper than 71 m. The high-velocit yzones may be due to conglomerates intercalated with sandstones and siltstones. No evidence for large-scale fracture zones or faults is detected near the borehole

  • Both surface seismic and far-offset VSP data were recorded alongtwo mutually perpendicular profiles in the Pungam basin. The first-arrival times were simultaneously inverted using the tomography method. For the surface data, seismic energy was generated by a 5-kg sledgehammer at 48 stations and detected by 21 surface geophones at 3 m intervals and one 3-component geophone in test borehole for the purpose of static corrections. For the VSP data, seismic waves generated by the sledgehammer on the ground were detected by a 3-component borehole geophone in a depth range of $9{\sim}99\;m$. Delay times of the hammer data were corrected using the seisgun data before the inversion to yield velocity tomograms. The tomograms indicates that the soil layer with velocities less than 750 m/s averages 1.8 m thick. The velocity varies from 5353 m/s at the depth range of $31{\sim}40\;m$ to 4262 m/s at the depth range of $65{\sim}73\;m$. Compared with core samples, the relatively large variation in velocity may due to lithology changes and fracture effects with depth.

  • Oilseed crop Camelina (Camelina sativa L.) is a suitable for biodiesel production that has high adaptability under low-nutrient condition like marginal land and requires low-input cost for cultivation. Enhanced abiotic stress tolerance of Camelina is very important for oil production under the wide range of different climate. CsRCI2s (Rare Cold Inducible 2) are related proteins in various abiotic stresses that predicted to localized at plasma membrane (PM) and endoplasmic reticulum (ER). These proteins are consist of eight-family that can be divided into tail (CsRCI2D/E/F/G) and no-tail (CsRCI2A/B/E/H) type of C-terminal. However, it is still less understood the function of C-terminal tail. In this study, CsRCI2D/H genes were cloned through gateway cloning system that used pCB302-3 as destination vector. And we used agrobacterium-mediated transformation system for generation of overexpression (OX) transformants. Overexpression of target gene was confirmed using RT-PCR and segregation ratio on selection media. We analyzed physiological response in media and soil under abiotic stresses using CsRCI2D and CsRCI2H overexpression plant. To compare abiotic stresses tolerance, wild type and CsRCI2D/H OX line seeds were sown on agar plate treated with various NaCl and mannitol concentration for 7 days. In the test of growth rate under abiotic stress on media, CsRCI2H OX line showed similar to NaCl and mannitol stress. In the other hand, CsRCI2D OX line showed to be improved stress tolerance that especially increased in 200mM NaCl but was similar on mannitol media. In greenhouse, WT and CsRCI2D/H OX lines for physiological analysis and productivity under abiotic stresses were treated 100, 150, 200mM NaCl. Then it was measured various parameters such as leaf width and length, plant height, total seed weight, flower number, seed number. CsRCI2H OX line in greenhouse did not show any changes in physiological parameters but CsRCI2D OX line was improved both physiological response and productivity under NaCl stress. Among physiological parameters of CsRCI2D OX line under NaCl stress, leaf length and width were observed shorter than WT but it were slightly longer than WT in 200mM NaCl stress. Furthermore, total seed weight of CsRCI2D OX line under stress displayed to decrease than WT in normal condition, but it was gradually raised with increasing NaCl stress then more than WT relatively. These results suggested CsRCI2D might be contribute to improve abiotic stress tolerance. However, function of CsRCI2H is need to more detail study. In conclusion, overexpression of CsRCI2s family can generate various environmental stress tolerance plant and may improve crop productivity for bio-energy production.

Effect of different plant densities on growth and yield of sorghum(Sorghum bicolor L. Moench.)

  • Sorghum is a crop with a various plant height depending on the planting density. If the height exceeds 1.8m, which is the harvestable height of the combine, loss is caused by clogging of the installation, entrance of the threshing section and the threshing section. The purpose of this study is to set the planting distance and number of plants per hill suitable for combine harvesting as the plant length does not exceed 1.8m. The experimental variety was Nampungchal. The experiment design was a split-plot design with three replications. The treatments were as follow: Main-plot were 1 and 2 plants as number of plants per hill and sub-plots were $60{\times}20cm$ (practice), $70{\times}15$, 20, 25, 30 cm as planting distance. The amount of nitrogen, phosphate and potassium fertilization were 100, 70, $80kg\;ha^{-1}$. Data were collected: (1) grain yield: weight of grain in $kg\;ha^{-1}$, (2) 1000 grain weight: average weight of 1000 grain, (3) plant height: distance from soil to top of panicle, (4) ear length: distance from top of stem to top of ear in cm, (5) stem diameter: diameter of second internode, (6) tiller number per hill. Analyses of variance were performed using R version 3.3.1(https://www. r- project. org). The Duncan's multiple range test(DMR) was used to separate treatment means at P < 0.05. As number of plants per hill increased, plant height and yield increased and tiller number decreased. As planting distance increased, plant height and yield decreased and tiller number increased. At 1 plant per hill, the plant height did not exceed 1.8m at all planting distance. At 2 plants per hill, the plant height did not exceed 1.8m from the planting distance of $70{\times}25cm$. At 1 plant per hill, the tiller number increased to 0.23, 0.27, 0.60 and 0.70 as the planting distance increased to $70{\times}15$, 20, 25 and 30 cm, respectively. At 2 plants per hill, the tiller number increased to 0.03, 0.03, 0.14 and 0.40 as the planting distance increased to $70{\times}15$, 20, 25 and 30 cm, respectively. At 1 plant per hill, the yield decreased to 6030, 4280, 3400 and $3230kg\;ha^{-1}$ as the planting distance increased. At 2 plant per hill, the yield decreased to 7850, 5770, 5720 and $4960kg\;ha^{-1}$ as the planting distance increased. We recommend that the optimum number of plants per hill and planting distance is 2 and $70{\times}25cm$ suitable for combine harvesting.

Effect of prohexadium-calcium on growth, lodging and yield of proso millet (Panicum miliaceum L.)

  • The proso millet is vulnerable to lodging due to high plant height and shallow root. A lodging results in a hard mechanical harvesting and yield loss. One of solutions on this problem is inhibition of internode elongation. The objective of this study was to set up use time and dose of prohexadium-calcium, is growth inhibitor. The experimental variety was Ibaekchal. The experiment design was a split-plot design with three replications. The treatments were as follow: Main-plots were 25 and 35 day after sowing(DAS) as use time and sub-plots were 0%, 50%, 100%(diluted solution of 1000 times, $1000{\ell}\;ha^{-1}$), 150% as dose. The amount of nitrogen, phosphate and potassium fertilization were 90, 70, $80kg\;ha^{-1}$, respectively. The size of high ridge and plant spacing were $90{\times}30cm$ and $60{\times}15cm$, respectively. Proso millet was sown on June 9, 2016 by hands and was adjusted at 2 plant per hill. The growth survey of vegetative growth stage was conducted at 1 day before treatment and with one week interval after treatment. Data were collected: (1) grain yield: weight of grain in $kg\;ha^{-1}$, (2) 1000 grain weight: average weight of 1000 grain, (3) plant height: distance from soil to top of panicle or leaf in cm, (4) ear length: distance from top of stem to top of ear in cm, (5) stem diameter: diameter of second internode (6) degree of lodging: percentage of lodging area, etc. Analyses of variance were performed using R version 3.3.1(https://www. r- project. org). The Duncan's multiple range test(DMR) was used to separate treatment means at P < 0.05. There was a significant difference in plant height and number of stem among the use time and dose of prohexadium-calcium during vegetative growth stage. At 25 DAS, the difference with no treatment increased until 25 day after treatment and decreased since then. The difference in number of stem increased until 18 day and decreased since 25 day. At 35 DAS, the difference with no treatment in plant height and number of stem increased until 22 day after treatment and decreased since then. We assumed that the effect of prohexadium-calcium was inhibition of internode elongation and promotion of tillering, continued untel 25day after treatment. At 25 DAS, the degree of lodging deceased to 100%, 30%, 10% and 0% as dose increased. At 35 DAS, the degree of lodging decreased to 100%, 20%, 0% and 0% as dose increased. At 25 DAS, the yield was 2910, 2710, 3190, $2310kg\;ha^{-1}$ among dose. At 35 DAS, the yield was 2750, 2630, 2220, $2050kg\;ha^{-1}$. We recommend that the optimum use time and dose of prohexadium-calcium for proso millet is 1000 times diluted solution of $1000{\ell}$ per ha at 25 day after sowing.

Preparation and Physical Properties of Eco-Friendly Biodegradable PLA/PBAT/HCO Blended Films (친환경 생분해성 PLA/PBAT/HCO 블랜드 필름 제조 및 물리적 특성)

  • In this study, eco-friendly biodegradable materials were prepared using poly(lactic acid) (PLA), poly(butylene adipate-co-terephthalate) (PBAT), and hydrogenated castor oil power (HCO) as an additive. The prepared PLA/PBAT/HCO blended films were characterized by the scanning electron microscope (SEM) and fourier-transform infrared spectroscopy (FT-IR). The results of SEM analysis indicated that PLA/PBAT (8 : 2) blended films added HCO showed no rough area, crack, or large agglomeration when compared with those adding various additives (12-hydroxy stearic acid (12HSA) and cellulose). The FT-IR results indicated the presence of specific peak of HCO in the PLA/PBAT blended films, and its peak intensity increased with increasing HCO content (0~5.0 wt%). Tensile strength, elongation at break, and water barrier and thermal properties of the prepared PLA/PBAT/HCO blended films were also investigated, indicating that the physical and thermal properties was improved more than three times by the addition of HCO. The biodegradability test in soil revealed that the prepared biodegradable materials were degraded by about 6.0~20% after 90 days.

Real-Time Micro-Weather Factors of Growing Field to the Epidemics of Rice Blast (벼 도열병 Epidemics에 미치는 재배 포장 실황기상 요인)

  • It was investigated on the relationship of the rice blast epidemics and the real-time meteorological factors, at the experimental paddy field in 1997. Weather factors(temperature, relative humidity, irradiation, precipitation, the direction of wind, wind speed, soil temperature and leaf-wetness, etc) were measured by using the automated weather station. The most influenced weather factor to blast epidemics, was the average max-temp($R^2$= 0.95) during 10 days before leaf blast epidemics, while the least thing was wind speed($R^2$= 0.24). The most potential weather factors correlated with the blast epidemics were T-ave(average temperature), T-max(maximum temperature), RH(Relative Humidity) and RD(Relative Humidity > 90% hrs). A statistics model(the regression equation) of the blast epidemics with the potential weather factors, was established as tallows ; Y = -3410.91 - 23.91 $\times$ T-ave + 28.56 $\times$ T-max + 41.0 $\times$ RH - 3.75 $\times$ RD, ($R^2$= 0.99). (T-ave >= 19$^{\circ}C$, T-max - T-ave >= 5.2$^{\circ}C$ and RH% >= 90.4%). According to the fitness test($\chi$$^2$) of the model, the observed blast disease severity was quite close to those expected.

Effect of Compaction Method on Induced Earth Pressure Using Dynamic Compaction Roller (진동롤러에 의한 다짐방법이 인접구조물의 다짐토압에 미치는 영향)

  • To increase the structural integrity of concrete box culvert good compaction by the dynamic compaction roller with bi9 capacity is as effective as good backfill materials. It is needed for effective compaction that a compaction roller closes to concrete structure with high frequency. However structural distress of the culvert could be occur due to the excessive earth pressure by great dynamic compaction load. To investigate the characteristics of Induced stress by compaction, a box culvert was constructed with changing cushion materials and compaction methods. Two types of cushion material such as tire rubber chip and EPS(Expanded Polystyrene) were used as cushion panels and they are set on the culverts before backfill construction. Laboratory test result of cushion material says that the value of dynamic elastic modulus of rubber is lesser than that of EPS. On the other hand, material damping of rubber material is greater than that of EPS. In most case, dynamic compaction rollers with 10.5 ton weights were used and vibration frequency was applied 30Hz for the great compaction energy. This paper presents the main results on the characteristics of dynamic earth pressures during compaction. The amounts of induced dynamic pressures$(\Delta\sigma\;h)$ by compaction are affected with construction condition such as compaction frequency, depth of pressure cell, distance between roller and the wall of culvert and roller direction. Based on the measured values dynamic lateral pressure on the culverts, it could be said that orthogonal direction of roller to the length of culvert is more effective to compaction efficiency than parallel direction.
