A report of 21 unreported bacterial species in Korea, belonging to the Betaproteobacteria

  • As a subset investigation to discover indigenous prokaryotic species in Korea, a total of 21 bacterial strains assigned to the class Betaproteobacteria were isolated from a wide range of environmental samples which collected from fresh water, roots of plants, mineral water and soil from ginseng farm. Phylogenetic analysis based on 16S rRNA gene sequences indicated that 21 isolated strains were most closely related to the class Betaproteobacteria, with high 16S rRNA gene sequence similarity (>99.1%) and constructed a robust phylogenetic clade with the closest species in the class Betaproteobacteria. These isolated species have no previous report or publication in Korea; therefore 17 species in 14 genera of 6 families in the order Burkholderiales, 1 species in the order Methylophilales, 2 species in 2 genera of 1 family in the order Neisseriales are reported for betaproteobacterial species found in Korea.   • An outcome of the study to discover indigenous prokaryotic species in Korea, a total of 26 bacterial species assigned to the classes Bacteroidetes and Firmicutes were isolated from diverse environmental samples collected from soil, tidal flat, freshwater, seawater, wetland, plant roots, and fermented foods. From the high 16S rRNA gene sequence similarity (>99.0%) and formation of a robust phylogenetic clade with the closest species, it was determined that each strain belonged to each independent and predefined bacterial species. There is no official report that these 26 species have been described in Korea; therefore 14 strains for the order Flavobacteriales and two strains for the order Cytophagales were assigned to the class Bacteroidetes, and 8 strains for the order Bacillales and 4 strains for the order Lactobacillales were assigned to the class Firmicutes are reported for new bacterial species found in Korea. Gram reaction, colony and cell morphology, basic biochemical characteristics, isolation source, and strain IDs are also described in the species description section.

알칼리성 Cellulase를 생산하는 호알칼리성 Bacillus sp. HSH-810의 분리 및 효소 특성 (Isolation and Characterization of an Alkaline Cellulase Produced by Alkalophilic Bacillus sp. HSH-810)

  • 일반 토양과 부엽토, 퇴비로부터 alkaline cellulase 생성이 우수한 균주를 분리한 후 형태적, 배양학적 및 생화학적 동정을 실시한 결과 호알칼리성 Bacillus sp. HSH-810 균주인 것으로 판명되었다. 분리균 Bacillus sp. HSH-810의 생육과 효소 활성은 $30^{\circ}C$와 pH 10.0에서 가장 높은 것으로 나타났다. 본 균주는 배지 중에 탄소원과 질소원, 무기염으로 1.0%의 CMC와 0.5%의 peptone, 0.02%의 $CaC1_2$, $CoC1_2$를 사용하였을 경우 최대의 alkaline cellulase생산성을 나타내었다. 효소의 최적 활성 pH와 온도는 각각 10.5와 $50^{\circ}C$였다. 이 효소는 pH 6.0-13.0과 $50^{\circ}C$의 온도에서 매우 안정하였다. 효소액에 계면활성제를 첨가하여 효소 활성을 측정한 결과, 효소액은 sodium-$\alpha$-olefin sulfonate (AOS)와 sodium dodecyl sulfonate (SDS), Tween 20, Tween 80에 대하여 안정성 이 높은 특징을 보였으나 0.1%의 linear alkyl-benzene sulfonate (LAS)를 첨가한 경우에는 효소 활성이 심하게 저해되었다.

충남 섬 지역 근권 토양의 수지상균근균 군집 구조 (Community Structure of Arbuscular Mycorrhizal Fungi in the Islands of Chungnam, Korea)

  • 본 연구에서는 충남 서해안에 분포하는 5개의 섬 지역(신진도, 마도, 대난지도, 원산도, 삽시도)과 육지 해안 지역인 무창포의 근권 토양내 수지상균근균 군집 구조의 차이점을 확인해보았다. 그 결과 충남 지역에서 국내 미기록종인 Acaulospora brasiliensis, Redeckera fulvum를 포함하여 14개 속 24종의 수지상균근균을 확인하였다. 또한 대부분의 섬 지역에서 Acaulospora 속이 우점하고 있음을 확인할 수 있었으며, 섬 지역의 근권에서는 육지 지역과는 다른 수지상균근균 군집 구조가 발달함을 보여주었다. 특히 섬 간의 수지상균근균 군집은 섬들 사이의 물리적 거리가 가까울수록 더 유사하게 발달하는 것을 확인하였다.

  • As a subset study to discover indigenous prokaryotic species in Korea in 2014, a total of 34 bacterial strains assigned to the phylum Actinobacteria were isolated from various environmental samples collected from activate sludge, biotite, freshwater, gut of marine organisms, mud flat, sediment, soil, spent mushroom compost and sea water. On the basis of high 16S rRNA gene sequence similarity and a tight phylogenetic association with the closest species, it was revealed that each strain was assigned to independent and previously described bacterial species, with the exception of one isolate. There is no official report that these 34 species included in the phylum Actinobacteria have been described in Korea: 6 species of 5 genera in the order Corynebacteriales, 1 species of 1 genus in the order Frankiales, 2 species of 2 genera in the Micromonosporales, 14 species of 10 genera in Micrococcales, 2 species of 2 genera in the Propionibacteriales, 1 species of 1 genus in the Pseudonocardiales, 4 species of 2 genera in the Streptomycetales, 2 species of 2 genera in the Streptosporangiales and 1 species of 1 genus in the Solirubrobacterales. Description of 39 unrecorded bacterial species in Korea, belonging to the class Alphaproteobacteria

  • During an investigation of the biodiversity of bacterial species in Korea, we discovered many indigenous prokaryotic species. A total of 39 bacterial strains in the class Alphaproteobacteria were isolated from various environmental samples collected from marine organisms, sea water, fresh water, tap water, mud flats, activated sludge, mineral water, tidal flats, soil and decayed plants. From the high 16S rRNA gene sequence similarity (>98.7%) and formation of robust phylogenetic clades with the most closely related species, it was determined that each strain belonged to each independent and predefined bacterial species. There is no official report that any of these 39 Alphaproteobacteria species have been described in Korea. Specifically, 18 species in 11 genera in the order Sphingomonadales, 11 species in 10 genera in the order Rhizobiales, two species in two genera in the order Caulobacterales, six species in six genera in the order Rhodobacterales and two species in two genera in the order Rhodospirillales were found in Korea.   • Within a comprehensive, widescale investigation of indigenous prokaryotic species in Korea, 29 bacterial strains in the phylum Bacteroidetes were isolated from diverse environmental habitats that included soil, plant roots, natural caves, tidal flats, freshwater from lakes, and seawater. Based on their high 16S rRNA gene sequence similarities (>99.1%) and the formation of robust phylogenetic clades with the closest type species, each strain likely belonged to an independent and predefined bacterial species. There are no publications or official reports of the isolation of these 29 species in Korea. Our study provides strong evidence that seven species in three genera in the order Cytophagales, 15 species in 13 genera in the order Flavobacteriales and seven species in five genera in the order Sphingobacteriales, all within the phylum Bacteriodetes, are new reports of bacterial species in Korea.   • During a comprehensive investigation of indigenous prokaryotic species in Korea, a total 31 bacterial strains assigned to the class Alphaproteobacteria were isolated from diverse environmental habitats including freshwater, seawater, brackish water, ginseng soil, plant roots, natural caves, and tidal flats. Based on their high 16S rRNA gene sequence similarities(>99.1%) and formation of robust phylogenetic clades with the closest type species, each strain was assigned to an independent and predefined bacterial species. Because there were no published or official reports regarding the isolation of these 31 species in Korea, this study identified three species in two genera in the order Caulobacterales, 12 species in 10 genera in the order Rhodobacterales, three species in two genera in the order Rhizobiales, two species in two genera in the order Rhodospirillales and 11 species in seven genera, all in the order Sphingomonadaceae within the Alphaproteobacteria are reported as new alphaproteobacterial species in Korea. Evaluation of Certain Plant Extracts and Antagonists Against Fusarium solani and Alternaria tenuissima, the Incitants of Root Rot and Die-Back Diseases of Mulberry

  • The studies have been conducted to control the soil borne fungal pathogens viz, Fusarium solani (Mart) Sacc. and Alternaria tenuissima the incitants of root rot and die-back diseases on mulberry stem cuttings planted in the mulberry nurseries and also in established mulberry gardens ten plant extracts with 10% concentration except Lantana camara (undiluted) were tested through poisoned food technique and four biofungicides were also screened by dual culture method under in vitro conditions. Plant extract of Prosopis juliflora showed the maximum inhibition on the mycelial growth (81.2% over A. tenuissima and 80.0% over F. solani) and followed by L. camara (66.7% over A. tenuissima and 68.9% over F. solani). Among the antagonists Pseudomonas fluorescens and Trichoderma viride showed maximum inhibition on the mycelial growth of both pathogenic fungi. The promising plant extracts (P. juliflora and L. camara) and antagonists (P. fluorescens and T. viride) were tested against both the pathogenic fungi under in vivo conditions along with the existing popular chemical Mancozeb. All the tested plant products and bio-fungicides showed inhibitory effect on both fungi. But the maximum survival percentage of mulberry cuttings was recorded in the treatment with T. viride (95% against F. solani and 90% against A. tenuisssima) followed by P. fluorescens (90% against both fungi) and T. harzianum (80% against F. solani and 85% against A. tenuisssima). Incase of the treatments with plant extracts and chemical fungicide the P. juliflora (60% against F. solani and 55% against A. tenuisssima) showed higher survival percentage and followed by L. camara (55% against F. solani and 50% against A. tenuisssima) and Mancozeb (55% against both fungi). In case of control only 10% of survival was recorded in F. solani inoculated cuttings and 15% survival in A. tenuissima inoculated cuttings.

디젤유 분해균주의 특성 및 토양배양 (Characterization of Diesel Oil-Degrading Bacteria)

  • 디젤유로 오염된 토양으로부터 분리한 디젤 분해 우수 균주를 HS 균주로 명명하고, 각 균주의 디젤유 분해능과 특성을 조사하였다. 분리된 HS균주의 동정결과 HSI 균주는 Acinetobacter sp. HS2, HS3 균주는Pseudomonas sp.로 동정되었다. 최소배지에서 디젤유2%, pH 7.0,$25^{\circ}C$, 교반속도 200 rpm의 조건으로 5일간 배양한 결과 HSI 균주는 88% 이상의 높은 분해효율을 나타내었다. 소수성과 유화능의 측정 결과 HSI 균주가 가장 높은 소수성을 나타내었고, 유화능은 HS3 균주가 가장 높게 나타났다. 위의 결과를 토대로 액체 배양시 분해효율이 가장 높은 HSI 균주를 선택하여 토양배양을 실시한 결과 30일이 경과된 후 80%이상의 디젤유 분해효율을 나타내었고, 디젤유 분해효율은 미생물 활성과 비례하는 것으로 확인되었다. 따라서 신규 분리된 디젤유 분해균주는 높은 디젤유 분해능과 토양 생존능으로 실제 유류오염 환경에 적용이 가능할 것으로 사료된다.
