Recycling of Sludgewater containing Set-retarder (지연제 첨가에 의한 회수수의 재이용)

  • Most of ready-mixed concrete plants have the problem of construction waste sludge, which pollutes environment and causes economic loss due to the discard and increasing the cost of concrete. Thus, a recycling of the cement sludge has been strongly desired as one of their solution. This research is to the study on the recycling of the cement sludge, especially the study on the hydration control by the sodium gluconate as a set-retarder. The set-retarder can delay the hydration of the cement included in the sludge water, so that the sludge water can be substituted with some of new cement without the property of the cement. And it invests the effect of the sodium gluconate to the hydration of the cement in suspension. The degree of hydration of cement may be controlled by adding the sodium gliconate. The hydration delay time is observed that depends on the concentration of residual sodium gloconate, not how long the cement has been hydrated before the addition of the sodium gluconate.

Textural Properties of Soygel with Added Alginate and Pectin (Alginate와 pectin 첨가에 의한 콩묵의 텍스쳐 특성)

  • Effect on alginate, pectin and Ca salts on textural properties of whole soybean gel(soygel) was investigated. The soygel was prepared by addition of sodium alginate, pectin and Ca salts into whole soy flour(300 mesh, WSF) suspension followed by through mixing. Addition of sodium alginate or pectin into WSF suspension significantly increased the hardness, adhesiveness and cohesiveness of the gel. The most uniform gel was obtained by $12{\sim}15%$ addition of sodium alginate or pectin, while their mixture rather decreased the textural properties. Sensory evaluation by ranking test for the gels showed that 12% sodium alginate and 0.125g $CaSO_4/g$ WSF produced the most desirable gel in terms of elasticity, uniformity, smoothness and total acceptability, followed by 12% sodium alginate and 0.12g Ca $gluconate-CaSO_4$(50 : 50). From the results, the optimal preparative conditions for soygel were suspension of WSF in 10 times of water(v/w), addition of 12% sodium alginate(w/w of WSF) and 12.5% $CaSO_4$ or Ca $gluconate-CaSO_4$(50 : 50) into WSF suspension followed by gel formation at $4^{\circ}C$.

Composition and microstructure of Lead-Tin alloy electrodeposits (납-주석합금 도금층의 조성 및 조직특성)

  • The composition and the microstructure of the lead-tin alloy electrodeposited in a gluconate bath were invesitigated according to electrolysis conditions. The tin content of the lead-tin alloy electrodeposits increased with increasing current density and EDTA addition, while it decreased with increasing temperature and sodium gluconate concentration. The preferred orientation of the alloy deposits changed from the (220) plane through (200) to (200) + (111) planes with increasing cathode overpotential. The surface morphology of the films was closely related to both the preferred orientation and the alloy composition.

Comparative Effects of Sodium Gluconate, Mannan Oligosaccharide and Potassium Diformate on Growth Performances and Small Intestinal Morphology of Nursery Pigs

  • This study was conducted to compare the effects of dietary supplementation of Sodium Gluconate (SG), Mannan Oligosaccharide (MOS) and Potassium Diformate (PDF) on growth performance and small intestinal morphology in nursery piglets. One hundred forty four female piglets ($11.69{\pm}0.71\;kg$) were divided into 4 treatments with six replicates of six pigs each. The pigs received a control diet or diets supplemented with SG, MOS and PDF at 2,500, 3,000 and 8,000 ppm; respectively, for 6 weeks. Supplementation of SG, MOS or PDF increased final body weight, average daily gain and tended to improve feed to gain ratio (p = 0.02, 0.04 and 0.16; respectively), other than average daily feed intake, intestinal pH and the bacterial populations were not influenced by the dietary treatments. SG significantly decreased the ammonia concentration in the caecum (p<0.05) and supplementation of SG, MOS or PDF tended to increase lactic acid and total short chain fatty acid concentration in the caecum (p = 0.08, 0.09; respectively), in addition SG, MOS or PDF slightly increased butyric acid concentration in the caecum (p = 0.14). SG highly significant increased the villous height in jejunum (p<0.01) and supplementing SG, MOS or PDF significantly increased crypt depth in jejunum (p<0.05), moreover, PDF significantly increased villous height and crypt depth ratio in jejunum (p<0.05) compared with control. The dietary treatments did not influence villous height and crypt depth in duodenum and villous height in jejunum (p>0.05). It can be concluded that supplementing SG, MOS or PDF as a feed additive has the potential to improve the growth performance, the intestinal lactic acid bacteria population, intestinal short-chain fatty acid concentration and the intestinal morphology of pigs.

Overproduction of Sodium Gluconate Using the Recombinant Aspergillus niger (재조합 Aspergillus niger에 의한 글루콘산나트륨의 산업적 생산)

  • Polymerase chain reaction(PCR) was conducted to obtain the gene encoding glucose oxidase(GOD) from Aspergillus niger(ATCC 2110) and the DNA sequence determined was coincided with published GOD sequence from A. niger. Recombinant transforming vector containing GOD and hygromycin B(hyg.B) resistant gene(hph) was constructed and used for further transformation of A. niger ATCC 2110. Selectivity of hyg.B against A. niger differed depending on which media were used i.e., nutrient-rich media such as potato dextrose agar(PDA) and complete medium(CM) showed only 50% growth inhibition at 400 $\mu$m ml$^-1$ of hyg.B while the minimal media inhibited mycelial growth completely at 200 $\mu$m ml$^-1$ of hyg.B. Twenty to sixty putative transformants were isolated from the hyg.B-containing minimal top agar, transferred successively onto alternating selective and nonselective media for a mitotic stability of hyg.B resistance and, then, single-spored. Among the stable transformants, the transformant(GOD1-6) grown by flask culture showed the considerable increase of extracellular GOD activity, which was estimated to the degree of 50% - 100% comparing to that of wild type. Transformation of tGOD1-6 was resulted from integration of the vectors into heterologous as well as homologous regions of the A. niger genome. Southern blot analysis revealed that there were two independent integrations of vector into fungal genome and one into the GOD gene due to homologous recombination. In addition, GOD activity and sodium gluconate production when tGOD1-6 was fed-batch fermented were enhanced 11 fold and 2.25 fold, respectively, compared to that of the wild type.

Possible Protective Effects of Quercetin and Sodium Gluconate Against Colon Cancer Induction by Dimethylhydrazine in Mice

  • Micronutrients in food have been found to have chemopreventive effects, supporting the conclusions from epidemiologie studies that consumption of fresh fruits and vegetables reduces cancer risk. The present study was carried out to evaluate the role of querctin (Q) and sodium gluconate (GNA) supplementation separately or in combination in ameliorating promotion of colon tumor development by dimethyl-hydrazine (DMH) in mice. Histopathological observation of colons in mice treated with DMH showed goblet cell dysplasia with inflammatory cell infiltration. This pathological finding was associated with significant alteration in oxidative stress markers in colon tissues and carcinoembryonic antigen (CEA) levels in plasma. Mice co-treated with GNA and Q showed mild changes of absorptive and goblet cells and inflammatory cell infiltration in lamina properia, with improvement in oxidative stress markers. In conclusion, findings of the present study indicate significant roles for reactive oxygen species (ROS) in pathogenesis of DMH-induced colon toxicity and initiation of colon cancer. Also, they suggest that Q, GNA or the combination of both have a positive beneficial effect against DMH induced colonic cancer induction in mice.

  • Acrylic copolymer was synthesized from 2-dimethylaminoethyl methacrylate and alkylmethacrylate containing long chain hydrocarbon group. To facilitate emulsification in water, acrylic copolymer was treated with acetic acid, and therefore acetated acrylic copolymer was produced. Acetated acrylic copolymer was perfectly emulsified in water and showed increased emulsion stability. Polymer as a cement dispersion agent(PDCM-PSD) was prepared by blending the newly synthesized acetates acrylic copolymer with sodium gluconate, oleic acid, and triethanolamine. The applicability of the blended polymer was examined, and it was found that the effects of dispersion and water-proof(0.3~0.5) were excellent.

  • DMA-co-DAMA were synthesized from 2-diethylaminoethyl metacrylate and dodecyl-metacrylate containing long chain hydrocarbon group with hydrophilic and hydrophobic radicals. To facilitate water emulsification,acrylic copolymer was cationized by acetic acid to produce acetated acrylic copolymer. The structures of the synthesized copolymer and acetated copolymers were confirmed by IR, NMR, and molecular weight was measured by GPC, and C. H. N elemental analysis. Acetated acrylic copolymers were perfectly emulsified in water and showed increased emulsion stability. Polymer dispersion for cement modifier(PDCM-PDD) was prepared by blending of the guaternized acrylic copolymer synthesized above sodium silicate sodium gluconate oleic acid and triethanol amine. The result with prepared polymer dispersion of cement modfier was examined, and it was found that excellent waterproffing effect; Water permeability ratio is 0.44 under the water pressure of $100g/cm^2$ and 0.55 under $3kg/cm^2$, and water absorption ratio is 0.36~0.47 and 1.02 compressive strength ratio at mixed ratio of water/PDCM-PDD is 45 times.

Optimization of Biotransformation Process for Sodium Gluconate Production by Aspergillus niger (Aspergillus niger를 이용한 글루콘산 나트륨 생산 생변환 공정의 최적화)

  • In order to produce high concentration of sodium gluconate, optimization of the fermentation conditions, such as glucose concentration, inoculum size, dissolved oxygen concentration and glucose feeding method, was examined. When the glucose concentration was maintained in the range of 30∼50 g/L during the batch fermentation, glucose conversion yield and productivity were 92.2% and 6.0 g/L/hr, respectively. In the case of the low concentration below 30 g/L, the yield decreased by about 25%. As the inoculum size increased above 20%(w/v), lag phase was shortened but the productivity decreased. The dissolved oxygen level of 60∼70% was shown to be the threshold point for 75% of increase in the productivity of sodium gluconate. Finally, optimal glucose feeding rate was determined using various feeding methods such as exponential feeding, feeding based on the average glucose consumption rate and was determined using various feeding methods such as exponential feeding, feeding based on the average glucose consumption rate and on the oxygen uptake rate and etc. Our result shows that glucose feeding, based on the oxygen uptake rate is a very simple, efficient and robust method, especially when oxygen is consumed as a substrate for the bioconversion. Using the above glucose feeding strategy under the optimized condition, 255 g/L of sodium gluconate concentration, 12 g/L/hr of productivity and 95% of glucose conversion yield were achieved with A. niger ACM53.
