Quercetin Down-regulates IL-6/STAT-3 Signals to Induce Mitochondrial-mediated Apoptosis in a Non-small-cell Lung-cancer Cell Line, A549

  • Objectives: Quercetin, a flavonoid compound, has been reported to induce apoptosis in cancer cells, but its anti-inflammatory effects, which are also closely linked with apoptosis, if any, on non-small-cell lung cancer (NSCLC) have not so far been critically examined. In this study, we tried to determine if quercetin had any demonstrable anti-inflammatory potential, which also could significantly contribute to inducing apoptosis in a NSCLC cell line, A549. Methods: In this context, several assays, including cytotoxicity, flow cytometry and fluorimetry, were done. Gene expression was analyzed by using a western blot analysis. Results: Results revealed that quercetin could induce apoptosis in A549 cells through mitochondrial depolarization by causing an imbalance in B-cell lymphoma 2/Bcl2 Antagonist X (Bcl2/Bax) ratio and by down-regulating the interleukine-6/signal transducer and activator of transcription 3 (IL-6/STAT3) signaling pathway. An analysis of the data revealed that quercetin could block nuclear factor kappa-light-chain-enhancer of activated B cells (NF-${\kappa}B$) activity at early hours, which might cause a down-regulation of the IL-6 titer, and the IL-6 expression, in turn, could inhibit p-STAT3 expression. Down-regulation of both the STAT3 and the NF-${\kappa}B$ expressions might, therefore, cause down-regulation of Bcl2 activity because both are major upstream effectors of Bcl2. Alteration in Bcl2 responses might result in an imbalance in the Bcl2/Bax ratio, which could ultimately bring about mitochondria mediated apoptosis in A549 cells. Conclusion: Overall, the finding of this study indicates that a quercetin induced anti-inflammatory pathway in A549 cells appeared to make a significant contribution towards induction of apoptosis in NSCLC and, thus, may have a therapeutic use such as a strong apoptosis inducer in cancer cells.

Root Extract of Scutellaria Baicalensis Increases Gefitinib Sensitivity in H1975 Human Non-small Cell Lung Cancer Cells (H1975 세포에서 황금추출물에 의한 gefitinib 저항성 억제 효과)

  • Gefitinib, a first generation epidermal growth factor receptor tyrosine kinase inhibitor (EGFR TKI), provides obvious clinical benefit in patients with EGFR-mutant non-small cell lung cancer (NSCLC). However, patients ultimately develop gefitinib resistance which mainly caused by EGFR T790M secondary mutation. In the current study, we investigated whether the root extract of Scutellaria baicalensis (SB) overcomes gefitinib resistance. Gefitinib-resistant H1975 human NSCLC cells (EGFR L858R/T790M double mutant) were treated with gefitinib and/or ethanol extract of SB (ESB) to evaluate the effect of ESB on the gefitinib sensitivity. The cell viability was measured by MTT assay and trypan blue exclusion assay. The colony-forming ability was evaluated by anchorage-dependent colony formation assay. Combined treatment with gefitinib and ESB markedly decreased the cell viability and colony formation than single treatment with gefitinib or ESB in H1975 cells. In addition, cells treated with both gefitinib and ESB exhibited a significant increase of sub-G1 DNA content which indicates apoptotic cells compared with those treated with gefitinib or ESB alone. As a molecular mechanism, combined treatment with gefitinib and ESB strongly downregulated the phosphorylation of ERK and JNK than single treatment with gefitinib or ESB. Taken together, our results demonstrate that ESB sensitizes H1975 cells to gefitinib treatment. We cautiously propose that ESB can be used in combination with gefitinib for the advanced NSCLC patients with acquired resistance to EGFR TKIs.

  • The present study was performed to clarify the histochemical compositions and fine structure of the mucus secreting cells in the gastrointestinal mucosa of normal mice. The mucus cells in the surface epithelium of stomach body had neutral mucin and some quantity of weak acid mucin. And the mucus cells in gastric pits and mucus neck cells had neutral mucin. The goblet cells in villial epithelium of small intestine contain strong acid sulfated mucin as their main content and a little of neutral mucopolysaccharide. However, the goblet cells in intestinal glands-Liberkuhn crypt were confirmed to contain non-sulfated weak acid mucin. The goblet cells in the surface epithelium of colon had the same component as the small intestine did. But the cells in the crypts of colon contained neutral and weak acid mucin as their main contents. The majority of secretory granules of the surface epithelial cells of the stomach body had high electron density, and some granules with low electron density appeared too. While the mucin granules in the mucus neck cells were low in its electron density, and some of those granules were frequently found to have dense core in them. Secretory granules in goblet cells of small and large intestines had low electron density. The mode of secretion in mucin-containing cells in gastro-intestinal tract was found to be merocrine.

The Endocrine Control of Corpus Luteum Function (황체기능의 내분비 제어)

  • The corpus luteum (CL) is formed by the action of a surge of luteinizing hormone (LH) on the pre-ovulatory follicle. Luteal cells derived from granulosa and theca interna cells continue to secrete progesterone for about two weeks. LH in domestic animals is essential for the normal secretion of progesterone at all stages of the luteal phase. For this process in the rodents, 20$\alpha$-hydroxysteroid dehydrogenase (20$\alpha$-HSD) is indispensable. 20$\alpha$-HSD is an enzyme to be a biologically inactive steroid. This enzyme plays a critical role in the regulation of the rat luteal function and reported to be present in steroid-producing tissues such as the testis and adrenal gland. We have purified 20$\alpha$-HSD and found two distinct 20$\alpha$-HSD molecules (HSD-1 and HSD-2). Their molecular weights are both estimated to be 33kd.The amino acid compositions of HSD-1 and HSD-2 are mostly similar, but there is a slight difference in the content of lysine. We demonstrated that 1) CL of previous generations contribute more to whole ovarian 20$\alpha$-HSD activity, 2) newly formed corpora lutea contain only 20$\alpha$-HSD-1 activity, and 3) old CL express activities of each HSD isozyme as shown in the luteal tissue of cycling rats on the day of diestrus where only degenerating old CL exist. The increase in 20$\alpha$-HSD activity identified seems to be related to the increase in the numbers of 20$\alpha$-HSD-positive cells. Interestingly, 20$\alpha$-HSD-1 activities were strongly found in the follicle fluids and theca interna cells by immunohistochemical study. Thus, the activity of 20$\alpha$-HSD may be related to a survival mechanism of those luteal cells and follicles remaining in the ovaries. Luteal cells arise from two sources. The small luteal cells are all of theca cell origin, while the large luteal cells are mainly of granulosa cell origin. CL of Korean Native Cattle, as those of other animal species, contains two morphologycally and functionally distinct luteal cell populations, such as small and large luteal cells as well as nonluteal cells. In all reproductive states except in the late luteal phase, the bovine CL also contained more small luteal cells than large luteal cells. Luteal tissue secretes a variety of growth factors (proteins) and the pattern of secretion changes during all stages of the luteal phase. These growth factors could be important in regulating the function of the bovine corpus luteum and may act in a potential endocrine autocrine and paracrine mechanisms. Therefore, further work has to be done to elucidate the role of growth factors in the ovary, especially in the corpus luterum. Interest should be focussed on interaction of these growth factors in the regulation of luteal cell and the localization of cytokine synthesis in differnet luteal cells.

Effect of small Black Soybean Fraction on the T cell-mediated Immune Responses in vivo and Proliferation of Leukemia Cells in vitro

  • We investigated effect of small black soybean fraction (SBSF) T cell-mediated responses for tumor surveillance and proliferation in leukemia cells in vitro. Each SBSF butanol fraction (SBSFBu) and SBSF chloroform fraction (SBSFCh) was administered p.o. once a day far 21 days in BALB/c mice and then levels of serum cytokines and subpopulation of lymphocytes were measured. Moreover, SBSF fraction was treated into the cultured various cell lines for proliferation in leukemia cell lines, NO production by RAW264.7 cells, and expression of p53 gene in U937 leukemia cells. These results showed that SBSFBu increased levels of serum IL-4but not IL-2 and IFN-${\gamma}$, and increased expression of CD4$^+$ T cells and CD8$^+$ T cells in splenocytes in vivo, while SBSFCh increased levels of serum IL-2 and IFN-${\gamma}$ but decreased IL-4, and increased CD8$^+$ T cells but not CD4$^+$ T cells. Moreover, both of SBSFBu and SBSFCh inhibited proliferation of HL60, U937, and L1210 leukemia cell lines in a dose-dependent manner, up-regulated NO production by RAW264.7 cells in a dose-dependent manner, and enhanced expression of p53 gene in U937 leukemia cells. Our findings indicate that SBSFBu and SBSFCh may enhance T cell-dependent immune responses, and that both of SBSFBu and SBSFCh may inhibit proliferation of leukemia cells by up-regulation of NO production and expression of p53 gene.

  • Coughing and breath shortness are common symptoms of nano (small) cell lung cancer. Smoking is main factor in causing such cancers. The cancer cells form on the soft tissues of lung. Deformation behavior and wave vibration of lung affected when cancer cells exist. Therefore, in the current work, phase velocity behavior of the small cell lung cancer as a main part of the body via an exact size-dependent theory is presented. Regarding this problem, displacement fields of small cell lung cancer are obtained using first-order shear deformation theory with five parameters. Besides, the size-dependent small cell lung cancer is modeled via nonlocal stress/strain gradient theory (NSGT). An analytical method is applied for solving the governing equations of the small cell lung cancer structure. The novelty of the current study is the consideration of the five-parameter of displacement for curved panel, and porosity as well as NSGT are employed and solved using the analytical method. For more verification, the outcomes of this reports are compared with the predictions of deep neural network (DNN) with adaptive optimization method. A thorough parametric investigation is conducted on the effect of NSGT parameters, porosity and geometry on the phase velocity behavior of the small cell lung cancer structure.

Degenerative Changes of the Rat Dorsal Root Ganglion (DRG) Cells Following a Tight Spinal Nerve Ligation (랫드 척수신경 결찰에 따른 척수신경절세포의 퇴행성변화)

  • This study aim to disclose a possible mechanism for the neuronal cell death induced by peripheral nerve injury following a spinal nerve ligation (SNL) as a neuropathic pain model. Male Sprague-Dawley rats (270~290 g) were used for this study. Pain threshold was evaluated for their response to mechanical (von Frey hairs) stimuli 1, 3, and 7 days after a tight ligation of L5 ventral ramus. In control group, the small ganglion cells were strongly stained with routine toluidine blue (TB), whereas the large ganglion cells showed a little bit weak stainity. Each large ganglion cell is surrounded by perineuronal satellite cells. In experimental groups, small ganglion cells showing apparent degenerative changes increased on 1 day, and showed a peak in degenerative cell number at 3 days group, and decreased gradually at 7 days group. We also found a small number of large-sized ganglion cells showing mild degenerative changes. However their satellite cells ware relatively intact with no typical findings throughout this experiment. Under the electron microscope, small ganglion cells showed various stage and typical features of the dark degeneration including mitochondrial swelling.

Small Cell Carcinoma of the Esophayus (식도의 소세포함 치험 1례)

  • Small cell carcinoma is a highly malignant esophageal tumor composed of anaplastic small cells with features very similar to those of its pulmonary counterpart. The prognosis is poorer than that of squamous carcinoma of the esophagus because of its propensity of generalized spread and metastasis. Once the diagnosis of small cell carcinoma was established, surgery should be undertaken as early as possible. We have described an experience of small cell carcinoma of the lower esophagus in a 72 year old male patient with a review of the literatures regarding treatment methods and prognosis.

Small RNAs: Classification, Biogenesis, and Function

  • Eukaryotes produce various types of small RNAs of 19-28 nt in length. With rapidly increasing numbers of small RNAs listed in recent years, we have come to realize how widespread their functions are and how diverse the biogenesis pathways have evolved. At the same time, we are beginning to grasp the common features and rules governing the key steps in small RNA pathways. In this review, I will summarize the current classification, biogenesis, action mechanism and function of these fascinating molecules.

Transmission of Continuous Media by Send-rate Control and Packet Drop over a Packer Network (패킷망에서 전송율 제어와 패킷 폐기에 의한 연속 미디어 전송방안)

  • When continuous media are transmitted over the communication networks, asynchrony which can not maintain temporal relationships among packets may occur due to a random transit delay. There exist two types of synchronization schemes ; for guaranteed or non-guaranteed resource networks. The former which applies a resource reservation technique maintains delay characteristics, however, the latter supply a best-effort service. In this paper, I propose a intra-media synchronization scheme to transmit continuous media on general networks not guaranteeing a bounded delay tome. The scheme controls transmission times of the packets by estimating next delay time with the delay distribution. So, the arriving packets may be maintained within a limited delay boundary, and playout will be performed after buffering to smoothen small delay variations. The continually increasing delay due to network overload causes buffer underflow at the receiver. To solve it, the transmitter is required to speed up instantaneously. Too much increase of transmission-rate may cause network congestion. At that time, the transmitter drops the current packet when informed excessive delay from the receiver.
