This study is aimed to investigate microbiological contamination of Angelica gigas Nakai. A total of 111 samples including root, soil, and irrigation water were collected from farms and market to detect aerobic bacteria, Bacillus cereus, coliform, Escherichia coli, Listeria monocytogenes,. Salmonella spp., and Staphylococcus aureus. The contaminations of aerobic bacteria, coliform, and Bacillus cereus in the root during cultivation were found 6.71 log CFU $g^{-1}$, 4.13 log CFU $g^{-1}$, and 3.54 log CFU $g^{-1}$, respectively. The contamination of coliform and B. cereus were detected in all steps from harvesting to processing, with the highest count recorded from the cutting step. In marketing, the contaminations of aerobic bacterial, coliform, and B. cereus were 5.5~6.0 log CFU $g^{-1}$, 2.4~2.6 log CFU $g^{-1}$, and 3.5~4.0 log CFU $g^{-1}$, respectively. Listeria monocytogenes, Salmonella spp, and Staphylococcus aureus were not detected in any of samples. This result indicated that hygienic soil management and post harvest management should be performed to reduce the contamination of hazard microorganisms and to produce safe agro-products.
To develop indicators for soil health evaluation, biological characteristics of native soils from the different parent materials were studied. Survival rate of fluorescent Pseudomonas in soils was the lowest as 0.1% while those of thermophilic Bacillus and alkaliphilic bacteria were over the 90% by the soil drying stress. There was positive relationship between soil microbial biomass and organic carbon exudated from the microbial biomass by the treatment. The average air-drying effect of soils was 39.7% with ranges of 9.7~95.0%. The propagules of mesophilic Bacillus and Gram negative bacteria were increased by the re-wetting of dried soils. Soil pH affected positively to the recovering rate of microbial number. Average recovering rate of microbes was 65.3%, and there was positive relationship between microbial biomass recovery and fluorescent Pseudomonas population.
A series of field and laboratory experimentes were conducted to find out the effects of kinds of crops residues and the different amount of fertilizer to the microbial interaction, chemical properties, plant growth and their interaction under continous cultivation of hot pepper, soybean, sesame and upland rice. The results obtained were summerized follows : 1. Total number of bacteria and actinomycetes were enhanced by cultivation of upland rice and soybean while no appreciable effects were obtained by the cultivation of hot pepper and sesame. 2. The number of bacteria, actinomycetes and fungi were increased by return of crop residues when the cultivation of hot pepper, soybean, sesame and upland rice. Specially, actinomycetes was remarkably increased by upland rice cultivation. 3. Increased amount of fertilizer were remarkably affected to decrease of number of soil microorganisms. Specially, actinomycetes succession was appearently affects while plant growing time. 4. The number of identified soil bacterial species were obtained high in order of Bacillus sp.>Rhizobium sp.>Agrobacterium sp.>Pseudomonas sp. The number of Gram positive bacteria were superior than that of Gram negative bateria. 5. Interaction between microbial succession and crops cultivation, the number of Bacillus sp. was increased in hot pepper, Rhizobium sp. was in soybean, and sesame, and Agrobacterium sp. were increased in soybean, respectively. 6. Survival and habitate of soil microorganisms were differ with kinds of crop, application of residue and fertilizers. Most high number of Bacillus sp. Rhizobium sp. and Pseudomonas sp. were obtained on the rhizoplane and rhizosphere while Agrobacterium sp. and Fusarium sp. were high in rhizosphere. 7. Factors in relation to change of soil microbial population was obtained high in order of climates>crops>organic>matter>fertilizer.
BACKGROUND: The urgency of feeding the world's growing population while combating soil pollution, salinization and desertification requires suitable biotechnology not only to improve crop productivity but also to improve soil health through interactions of soil nutrient and soil microorganism. Interest in the utilization of microbial fertilizer has increased. A principle of nature farming is to produce abundant and healthy crops without using chemical fertilizer and pesticides, and without interrupting the natural ecosystem. Beneficial microorganisms may provide supplemental nutrients in the soil, promote crop growth, and enhance plant resistance against pathogenic microorganisms. We mixed beneficial microorganisms such as Bacillus sp. Han-5 with anti-fungal activities, Trichoderma harziaum, Trichoderma longibrachiatum with organic material degrading activity, Actinomycetes bovis with antibiotic production and Pseudomonas sp. with nitrogen fixation. This study was carried out to investigate the mixtures on the soil microflora and soil chemical properties and the effect on the growth of lettuce and cucumber under greenhouse conditions. METHODS AND RESULTS: The microbial mixtures were used with each of organic fertilizer, swine manure and organic+swine manure and compared in regard to changes in soil chemical properties, soil microflora properties and crop growth. At 50 days after the treatment of microorganism mixtures, the pH improved from 5.8 to 6.3, and the EC, $NO_3$-Na and K decreased by 52.4%, 60.5% and 29.3%, respectively. The available $P_2O_5$ and $SiO_2$ increased by 25.9% and 21.2%, respectively. Otherwise, the population density of fluorescent Pseudomonas sp. was accelerated and the growth of vegetables increased. Moreover, the population density of E. coli and Fusarium sp., decreased remarkably. The ratio of bacteria to fungi (B/F) and the ratio of Actinomycetes bovis to fungi (A/F) increased 2.3 (from 272.2 to 624.4) and 1.7 times (from 38.3 to 64), respectively. Furthermore, the growth and yield of cucumber and lettuce significantly increased by the treatment of microorganism mixtures. CONCLUSION(S): These results suggest that the treatment of microorganism mixtures improved the chemical properties and the microflora of soil and the crop growth. Therefore, it is concluded that the microorganism mixtures could be good alternative soil amendments to restore soil nutrients and soil microflora.

We investigated the effects on soil microbial diversity and the growth promotion of red pepper resulting from inoculation with a microbial agent composed of Bacillus subtilis AH18, B. licheniformis K11 and Pseudomonas fluorescens 2112 in a red pepper farming field. Photosynthetic bacteria, Trichoderma spp., Azotobacter spp., Actinomycetes, nitrate oxidizing bacteria, nitrite oxidizing bacteria, nitrogen fixing bacteria, denitrifying bacteria, phosphate solubilizing bacteria, cellulase producing bacteria, and urease producing bacteria are all indicator microbes of healthy soil microbial diversity. The microbial diversity of the consortium microbial agent treated soil was seen to be 1.1 to 14 times greater than soils where other commercial agent treatments were used, the latter being the commercial agent AC-1, and chemical fertilizer. The yield of red pepper in the field with the treated consortium microbial agent was increased by more than 15% when compared to the other treatments. Overall, the microbial diversity of the red pepper farming field soil was improved by the consortium microbial agent, and the promotion of growth and subsequent yield of red pepper was higher than soils where the other treatments were utilized.

Liquid manure fertilizer drived from pig slurry is a valuable source of nutrients for crop production. However, there is no study for environmental assessment regarding microbial quality to apply liquid manure fertilizer. Therefore, this study aimed at quantifying the level of environmental impact on fecal coliform (Escherichia coli or E. coli ) survival in saturated soil such as paddy field. Surface water samples were collected up to 168 and 11 hours under natural sunlight and artificial ultraviolet radiation, respectively. The inactivation rate of E. coli under natural sunlight increased gradually after 48 hours. However, the inactivation rate of E. coli under artificial ultraviolet radiation increased linearly over time. Our findings suggested that the ultraviolet radiation is the limited factor on E. coli survival in surface water of saturated soil. This result will provide useful and practical guideline to applicators of agricultural soil in deciding appropriate handling and time frames for preventing pollution of water quality for sustainable agriculture.
The relationships between microbial activity and physico-KDICical characteristics of soils were investigated in three golf courses of Kwanak, Gold and Korea Country Clubs, with different open years. The soil samples were collected in tee, fairway and rough. There were ranges of 4.80-5.55 in pH, $25.55-98.50{\mu}S$ / cm in conductivity, 10.96-16.73% in moisture content, 0.18-0.36g / g in water holding capacity, 3.68-5.39% in organic matter, and 0.10-0.25% in total nitrogen. Dehydrogenase activity(DHA) as an index of soil microbial activity was determined. DHA values of soil were $69.83-314.43{\mu}$g / g in three courses and showed the order of Kwanak>Gold>Korea Country Club with open year. This indicates that DHA was affected by several fertilizer treatments rather than herbicide and pesticide treatments. DHA was significantly different with golf clubs as well as golf courses and positively correlated with water holding capacity and total nitrogen.

Mine soils are usually unfavorable for plant growth due to their acidic condition and low contents of organic matter and nutrients. To investigate the effect of organic material and lime on nitrogen processes in an acidic metal mine soil, we conducted an incubation experiment with treating livestock manure compost, dolomite, and oyster shell and measured soil pH, dehydrogenase activity, and concentration of soil inorganic N ($NH_4{^+}$ and $NO_3{^-}$). Compost increased not only soil inorganic N concentration, but also soil pH from 4.4 to 4.8 and dehydrogenase activity from 2.4 to $3.9{\mu}g\;TPF\;g^{-1}day^{-1}$. Applying lime with compost significantly (P<0.05) increased soil pH (5.9-6.4) and dehydrogenase activity ($4.3-7.0{\mu}g\;TPF\;g^{-1}day^{-1}$) compared with applying only compost. Here, the variation in dehydrogenase activity was significantly (P<0.05) correlated with that in soil pH. Soil inorganic N decreased with time by 14 days after treatment (DAT) due to N immobilization, but increased with time after 14 DAT. At 28 DAT, soil inorganic N was significantly (P<0.05) higher in the lime treatments than the only compost treatment. Especially the enhanced dehydrogenase activity in the lime treatments would increase soil inorganic N due to the favored mineralization of organic matter. Although compost and lime increased soil microbial biomass and enzyme activity, ammonia oxidation still proceeded slowly. We concluded that compost and lime in acidic mine soils could increase soil microbial activity and inorganic N concentration, but considerable ammonium could remain for a relatively long time.
Effects of chitin and chitosan treatments on soil microorganisms and the mesofauna were investigated in a microcosm and a fumigated field experiment. Responses of microorganisms were determined using microbial phospholipid fatty acid (PLFA) analysis, whereas responses of the mesofauna were measured in terms of the abundances of nematodes and microarthropods. Soil nitrate concentration increased on the application of chitin. Overall, chitin promoted bacterial and fungal abundance, leading to an increase in abundance of free-living soil nematodes that feed on decomposers. The ratio of saturated to unsaturated fatty acids was highest in the chitin-treated soil. Chitosan had a minimal effect on the abundance of microorganisms; however, it reduced the abundance of collembolans in the microcosm experiment. These results indicate that the application of chitin has beneficial effects on the supply of nutrients and promotion of the abundance of soil organisms.
Fertilization effects on changes in soil $CO_2$ flux and organic C in switchgrass (Panicum virgatum L.) land managed for biomass production were investigated. The mean daily soil $CO_2$ flux in the manure treatment was 5.63 g $CO_2-C\;m^{-2}\;d^{-1}$, and this was significantly higher than the mean value of 3.36 g $CO_2-C\;m^{-2}\;d^{-1}$ in the control. The mean daily $CO_2$ fluxes in N and P fertilizer treatments plots were not different when compared to the value in the control plots. Potentially mineralizable C (PMC), soil microbial biomass C (SMBC), and particulate organic C (POC) were highest at the 0 to 10 cm depth of the manure treatment. Potentially mineralizable C had the strongest correlation with SMBC (r = 0.91) and POC (r = 0.84). There was also a strong correlation between SMBC and POC (r = 0.90). Our results indicated that for the N and P levels studied, fertilization had no impact on temporal changes in soil organic C, but manure application had a significant impact on temporal changes in soil $CO_2$ evolution and active C constituents such as PMC, SMBC, and POC.
