The purposes of this research are to investigate the pollution level of heavy metals due to the urbanization and industrialization in the satellite cities of Seoul, and to assess the chemical species and the sources of heavy metals in highly contaminated soils and dusts. Soil and dust samples were collected from the Uijeongbu and the Koyang cities, which are northern and the northwestern satellite cities of Seoul metropolitan city, respectively. Relatively high pH values($6.3{\sim}9.9$) were found in roadside soils compared with agricultural and forest soils. Difference in pH values of soils was not identified between before and after rainy seasons. In spite of no specific pollution sources in the above cities, the contents of Cu, Pb, and Zn in soils and dusts were much higher than the world average contents. The metal levels in dusts were higher than those in soils, but the metal concentration in dusts was significantly decreased after rainy season. Pollution index was high(> 1.0) in the areas of heavy traffic, industrial complex, and city centres. There is an appreciable proportion of total Zn in exchangeable/water-acid soluble fraction. Copper is predominantly associated with reducible and oxidizable phases, whereas Pb is largely in reducible association. It is concluded that the mobility and bioavailability of metals are high in the order of Zn >> Cu > Pb, on the basis of characteristic particle morphology and chemical composition, Pb-containing particles are originated probably from the automobile exhaust, particularly in heavy traffic areas. The metallic forms and iron-oxide associated forms of Cr, Ni, Cu, Zn, and Pb can be assessed as industrial origin.
This study was performed to investigate the effects of various media compositions in regeneration of Lilium lancifolium. The adventitious bud initiation from microscale was the best on MS medium supplemented with BAP 1.0 mg/L and NAA 0.1 mg/L after 4 weeks of culture. However, from bulbscales, adventitious bud initiation was the best in dark condition on MS medium supplemented with BAP 0.5 mg/L and NAA 0.1 mg/L. On the other hand, callus induction was found to be the best from the microscales incubated in complete dark condition for 8 weeks on MS medium supplemented with 2,4-D 1.0 mg/L and BAP 0.1 mg/L. The highest plantlet regeneration from callus was obtained after incubation in the light condition for 8 weeks on MS medium supplemented with NAA 0.5 mg/L and BAP 0.1 mg/L. Rooting of shoots was obtained easily on MS medium and the plantlets were transferred to soil pots after 8 weeks. The chromosome analysis of the root tip cells was revealed that the callus-derived plantlets had normal chromosome number, 2n=24. No variation was observed in the morphology of the plantlets.
$\alpha$-Amylase producing bacteria were isolated from activated sludge of corn processing wastewater plant and paddy field soil samples and selected by the direct iodine reaction. The isolate was identified as Bacillus sp. after morphology, API system and fatty acid analyses. To enchance $\alpha$-amylase productivity, a successive mutation of Bacillus sp. AIV 19 was performed using the treatment of nitrosoguanidine(NTG).The mutant, Bacillus sp. AIV 1940, showed about 1.8-fold level of amylase activity compared with parental strain. The isolate was Gram-positive and rod (2.8-3.0 $\mu$m long, 0.5-0.6 $\mu$m wide) type. The strain increased the bacterial mass at 3000 mg/l starch concentration. Organic substance removal rate was 40.2, 72.3% respectively after 1 and 3 day reaction using starch synthetic wastewater (intial CODcr was 4,455 mg/l).
A strain WL-2 was isolated from soil as a producer of the extracellular xylanase, which catalyzes the hydrolysis of oat spelt xylan. The strain WL-2 was identified as Streptomyces sp. on the basis of its 16S rRNA sequence, morphology, cultural and physiological properties. The xylanase of culture filtrate was the most active at $60^{\circ}C$ and pH 6.0, and retained $90{\%}$ of its maximum activity at range of pH $4.5{\~}6.5$. In order to optimize the culture medium for xylanase production, ingredients of G.S.S medium were replaced by several carbohydrates. The carbohydrates such as ${\alpha}-cellulose$, oat spelt xylan and maltose increased dramatically the xylanase productivity of Streptomyces sp. WL-2. The maximum xylanase productivity was reached to 120 U/ml in the modified medium containing $1{\%}\;\alpha-cellulose$ and $1\%}$ maltose.
In this study, we aimed to characterize fungal samples from necrotic lesions on collar regions observed in different sweetpotato growing regions during 2015 and 2016 in Korea. Sclerotia appeared on the root zone soil surface, and white dense mycelia were observed. At the later stages of infection, mother roots quickly rotted, and large areas of the plants were destroyed. The disease occurrence was monitored at 45 and 84 farms, and 11.8% and 6.8% of the land areas were found to be infected in 2015 and 2016, respectively. Fungi were isolated from disease samples, and 36 strains were preserved. Based on the cultural and morphological characteristics of colonies, the isolates resembled the reference strain of Sclerotium rolfsii. Representative strains were identified as S. rolfsii (teleomorph: Athelia rolfsii) based on phylogenetic analysis of the internal transcribed spacer and large subunit genes along with morphological observations. To test the pathogenicity, sweetpotato storage roots were inoculated with different S. rolfsii strains. 'Yulmi' variety displayed the highest disease incidence, whereas 'Pungwonmi' resulted in the least. These findings suggested that morphological characteristics and molecular phylogenetic analysis were useful for identification of S. rolfsii.
Many plants take up soluble silica from the soil, and they deposit in plant tissues. Silica bodies are most commonly found in the leaf epidermis. Silica body is important factor in growth and development of plant. It help to maintain rigidity in stem and linear leaves, and its content may be correlated with resistance to fungal disease. In this study, several morphologically distinctive forms of silica bodies on Cyperaceae in Korea are recognized, which are eventually classified into two groups. Silica body type of first group (Rhynchospora, Fuirena, Scleria, Carex, Eriophorum) is A-type, which is conical in shape. Silica body type of second group (Cyperus, Scirpus, Eleocharis, Fimbristylis, Kyllinga, Bulbostylis, Lipocarpha) is B-type, which is conical in shape with numerous satellite body. These types are consistent enough to use as characters in taxonomic studies within genus. Silica body type may be associated with habitat: the species in dry region have A-type, whereas the species in wet region have B-type. These results suggested that shapes and presence or absence of silica body might be valuable to systematic analysis.
A new species of Xiphinemella is described and illustrated from soil around the roots of larch in Korea. Xiphinemella maiensis n. sp. has stylet length of 63-68$\mu\textrm{m}$, body length of 2.5-3.2 mm, and vulva position of 49-50%. It differs from X. esseri in more posteriorly positioned vulva (42-47% in X. esseri), shorter stylet (72-75$\mu\textrm{m}$ in X. esseri), and more anteriorly located guide ring (25-27 vs. 35-38$\mu\textrm{m}$). It also differs from X. ornata in having longer body (1.94-2.32 mm in X. ornata) and shorter stylet (82-92$\mu\textrm{m}$ in X. ornata). A key to species of the genus is provided. Xiphnema brevicolle, X. diffusum, X. chambersi, and X. insigne are firstly recorded from Korea.
Background: Ginseng black spot disease resulting from Alternaria panax Whuetz is a common soil-borne disease, with an annual incidence rate higher than 20-30%. In this study, the bacterial strains with good antagonistic effect against A. panax are screened. Methods: A total of 285 bacterial strains isolated from ginseng rhizosphere soils were screened using the Kirby-Bauer disk diffusion method and the Oxford cup plate assay. We analyzed the antifungal spectrum of SZ-22 by confronting incubation. To evaluate the efficacy of biocontrol against ginseng black spot and for growth promotion by SZ-22, we performed pot experiments in a plastic greenhouse. Taxonomic position of SZ-22 was identified using morphology, physiological, and biochemical characteristics, 16S ribosomal DNA, and gyrB sequences. Results: SZ-22 (which was identified as Brevundimonas terrae) showed the strongest inhibition rate against A. panax, which showed 83.70% inhibition, and it also provided broad-spectrum antifungal effects. The inhibition efficacies of the SZ-22 bacterial suspension against ginseng black spot reached 82.47% inhibition, which is significantly higher than that of the 25% suspension concentrate azoxystrobin fungicide treatment (p < 0.05). Moreover, the SZ-22 bacterial suspension also caused ginseng plant growth promotion as well as root enhancement. Conclusion: Although the results of the outdoor pot-culture method were influenced by the pathogen inoculum density, the cropping history of the field site, and the weather conditions, B. terrae SZ-22 controlled ginseng black spot and promoted ginseng growth successfully. This study provides resource for the biocontrol of ginseng black spot.

Vaccinium oldhami Miq. is a Korean native tree, which is deciduous and shrub tree with broad leaf. It grows 1~4m in height generally. Ecologically, this tree grows well in shady place even in barren soil. Also, the tree has resistance to cold and dry, which tend to form a little community. This research investigates quantitative morphological characteristics of leaf and fruit among the V. oldhami in South Korea and then considers its relationship on the basis of raw data among the 10 populations. This study will give us invaluable information about growing conditions, reasonable management and breeding by selection of V. oldhami in South Korea. The main results obtained from this study are summarized as follows; Leaf size of Mudeung population was larger than other populations. Naebyeon population was smaller in size of the leaf than other populations. Anmyeondo population was larger in fruit characteristics compared with other populations and Deogyu population was the smallest among populations. According to cluster analysis based on the leaf and fruit morphological characteristics, the natural V. oldhami populations were classified into four groups such as the first group of Kumo population, the second group of Mudeung population, the third group of Anmyundo, Daedun, Doolyun population and the fourth group of the other five populations.
While screening indigenous prokaryotic species in Republic of Korea in 2017, a total of 17 bacterial strains assigned to the phylum Bacteroidetes were isolated from a variety of environmental habitats including water of fountain, tidal flat, plant root, soil, the gut of Russian grayling butterfly, ginseng field, seawater, lagoon and seashore sand. From the 16S rRNA gene sequence similarity of more than 98.7% and the formation of a robust phylogenetic clade with the closest species, it was found that the 17 strains belong to independent and recognized bacterial species. There has been no official report that the identified 17 species have been previously isolated in the Republic of Korea. Thus, 15 species in 10 genera of one family in the order Flavobacteriales, one species in one genus of one family in the order Cytophagales, and one species in one genus of one family in the order Sphingobacteriales are proposed as unrecorded species of the phylum Bacteroidetes found in the Republic of Korea. Gram reaction, colony and cell morphology, basic phenotypic characteristics, isolation source, taxonomic status, strain ID and other information are described in the species descriptions.
