Pilot 규모 biopile에 의한 유류오염토양의 정화 (Bioremediation of Petroleum Contaminated Soils by Pilot Scale Biopile)

  • The pilot scale biopile system was designed and operated for evaluation of bioremediation efficiency for petroleum contaminated soil. The pilot scale biopile consisted of biopile dome, aeration system and monitoring system and two biopiles were operated with nutrients and inoculum for more 100 days. The test pile A and B were analyzed with regard to pH, total carbon contents, water contents, nutrients (N, P) and TPH. The initial TPH concentrations for pile A and pile B were about 10,000 mg/kg and 2,300 mg/kg, respectively. After 100 days, the TPH contents decreased about 70% in the pile A and 30% in the pile B. Also, n-$C_{17}$/pristane and n-$C_{18}$/phytane ratios in all pile were significantly changed. The microbial densities in the pile A was increased by approximately $10^7$ CFU/g-soil~$10^8$ CFU/g-soil, but there was almost no changed in the pile B. The average biodegradation rates were calculated about 66.8 mg/kg-day in the pile A and 10.9 mg/kg-day in the pile B. Over the course of operation period, pile temperature was considered the major limiting factor for the efficiency of all biopiles.

우리나라에서의 유기물(有機物) 시용효과(施用效果) (Effects of Organic Material Application on the Growth and Yield of Crops in Korea)

  • The soil organic matter contents in arable land are generally low in Korea. Thus it is generally agreed that the application of organic materials to soils would be much beneficial. Present paper is a review on the effectiveness of organic mat ter application in uplands and lowlands. 1. The effect of organic matter application in uplands are of more clear and simple to explain as compared to that in lowlands. In uplands, appropriate application of organic matters such as compost and various crops residues improves the physical properties of soils leasing to increased water holding capacity, better aeration, and decrease in soil erosion. 2. In lowland, rice soils under water logged conditions the effect of organic matter application on rice yield is not straight borward and demands more refined knowledges for the interpretation of it. 3. It is found that the application of compost in rice soils is more effective when nitrogen fertilizer application is limited it dicating that nitrogen contained in the organic maerials can become available to rice plant and plays an important role for increased yield of rice under the condition where nitrogen fertilizer supply is limited. 4. Application of organic matter does not always bring about the desirable effects. Very often the organic matter application results in more intensive soil reduction leading to the accumulation of harmful substances which would can cancel out the positive effects of organic matter. This is partiunlarly true in poorly drained soils. 5. Rice straw or compost, when applied rice soils, supply sizeable amounts of available silicate to rice plant resulting in yield increase. 6. Although the effectiveness of organic matter application on rice yield in short term experiments is not consistent due to many reasons, the long term effect of organic matter is significant. 7. The term of the $O.M/SiO_2$ ratio in rice soils can serve as a criterion for the judgement of whether organic matter or silicate fertilizer is needed to be applied in a certain soil.

폭기시간과 유기질비료 농도에 따른 혼합유기질비료의 액비화 특성비교 (Comparison of Liquefying Efficiency of Mixed Organic Fertilizer as Affected by Aeration Time and the Ratio of Organic Fertilizer to Water)

  • 본 연구는 혼합유기질비료를 액비화할 때 액비화 효율을 증진시키기 위하여 폭기시간과 유기질비료 농도에 따른 무기성분의 특성변화를 조사하였다. 혼합 유기질비료는 참깻묵, 쌀겨, 어분, 혈분 등의 유기질 재료와 일라이트, 패화석 등으로 이루어져 있으며 약 2개월간의 발효과정을 거쳤고 N, $P_2O_5$, $K_2O$ 및 유기물함량은 각각 $23.0g\;kg^{-1}$, $17.0g\;kg^{-1}$, $23.9g\;kg^{-1}$, $290g\;kg^{-1}$이었다. 폭기시간은 일 2시간, 8시간, 연속 폭기 및 무폭기의 처리를 두었으며 물량에 대한 유기질비료 농도는 10%로 하였다. 유기질비료 농도에 따른 특성변화는 전체 물 함량에 대한 유기질비료 농도를 5%, 10% 및 20%로 하고 일 2시간씩 폭기를 하였다. 액비제조 기간이 경과함에 따라 폭기시간에 관계없이 pH, EC 및 $NH_4-N$ 농도는 증가하였다. 일 2시간 폭기한 처리구에서 액비제조 10일 후의 N, $P_2O_5$$K_2O$ 농도는 각각 $646mg\;kg^{-1}$, $68.1mg\;kg^{-1}$, $453mg\;kg^{-1}$으로 침출률은 각각 27.6%, 4.0% 및 18.9%이었으며 일 8시간 폭기 및 연속폭기조와 유의적인 차이가 없었다. 연속폭기조에서 호기성 세균, 포자형성 세균 및 사상균의 생균수가 다른 처리구보다 많았다. 물에 대한 유기질비료의 농도를 증가시키면 EC, $NH_4-N$ 및 무기성분 함량은 증가하였으나 유기질비료의 침출률은 감소하였다. 액비제조 10일 후 유기질비료 20% 처리구의 N, $P_2O_5$$K_2O$ 농도는 각각 $1,140mg\;kg^{-1}$, $35.4mg\;kg^{-1}$, $544mg\;kg^{-1}$이었고 침출률은 각각 24.8%, 2.4% 및 13.6%이었다. 유기질비료 농도의 증가는 포자형성 세균과 Pseudomonas spp. 균의 생균수를 증가시켰다.

녹색기술 무배수구 용기깊이와 슬릿(Slit)차이에 따른 디펜바키아 마리안느와 렉스 베고니아의 생육반응 (Growth Responses of Dieffenbachia amoena 'Marianne' and Begonia rex to Different Lengths and Numbers of Slitwalls in Drainless Containers for Green Technology)

  • The effects were investigated of different lengths and numbers of slitwalls in drainless containers on growth and change in soil moisture volumes on the growth of Diffenbachia amoena 'Marianne' and Begonia rex. Drainless containers filled with amended soil, with square shape ($240mm{\times}240mm$) were used, as well as three different sets of slitwalls (2, 4 or 8, respectively) in addition to non-slitwall containers. Two indoor foliage plants were grown in slitwall containers in randomized blocks with 3 replications in greenhouse conditions, from March to September, 2009. Soil moisture volumes per container were measured by weighing containers every 2 hours during the day. The change in soil moisture volumes showed considerable differences among slitwalls tested in comparison to control containers before and after twice-weekly irrigation. Particularly, the differences in the S2 (195mm, slitwall 2) containers were significantly greater than other containers tested. For Diffenbachia amoena 'Marianne', plant height, length of leaf, dry weight and fresh weight were higher with S2 containers than with those grown in other containers tested. The Begonia rex with the best quality in terms of plant height, length of leaf and width of leaf was grown in S8 (360mm, slitwall 8) containers. Particularly, statistical analysis has indicated that shoot fresh weights of Begonia rex grown in S8 were 3-fold higher than those grown in CS8 containers. The different results obtained within the two species led us to hypothesize a species-specific influence on indoor foliage plant performance. However, plants of both species grown in slitwall containers showed good results compared with plants grown in non-slitwall containers.

Pythium myriotyrum에 의한 생강뿌리썩음병의 포장내 발병진전에 미치는 토양소독, 살균제 시용, 좁은 이랑재배 효과

  • Effects of soil disinfection, fungicide application, and narrow ridge cultivation on ginger rhizome rot development were examined in two naturally-infested fields at Seosan, Choongnam province. Soil disinfection treatments were assigned to main plots, and fungicide and ridge treatments to sub-plots in a split plot design with three replications. The rhizome rot started in late July, and progressed rapidly until late September with the peak incidence in mid-august to early September. Soil disinfection by dazomet application showed the most prominent inhibition effects in both fields, where the disease was reduced by the treatment from 17.5% to 4.8% in one field, and from 51.0% to 2.2% in the other field. Three to five applications of fungicide metalaxyl-copper during the growing season inhibited the disease by 89.7% in one field, but less effectively in the other field. Narrow ridge cultivation reduced the disease effectively by 78.1% and 63.9%, compared to the unridged control plots in each field, respectively. Germination rate of seed-rhizomes and growth of ginger plants were similar between treatments, except when the plots received improper aeration after applying dazomet, and then the germination rate was significantly reduced. The greatest yields were obtained in the disinfected plots, regardless of rhizome rot incidence, except one control plot with very little disease. Ginger yield was negatively correlated with disease severity. However, the yield of ridge plots averaged 58∼59% compared to those of the unridged plots, due mainly to the half planting rate of the ridge plots. In spatial progress, the disease in the disinfected plots started from a single focus of the inoculum, and spread into the adjacent areas only, whereas in the untreated plots, the disease started from many foci that were distributed over the plot, and rapidly progressed to make an epidemic during the season. The soil density of P. myriotylum in the disinfected plots was not changed or, if not, increased slightly during the season. However, in the untreated plots it increased rapidly to reach the density 3 to 5 times greater by the end of the season.

  • 토양에서 질소변화과정에 관한 보다나은 이해력 증진은 질소비료의 효용성을 증대시키기 위한 필수적 요인이다. 실험실 실험은 대표적인 Piedmont토양에 있어 질소무기화 및 질산화작용을 구명하기 위해 수행하였다. 질소무기화 및 질산화 량은 Enon, Mecklenburg, Chewcla토양의 표토층에서 60 일까지 증가되었으며, 그후로부터 90일까지는 감소하는 경향을 보였다. 그러나 Wehadkee토양에서 질소의 무기화 및 질산화 량은 숙성시간에 따라 차이가 없었다. 위에서 열거한 모든 토양의 토심이 15-30cm의 토층에서 질소무기화 및 질산화 량은 90 일까지 시간이 경과함에 따라 증가하였다. 토양에 있어 질소무기화 및 질산화 량은 토심별로 토양종류에 따라 다르게 나타났으며, 이러한 차이점은 토양통풍 및 질산화 bacteria의 수에 영향을 받은 것으로 사료된다. 전반적으로, 질소의 무기화 및 질산화 량은 subsurface lagers보다는 표토층에서 많은 것으로 나타났다.

폭약(爆藥)에 의(依)한 과수원토양(果樹園土壤)의 물리성개량(物理性改良)에 관(關)한 연구 (The effect of Dynamite Explosion on Physical Properties of Orchard Soil)

  • 토양(土壤)의 물리성(物理性)이 심근성작물(深根性作物) 재배(栽培)에 극(極)히 불량(不良)한 토양(土壤)의 물리성(物理性)을 개량(改良)하여 우량과원(優良果園)으로 조성(造成)할 수 있는 손쉽고도 경제적(經濟的)인 방법(方法)을 찾기 위하여 저구릉지(低丘陵地)에 많이 분포(分布)되어 있는 전남식양토(全南埴壤土)에 대하여 1m 깊이에서 폭파처리하여 토양물리(土壤物理) 성화(性化)를 조사(調査)한 결과(結果)를 요약(要約)하면 아래와 같다. 1. 무처리구의 물리성(物理性)은 가비중(假比重), 경도(硬度)가 과도(過度)하게 크고 공극률(孔隙率) 및 기상이 근신장(根伸張)에 상당한 저해를 줄 정도로 낮았다. 이런 현상(現象)은 표토(表土)보다 심토(深土)에서 더욱 심(甚)하였다. 2. 화약폭파는 토양(土壤)을 파쇄시키고 균열을 만들어 줌으로서 가비중(假比重)과 경도(硬度)를 낮추고 공극량(孔隙量) 특히 대공극(大孔隙)을 늘려주는 효과가 크다고 인정(認定)되었다. 3. 물리성(物理性)을 적정수준(適正水準)으로 개량(改良)하는 효과범위는 폭약 두개를 사용(使用)한 경우 60cm 깊이에서 1m 반경이고 80cm 깊이에서는 30cm 반경이었다. 그러나 폭약 3개를 사용(使用)할 경우는 80cm 깊이에서도 1m 반경까지도 적정수준(適正水準)으로 물리성(物理性)이 개량(改良)되었다. 4. 파쇄 및 균열된 정도는 두개 사용시(使用時)는 불균일(不均一) 하였으나 3개 사용시(使用時)는 효과범위내에서 균일(均一)한 물리성(物理性) 분포(分布)를 나타내어 더욱 효과적이라고 볼 수 있다. 5. 폭약 2개를 사용(使用)하였을 경우 폭속(爆速)이 4,000m/sec, 폭발위력(爆發威力)이 50%인 초안폭약의 효과가 폭속(爆速)과 위력(威力)이 각각 5,000m/sec, 60%인 Gelatin 폭약보다 좋은 효과를 나타내었다.

토양미생물을 이용한 축산폐수 처리의 공정별 제거효율 및 동력학적 상수 (Swine wastewater treatment with soil microbes and its efficiency and dynamic parameters)

  • 토양 미생물을 이용한 고농도 축산폐수의 과부하 처리를 위한 공정개발 연구결과 유기물 제거율은 99%이었다. 상온 혐기조에서 $0.7kg\;BOD/m^3/d$ 이하에서 처리수의 BOD 농도는 150mg/L 이하를 이었다. 상온 혐기조를 운전한 실험에서 후속 폭기조의 물질 대사계수(km)는 $0.73hr^{-1}$, 세포증식계수(Y)는 0.433 g/VSS/g BODrm/d 였으며 중온으로 운전한 경우 후속 폭기조에서 각각 $1.45hr^{-1}$과 0.776 g VSS/g BODrm/d 로 반응계수 값이 두 배에 가깝게 개선되었다.

Optimization of Indole-3-acetic Acid (IAA) Production by Bacillus megaterium BM5

  • One of the important phytohormones produced by plant growth promoting bacteria is the auxin; indole-3-acetic acid (IAA), with L-tryptophan as the precursor. In this study, we focused on the investigation of optimal conditions for the production of IAA by Bacillus megaterium BM5. We investigated culturing conditions, such as incubation temperature, pH of the culture medium and incubation period, with varying media components such as inoculation volume, tryptophan concentration and carbon and nitrogen source. Besides, optimization study intended for high IAA production was carried out with fermentation parameters such as rpm and aeration. The initial yield of $42{\mu}g\;IAA\;ml^{-1}$ after 24 hr increased to $85{\mu}g\;ml^{-1}$ when 5% (v/v) of L-tryptophan was used in the culture broth. The maximum yield of $320{\mu}g\;IAA\;ml^{-1}$ was observed in trypticase soy broth (TSB) supplemented with starch and soybean meal as C and N sources with a C/N ratio of 3:1 (v/v) at $30^{\circ}C$, pH 8.0 for 48 hrs with 1.0 vvm and 250 rpm in 5 L working volume using 10 L scale fermenter. The bacterial auxin extracted from the culture broth was confirmed by thin layer chromatography and high-performance liquid chromatography and effect on plant growth was confirmed by root elongation test.

Composting and trickling filter for treatment of olive mill waste

  • Agricultural practice and improper waste disposal in developing regions have resulted in environmental degradation in land and waters, for which low-cost, proven solutions are needed. We demonstrate in the laboratory the applications of composting and trickling filter techniques to treat olive mill wastes that can be implemented in the West Bank and other regions of the world. To a pomace waste sample from a California mill, we amended with saw dust (wood carbon source) and baking soda ($NaHCO_3$ alkalinity) at weight ratios of waste/wood/$NaHCO_3$ at 70:27:1 and composted it for periods of 11 and 48 days; the compost was used as an additive to potting soil for transplanting. The pomace sample was also blended into slurry and introduced to a water-circulating pond and trickling filter system (P/TF) to examine any inhibitive effect of the pomace on biological removal of the organic waste. The results showed the compost-amended potting soil supported plant growth without noticeable stress over 34 days and the P/TF system removed BOD and COD by >90% from the waste liquid within 2 days, with a first-order rate constant of 1.9 $d^{-1}$ in the pond. An onsite treatment design is proposed that promises implementation for agricultural waste disposal in developing regions.
